Studies on the Classification of Aspergillus spp. by Fluorence Antibody Reaction (형광항체반응(螢光抗體反應)에 의(依)한 Aspergillus spp.의 분류(分類)에 관(關)한 연구(硏究))

  • Author investigated fluorence antibody reaction for the antigenic relationships between Asp niger group, Asp flavus and Asp parasiticus which was indicated as follows: 1. It was concluded that there are complete differences in the antigenic properties each other because it has not cross reaction, therefore identification of strains will be simpley classified. 2. A complete cross reaction between Asp flavus and Asp parasitic us in the Asp flavus groups existed, accordingly this reaction could not identified the strain and classified between Asp. flavus and Asp. parasiticus. 3. This experiment also followed with the separated each strains from the origin (Meju, Nuruk, ATCC, NRRL), but there no differences. From the above results, this method could be classified between Asp flavus group and Asp niger group in the genus Aspergillus, but classification of Asp. flavus and Asp. parasiticus should hardely conclude with this method.

Studies on the Serological Classification for Korean aspergilli (한국산 Aspergilli의 혈청학적 분류방법)

  • Of the Asp. spp. isolated by the Institute of Applied Microbiology, Kon-Kuk University, 7 strains were selected for the study of the immunological differencences among them using gel precipitation test. The strains were the following types : 1 type of flavus and 2 types of oryzae were isolated from Meju ; 1 type of flavus from Nuruk ; and each one type of flavus, parasiticus and oryzae from Kokja.Asp.flavus from ATCC, Asp. parasiticus nad Asp. niger NRRL strains were also used in the study as a standard. From this study, several points can be raised ; 1) There was no common antigenic property between Asp. niger and Asp. flavus, because of no formation of reaction line. Therefore, all strains could be easily distinguished. 2) There was common antigenic property, that is, the formation of reaction line between Asp. flavus and Asp. parasticus. Accordingly two strains could not be easily distinguished by the gel precipitation test. 3) Each type of oryzae, parasiticus and flavus of Asp. flavus group had common antigen one another as well as specific antigens only in the difference of the reaction lines, so they could be easily identified in the gel precipitation test. 4) Each isolated strain from Meju and Nuruk appeared to be identical. 5) It was shown that the gel precipitation test of serological methods was very useful for the classification of Asp. spp.

Effect of Various Fungi on the Aflatoxin Productivity in tile Culture of Asp. Flavus (진균류(眞菌類)의 상호작용(相互作用)에 의한 Aflatoxin 생성능(生成能)에 관(關)한 연구(硏究))

  • This study was attempted to know that the interactions of various fungi, and methionine and $MgSO_4$ introduced as the substrate of culture media for fungi were affected to produce aflatoxins by Asp. flavus. 5 different fungi were isolated from the fermented soybean mash and were cultured in Chemically Defined medium (C. D. media) and soybean mash at $25^{\circ}C$ for 10 days. (1) It was confirmed that Asp. flavus produced aflatoxins in the C. D. medium and soybean mast, but that Asp. niger, Asp. oryzae, Asp. awamori and Asp. terreus did not produced them respectively. (2) Asp. flavus cultured with Asp. niger did not produce aflatoxins in C. D. medium, but produced in soybean mash, in other hand, Asp. flavus with other fungi except Asp. niger produced aflatoxins in C. D. medium and soybean mash. (3) The growth of fungi were more prosperous in the seperate culture than in the mixed culture. (4) In the C. D. medium added 20% of cultured medium of Asp. niger, Asp. flavus did not produce aflatoxins but other cultured medium did not prohibit the production of aflatoxins by Asp. flavus. (5) On the contrary, $MgSO_4$ increasing the productivity of aflatoxins by Asp. flavus in the C. D. medium, methionine known as one of precursor of aflatoxins did not affected the increasing productivity with significance.

The taxonomical studies of Korean Aspergilli (한국산 Aspergilli 에 대한 분류학적 연구)

  • Through the present experiment the various Aspergilli have been isolated from 53 Meju specimens (fermented soy bean cake) which were collected from 43 local areas in Korea and those of them are arranged as 6 different groups: Asp. flavus-oryzae group (38 strains), Asp. niger group (11 strains) Asp. nidulans group (5 strains), Asp. clavatus group (4 strains), Asp. fumigatus group (3 strains), and Asp. ochraceus group (2 strains), as the result of identification of them by the group key of Thom and Raper. Further identifications of them according to the species keys of Thom and Raper, Yamata and Sakakuchi have been also conducted and the results obtained are following as: 1. Group members (38) of Asp. flavus-oryzae group are identified as. Asp. oryzae, Asp. oryzae var. fulvus, Asp. flavus. and Asp. parasiticus, not identified two different strains belong to this group. 2. Group members (11) of Asp. niger group are identified as Asp. niger and Asp. phoenicus. 3. Group members (5) of Asp. nidulans groups are identified as the same species, Asp. nidulans. 4. Group members (4) of Asp. clavatus group are identified as the same species, Asp. clavatits. 5. Group members (3) of Asp. fumigatus group are identified as the same species, Asp. fumigatus. 6. Group members (2) of Asp. ochraceits group are identified as the same species, Asp. sulphereus.

Control of Aflatoxin and Characteristics of the Quality in Doenjang(soybean paste) Prepared with Antifungal Bacteria (길항미생물에 의한 된장 중 아플라톡신 제어 및 그 품질특성)

  • In oder to acquire microbial agents that can be utilized for control of aflatoxin produced by Aspergillus. flavus and Asp. parasiticus, antifungal bacteria were isolated. Antifungal bacteria was identified as Bacillus spp. based on morphology and physico-biochemical characteristics. Amount of aflatoxin $B_1$ from Doenjang(soybean paste) prepared with Asp. flavus, Asp. parasiticus, antifungal bacteria(Bacillus sp.), or mixture of Asp. flavus and Asp. parasiticus was 27.2 ppb, 30.3 ppb, 3.4 ppb, and 3.7 ppb, respectively. Aflatoxin $B_1$ was not detected from Doenjang(control) and Doenjang prepared with antifungal bacteria. Content and compositions of free sugars, fatty acid, organic acid and free amino acid in Doenjang prepared with Asp. flavus and Asp. parasiticus, antifungal bacteria and mixture of Asp. flavus and Asp. parasiticus were not significantly different. For volatile flavor compounds of Doenjang prepared with antifungal bacteria, 2-pentyl furan and butanoic acid were disappeared or reduced, while octadecene compounds were produced. However, those of Doenjang prepared with Asp. flavus or Asp. parasiticus and Doenjang(control) were not significantly different. These results suggested that the antifungal bacteria(Bacillus sp.) inhibited production of aflatoxin and that antifungal bacteria did not effect the quality of Doenjang.

Radiation Sensitivity of Some Food Decay Fungi (몇몇 음식 부식 균류의 방사선 감수성에 대한 연구)

  • Five species of food decay fungl, Aspergillus flavus, Asp. uiger, Penicillum sp., Botrytis cinerea, and Rhizopus stolonifer, were examined for their radio sensitivity in several suspension media. Asp. flavus, Asp. niger and Penicillum sp. have almost the same sensitivity toward gamma rays. with D value in the range of 30 to 35 K rad, whereas Botrytis cinerea has a D value of approximately 55K rad and Rhizopus stolonifer, the most re4sistant fungus studied, has a crease in their radioresistnace when compared with spores irradiated in water. Asp. flavus and penicillium sp. spores irradiated in citrate buffer at pH3-7 showed almost no change in their radisensitiity with pH, but Botryis cinerea spores showed a distinct decrease in their radioresistnace at pH 6 and 7. Penicillum sp. spores irradiated in sucrose solutions showed no sinificant change in their radioresistance. Botrytis cinerea spores displayed a higher radioresistance when they were irradiated in sucrose solution than in water.

Radiation Sensitivity of Some Toxigenic Molds Isolated from Deteriorated Rice (변질미(變質米)에서 분리된 독소생성(毒素生成)곰팡이의 방사선(放射線) 감수성(感受性))

  • Seven toxigenic molds isolated from deteriorated rice in Korea, Aspergillus clavatus, Asp. flavus var. columnaris, Asp. fumigatus, Asp. ochraceus, Penicillium citrinum, Pen. implicatum, and Pen. islandicum, were examined for their sensitivity toward Co-60 gamma rays for survival. Conidia of all tested species showed survival curves of sigmoidal type, from which decimal reduction doses were found to vary in the range of $14{\sim}33$ krad, induction doses, in the range of $12{\sim}56$ krad and inactivation factors at 200 krad, in the range of $4.6{\sim}12.8$, all at a dose rate of 11.56 krad/min. Dose rate effects on the radiosensitivity of Asp. flavus and Pen. islandicum conidia indicated that the higher dose rate (11.56 krad/min) caused decreases in decimal reduction doses as compared with the lower dose rate (2.67 krad/min).

  • Effects of combined treatments of radiation-heat and radiation-chemicals on the survival of mycotoxin-producing molds Aspergillus flavus and Penicillum islandicum conidia were investigated. Combined treatments of gamma-radiation and heat (10 minutes at $50^{\circ}C\;or\;55^{\circ}C$) showed a synergistic effect, causing remarkable decreases in $D_{10}$ values and induction doses in the two molds. Combined treatments of gamma-radiation and chemicals (sorbic acid) had no synergistic effect. Asp. flavus was more resistant to heat and sorbic acid than was Pen. islandicum.

Isolation of Microorganisms from Red Pepper Powder and their Radiosensitivity (고추가루중 미생물(微生物)의 분리(分離) 및 방사선 감수성(感受性))

  • From samples of red pepper powder sold in Korea were isolated and identified 13 species of molds (Aspergillus amstelodami, Asp. chevalieri, Asp. clavatus, Asp. flavus, Asp. janus var. effusus, Asp. oryzae, Asp. oryzae var. brevis, Asp. repens, Asp. sydowi, Asp. thomii, Asp. tubingensis, Penicillium thomii, Scopulariopsis brevicaulis) and 5 species of bacteria (Bacillus pumilus, Bac. subtilis, Micrococcus luteus, M. varians, Staphylococcus aureus). Radiosensitivity of these microorganisms was examined to give $D_{10}$ values of $14{\sim}41\;krad$ for molds, $11{\sim}24\;krad$ for bacterial vegetative cells and $190{\sim}250\;krad$ for bacterial spores. The red pepper powder was contaminated with $2{\sim}3{\times}10^2$ mold counts/g and $3{\sim}6{\times}10^7$ bacterial counts/g, which would be sufficiently destroyed by irradiating 200 krad ${\gamma}-rays$.

  • 62 specimens of deteriorated rice which were collected all over Korea, were classified according to their color outlooks, and the causitive microorganisms were isolated and identified. The following results were observed; 1. 62 specimens of deteriorated rice were classified according to color outlooks into 7 types; reddish yellow, light reddish yellow, light gray yellow, light reddish yellow, light gray yellow, light red, ligt gray, dark gray and rice weevil type. 2. The isolated microorganisms from 62 specimens of deteriorated rice were 44 species, 5 genera of molds, 1 species of yeast, and 14 species, 4 genera of bacteria. 3. The frequently observed microorganisms which caused the deterioration were Asp. glaucus group, Asp. oryzae, Asp. candidus, and Asp. versicolor. Among bacteria, Bacillus was dominant. 4. The relationship between color outlook types and the deterioration causing microrrganisms was not definite, but Pen. islandicum from reddish yellow, Asp. candidus and Asp. clavatus from light reddish yellow, Asp. glaucus group, Asp. candidus, Asp. versicolor and Asp. fumigatus from light red, Asp. oryzae from light gray yellow, and Asp. glaucus group and Pencillium species from light gray and dark gray were chiefly isolated. 5. As mycotoxin producing fungi, Asp. fumigatus, Pen. citrinum, Pen. islandicum, Asp. flavus, and Asp. ochraceus were detected, but their growth frequencies were not very high to be problems except Pen, islandicum which infected an imported rice sample heavily.
