In this study, in order to provide an information for artificial cultivation of wild mushroom, the meteorological effects on wild mushroom appearance were examined using daily meteorological observations in Chiak National Park. The survey of wild mushroom appearance was carried out once a month from June to October. Under high temperature and humidity conditions in July and August, the appearance of wild mushroom was frequent. In contrast, lower number of wild mushroom appeared in October. Wild mushroom appearance was affected by solar radiation, relative humidity, precipitation, and soil water content whereas the impact of air and soil temperature was lower than that of other meteorological elements.
Cordyceps militaris, a member of Ascomycota, a mushroom referred to as caterpillar Dong-chung-ha-cho, is commercially valuable because of its high content of bioactive substances, including cordycepin, and its potential for artificial cultivation. Cordycepin (3'-deoxyadenosine) is highly associated with the pharmacological effects of C. militaris. C. militaris is heterothallic in that two mating-type loci, idiomorph MAT1-1 and MAT1-2, exist discretely in two different spores. In this study, nine C. militaris strains were mated with each other to prepare newly bred strains that produced a larger amount of cordycepin than the parent strains. Nine strains of C. militaris were identified by comparing the internal transcribed spacer sequence, and a total of 12 single spores were isolated from the nine strains of C. militaris. After the MAT idiomorph was confirmed by PCR, 36 mating combinations were performed with six single spores with MAT1-1 and the others with MAT1-2. Eight mating combinations were successfully mated, producing stroma with perithecia. Cordycepin content analysis of all strains by high-performance liquid chromatography revealed that the KASP4-bred strain produced the maximum cordycepin among all strains, regardless of the medium and stroma parts. Finally, universal rice primer-PCR was performed to demonstrate that the bred strains were genetically different from the parental strains and new C. militaris strains. These results may be related to the recombination of genes during mating. The newly produced strains can be used to meet the industrial demand for cordycepin. In addition, breeding through mating suggests the possibility of producing numerous cordycepin-producing C. militaris strains.

Present expriments were conducted to determine the possibility of cultivation of Coriolus versicolor selected among the higher fungi growing in Korea. Dried apple sawdust used in this study consisted of 31.5% C, 0.54% N, 2.41 % CaO, 0.05% $P_2O_5$, 0.48% MgO and 0.81% $K_2O$(pH 5.6). Coriolus versicolor mushrooms were cultivated on the sawdust media; apple sawdust; rice bran=80:20 in 850cc polypropylene bottles. The isolate of Coriolus versicolor used was YCV collected from an Apple farm in Youngchun district. It took 34 days to make fruitbody from spawning. The fruit bodies produced the total fresh weight 28 g in a bottles, and converted to 64% of fresh weight.
Cylindrocarpon destructans is a soil-borne plant pathogenic fungus causing root rot on ginseng and trees. Rapid and exact detection of this pathogen was practiced on ginseng seedlings by nested PCR using speciesspecific primer set. The second round of PCR amplification by Dest 1 and Dest 4 primer set formed 400 bp of species-specific fragment of C. destructans from the product of first round of amplification by ITS 1 and ITS 4 primer set. In the PCR sensitivity test based on DNA density, nested PCR detected to the limit of one fg and it meant the nested PCR could detect up to a few spores of C. destructans. Also, nested PCR made it possible to detect the pathogen from ginseng seedlings infected by replantation on artificial infested soil. Our nested PCR results using species-specific primer set could be utilized for diagnosis of root rot disease in ginseng cultivation.

Background: Panax ginseng C. A. Meyer is wood-cultivated ginseng (WCG) in Korea which depends on an artificial forest growth method. To produce this type of ginseng, various P. ginseng cultivars can be used. To obtain a WCG similar to wild ginseng (WG), this method is usually performed in a mountain using seeds or seedlings of cultivated ginseng (CG) and WG. Recently, the WCG industry is suffering a problem in that Panax notoginseng (Burk.) F. H. Chen or Panax quinquefolium L. are being sold as WCG Korean market; These morphological similarities have created confusion among customers. Methods and Results: WCG samples were collected from five areas in Korea. After polymerase chain reaction (PCR) amplification using the primer pair labeled with fluorescence dye (FAM, NED, PET, or VIC), fragment analysis were performed. PCR products were separated by capillary electrophoresis with an ABI 3730 DNA analyzer. From the results, WCG cultivated in Korea showed very diverse genetic background. Conclusions: In this study, we tried to develop a method to discriminate between WCG, P. notoginseng or P. quinquefolium using simple sequence repeat (SSR) markers. Furthermore, we analyzed the genetic diversity of WCG collected from five cultivation areas in Korea.

Humic acid (HA) is a complex organic matter found in the environments, especially in grassland soils with a high density. The bioactivity of HA to promote plant growth depends largely on its extraction sources. The quality-control of HA and the quality improvements via an artificial synthesis are thus challenging. We recently reported that a polymeric product from fungal laccase-mediated oxidation of catechol and vanillic acid (CAVA) displays a HA-like activity to enhance seed germination and salt stress tolerance in a model plant, Arabidopsis. Here, we examined whether HA or CAVA enhances the growth of Italian ryegrass seedling. Height and fresh weight of the plant with foliar application of HA or CAVA were bigger than those with only water. Interestingly, enhanced root developments were also observed in spite of the foliar treatments of HA or CAVA. Finally, we proved that HA or CAVA promotes the regrowth of Italian ryegrass after cutting. Collectively, CAVA acts as a HA mimic in Italian ryegrass cultivation, and both as a biostimulant enhanced the early growth and regrowth after cutting of Italian ryegrass, which could improve the productivity of forage crops.
This study was carried out to investigate soil and environment characteristics of Schizandra Chinensis Baillon habitat as a part of establishment of conservation plan for important medicinal crop resources in Jangsu-gun, Jeollabuk-do. These habitats are mostly located on the slopes of mountains facing northwest at an altitude of 500 m to 700 m with angles of inclination ranging from 17.6 to 36.4%. Average yearly precipitation and temperature of Schizandra Chinensis Baillon habitat were 1,493.4 mm and $16.9^{\circ}C$, respectively. The number of plant species growing in Schizandra Chinensis Baillon habitat was found 55 species. Salix gracilistyla, Lespedeza bicolor, and Artemisia princeps var. were shown higher density and coverage. Chemical quality of Schizandra Chinensis Baillon habitats soils was relatively higher than artificial cultivation soil.
Korean ginseng (Panax ginseng) has long been cultivated as an important economic medicinal plant. Owing to the seasonal and long-term agricultural cultivation methods of Korean ginseng, they are always vulnerable to various environmental stress conditions. ABSCISIC ACID (ABA) is an essential plant hormone associated with seed development and diverse abiotic stress responses including drought, cold and salinity stress. By modulating ABA responses, plants can regulate their immune responses and growth patterns to increase their ability to tolerate stress. With recent advances in genome sequencing technology, we first reported the functional features of genes related to canonical ABA signaling pathway in P. ginseng genome. Based on the protein sequences and functional genomic analysis of Arabidopsis thaliana, the ABA related genes were successfully identified. Our functional genomic characterizations clearly showed that the ABA signaling related genes consisting the ABA receptor proteins (PgPYLs), kinase family (PgSnRKs) and transcription factors (PgABFs, PgABI3s and PgABI5s) were evolutionary conserved in the P. ginseng genome. We confirmed that overexpressing ABA related genes of P. ginseng completely restored the ABA responses and stress tolerance in ABA defective Arabidopsis mutants. Finally, tissue and age specific spatio-temporal expression patterns of the identified ABA-related genes in P. ginseng tissues were also classified using various available RNA sequencing data. This study provides ABA signal transduction related genes and their functional genomic information related to the growth and development of Korean ginseng. Additionally, the results of this study could be useful in the breeding or artificial selection of ginseng which is resistant to various stresses.

A field survey was carried out targeting 59 pine wilt disease control areas distributed in Andong, and the ecological characteristics of the areas affected by pine wilt disease were analyzed using vegetation information, ecological information, and land use information. Vegetation characteristics of Pine wilt disease affected forest area showed a 3-layer vegetation structure, high percentage of accidental occurrence species (37%) and secondary vegetation species (59.6%), appearing 12 taxa naturalized plants and 3 taxa ecosystem disturbance organisms designated by the Ministry of Environment. Ecological information of Pine wilt affected area showed frequent occurrence of water stress in south and west slopes, low lying grounds in mountains, and in well-drained soils. Also, surrounding the area has been used as roads, tombs, and cultivation where intensive human activities were the cause of disturbance and stress to the pine forest. It was analyzed that the pine forest in Andong city suffered extensive damage due to the onset of pine wilt disease while the pine trees were weakened due to continuous human activities. Conclusively, the spread and onset of pine wilt disease are worsened by artificial factors than natural environmental conditions.
Background: Panax ginseng, as one of the most widely used herbal medicines worldwide, has been studied comprehensively in terms of the chemical components and pharmacology. The proteins from ginseng are also of great importance for both nutrition value and the mechanism of secondary metabolites. However, the proteomic studies are less reported in the absence of the genome information. With the completion of ginseng genome sequencing, the proteome profiling has become available for the functional study of ginseng protein components. Methods: We optimized the protein extraction process systematically by using SDS-PAGE and one-dimensional liquid chromatography mass spectrometry. The extracted proteins were then analyzed by two-dimensional chromatography separation and cutting-edge mass spectrometry technique. Results: A total of 2,732 and 3,608 proteins were identified from ginseng root and cauline leaf, respectively, which was the largest data set reported so far. Only around 50% protein overlapped between the cauline leaf and root tissue parts because of the function assignment for plant growing. Further gene ontology and KEGG pathway revealed the distinguish difference between ginseng root and leaf, which accounts for the photosynthesis and metabolic process. With in-deep analysis of functional proteins related to ginsenoside synthesis, we interestingly found the cytochrome P450 and UDP-glycosyltransferase expression extensively in cauline leaf but not in the root, indicating that the post glucoside synthesis of ginsenosides might be carried out when growing and then transported to the root at withering. Conclusion: The systematically proteome analysis of Panax ginseng will provide us comprehensive understanding of ginsenoside synthesis and guidance for artificial cultivation.
