단순흉부영상의 Template-Matching을 이용한 폐 결절 자동 추출 (Automated Detection of Pulmonary Nodules in Chest Radiography Using Template Matching)

  • This paper proposes some technical approaches for automatic detection of pulmonary nodules in chest X-ray images. We applied threshold technique for the lung field segmentation and extended the lung field by using morphological methods. A template matching technique was employed for automatic detecting nodules in lung area. Genetic algorithm(GA) was used in template matching(TM) to select a matched image from various reference patterns(simulated typical nodules). We eliminated the false-positive candidates by using histograms and contrasts. We used standard databases published by Japanese Society of Radiological Technology (JSRT) for correct results. Also we employ two-dimensional Gaussian distribution for some reference images because the shadow of lung nodules in radiogram generally shows the distributions. Nodules of about 89% were correctly detected by our scheme. The simulation results show that it is an effective method to indicate lesions on chest radiograms.

  • Bone age assessment has been widely used to measure the ossification in pediatric radiology. For the assessment, first bone age of each epiphyseal plate is estimated using DCT/LDA, then the bone age of a patient is calculated by using the median of 9 estimations. For some patients, however, due to various reasons such as X-ray image quality or the pose of fingers, it is common to miss couple of plates in automated systems. In this paper, we investigate the relationship between the number of detected plates and the accuracy of bone age assessment. In the experimental results, we confirmed the similarity between bone age assessed using more than 7 epiphyseal plates and that assessed using 9 epiphyseal plates.

  • The crystal and molecular structure of 2-methyl-3-(N-trimethyl ammonium)phenol iodide, $C_{10}H_{16}NOI, was determined by X-ray diffraction method. The compound crystallizes in the orthorhombic space group $P_{na2}_{1}$ with a=13.327(3), b=12.496(3), C=7.227(2)A and Z=4. A total of 489 independent observed reflections were collected by the automated Four-circle diffractometer and was solved by heavy atom method and refined by anisotropic block-diagonal least-squares method to the R value of 0.04. The benzene ring is slightly distorted from regular hexagon. The I atom and 2-methyl-3-(N-trimethyl ammonium)phenol group is held together by van der Waals forces in the crystal. Intermolecular hydrogen bond is of the type O-H....I with the length 3.35.angs.. Apart from the hydrogen bonding system the molecules are held together by van der Waals forces in the crystal.

  • We have developed a new HD-2700 (Hitachi High-Technologies Corp., Japan) scanning transmission electron microscope (STEM) that includes an automatic aberration correction function, and a large-solid-angle energy-dispersive X-ray spectroscopy detector that enables high-resolution and sensitive analysis. For observation with atomic resolution, using spherical-aberration-corrected STEM, in order that satisfactory performance of the device can be achieved readily, and within a short time, irrespective of the operator's skill level, a spherical-aberration-correction device with an automatic aberration-correction function was developed. This automatic aberration-correction function carries out the entire correction-related process (aberration measurement, selection and correction) automatically, with automatic selection of the aberrations that require correction, and automatic measurement of the appropriate corrections.

전자현미분석에서 발생하는 규산염 유리 시료의 Na 이동 효과 보정 (Correction for Na Migration Effects in Silicate Glasses During Electron Microprobe Analysis)

  • 전자현미분석기를 이용한 규산염 유리의 정량화학조성 분석 시 알칼리 원소, 특히 Na이 전자빔에 의해 분석 영역 밖으로 이동(migration)하면서 X선 측정 세기가 시간에 따라 감소하는 현상은 오래 전부터 알려져 왔다. EPMA로 규산염 유리의 Na 함량을 정확하게 측정하기 위해서는 전자빔에 의한 Na 이동을 최소화하는 분석방법을 적용하거나 Na 이동에 따른 X선 측정 세기의 감소 효과를 보정해 주어야 한다. 본 연구에서는 X선 세기의 시계열 변화 곡선을 이용해 Na 이동에 의한 X선 감소를 보정할 수 있는 Probe for EPMA 소프트웨어의 Time Dependent Intensity(TDI) 보정 기능을 이용해 규산염 유리 표준시료 8종의 Na을 분석하였다. 일반적인 지질시료 분석 조건인 15 kV 가속전압, 20 nA 전류 세기 하에서 TDI 보정의 정확도를 검증하였다. 연구 결과 20 ㎛ 크기의 큰 전자빔을 사용했을 때는 Na 감소가 거의 일어나지 않아 보정 없이도 Na의 함량을 정확하게 측정할 수 있었다. 빔의 크기가 10 ㎛ 이하일 때는 Na 감소가 일어나 최대 -55%에 달하는 큰 오차가 발생하는데 TDI 보정을 적용해 이 오차를 ±10% 이내로 줄일 수 있다. Na X선 시계열 변화의 초기 측정값에 가중치를 주고 변화 추세를 선형적으로 가정하는 방법을 사용하면 상대오차를 ±6% 이내로 더 줄여 정확한 Na2O 함량을 얻을 수 있었다. 따라서 알칼리 원소가 많이 포함된 유리질 시료에서 분석 영역이 충분히 크지 못해 작은 전자빔을 사용해야만 한다면 적절한 TDI 보정을 반드시 해주어야 정확한 조성을 얻을 수 있다.

Structural Similarity Index 인자를 이용한 방사선 분할 조사간 환자 체위 변화의 자동화 검출능 평가: 초기 보고 (Feasibility of Automated Detection of Inter-fractional Deviation in Patient Positioning Using Structural Similarity Index: Preliminary Results)

  • 현대 방사선치료는 고선명 X선 투사영상을 이용하여 환자 및 종양의 위치를 확인하는 기술이 요구되지만, 3차원 영상 촬영을 위한 피폭량 및 영상정보의 급격한 증가는 환자에게 추가적인 부담이 될 수 있다. 본 연구에서는 영상의 구조 정보를 효과적으로 추출할 수 있는 Structural similarity (SSIM) 인자를 도입하여 매일 촬영하는 환자의 2차원 X선 영상간 차이를 자동 분석하여 환자의 위치 정확성의 검증 가능성을 제시하였다. 먼저 종양을 모사하기 위하여 구형 전산 팬텀의 크기와 위치를 변화시키면서 각각의 투사 영상을 시뮬레이션하고, SSIM 인자를 통해 영상 간 차이를 검출하여 분석하였다. 또한 12일간 매일 촬영한 방사선 치료 환자의 2차원 X선 영상들 간 차이를 동일한 방법으로 검출하였다. 그 결과 산출된 팬텀 변화에 따른 SSIM 값은 0.85~1 범위로, 관심영역(ROI)을 영상 전체가 아닌 팬텀으로 한정하였을 때는 0.006~1 범위로 나타나서 ROI 적용 시 민감도가 크게 상승하는 것을 확인하였다. 또한 임상 영상의 SSIM은 0.799~0.853 범위의 값을 나타냈으며 영상 간 차이가 SSIM 분포 상에 검출되는 것을 확인하였다. 본 연구결과는 소요 시간 및 피폭 등의 우려로 매일 사용하기 어려운 3차원 영상기법 대신 간단한 2차원 영상만을 이용하여 객관적이고 정량적인 환자 위치 정확성의 자동 평가 기법을 제공할 수 있을 것으로 기대된다.

Uranium Particle Identification with SEM-EDX for Isotopic Analysis by Secondary Ion Mass Spectrometry

  • Secondary ion mass spectrometry (SIMS) is a promising tool to measure isotope ratios of individual uranium particles in environmental samples for nuclear safeguards. However, the analysis requires prior identification of a small number of uranium particles that coexist with a large number of other particles without uranium. In the present study, this identification was performed by scanning electron microscopy - energy dispersive X-ray analysis with automated particle search mode. The analytical results for an environmental sample taken at a nuclear facility indicated that the observation of backscattered electron images with × 1000 magnification was appropriate to efficiently identify uranium particles. Lower magnification (less than × 500) made it difficult to detect smaller particles of approximately 1 μm diameter. After identification, each particle was manipulated and transferred for subsequent isotope ratio analysis by SIMS. Consequently, the isotope ratios of individual uranium particles were successfully determined without any molecular ion interference. It was demonstrated that the proposed technique provides a powerful tool to measure individual particles not only for nuclear safeguards but also for environmental sciences.

Mining Structure Elements from RNA Structure Data, and Visualizing Structure Elements

  • Most currently known molecular structures were determined by X-ray crystallography or Nuclear Magnetic Resonance (NMR). These methods generate a large amount of structure data, even far small molecules, and consist mainly of three-dimensional atomic coordinates. These are useful for analyzing molecular structure, but structure elements at higher level are also needed for a complete understanding of structure, and especially for structure prediction. Computational approaches exist for identifying secondary structural elements in proteins from atomic coordinates. However, similar methods have not been developed for RNA due in part to the very small amount of structure data so far available, and extracting the structural elements of RNA requires substantial manual work. Since the number of three-dimensional RNA structures is increasing, a more systematic and automated method is needed. We have developed a set of algorithms for recognizing secondary and tertiary structural elements in RNA molecules and in the protein-RNA structures in protein data banks (PDB). The present work represents the first attempt at extracting RNA structure elements from atomic coordinates in structure databases. The regularities in the structure elements revealed by the algorithms should provide useful information for predicting the structure of RNA molecules bound to proteins.

3D Structure of Bacillus halodurans O-Methyltransferase, a Novel Bacterial O-Methyltransferase by Comparative Homology Modeling

  • Bacillus halodurans O-methyltransferase (BhOMT) is a S-adenosylmethionine (SAM or AdoMet) dependent methyltransferase. Three dimensional structure of the BhOMT bound to S-adenosyl-L-homocysteine (SAH or AdoHcy) has been determined by comparative homology modeling. BhOMT has 40% sequence identity with caffeoyl-CoA 3-O-methyltransferase (CCoAOMT) from alfalfa. Based on x-ray structure of CCoAOMT, three dimensional structure of BhOMT was determined using MODELLER. The substrate binding sites of these two proteins showed slight differences, but these differences were important to characterize the substrate of BhOMT. Automated docking study showed that four flavonoids, quercetin, fisetin, myricetin, and luteolin which have two hydroxyl groups simultaneously at 3'- and 4'-position in the B-ring and structural rigidity of Cring resulting from the double bond characters between C2 and C3, were well docked as ligands of BhOMT. These flavonoids form stable hydrogen bondings with K211, R170, and hydroxyl group at 3'-position in the Bring has stable electrostatic interaction with Ca2+ ion in BhOMT. This study will be helpful to understand the biochemical function of BhOMT as an O-methyltransferase for flavonoids.

Phospholipid polymer can reduce cytotoxicity of poly (lactic acid) nanoparticles in a high-content screening assay

  • The objective of this study was to evaluate the cytotoxicity of poly (lactic acid) (PLA) nanoparticles. We used a water-soluble, amphiphilic phospholipid polymer, poly (2-methacryloyloxyethyl phosphorylcholine-co-n-butyl methacrylate) (PMB30W), as a stabilizer for the PLA nanoparticles. The PLA nanoparticles and PMB30W-modified PLA (PLA/PMB30W) nanoparticles were prepared by evaporating tetrahydrofuran (THF) from its aqueous solution. Precipitation of the polymers from the aqueous solution produced PLA and PLA/PMB30W nanoparticles with a size distribution of $0.4-0.5{\mu}m$. The partial coverage of PMB30W on the surface of the PLA/PMB30W nanoparticles was confirmed by X-ray photoelectron spectroscopy (XPS) and dynamic light-scattering (DLS). A high-content automated screening assay (240 random fields per group) revealed that the PLA nanoparticles induced apoptosis in a mouse macrophage-like cell line (apoptotic population: 73.9% in 0.8 mg PLA/mL), while the PLA/PMB30W nanoparticles remained relatively non-hazardous in vitro (apoptotic population: 13.8% in 0.8 mg PLA/mL). The reduction of the apoptotic population was attributed to the phosphorylcholine groups in the PMB30W bound to the surface of the nanoparticle. In conclusion, precipitation of PLA in THF aqueous solution enabled the preparation of PLA nanoparticles with similar shapes and size distribution but different surface characteristics. PMB30W was an effective stabilizer and surface modifier, which reduced the cytotoxicity of PLA nanoparticles by enabling their avoidance of the mononuclear phagocyte system.
