Isolation and Identification of the Fungi Producing a Soybean Milk Clotting Enzyme (두유 응고효소 생산 곰팡이의 분리 및 동정)

  • Twenty-five fungal strains producing an extracellular soybean milk clotting enzyme were isolated from 146 soil samples, and identified as 11 species belonging to six genera of Aspergillus oryzae (5 strains), Aspergillus flavus (2 strains), Aspergillus parasiticus (1 strain), Aspergillus tamarii (2 strains), Aspergillus niger (4 strains), Aspergillus fumigatus (2 strains), Mucor hiemalis (2 strains), Wallemia sebi (4 strains), Scopulariopsis condida (1 strain), Fusarium redolens(1 strain) and Verticillum lecanii (1 strain). Among them, Aspergillus oryzae 020 and Aspergillus tamarii 287 showed relatively high soybean milk clotting activity. The coagulabilities of the enzyme from representative strains of those species decreased as the pH of soybean milk increased from 6.0 to 7.0 The optimum temperature for soybean milk clotting enzymes of those strains were 65$^{\circ}C$.

Inhibitory Effect of the Culture Broth Extract of Aspergillus tamarii on Nitric Oxide Production and Its Antioxidative Activity

  • Many studies have explored suppression of aflatoxin produced by Aspergillus Genus. On the other hand, this study examined the inhibitory effect of the culture broth extract (CE) of A. tamarii obtained from dead silkworm on nitric oxide (NO) production and its antioxidative activity. The culture broth was extracted with EtOAc, dried, and then used in this experiment. As a result, CE did not show cytotoxicity on RAW 264.7 cells at any concentration. Moreover, CE suppressed lipopolysaccharide (LPS)-induced NO production of RAW 264.7 cells in a dose-dependent manner. The total phenol content according to the Folin-Dennis method, the antioxidative activity by DPPH, and the nitrate radical scavenging capacity of CE were increased in a dose-dependent manner. Thus, many of the phenolic compounds were considered to represent the antioxidative activity.

Roles of Endophytic Fungi Isolated from Mangifera indica L. in Promoting Plant Growth

  • Endophytic fungi have been shown to synthesize bioactive secondary metabolites, some of which promote plant growth through various mechanisms. In our previous study, endophytic fungi were isolated from mango trees (Mangifera indica L.). The present study examined fifty endophytic fungal isolates for mineral solubilization activity, ammonia production, and siderophore production. It was shown that these isolates could produce phytohormones indole-3-acetic acid and gibberellic acid, as well as inhibit plant pathogens, specifically Colletotrichum gloeosporioides and Lasiodiplodia theobromae. The results showed that all the isolated fungal endophytes exhibited various activities. Based on the findings, two fungal endophytes-Aureobasidium pullulans CY.OS 13 and Aspergillus tamarii CY.OS 144-were selected for dual inoculation in chili plants under pot-scale conditions to investigate their potential to improve growth-related traits such as seed germination, shoot and root length, biomass, and chlorophyll content. Seed treated with A. pullulans CY.OS 13 and/or A. tamarii CY.OS 144 showed a significant (p < 0.05) increase in seed germination and growth parameters of chili plants grown under pot-scale conditions. Particularly, chili plants whose seeds were injected with a combination of the two selected endophytic fungi showed the highest plant development traits. Therefore, the selected endophytic fungi have the potential to be used as biofertilizers, especially when combined. They could eventually replace chemical fertilizers because they are environmentally friendly, beneficial to humans, and can even promote sustainable agriculture.

  • The growth and multiplication of Aspergillus flavus Link and A. tamarii Kita were observed in vitro under variable temperatures of $22-31^{\circ}C$. The matte weight of mycelium and number of conidia/ml were significantly higher (P < 0.01) at the higher temperature than the lower temperature in both the species of Aspergillus. In vivo the mortality in silkworm, Bombyx mori L. with the infection of Aspergillus species was significantly (P < 0.01) different at different temperature and relative humidity conditions.

Re-Identification of Aspergillus Subgenus Circumdati Strains in Korea Led to the Discovery of Three Unrecorded Species

  • Aspergillus is one of the largest and diverse genera of fungi with huge economical, biotechnological, and social significance. Taxonomically, Aspergillus is divided into six subgenera comprising 27 sections. In this study, 235 strains of Aspergillus subgenus Circumdati (section: Candidi, Circumdati, Flavi, Flavipedes, Nigri, and Terrei) preserved at the Korean Agricultural Culture Collection (KACC) were analyzed and re-identified using a combined dataset of partial b-tubulin (BenA), Calmodulin (CaM) gene sequences and morphological data. We confirmed nineteen species to be priorly reported in Korea (A. neotritici, A. terreus, A. floccosus, A. allahabadii, A. steynii, A. westerdijkiae, A. ochraceus, A. ostianus, A. sclerotiorum, A. luchuensis, A. tubingensis, A. niger, A. welwitschiae, A. japonicus, A. nomius, A. tamarii, A. parasiticus, A. flavi, and A. oryzae). Among the studied strains, three species (A. subalbidus, A. iizukae, and A. uvarum), previously unreported or not officially documented, were discovered in Korea, to the best of our knowledge. We have given a detailed description of the characteristic features of the three species, which remain uncharted in Korea.

Investigation of Enzymatic Activities in Marine Algae-Derived Fungi

  • Marine macroalgae are important in coastal ecosystems and interact with marine microorganisms. In this study, we isolated fungi from seven types of marine macroalgae including Cladophora sp., Gloiopeltis furcate, Gracilariopsis chorda, Hydroclathrus clathratus, Prionitis crispata, Sargassum micracanthum, and Ulva lactuca collected in Korea. Morphological and phylogenetic analyses identified the isolates as four Aspergillus spp. (A. fumigatus, A. sydowii, A. tamarii, and A. terreus), three Penicillium spp. (P. crustosum, P. jejuense, and P. rubens), and Cladosporium tenuissimum. Among them, A. fumigatus TOP-U2, A. tamarii SH-Sw5, and A. terreus GJ-Gf2 strains showed the activities of all enzymes examined (amylase, chitinase, lipase, and protease). Based on the enzymatic index (EI) values in solid media, A. terreus GJ-Gf2 and C. tenuissimum UL-Pr1 exhibited the highest amylase and lipase activities, respectively. Chitinolytic activity was only observed in A. terreus GJ-Gf2, A. tamarii SH-Sw5, and A. fumigatus TOP-U2. Penicillium crustosum UL-Cl2 and C. tenuissimum UL-Pr1 showed the highest protease activities. To the best of our knowledge, this is the first report of lipolytic and proteolytic activities in a marine-derived C. tenuissimum strain. Overall, the fungal strains isolated from the marine macroalgae in this study actively produced industrially important enzymes.

Screening of Fungicides and Natural Plant Products and Their Efficacy on Control of Aspergillosis in Silkworm, Bombyx mori L.

  • Seven fungicides viz., salcylic acid, bacistin (Carbandazim 50% WP), bayleton (Triadimefon 25% WP), Dithane M-45 (Mancozeb 75% WP), captan (Captaf 50% WP) formaldehyde and benzoic acid at three concentrations (0.50,0.75 and 1.0%) and ten plant products viz., Hena leaf, garlic bulb, tomato leaf, mango bark, cotton leaf, turmeric powder, onion, tulsi leaf, neem leaf and ginger at 1.0,2.0 and 3.0% concentrations were screened against Aspergillus flavus and A. tamarii in vitro. Among fungicides, salcylic acid and bavistin and among plants Hena and Mango bark powder were found to be very effective at all concentrations tested. Based on in vitro screening, only selected six fungicides at 1.0, 1.5 and 2.(0% and six plants at 2.0,4.0 and 6.0% concentrations were tested in vivo for controlling Aspergillosis in silkworm. Salcylic acid and bavistin fungicides and Hena leaf powder and Mango bark powder have shown considerable effect in controlling Aspergillus infection in silkworm at all concentrations tested.

Studies on Mycoflora and Mycotoxins of Cowpea Cultivars (동부 콩열매에 있는 진균류와 균독소에 관한 연구)

  • Thirty three species and two species varieties belonging to 14 genera of fungi were collected from 20 cowpea cultivars on glucose Czapek's agar (11 genera and 25 species+1 var.) and glucose-Czapek's agar supplemented with 10% NaCl (7 genera and 18 species+2 var.) at $28{\pm}2^{\circ}C$. The total count of fungi were 6716 colonies/g in all cowpea cultivars. On glucose-Czapek's agar and identified; Aspergillus flavus, A. niger, A. sydowii, A. flavus var. columnaris, A. terreus, Penicillium chrysogenum, Emericella nidutans and Rhizopus stolonifer. The total count of halotolerant or halophilic fungi was 3515 colonies/g on 10% NaCl-glueose-Czapek's agar and identified; the most common species were: A. flavus, A. sydowii, A. tamarii A. flavipes, A. niger, A. flavus var. columnaris, A. ochraceus, A. oryzae and P. chrvsogenusm. Thin layer chrormatographic analysis of chloroform extracts of the different seed samples revealed that four cultivars were naturally contaminated with aflatoxins $B_1,\;B_2,\;G_1$ and $G_2$, $(45-112\;{\mu}g/kg)$.

Insecticidal Activity of Ethyl Acetate Extracts from Culture Filtrates of Mangrove Fungal Endophytes

  • In the search for novel potent fungi-derived bioactive compounds for bioinsecticide applications, crude ethyl acetate culture filtrate extracts from 110 mangrove fungal endophytes were screened for their toxicity. Toxicity tests of all extracts against brine shrimp (Artemia salina) larvae were performed. The extracts with the highest toxicity were further examined for insecticidal activity against Spodoptera litura larvae and acetylcholinesterase (AChE) inhibition activity. The results showed that the extracts of five isolates exhibited the highest toxicity to brine shrimp at 50% lethal concentration ($LC_{50}$) values of 7.45 to 10.24 ppm. These five fungal isolates that obtained from Rhizophora mucronata were identified based on sequence data analysis of the internal transcribed spacer region of rDNA as Aspergillus oryzae (strain BPPTCC 6036), Emericella nidulans (strains BPPTCC 6035 and BPPTCC 6038), A. tamarii (strain BPPTCC 6037), and A. versicolor (strain BPPTCC 6039). The mean percentage of S. litura larval mortality following topical application of the five extracts ranged from 16.7% to 43.3%. In the AChE inhibition assay, the inhibition rates of the five extracts ranged from 40.7% to 48.9%, while eserine (positive control) had an inhibition rate of 96.8%, at a concentration of 100 ppm. The extracts used were crude extracts, so their potential as sources of AChE inhibition compounds makes them likely candidates as neurotoxins. The high-performance liquid chromatography profiles of the five extracts differed, indicating variations in their chemical constituents. This study highlights the potential of culture filtrate ethyl acetate extracts of mangrove fungal endophytes as a source of new potential bioactive compounds for bioinsecticide applications.
