The Inhibitory Effect of Grapefruit Seed Extracts on the Physiological Function of Enterobacter pyrinus (Grapefruit 종자추출물이 Enterobacter pyrinus의 생리기능에 미치는 영향)

  • Grapefruit seed extracts(GFSE) have some unknown compounds which exhibit the antibiotic activities aganist microorganisms including bacteria and fungi. We have examined the effects of GFSE on the growth of Enterobacter pyrinus which was isolated from necrotic lesions of pear trees. During the cultivation, the growth of the bacteria was strongly inhibited at the low concentration(0.01%, w/w) of GFSE. Hydrophobic fraction extracted from GFSE by mixed solvents (chloroform : methanol : water, 1 : 2 : 0.8, v/v/v) had components which inhibited the growth of bacteria. There was, however, no inhibitory effect of GFSE on the activities of several enzymes including hexokinase, glucose 6-phosphate dehydrogenase, malate dehydrogenase and succinate dehydrogenase. $O-nitrophenyl-{\beta}-D-galactopyranoside(ONPG)$, the artificial substrate of ${\beta}-galactosidase$ was hydrolyzed in the presence of GFSE, indicating that the membrane was pertubated by the GFSE. From the results it was suggested that the antibiotic activity of GFSE is due to the change of membrane permeability of cell. GFSE was fractionated by high performance liquid chromatography equipped with $C_{18}$ reverse phase column. Among active fractions, three peaks were identified as 1-chloro-2-methyl-benzene (o-toluene), N,N-dimethyl-benzenemethaneamine, 1-[2-(2-ethylethoxy)ethoxy]-4- (1,1,3,3-tetramethyl)-bezene, respectively, while the other three remained unidentified.

Distributional Patterns and the Evaluation of Hydrophytic Plants of Urban Wetlands in Seongnam City, Gyunggi-do Province, Korea (경기도 성남시 도시지역 습지의 유형 분포 및 습지식물의 특성 평가)

  • This study was carried out to obtain ecological information necessary for a conservation plan based on the distributional patterns, wetland types, and hydrophytic characteristics of urban wetlands in Seongnam City, Kyunggi Province where representing the various patterns of land use made by rapid urbanization since 1970s. Total 162 sites of four wetland types were identified as urban wetlands during the first survey. The sites were classified into 55 forested swamps, 4 riverine wetlands, 62 abandoned paddy fields, 37 small ponds, and 4 reservoirs, etc. The second survey targeted 107 sites which were identified as good wetlands. It showed that 42 sites(about 39%) were already degraded due to drainage, landfill, and crop cultivation at 6 months intervals. Both hydrologic conditions and hydrophytic characteristics of 27 good wetlands help maintain current ecological status, but most wetlands have been degraded by artificial impacts. Among 184 species identified, only 75 species(about 40.7%) were hydrophytes. Prevalence Index of hydrophyte based on three categories of OBL(obligatory wetland plant), FAC(facultitative plant), UPL(obligatory upland plant) was 3.7, indicating that vegetation data alone is inadequate to designate as wetlands. This study revealed that as critical habitats for wildlife they playa vital role in ecotone between both terrestrial and aquatic ecosystem with its proper distributional pattern in spite of their small areas compared to the entire geographic region of the City.

Phylogenetic relationships of Lyophyllum decastes on the based of ITS region sequences and RAPD (잿빛만가닥버섯(Lyophyllum decastes)의 ITS 영역염기서열 및 RAPD에 의한 계통학적 유연관계 분석)

  • Phylogenetic relationships of Hypsizygus mamoreus and Lyophyllum decastes artificial cultivated using ITS sequences and RAPD polymorphism have been analyzed. Based on ITS region sequences of 14 strains, we could divide into 2 group as group1 to Hypsizygus mamoreus and the control isolated group2 to Lyophyllum decastes were identified as the same species. Restrict analysis of rDNA ITS region which was amplified by PCR, 14 collected strains could be classified into 4 clusters. There was approximately 58% genetic similarity between cluster I(SPA 100, 101, 102) and cluster II(SPA 200, 208 and SPA 201, 202), 41% between cluster III(SPA 104, 103, 203) and cluster IV(SPA 204, 206, 207, 205) by BLAST analysis. RAPD polymorphisms were used to analyze the species diversity of artificially cultivated Lyophyllum decastes SPA 202. As a result, similarity between SPA 202 and SPA 203 was 75%, at the same time, similarity between SPA 202 and Pleurotus eryngii SPA 103 and SPA 104 was 65%.

Mycelial characteristics for the artificial cultivation of Inonotus obliqua (Pers.) Pilot (차가버섯(Inonotus obliqua) 인공재배를 위한 균사 배양적 특성)

  • The results of examining characteristics of mycelial growth and culture condition for determining the condition of artificially culturing Inonotus obliqua (chaga) are as follows. 1) Mycelial growth and density oft, oblique were the highest in the medium of BDA (Birch Dextrose Agar; 66.3mm/10d) followed by the order of GDA, PDA, CDA, PODA, ODA, YM, MCM, MEA (pH 7.0), CHA, and MEA (pH 4.7). 2) Optimal temperature for the mycelial growth and density of 1. obliqua were shown to be 30$^{\circ}C$, but the mycelia were dead at 40$^{\circ}C$. the mycelial growth and density of KNAC3005 strains was the highest at 30$^{\circ}C$(66.3mm/10d) followed by the order of 25, 20, 15, 35, 10, and 5$^{\circ}C$. 3) Optimal pH for the mycelial growth and density were revealed to be 6.0 (88.4mm/10d). Above or below pH 6.0, the mycelial growth and density were shown to be retarded. 4) Optimal carbon, nitrogen and organic acid sources for the spawn growth of 40 were maltose (331mg/25$m\ell$/15d), peptone (347mg/25$m\ell$/15d), and glutamic acid (357mg/25$m\ell$/15d), respectively. Optimal level of biotin was 370mg/15d and optimal C/N ratio was 40.

Mycelial Growth and Fairy-Ring Formation of Tricholoma matsutake from Matsutake-Infected Pine Trees (송이 감염묘로부터 송이균의 생장과 균환 형성)

  • Transplantation of matsutake-infected pine tree into a pine forest has great potential as an artificial cultivation method of Tricholoma matsutake. Matsutake-infected pine trees had previously transplanted by this research group into the experimental area from 2001 to 2004 and its survival rate determined in 2006 was 20%. For the survived samples, the characteristics of mycelial growth and the development of fairy-ring formation of T. matsutake have been evaluated until 2009. It was found that size of the indeterminate ring showed significant differences among the individual trees and varied from $4\;cm\;{\times}\;4\;cm$ to $52\;cm\;{\times}\;35\;cm$. The variation was possibly resulted from the differences in production area of the matsutake-infected pine tree and those in site characteristics of the transplanted spots. For the characteristics of mycelial growth, it grew in the shape of a cudgel or an acute-angled 'V' in early stage, and then the shape became more wider and changed into an obtuse-angled 'V' as time passed. We expect that matsutake mushrooms may occur from the fairy-rings of some of these matsutake-infected pine trees in the autumn of 2010.

  • To give basic information on the agricultural mechanization and multiple cropping adoptability of the paddified clayey terrace soils which have poor permeability and poor adoptability to mechanization, a scheme of drainage classification as well as taxonomic classification was intended. 1. The degrees of gleization of terrace paddy soils were well distinguished by the comparison of "Redness rating" of their profiles. 2. When the criteria of "Imperfectly drained" soils were defined as follows; Soils have more than 50cm of accumulated depth which has less than 0.5 in Redness rating within 1.2m of the profile, the Geugrag series could be classified to "Imperfectly drained." The tentative classification of drainage class of Geugrag soils seemed to well matching with land suitability groups, and give possibility of drainage recommendation in the case of dry land crop cultivation. 3. The Geugrag soil which was well paddified by artificial surface irrigation, could be proposed to classify "Anthroaquic Ochraqualfs."

Use of Pellet or Cube-type Phenolic Foam as an Artificial Medium for Production of Tomato Plug Seedlings (토마토 플러그 묘 생산을 위한 펠릿 및 큐브형 phenolic foam 인공배지의 이용)

  • Growers in plug seedling production think that root media in which rockwool is a component has given rise to several environmental problems. Therefore, the demand for new materials as a substitute for rockwool has been increased. This study examined the possibility of cultivation of tomato plug seedlings using a newly developed growing medium with phenolic foam. Plug seeds of tomato cultivar 'Madison' were sown in four pellet-type growing media: Grodan rockwool (GRW), UR rockwool (URW), phenolic foam LC (LC) or phenolic foam LC-lite (LC-lite). Then, the seedlings were transplanted to the four cube-type growing media 19 days after sowing. Seeds were germinated in a growth chamber ($25{\pm}2^{\circ}C$, 80% relative humidity, and dark) for 4 days and then the seedlings were grown with a nutrient solution supplied by an overhead irrigation system in a greenhouse. Plant height, number of leaves, leaf area, and fresh or dry weight of tomato seedlings were the greatest for the seedlings transplanted to URW cube media after being grown on LC-lite pellets. Root grade was the greatest for the seedlings transplanted on LC or LC-lite cube media after being grown on LC pellets. Chemical properties of all media tested for tomato growth were maintained within a stable range, while physical properties of URW showed high values in container capacity, air space, and total porosity. These results demonstrated that the phenolic foam media were effective for seedling growth and can substitute for rockwool as a root medium.

The Morphological Characteristics of Entomopathogenic Fungus, Isaria sinclairii (Berk.) Lloyd (곤충기생균 Isaria sinclairii(Berk.) Lloyd의 형태학적 특성)

  • NIAST has developed the artificial cultivation technique of entomopathogenic fungus, Isaria sinclairii by using the silkworm. NIAST is planning to provide the silkworm farmers the guidance of the technique. Screening of pharmacologically active principles of I. sinclairii is needed to register the fungal breeding, attain the approval of food materials, and ultimately position the fungus as a high value-added product. The result of investigating the characteristics of Isaria sinclairii was showed that conidia were irregularly long oval-shaped and measured 6.33$\times$3.33 $\mu\textrm{m}$ in average The optimal condition of cultivating hyphae was pH 5.0 in acidity and 21- $25^{\circ}C$ in temperature. And the best condition of producing conidia was pH 6.0 in acidity and 16-$20^{\circ}C$ in temperature.

Studies on Antioxidant, Anti-inflammation and Moisturizing Activities of Gastrodia elata Flower Extract (천마꽃 추출물의 항산화, 항염, 보습 활성에 대한 연구)

  • Gastrodia elata has a very low pollination rate in natural state, and even in artificial cultivation, there are very few individuals that bloom due to the degeneration, so little studies have been conducted. This study confirmed that the potential as a cosmetic ingredient by evaluating the antioxidant activity through the evaluation of DPPH radical scavenging activity, anti-inflammatory activity through the inhibitory effect on nitric oxide production, and the moisturizing activity through the effect on promoting hyaluronic acid production by artificially flowering G. elata flower. It was also confirmed that the appearance rate and flowering rate of G. elata harvested in spring were high, and the content of gastrodin was 0.36%. The IC50 value of G. elata flower extract was 0.045% and it was confirmed that G. elata flower extract had higher radical scavenging activity than G. elata root extract. The NO production inhibitory activity against the flower extract showed a significant inhibitory effect from 1% to 83.2%. Hyaluronic acid production promotion efficacy was not confirmed in the G. elata root extract, but the production rate increased with concentration dependence in the flower extract, and it was the highest at 46.9% when 0.02% treatment was performed. Based on the above research results, it is judged that G. elata flower extract has high potential for use as an antioxidant, anti-inflammatory, and skin moisturizing cosmetic ingredient.

News Article Analysis of the 4th Industrial Revolution and Advertising before and after COVID-19: Focusing on LDA and Word2vec (코로나 이전과 이후의 4차 산업혁명과 광고의 뉴스기사 분석 : LDA와 Word2vec을 중심으로)

  • The 4th industrial revolution refers to the next-generation industrial revolution led by information and communication technologies such as artificial intelligence (AI), Internet of Things (IoT), robot technology, drones, autonomous driving and virtual reality (VR) and it also has made a significant impact on the development of the advertising industry. However, the world is rapidly changing to a non-contact, non-face-to-face living environment to prevent the spread of COVID 19. Accordingly, the role of the 4th industrial revolution and advertising is changing. Therefore, in this study, text analysis was performed using Big Kinds to examine the 4th industrial revolution and changes in advertising before and after COVID 19. Comparisons were made between 2019 before COVID 19 and 2020 after COVID 19. Main topics and documents were classified through LDA topic model analysis and Word2vec, a deep learning technique. As the result of the study showed that before COVID 19, policies, contents, AI, etc. appeared, but after COVID 19, the field gradually expanded to finance, advertising, and delivery services utilizing data. Further, education appeared as an important issue. In addition, if the use of advertising related to the 4th industrial revolution technology was mainstream before COVID 19, keywords such as participation, cooperation, and daily necessities, were more actively used for education on advanced technology, while talent cultivation appeared prominently. Thus, these research results are meaningful in suggesting a multifaceted strategy that can be applied theoretically and practically, while suggesting the future direction of advertising in the 4th industrial revolution after COVID 19.
