생물반응기에서 배양한 가시오갈피 유식물체의 순화 및 생육특성 (Acclimatization and Growth Characteristics of Plantlets of Eleutherococcus senticosus Maxim Cultured by Bioreactor)

  • 가시오갈피 체세포 배를 이용한 우량종묘의 대량생산체계를 확립할 목적으로 체세포 배의 생물반응기 배양을 통해 얻은 식물체의 순화에 관한 기초시험을 수행하였다. 가시오갈피 기내배양 식물체의 토양활착은 밀폐된 공간보다 비가림 하지 않은 차광막조건에서 양호하였고 순화에는 Klasmann 상토와 원예용상토가 좋았다. 배양실 10도와 차광막에서 3월20일 순화한 기내식물체의 순화 율은 모두 90% 이상으로 양호하였다. 배양실조건에서는 $26^{\circ}C$의 순화온도에서는 10% 이하의 극히 저조한 순화 생존율을 보였으나 포장 차광막조건에서는 최고 온도가 $30^{\circ}C$를 넘는 5월20일 순화 처리구에서도 68%의 높은 순화생존율을 보여 불리한 환경조건에서 차광막순화가 순화에 더욱 적합하였다. 차광조건으로는 50% 차광망 처리가 적합하였다. 차광막에서 순화한 식물체의 묘 소질은 3월10일 순화를 시작한 식물체가 가장 좋았다.

  • In this paper we have implemented and tested TPN which is system to supply sufficent nutrition to nutritionally deficient patient by means of ES (expert system) a kind of A.1 (artificial intelligence) . This system affords to evaluation of nutritional state of patient which is essential to physi- cian. who performs TPN, decision of performing TPN and management of patient-data & calculation of information needing to making TPN fluid. The features were as follolv 1. we input data, take ideal weight of patient and 24hr's creatlnln In urine according to chart in system compare TSF (triceps skin fold), MAC (mid-arm circumference), AMC (arm muscle circumference) to 5th, 15th, 50th percentile and evaluate the nutritional state of patient. 2. Calculation of protein & nonprotein calorie needing to treament of patient can be made exactly by stress factor, activity factor and body temperature. 3. patient's personal recording needing to management of patient date name of chief doc- tor, name of department of admission, chart number, history can by taken very easily. 4. The way of system operating is pull-down Menu one, It can be processing very efficiently. 5. Date processing in system, we can manage memory volume of computer verlr efficiently using of dynamic allocation variables. 6. We can make it very easy to edit & revise the input data, processed data is saved to diskette in 2 files (TDF, THF) , these are semipermanent preservation.

원행을묘정리의궤(園行乙卯整理儀軌) 중(中) 조리면(調理面)에서 본 반과상고(盤果床考) (A Study of Cookery of Daily Meal (Bankwa Sang: Fruit Table) in Wonheng Ulmyo Jungri Euigwae (1795))

  • To analyze dialy meal of royal meal, studied Bankwa Sang(Fruit Table) were on record Wonheng Ulmyo Jungri Euigwae (1795). Historic book 'Jungri Euigwae' described the king's visit to his father's royal tomb 'Hyun Neung Won', during the domain of Cheung Jo, the 22nd king of Choson Dynasty. The results obtained from this study areas follows. The fruit table, which similar in concept to desert in the west but quite different in service, was prepared for a guest. The table consisted of two kinds of trays, on which serveral kinds of fruit were stacked to a height of between 4 chon (4寸 : about 12 cm) and 1 chuk(1尺 : about 30.3 cm) according to Korean measurement system. The table was decorated with beautiful artificial flowers made of paper and silk. The number of sets to be arranged on the table were different according to the royal status of the eater: 12 sets-25 sets for king's mother, 7 sets-11 sets for the king. Soy sauce mixed with vinegar and pine-nu meal, mustard were ruled out from kind of sets. Kinds of dishes served with a meal generally were noodles (麵), soup (湯), fried fish (煎油花), fried meats and vegetables (花陽灸), slices of raw fish (魚膾), minced raw meat (肉膾), slices of boiled beef (片肉), stew (蒸), rice cake (餠), sweet rice dish (藥飯), patterned savory cake (茶食), fried cake made of wheat flour, honey and oil (藥果), fried glutinous rice cake (强精), various fruits preserved in honey (正果), sugar candies (各色糖), fruits (果物) honey (淸), soy sauce mixed with vinegar and pine-nut meal (醋醬), mustard (莽子).

느타리버섯 자실체 및 균사체 추출물의 항산화효과 (Antioxidative Effect of Fruit Body and Mycelial Extracts of Pleurotus ostreatus)

  • 느타리버섯 자실체 및 액체배양한 균사체를 핵산, 에탄올, 물 순으로 순차하여 얻은 추출물에 대하여 천연항산화제로 이용할 수 있는 가능성을 검토하였다. 용매별 추출수율은 자실체 및 액체배양 균사체 모두 물추출 분획의 추출 수율이 $35{\sim}48%$로 가장 높았다. 총 당의 함량은 ethanol추출 분획과 물추출 분획의 ethanol가용부에서 높은 함량을 나타내었으며, phenol성 화합물의 함량은 자실체 및 균사체 모두 hexane 분획에서 높은 함량을 보였다. 용매별로 추출한 추출물을 대두유에 500-3000ppm 첨가하여 일정 시간마다 과산화물가를 측정한 결과, 자실체 및 균사체 모두에서 ethanol 분획이 물 및 hexane 분획에 비해 강한 항산화능을 보였으며 BHA 및 BHT를 처리한 비교구 및 무처리구에 비해서도 항산화성이 우수하였으며 열안정성도 확인되었다.

  • 자색 고구마 뻥튀기의 실험결과는 색도 중 $L^*$ 값이 팽화시간이 증가할수록 $L^*$값이 감소하는 유의적인 차이를 나타내었으나 수분함량이나 팽화온도에 따른 차이는 나타내지 않았다. $a^*$값은 팽화시간, 팽화온도가 증가함에 따라 함께 증가하는 유의적 차이를 나타냈으며, $b^*$값은 몇몇 시험구에서 팽화시간과 팽화온도가 증가할수록 $b^*$값도 증가하였다. 비체적은 수분함량과 팽화온도가 높아질수록 증가하였고, 수분함량 11%, 팽화온도 $246^{\circ}C$, 팽화시간 4초에서 제조한 시험구가 가장 높은 값을 나타내었다. 또한 자색 고구마 뻥튀기의 파괴력은 팽화온도와 팽화시간이 증가함에 따라 함께 증가하며 수분함량이 낮을수록 높은 파괴력을 나타내는 유의적인 차이를 보였다. 관능평가에서 7% 수분함량, 팽화온도 $246^{\circ}C$, 팽화시간 5초에서 제조한 뻥튀기가 패널들에게 높은 점수를 얻었다.

백김치 유래 유산균을 이용한 요구르트의 Anti-Helicobacter pylori 활성 (Anti-Helicobacter pylori Activity of Yogurt Fermented with Lactic Acid Bacteria from Baikkimchi)

  • 백김치로부터 분리된 유산균으로 제조된 요구르트를 냉장 보관하는 동안 미생물학적 및 물리화학적 특성 및 Helicobacter pylori ATCC 43504에 대한 항균 활성을 조사하였다. 요구르트의 유산균수, 적정산도, 점도 및 총 고형물 함량은 사용된 균주에 따라 유의적인 차이가 있었으나, 발효 직후부터 7일간 저장하는 동안 유의할만한 차이 없이 일정하게 유지되었다. Lactobacillus brevis BK11과 Lactobacillus paracasei BK57 균주로 발효시킨 요구르트는 인공 위액과 담즙액에 대해 다른 균주들 보다 강한 저항성을 보였다. 한편, 이들 유산균으로 제조한 요구르트 내에 존재하는 유산 생성량은 상대적으로 높았으므로 H. pylori와 혼합 배양한 결과 대조구에 비해 유의적인 항균 효과를 나타낸 것으로 추정되었다. 특히, L. brevis BK11에 의해 발효시킨 요구르트에 의해선 AGS 세포에 대한 H. pylori의 부착을 억제할 수 있었고, 이들이 생산하는 urease의 활성을 낮추는데도 효과적이라는 것을 확인하였다.

Isolation and Characterization of an Agarase-Producing Bacterial Strain, Alteromonas sp. GNUM-1, from the West Sea, Korea

  • The agar-degrading bacterium GNUM-1 was isolated from the brown algal species Sargassum serratifolium, which was obtained from the West Sea of Korea, by using the selective artificial seawater agar plate. The cells were Gram-negative, $0.5-0.6{\mu}m$ wide and $2.0-2.5{\mu}m$ long curved rods with a single polar flagellum, forming nonpigmented, circular, smooth colonies. Cells grew at $20^{\circ}C-37^{\circ}C$, between pH 5.0 and 9.0, and at 1-10% (w/v) NaCl. The DNA G+C content of the GNUM-1 strain was 45.5 mol%. The 16S rRNA sequence of the GNUM-1 was very similar to those of Alteromonas stellipolaris LMG 21861 (99.86% sequence homology) and Alteromonas addita $R10SW13^T$(99.64% sequence homology), which led us to assign it to the genus Alteromonas. It showed positive activities for agarase, amylase, gelatinase, alkaline phosphatase, esterase (C8), lipase (C14), leucine arylamidase, valine arylamidase, ${\alpha}$-chymotrypsin, acid phosphatase, naphthol-AS-BI-phosphohydrolase, ${\alpha}$-galactosidase, ${\beta}$-galactosidase, ${\beta}$-glucosidase, catalase, and urease. It can utilize citrate, malic acid, and trisodium citrate. The major fatty acids were summed feature 3 (21.5%, comprising $C_{16:1}{\omega}7c/iso-C_{15:0}$ 2-OH) and C16:0 (15.04%). On the basis of the variations in many biochemical characteristics, GNUM-1 was considered as unique and thus was named Alteromonas sp. GNUM-1. It produced the highest agarase activity in modified ASW medium containing 0.4% sucrose, but lower activity in rich media despite superior growth, implying that agarase production is tightly regulated and repressed in a rich nutrient condition. The 30 kDa protein with agarase activity was identified by zymography, and this report serves as the very first account of such a protein in the genus Alteromonas.

Utilization and Isolation of new active substances from Sericulture Related MaterialsII. Development of an oral antihyperglycemic agent from silkworm powder

  • Since 1992, Sericulture & Entomology Research Institute(NSERI) and Kyung Hee University group screened an activity of lowering blood-glucose levels with silkworm related materials such as silkworm larvae powder, dried feces, pupae and silkworm moth in other to guide laymans to rational and educated utilization of silkworm-related materials for the treatment of diabetes mellitus. In experiments examining several silkworms in different stages and prepared in different conditions, a freeze dried silkworm powder of 5th instar 3rd day showed a higher glucose lowering activity about 20% than heat dried matured silkworm powder. Among the three dosage of 500mg, 830mg and 1,160mg in ate preliminary clinical trial, the 830mg exhibited a significant effect on postprandial blood glucose level and did and did not cause any hypoglycemic side effect. In the blood glucose lowering activity of mulberry and silkworm varieties, the Yongcheonppong and Samkwangjam showed the highest activity for lowering blood glucose levels. In experiments to see the difference in blood-glucose lowering activity between either male and female or larvae and pupae, activity of larvae was higher than that of pupae and the male was higher than female. The heating dry and artificial diet showed lower than mulberry diet and normal freeze dry of the 5th instar 3rd day. Among the sericultural products, larvae showed the highest activity. We find out the fact that effect of silkworm powder attributed to the inhibition of ${\alpha}$-glucohydrolase catalyzed reaction in the small intestine.

Spatial protein expression of Panax ginseng by in-depth proteomic analysis for ginsenoside biosynthesis and transportation

  • Background: Panax ginseng, as one of the most widely used herbal medicines worldwide, has been studied comprehensively in terms of the chemical components and pharmacology. The proteins from ginseng are also of great importance for both nutrition value and the mechanism of secondary metabolites. However, the proteomic studies are less reported in the absence of the genome information. With the completion of ginseng genome sequencing, the proteome profiling has become available for the functional study of ginseng protein components. Methods: We optimized the protein extraction process systematically by using SDS-PAGE and one-dimensional liquid chromatography mass spectrometry. The extracted proteins were then analyzed by two-dimensional chromatography separation and cutting-edge mass spectrometry technique. Results: A total of 2,732 and 3,608 proteins were identified from ginseng root and cauline leaf, respectively, which was the largest data set reported so far. Only around 50% protein overlapped between the cauline leaf and root tissue parts because of the function assignment for plant growing. Further gene ontology and KEGG pathway revealed the distinguish difference between ginseng root and leaf, which accounts for the photosynthesis and metabolic process. With in-deep analysis of functional proteins related to ginsenoside synthesis, we interestingly found the cytochrome P450 and UDP-glycosyltransferase expression extensively in cauline leaf but not in the root, indicating that the post glucoside synthesis of ginsenosides might be carried out when growing and then transported to the root at withering. Conclusion: The systematically proteome analysis of Panax ginseng will provide us comprehensive understanding of ginsenoside synthesis and guidance for artificial cultivation.

재래식 된장으로부터 아민 산화 효소를 생산하는 프로바이오틱 바실러스균의 분리 동정 (Isolation and Identification of Probiotic Bacillus strain Forming Amine Oxidase from Traditional Fermented Soybean Paste)

  • 본 연구의 목적은 3년 이상 숙성된 재래식 된장으로부터 아민 산화 효소를 생산하는 프로바이오틱 바실러스균을 분리 동정하는 것이다. 시료로부터 분리된 바이오제닉 아민(biogenic amines, BA) 생성균은 Bacillus sp. TS09, Bacillus licheniformis TS17, Bacillus subtilis TS19, Bacillus cereus TS23, Bacillus sp. TS30, Bacillus megaterium TS31, B. subtilis TS44, Bacillus coagulans TS46 및 Bacillus amyloliquefaciens TS59 등으로 동정되었다. 한편 동일한 시료로부터 분리된 B. subtilis TS04와 TS50은 인공 위액 및 담즙액에 대한 저항성, 장내 상피세포에 대한 부착능 및 BA 생성균(Bacillus sp. TS30 및 B. subtilis TS44)에 대한 박테리오신 생산 등의 프로바이오틱 활성을 나타내었다. 게다가 B. subtilis TS04와 TS50 균주가 생산한 아민 산화 효소에 의하여 카다베린, 푸트레신 및 티라민의 생성량을 감소시킬 수 있었으므로 이들은 BA 중독 위험을 낮출 수 있는 프로바이오틱스 소재로서의 활용 가치가 높을 것으로 판단된다.
