Production of Pups Following Artificial Insemination by Canine Intrauterine Inseminator (개 자궁내 인공수정기에 의한 인공수정 후 산자생산)

  • This study was conducted to develop an intrauterine inseminator (IUI) to deposit of frozen semen into uterus and to evaluate the results obtained after artificial insemination by IUI. Two Japanese spitzs (2 to 4 years of age) were used as semen donors. Semen was collected by manual masturbation into sterile glass collection tubes and separated into 3 fractions with only the sperm-rich fractions retained for further examination. Sperm motility >70%, sperm concentration of 200 to $400{\times}10^6 cells/ml$$\times$g for 5 min and poured out the suspended solution, and then diluted with 2 ml Tris-buffer which was consisted of 2.4 g Tris, 1.4 g citric acid, 0.8 g glucose, 0.1 $\mu\textrm{g}$/ml streptomycin, 100 IU/ml penicillin, 20 ml egg yolk to 100 ml mili-Q water (Ext I) or supplemented with 8 ml glycerol and 1 ml Equex STM paste to 100 rnl (Ext II). The diluted semen was cooled to 5$^{\circ}C$ in cold room, where the temperature in the sample reached 5$^{\circ}C$. Two h after beginning the cooling procedure, 2 ml of Ext II, also at 5$^{\circ}C$, was added and mixed by gently reversing the tubes several times during 1 h. The final sperm concentration for freezing was approximately $50{\times}10^6 cells/ml$. After equilibration, the semen was loaded into 0.5 ml straw and frozen on the liquid nitrogen vapour in styrofoam box. The straws were thawed at 7$0^{\circ}C$ for precisely 6 sec. After thawing of each straw, the frozen semen can survived over 50% motility. All the females were inseminated twice with 1 ml of $25{\times}10^6 cells/ml$

Negative Selection Algorithm for DNA Pattern Classification

  • We propose a pattern classification algorithm using self-nonself discrimination principle of immune cells and apply it to DNA pattern classification problem. Pattern classification problem in bioinformatics is very important and frequent one. In this paper, we propose a classification algorithm based on the negative selection of the immune system to classify DNA patterns. The negative selection is the process to determine an antigenic receptor that recognize antigens, nonself cells. The immune cells use this antigen receptor to judge whether a self or not. If one composes ${\eta}$ groups of antigenic receptor for ${\eta}$ different patterns, these receptor groups can classify into ${\eta}$ patterns. We propose a pattern classification algorithm based on the negative selection in nucleotide base level and amino acid level. Also to show the validity of our algorithm, experimental results of RNA group classification are presented.

  • Hollow fiber membrane has been successfully developed as an artificial kidney device in the 1970's. In the early 1970's animal cells were introduced into a hollow fiber membrane cartridge and well propagated in the cartridge. Since then, hollow fiber membrane was utilized as a bioreactor in order to immobilize enzymes as well as to culture microbial cells and plant cells. In this review, the present status and the prospect of hollow fiber membrane bioreactor are investigated in view of cell density and product productivity.

  • To improve antithrombogenicity of polymer that used in vascular graft and artificial organs, seeding of human endothelial cells on the polyurethane was studied. Human endothelial cells were ismlated from human umbilical veins, using type I collagenase, and identified with goat anti vWF antibodies. Human endothilial cell seeding was tried upon the polyurethane which has good mechanical property and resists stresses. The hydrophobic polyurethane surface was changed hydrophilic by corona discharf:e treatment. Surface hydrophilicity was measured with Wilhemly plate method and the goniometer. To evaluate matrix protein adsorption, fibronectin adsorption test was done. To eveluate cell adhesion, human endothelial cell attachment forces were measured rising a perfusion chamber of , ism diamter. Less cells were detached from the hydrophilically treated polyurethane. This showed that corona discharge on the polyurethane could improve matrix adsorption and endothelial cell attachment.

  • As part of the cell division method, we proposed a method for segmenting images generated by topography microscopes through deep learning-based feature generation and graph segmentation. Hybrid vector shapes preserve the overall shape and boundary information of cells, so most cell shapes can be captured without any post-processing burden. NIH-3T3 and Hela-S3 cells have satisfactory results in cell description preservation. Compared to other deep learning methods, the proposed cell image segmentation method does not require postprocessing. It is also effective in preserving the overall morphology of cells and has shown better results in terms of cell boundary preservation.

Effects Of Cultured Bone Cell On The Regeneration Of Alveolar Bone (배양골세포 이식이 치조골재생에 미치는 영향)

  • This study was performed to estimate the effects of cultured bone cell inoculated on porous type hydroxyaptite for the regeneration of the artificial alveolar bone defect. In this experiment 3 beagle dogs were used, and each of them were divided into right and left mandible. Every surgical intervention were performed under the general anesthesia by using with intravenous injection of Pentobarbital sodium(30mg/Kg). To reduce the gingival bleeding during surgery, operative site was injected with Lidocaine hydrochloride(l:80,000 Epinephrine) as local anesthesia. After surgery experimental animal were feeded with soft dietl Mighty dog, Frisies Co., U.S.A.) for 1 weeks to avoid irritaion to soft tissue by food. 2 months before surgery both side of mandibular 1st premolar were extracted and bone chips from mandibular body were obtained from all animals. Bone cells were cultured from bone chips obtained from mandible with Dulbecco's Modified Essential Medium contained with 10% Fetal Bovine Serum under the conventional conditions. Porous type hydroxyapatite were immerse into the high concentrated cell suspension solution, and put 4 hours for attachin the cells on the surface of hydroxyapatite. Graft material were inserted on the artificial bone defect after 3 days of culture. Before insertion of cellinoculated graft material, scanning electronic microscopic observation were performed to confirm the attachment and spreading of cell on the hydroxyapatite surface. 3 artificial bone defects were made with bone trephine drill on the both side of mandible of the experimental animal. First defect was designed without insertion of graft material as negative control, second was filled with porous replamineform hydroxyapatite inoculated with cultured bone marrow cells as expermiental site, and third was filled with graft materials only as positive control. The size of every artificial bone defect was 3mm in diameter and 3mm in depth. After the every surgical intervention of animals, oral hygiene program were performed with 1.0% chlorhexidine digluconate. All of the animals were sacrificed at 2, 4, 6 weeks after surgery. For obtaining histological section, tissus were fixed in 10% Buffered formalin and decalcified with Planko - Rycho Solution for 72hr. Tissue embeding was performed in paraffin and cut parallel to the surface of mandibular body. Section in 8um thickness of tissue was done and stained with Hematoxylin - Eosin. All the specimens were observed under the light microscopy. The following results were obtained : 1. In the case of control site which has no graft material, less inflammatory cell infiltration and rapid new bone forming tendency were revealed compared with experimental groups. But bone surface were observed depression pattern on defect area because of soft tissue invasion into the artificial bone defect during the experimental period. 2. In the porous hydroxyapatite only group, inflammatory cell infiltration was prominet and dense connective tissue were encapsulated around grafted materials. osteoblastic activity in the early stage after surgery was low to compared with grafted with bone cells. 3. In the case of porous hydroxyapatite inoculated with bone cell, less inflammatory cell infiltration and rapid new bone formation activity was revealed than hydroxyapatite only group. Active new bone formation were observed in the early stage of control group. 4. The origin of new bone forming was revealed not from the center of defected area but from the surface of preexisting bony wall on every specimen. 5. In this experiment, osteoclastic cell was not found around grafted materials, and fibrovascular invasion into regions with no noticeable foreign body reaction. Conclusively, the cultured bone cell inoculated onto the porous hydroxyapatite may have an important role of regeneration of artificial bone defects of alveolar bone.

  • This study was aimed to characterize osteogenic potential of rat bone marrow stromal cells (BMSC) isolated with standard flushing method and investigate the plasticity of transdifferentiation between osteoblastic and adipocytic lineage of cultured BMSC. Unlike aspiration method in human, rat bone marrow was extracted by means of irrigation with culture media that elevates the possibility of co-extraction of committed osteoprogenitor, or preosteoblast or other progenitor cells of several types present inside bone marrow. The cultured stromal cells showed high ALP activity which is representative marker of osteoblast without any treatment. Osteogenic inducers such as Dex and BMP-2 were examined for the evaluation of their effect on osteogenic and adipocytic differentiation of stromal cells, because they function as osteoinductive agent in stromal cells, but simultaneously induce adipogenic differentiation. Osteogenic differentiation was evaluated by measuring alkaline phosphatase activity or mRNA expression of osteoblast markers such as osteopontin, bone sialoprotein, collagen type I and CbfaI, and in vitro matrix mineralization by von Kossa staining. Oil red staining method was used to detect adipocyte and adipocytic marker, aP2 and $PPAR{\gamma}2$ expression was examined using RT-PCR. It can be supposed that irrigation procedure resulted in high portion of already differentiation-committed osteoprogenitor cell showing elevated ALP activity and strong mineralization only under the supplement of $100{\mu}M$ ascorbic 2-phosphate and 10mM ${\beta}$-glycerophosphate without any treatment of osteogenic inducers such as Dex and BMP-2. Dex and BMP-2 seemed to transdifferentiate osteoprogenitor cells having high ALP activity into adipocytes temporarily, but continuous treatment redifferentiated into osteoblast and developed in vitro matrix mineralization. This property must be considered either in tissue engineering for bone regeneration, or in research of characterization of osteogenic differentiation, with rat BMSC isolated by the standard irrigation method.

Physicochemical Analysis of Yogurt Produced by Leuconostoc mesenteroides H40 and Its Effects on Oxidative Stress in Neuronal Cells

  • Leuconostoc mesenteroides H40 (H40) was isolated from kimchi, and its probiotic properties and neuroprotective effect was evaluated in oxidatively stressed SH-SY5Y cells. H40 was stable in artificial gastric conditions and can be attached in HT-29 cells. In addition, H40 did not produce β-glucuronidase and showed resistant to several antibiotics. The conditioned medium (CM) was made using HT-29 cells refined with heat-killed probiotics (Probiotics-CM) and heated yogurts (Y-CM) to investigate the neuroprotective effect. Treatment with H40-CM not only increased cell viability but also significantly improved brain derived neurotropic factor (BDNF) expression and reduced the Bax/Bcl-2 ratio in oxidatively stress-induced SH-SY5Y cells. Besides, probiotic Y-CM significantly increased BDNF mRNA expression and decreased Bax/Bcl-2 ratio. The physicochemical properties of probiotic yogurt with H40 was not significantly different from the control yogurt. The viable cell counts of lactic acid bacteria in control and probiotic yogurt with H40 was 8.66 Log CFU/mL and 8.96 Log CFU/mL, respectively. Therefore, these results indicate that H40 can be used as prophylactic functional dairy food having neuroprotective effects.

Preparation and Properties of DMF-Based Polyurethanes Containing Bio-Polyol/Ester-Polyol for Wet-Type Polyurethane Artificial Leather (습식 인조피혁용 바이오 폴리올/폴리에스터 폴리올을 함유한 DMF 기반 폴리우레탄의 제조 및 물성)

  • Recently, attention has been paid to obtaining bio-polyols from renewable resources. Successful use of these natural ingredients successfully produced in the industry for the synthesis of various polyurethanes is a very important task. In this study, a series of dimethylformamide (DMF) based polyurethanes were synthesized from methylene diphenyl diisocyanate (MDI)/1, 4-butanediol and bio-polyol (polytrimethylene ether glycol based on 1, 3-propanediol : B-POL)/polyester polyol (polyadipate diol based on 1,4-butandiol : H-PET). The effect of different ratio of bio-polyol (B-POL)/polyester polyol (H-PET) on the physical properties of polyurethane was investigated. As the B-POL content in B-POL/H-PET mixture increased, the glass transition of soft segment (Tgs) and tensile strength of polyurethane decreased, however, the elongation at break and tear strength increased. On the other hand, artificial leather was produced by wet process using synthesized DMF-based polyurethanes. It was found that there was almost no difference in the effect of the B-POL/H-PET composition on the average size and density (the number of cells per unit volume) of the porous cells formed in artificial leather. These results show that there is no problem in using bio-polyol (B-POL) based polyurethane for artificial leather produced by wet process.

Research on Hyperparameter of RNN for Seismic Response Prediction of a Structure With Vibration Control System (진동 제어 장치를 포함한 구조물의 지진 응답 예측을 위한 순환신경망의 하이퍼파라미터 연구)

  • Recently, deep learning that is the most popular and effective class of machine learning algorithms is widely applied to various industrial areas. A number of research on various topics about structural engineering was performed by using artificial neural networks, such as structural design optimization, vibration control and system identification etc. When nonlinear semi-active structural control devices are applied to building structure, a lot of computational effort is required to predict dynamic structural responses of finite element method (FEM) model for development of control algorithm. To solve this problem, an artificial neural network model was developed in this study. Among various deep learning algorithms, a recurrent neural network (RNN) was used to make the time history response prediction model. An RNN can retain state from one iteration to the next by using its own output as input for the next step. An eleven-story building structure with semi-active tuned mass damper (TMD) was used as an example structure. The semi-active TMD was composed of magnetorheological damper. Five historical earthquakes and five artificial ground motions were used as ground excitations for training of an RNN model. Another artificial ground motion that was not used for training was used for verification of the developed RNN model. Parametric studies on various hyper-parameters including number of hidden layers, sequence length, number of LSTM cells, etc. After appropriate training iteration of the RNN model with proper hyper-parameters, the RNN model for prediction of seismic responses of the building structure with semi-active TMD was developed. The developed RNN model can effectively provide very accurate seismic responses compared to the FEM model.
