The production of media and optimal additive rate using the cultivation media wastes of Pleurotus eryngii (큰느타리버섯 폐배지 이용 배지 제조 및 적정 첨가비율)

  • After bottle culture of Pleurotus eryngii, media were taken out the bottle and normally utilized compost. However, nutritional elements were remained in the waste media. This study was carried out to investigate the reusable possibility and the optimal additive rate of waste media in an artificial cultivation of Pleurotus eryngii. The pH had tendency to decrease as the waste media was added from 6.0 to 4.8. Based on the additive rate of 10, 20, 30, 40, and 50%, each treatment waste media was added to new media for culturing Pleurotus eryngii. Among various treatments, the mycelial growth and primordia formation of Pleurotus eryngii were more favorable in the addition of 10-30% waste median than in the addition of 40 and 50%. The yield of its fruiting body was increased slightly in the treatment of 10-30% waste media.

Inhibition of Floral Induction and Variation of Yield in Angelica gigas Nagai (참당귀의 화성억제와 수량변화)

  • Since the inhibition of floral induction was considered to be an important subject to get high yield of Angelica gigas, the effects of low temperature, daylengh and shading were investigated in this study. The yield of A. gigas root grown by inhibition of floral induction was compared with the yield obtained by traditional cultivation method. When the seedings were exposed to the natural low air temperature condition until late of November, the plants which had 6 to 8 leaves showed vernalizing effect, and when the low temperature treatment was prolonged until mid of January, 4 to 5 leaf plants showed vernalizing effect. But 2 to 3 leaf plants did not show vernalizing effect regardless of the length of chilling treatment. The effectiveness of exposure to low temperature was markedly dependent upon the age of plants. In the artificial low temperature treatment the range of temperature varied with plant age. Vernalizing effect at 1,5 and 9$^{\circ}C$ of temperature did not show 3,5,6 leaf plants, respectively. The bolting percent in 8 leaf plant at 1,5 and 9$^{\circ}C$ for 8 weeks decreased by 80,45 and 5%, respectively. As daylength became longer, the bolting percent of A. gigas was increased. plants which were grown in the full sunlight showed the highest value in bolting percent, and the bolting percent was not decreased significantly with changing from full sunlight to 50% shading. When A. gigas were grown by the inhibition of floral induction, dry root weight per l0a showed the greatest value in 3 year old plant which 3.6 and 2.4 times higher than that of 1 and 2 year old plant, respectively. The yield of root was increased by 73.7% in 2 year old plant and 159.6% in 3 year old plant compared with the yield obtained by traditional cultivation method.

Effect of Peeling Blades and Abrasives on Mechanical Peeling of Cynanchum wilfordii Hemsley (박피(剝皮)날 및 연마재(硏磨材)가 백하수오(白何首烏) 기계박피(機械剝皮)에 미치는 영향(影響))

  • A rapid increasing in cultivation area due to favorable price temporarily and decreasing in consumer's demands caused by economic depressions recently, lead the price of Cynanchum wilfordii Hemsley to decline sharply. Thus, it may give rises to weakening of cultivation bases in Cynanchum wilfordii Hemsley. To investigate optimal mechanical peeling conditions of Cynanchum wilfordii Hemsley, protruded rubber, plastic pad, diamond shape's steel and palstic brush were introduced as blades and artificial stone, sand and small pebble were done as abrasives. The main results obtained were summarized as follows; 1. It took 2 minutes per 1kg in mechanical peeling of Cynanchum wilfordii Hemsley's raw root whereas 36 minutes in manual peeling and values of lightness showed more higher in manual peeling than in mechanical peeling. 2. Yield in combination of diamond shape's steel blade and sand abrasive showed the lowest at 89.9% among treatments and peeling rates in combination of diamond shape's steel blade and small pebble showed the highest at 71.3% in mechanical peeling. 3. Lightness, one of the most important factors in determining quality of Cynanchum wilfordii Hemsley, showed the highest in combination of diamond shape's steel blade and small pebble brasive at 61.90 in mechanical peeling. 4. As the speed of rotation gets faster, yields tend to lower in mechanical peeling. Peeling rates and lightness showed the highest at 66.8%, 57.96 respectively among treatments at 30 r.p.m. in mechanical peeling.

Isolation of a Phenol-degrading Bacterial Strain and Biological Treatment of Wastewater Containing Phenols (Phenol 분해균주의 분리 및 페놀함유 폐수의 생물학적 처리)

  • Aromatic hydrocarbons, such as phenol, have been detected frequently in wastewater, soil, and groundwater because of the extensive use of oil products. Bacterial strains (56 isolates) that degraded phenol were isolated from soil and industrial wastewater contaminated with hydrocarbons. GN13, which showed the best cell growth and phenol degradation, was selected for further analysis. The GN13 isolate was identified as Neisseria sp. based on the results of morphological, physiological, and biochemical taxonomic analyses and designated as Neisseria sp. GN13. The optimum temperature and pH for phenol removal of Neisseria sp. GN13 was $32^{\circ}C$ and 7.0, respectively. The highest cell growth occurred after cultivation for 30 hours in a jar fermentor using optimized medium containing 1,000 mg/l of phenol as the sole carbon source. Phenol was not detected after 27 hours of cultivation. Based on the analysis of catechol dioxygenase, it seemed that catechol was degraded through the meta- and ortho-cleavage pathway. Analysis of the biodegradation of phenol by Neisseria sp. GN13 in artificial wastewater containing phenol showed that the removal rate of phenol was 97% during incubation of 30 hours. The removal rate of total organic carbon (TOC) by Neisseria sp. GN13 and activated sludge was 83% and 78%, respectively. The COD removal rate by Neisseria sp. GN13 from petrochemical wastewater was about 1.3 times higher than that of a control containing only activated sludge.

Studies on the Morphological and Physiological Characteristics of Pleurotus cystidiosus O.K.Miller, the Abalone Mushroom (전복느타리버섯(Pleurotus cystidiosus O.K.Miller)의 형태적 및 생리적 특성에 관한 연구)

  • The pileus of Pleurotus cystidiosus was attached at the side and was spatular-shape. It grew to the size of $7.6{\sim}9.8\;cm$ with dark brown color. The length of stem was $1.4{\sim}2.9\;cm$. The individual weight of fruiting-body was 25.4 g in average. This study was conducted to Investigate the physiological characteristics and to develop the artificial cultivation technique of P. cystidiosus. To find out the optimum condition of mycelial growth of P. cystidiosus, physiological conditions such as media, temperature, pH carbon sources and nitrogen sources were investigated. Various cultural media were used for the cultivation of P. cystidiosus and compost dextrose agar was favorable for mycelial growth. The optimal growing temperature and pH were in range of 25 to $30^{\circ}C$, pH $7{\sim}9$, respectively. When different carbon sources were compared, glucose was found to be best. However nitrogen sources were different with each strains. When different C/N ratio were compared, 20:1 of C/N ratio was found to be best.

  • In the traditional medicine, the basidiocarps of Poria cocos (Fr.) Wolf naturally collec­ted have been widely used as a traditional remedy for dropsy, diarrhea, gonorrhea and nervous disease more than 3500 years ago. It was recently found to be composed of the particular compounds having antitumor effects. Therefore, its demand has been gradually increased, whereas it is still dependent upon the natural harvest. The artificial cultivation of P. cocos was carried out to observe the morphological characteristics and to investigate the cultural characteristics of P. cocos isolates collected. The morphological characteristics of its basidiocarps was observed. The physiological aspects of its isolates were also investigated.

  • Mushroom is a high in protein, low calorie food and has dietary fiber, vitamins, iron and minerals such as zinc. It is called that mushroom is one of the biggest concerns for healthy foods. When we make the artificial cultivation of mushroom, one of the greatest influence element is temperature. In this regard, farmers passively measure temperatures in the greenhouse as inaccurate way such as by the naked eyes. In this paper, we constructed a display system in order to improve the efficiency of manual management of temperature based on the influence of temperature on the mushroom. In related to the methods of mushroom cultivation, the recent technology apply the new technology such as sensors and IT convergence services. And then cultivating mushroom is managed effectively. In this paper, we implement an automatic display system for sensing data. By using this function, farmers could effectively manage environment needed to be grown mushroom, and anticipate the improvement of sales by increasing quality of mushrooms as well.

Effects of Temperature on Survival, Development, and Reproduction of the Non-diapause Asian Corn Borer, Ostrinia furnacalis (Lepidoptera: Crambidae) (조명나방(나비목: 포충나방과) 비휴면태의 생존과 발육, 생식에 미치는 온도의 영향)

  • We analyzed the effects of temperature on the survival, development, and reproduction of the non-diapause Asian corn borer, Ostrinia furnacalis (Lepidoptera: Crambidae). A 16:8 h light:dark photoperiod was supplied to the non-diapause stages of O. furnacalis. The insects were raised on an artificial diet at seven constant temperatures between 15 and 35℃ for immature stages, and eight temperatures between 13 and 33℃ for the adult stage. The survival rate of eggs exceeded 70% at all temperatures, whereas survival of larvae was as low as 7.4% at 15℃. The developmental periods of the immature stages decreased with increasing temperature, but the larval period did not decrease further at 35℃. The pupal weight increased with increasing temperature, but the female weight decreased at 35℃. Variations with different last larval instars within the colonies at different temperatures were observed. Adults produced offspring at all tested temperatures. Adult longevity and the pre-oviposition and oviposition periods decreased with increasing temperature, but the pre-oviposition period increased at 33℃. Total fecundity exceeded 400 viable eggs at 22℃ and 31℃. The mean daily fecundity during the oviposition period and the mean daily fecundity on the day of oviposition increased with increasing temperature, but decreased at 33℃. Daily fecundity sharply increased at earlier adult ages and slowly decreased thereafter. The simulated oviposition frequency was greatest at 22℃. The lower developmental threshold temperatures were estimated to be the lowest at 9.7℃ for the first instar larva and the highest at 14.7℃ for the 5th to last instar larval stage, using a linear model.

Comparion of Rockwool, Reused Rockwool and Coir Medium on Tomato (Solanum lycopersicum) Growth, Fruit Quality and Productivity in Greenhouse Soilless Culture (시설 내 수경재배에서 암면, 재사용암면, 코이어 배지에 따른 토마토의 생육 및 생산성 비교)

  • This experiment was conducted to find out the possibility of use of reused rockwool and comparison of growth, productivity and quality of tomatoes according to the use of rockwool and coir medium. The experiment was conducted in an automatic controlled greenhouse at Andong National University, College of Life Science, located in Andong, Gyeongsangbuk-do.. As a result of the experiment, there was no difference in the number of leaves, plant height, and leaf area between treatments, and the crown diameter was slightly higher in rockwool medium, also there was no difference between reused rockwool and coir medium. Fruit productivity showed different responses depending on the cultivation environment, but there was no significant difference between rockwool, reused rockwool and coir medium. In addition, the quality of fruit was observed to be different according to the concentration of EC in the medium. Therefore, in tomato hydroponic cultivation, there was no difference in the type of medium in growth, productivity, fruit quality and the environmental and water management had a great effect, and it is expected that the reuse of rockwool will have a positive effect on the economic point of view.

'Arihyang', a Strawberry Variety with Highly Firm and Large-Sized Fruit for Forcing Culture (촉성재배용 고경도 대과성 딸기 품종 '아리향')

  • A strawberry variety 'Arihyang' was derived as an artificial cross between 'Tochiotome' and 'Seolhyang' in 2014. The seedling and line selections were conducted from 2014 to 2015. Preliminary and advanced yield trials of '14-5-5,' which was the final selected line, were conducted from 2015 to 2017. 'Arihyang' is suitable for forced cultivation and has strong plant vigor, uniformly large-sized fruit, and a high yield compared to those of the check variety, 'Seolhyang' and 'Maehyang.' Especially, vitamin C was at a significant level, which was approximately 15% higher than that of 'Seolhyang.' The average number of flowers per first flower cluster was 10.5, which could reduce the labor of thinning fruit. Its fruit has a conical shape, dark red color, and glossy skin. The fruit was of good quality but has recommendations for harvest at the fully ripened stage. 'Arihyang' has intermediate resistant to phytophthora crown rot, but is susceptible to powdery mildew, gray mold, anthracnose, and fusarium wilt. It is reguired to manage major diseases and pests using optimum cultivation techniques and chemical control.
