Vitrification solution without sucrose for cryopreservation in mouse blastocysts

  • Objective: This study was designed to investigate the survival rate of vitrified mouse blastocysts depending on the presence or absence of sucrose in vitrification solution. Methods: Mouse two-cell embryos were collected and cultured to blastocysts. Two vitrification solutions were prepared. The control solution was composed of 25% glycerol, 25% ethylene glycol, and 0.5 M sucrose (G25E250.5S) containing 2.5 mL glycerol, 2.5 mL ethylene glycol, 2 mL SSS, and 0.855 g sucrose in 5 mL PB1. The experimental solution was composed of 25% glycerol and 25% ethylene glycol (G25E25) and contained 2.5 mL glycerol and 2.5 mL ethylene glycol in 5 mL PB1. Artificial shrinkage was conducted by aspirating the blastocoelic fluid using an ICSI pipette. To examine the effect of sucrose in the vitrification solution on the survival rate of mouse blastocysts, the shrunken-equilibrated blastocysts were rehydrated or vitrified after being exposed to one of the two vitrification solutions. After exposure and the vitrification-thawing process, the re-expansion rate and hatching rate were evaluated after 6 hours of in vitro culture. Results: The re-expansion rate of mouse blastocysts exposed to vitrification solution with and without sucrose were not different in the experimental solution (without sucrose) (98%) and the control solution (with sucrose) (92%) (p>0.05). The hatching rate was higher in the experimental solution (95%) than in the control solution (88%), but did not differ across two treatments (p>0.05). The re-expansion rate of mouse blastocysts vitrified in the control solution was 92% and 94%, respectively (p>0.05), and the hatching rate was higher in the experimental solution (90%) than in the control solution (74%) (p<0.05). Conclusion: Sucrose need not be added in vitrification solution for freezing of artificially shrunken mouse blastocysts.

Evaluation on Strain Properties of 60 MPa Class High Strength Concrete according to the Coarse Aggregate Type and Elevated Temperature Condition (60MPa급 고강도 콘크리트의 굵은골재 종류와 고온상태에 따른 변형특성 평가)

  • Strain properties of concrete member which acts as an important factor in the stability of the concrete structure in the event of fire, significantly affected the characteristics of the coarse aggregate, which accounts for most of the volume. For this reason, there are many studies on concrete using artificial lightweight aggregate which has smaller thermal expansion deformation than granite coarse aggregate. But the research is mostly limited on concrete using clay-based lightweight aggregate. Therefore, in this study, the high temperature compressive strength and elastic modulus, thermal strain and total strain, high temperature creep strain of concrete was evaluated. As a result, remaining rate of high-temperature strength of concrete using lightweight aggregate is higher than concrete with general aggregate and it is determined to be advantageous in terms of structural safety and ensuring high-temperature strength from the result of the total strain by loading and strain of thermal expansion. In addition, in the case of high-temperature creep, concrete shrinkage is increased by rising loading and temperature regardless of the type of aggregate, and concrete using lightweight aggregate shows bigger shrinkage than concrete with a granite-based aggregate. From this result, it is determined to require additional consideration on a high temperature creep strain in case of maintaining high temperature like as duration of a fire although concrete using light weight aggregate is an advantage in reducing the thermal expansion strain of the fire.

Applicability of Steel Slag Aggregate for Artificial Armor Unit (제강슬래그 골재의 소파블록 적용성 평가)

  • In order to evaluate the applicability of steel slag aggregates for tetrapod concrete, the properties of concrete as structural material were investigated. The biochemical research of marine concrete using steel slag aggregates was also carried out. The tested concrete properties are slump, ai content, compressive strength, splitting tensile strength, elastic modulus, carbonation, hydration heat, freezing and thawing, sulfate attack, drying shrinkage, etc. The biochemical experiments are carried to research the propagation and reproduction of seaweeds and survival of bottom dwelling species. According to this experiment results, the steel slag aggregate content did not have a significant effect on compressive strength, splitting tensile strength and elastic modulus. The durability of concrete was not influenced by the steel slag aggregate content. From the biochemical research, steel slag aggregate can be evaluated as the material that is ideally suited for promoting propagation and reproduction of seaweeds and sessile benthos.

  • Purpose: In this study, Spreadability of denture adhesive in accordance with the saturation level of saliva, respectively, by using the resin plate and the glass was measured thickness and Spreadability. Methods: Examine the spreadability of denture adhesive in accordance with the saturation level of saliva, respectively, by using the resin plate and the glass was measured thickness and Spreadability. Also, by measuring the adhesive strength according to the amount of saliva, and the edentulous patients using denture adhesive and dry mouth patients attempt to provide clinical information of the denture adhesive. Therefore, by using the relatively low shrinkage cold curing resin, after fabricating specimen of plate form, for 7 days, it was immersed in water. Results: For the control group only denture adhesive, an artificial saliva for the experimental group were injected in $0.1m{\ell}$, $0.2m{\ell}$ and $0.3m{\ell}$ of the denture adhesive on the surface, experimental results of the investigation of the 10 times the tensile bond strength of the specimens in each group was obtained the following results. Conclusion: As the time to pressure increase in the same amount of saliva was found that the amount coming out of the denture adhesive, as the amount of saliva in the same pressure increase coming out of many denture adhesives. And the greater the pressure came out a lot of denture adhesives. Spreadability in measuring saliva contact with $0.1m{\ell}$ and $2m{\ell}$ and $3m{\ell}$ group pressured the diameter of the circular was the denture adhesive is small when compared to the group without adding the pressure of 2kg and 3kg put the saliva. The size of the circle is the same amount of saliva denture adhesive spread more pressure showed a greater increase.

  • When the complete denture is made, the record base for jaw relation is divided into temporary record base and permanent record base. However, The temporary record base include some disadvantages such as, the lackness of intimate contact between model and base, the lackness of retention during the jaw relation registration, When we obtained jaw relation the permanent record bases made from heat curing resin were utilized. We could get several advantages as follows : 1. The permanent record base provided intimate contact between the model and record base. 2. In fabricating occlusal rim on record base, the dimensional change of record base was little because the permanent record base was lesser influenced to thermal change of occlusal rim than the temporary record base. 3. At the stage of jaw relation, the retention of final denture could be early checked. 4. It could be able to get more accurate registration of jaw relation because all procedure were done on the same base during the jaw relation, artificial teeth arrangement, try-in, and final denture construction. 5. Although there was an inconvenience due to double curing procedure, the shrinkage rate in resin polimerization was relatively reduced so that more dimensional stability could be taken.

Effect of outliers on the variable selection by the regularized regression

  • Many studies exist on the influence of one or few observations on estimators in a variety of statistical models under the "large n, small p" setup; however, diagnostic issues in the regression models have been rarely studied in a high dimensional setup. In the high dimensional data, the influence of observations is more serious because the sample size n is significantly less than the number variables p. Here, we investigate the influence of observations on the least absolute shrinkage and selection operator (LASSO) estimates, suggested by Tibshirani (Journal of the Royal Statistical Society, Series B, 73, 273-282, 1996), and the influence of observations on selected variables by the LASSO in the high dimensional setup. We also derived an analytic expression for the influence of the k observation on LASSO estimates in simple linear regression. Numerical studies based on artificial data and real data are done for illustration. Numerical results showed that the influence of observations on the LASSO estimates and the selected variables by the LASSO in the high dimensional setup is more severe than that in the usual "large n, small p" setup.

  • Purpose: The aim of this study was to evaluate accuracy of three type complete mandibular denture of before and after polymerization. Methods: Mandibular edentulous model was selected as the master model. 15 study models were made by Type IV stone. Wax complete mandibular dentures were produced by the denture base and artificial teeth. Before and after curing, STL files were obtained using a blue scanner. By superimposing the digitized complete mandibular denture data(after curing) with the CAD-reference(before curing) three-dimensionally, visual fit-discrepancies were drawn by calculating the root mean square (RMS) and visualized on a color-difference map. Each calculated RMS-value was statistically analyzed by 1-way analysis of variance(ANOVA) (${\alpha}=.05$). Results: Mean(SD) RMS-values was OM group $88.98(6.10){\mu}m$, BM group $82.35(13.46){\mu}m$, BDM group $77.83(9.46){\mu}m$. The results of the 1-way ANOVA showed no statistically significant differences in the RMS values of the Three groups for the material (P > .241). Conclusion : Deformation of artificial teeth position was observed in all groups after resin polymerization. But the values, all group were within the clinically acceptable range. The values of BDM group showed the least deformation than the other two groups.

Strength and Durability Characteristics of Low-alkali Mortar for Artificial Reefs Produced by 3D Printers (인공어초 3D 프린터 출력을 위한 저알칼리 모르타르의 강도와 내구성능)

  • Concrete prevents corrosion of reinforcing bars due to its strong alkalinity. However, in the sea, strong alkali components with a pH of 12 to 13 are eluted, which adversely affects the ecological environment and growth of marine organisms. In this study, the mechanical properties and durability of the low alkali mortar were evaluated for the development of a low alkali mortar for the 3D printed artificial reefs. As a result of evaluation of strength characteristics, the α-35 mixture, which were produced with fly ash, silica fume and α-hemihydrate gypsum, satisfied the strength requirement 27 MPa in terms of compressive strength. As a result of pH measurement, it was found that mixing with alpha-type hemihydrate gypsum resulted in minimizing pH due to the the formation of calcium sulfate instead of calcium hydroxide production. As a result of the chloride ion penetration resistance test, the α-35 mixture exhibited the best performance, 3844C. As a result of measuring the length change over time, the α-35 mixture showed the shrinkage 33.5% less compared to the Plain mix.

Preparation of shotcrete coarse aggregate with low grade clay and coal ash (저급 점토와 석탄회를 이용한 숏크리트용 골재의 제조)

  • In this study, the artificial coarse aggregate was manufactured by using coal ash and low grade clay. The characteristics of a coal ash-clay system were investigated using XRD, XRF, TG-DTA, SEM and Dilatometer with various coal ash contents. The chemical compositions are the fly ash, bottom ash and clay, $Al_2O_3$ are 28.5 wt%, 32.4 wt% and 18.1 wt%, and $SiO_2$ are 33.0 wt%, 53.7 wt% and 68.4 wt% in weight ratio, respectively. The shrinkage of specimens started at around $850^{\circ}C$ and changed little up to $1100^{\circ}C$, but increased markedly at above $1100^{\circ}C$. The shrinkage rate is strongly related to the decarbonization amount of coal ash. At the sintering temperature $1150^{\circ}C$, it was found that quartz, mullite, anorthite and albite phase exist in all specimens. It was found that bottom-clay system specimen sintered at $1150^{\circ}C$ had a good compressive strength of 87.5 kg/$cm^2$, and the compressive strength of bottom-clay specimen was higher than that of fly-clay system specimen. The reusability of coal ash as a raw material in the process of shotcrete resources such as artifical coarse aggregate is highly expected.

  • In patients with fully edentulous jaw, treatment of complete dentures should be carried out in many stages when following the conventional methods. Therefore there were disadvantages such as multiple visits to dental clinic is inevitable. In addition, errors caused by polymerization shrinkage, which happens during the fabrication of denture, and difficulties in reproduction of damaged or lost denture were considered as disadvantages. But nowadays, computer-aided design and computer-aided manufacturing (CAD/CAM) system is widely used in dentistry and it has begun to expand its spectrum in manufacturing complete dentures. Using CAD/CAM system to fabricate complete dentures can reduce the number of patient's visit and clinical chair time, since taking impression, recording jaw relation, and selection of artificial teeth are performed at the same time during the first visit, and delivering of dentures during the second visit is possible. In addition, because 3D-Printing technology is used, errors by polymerization shrinkage can be reduced. Among the companies that fabricate complete dentures using CAD/CAM system, DENTCA CAD/CAM denture (DENTCA Inc., Los Angeles, CA, USA) is the most commercialized company. In this case, we treated patients of complete dentures using conventional complete denture method and DENTCA CAD/CAM denture system in the same patient. We would like to report this case because we have achieved good results not only in functional aspects of pronunciation, chewing, and swallowing but also in aesthetic aspects.
