AFLP Fingerprinting of Brassica campestis L. ssp. napus var. nippo-oleifera Makino from Korea

  • AFLPS (amplified fragment length polymorphisms) were used to estimate the genetic diversity of seven populations of Brassica campestis L. ssp. napus var. nippo-oleifera Makino between naturalized and cultivated populations. The seven Korean populations maintained a high level of genetic diversity. For example, all eight primers were high polymorphic, with an average of 3.2 effective alleles per primer set, and the expected heterozygosity was also high. The majority of genetic variance resided within populations The combinations of an insect-pollinated, outcrossing breeding system, large populations sizes, a high degree of gene flow and a propensity for high fecundity may explain the high level of genetic diversity within cultivated populations. Estimates of genetic similarity on the proportion of shared fragments ranged from 0.952 to 0.999. The high level of gene flow In Korean naturalized populations is mainly caused by seed dispersal via sea tide and the gene flow of cultivated populations may be enhanced in part by artificial pollen dispersal.

A Monitoring for Citizen Participation in Artificial Nest Boxes Using Mobile Applications (모바일 애플리케이션을 활용한 시민참여 인공새집 모니터링 방안 연구)

  • Great tit (Parus major) is a bioindicator species that can measure environmental changes in urban ecosystems and plays an important role in maintaining health as a representative insectivorous bird. Researchers have utilized artificial nest box surveys to understand the reproductive ecology of the Paridae family of birds, including the Great tits, but it is difficult to conduct a macroscopic study due to spatial and temporal limitations. This study designed and applied a citizen-participatory monitoring of artificial nest boxes project to transcend the limitations of expert-centered monitoring methods. The Suwon Front Yard Bird Monitoring Team installed artificial nest boxes in green spaces in Suwon, Gyeonggi Province and observed the reproductive ecology of the Paridae family through the participation of voluntary citizen surveyors. Participants were recruited through an online survey from February 9 to February 22, 2021, and they directly performed from installation to observation of artificial next boxes from February 23 to August 31, 2021. Online education was provided to the volunteers for the entire monitoring process to lower the entry barrier for non-expert citizen surveyors and collect consistent data, and observation records were collected through a mobile app. A total of 98 citizen surveyors participated in the citizen-participatory monitoring of artificial nest boxes project, and 175 (84.95%) of the 256 distributed artificial nest boxes were installed in green spaces in Suwon City. Among the installed artificial nest boxes, the results of the citizen science project were confirmed for 173 (83.98%), excluding two boxes with position coordinate generation errors. A total of 987 artificial nest box observation records were collected from citizen surveyors, with a minimum of one time, a maximum of 26 times, and an average of 5.71±4.37 times. The number of observations of artificial birdhouses per month was 70 times (7.09%) in February, 444 times (44.98%) in March, 284 times (28.77%) in April, 133 times (13.48%) in May, 46 times (4.66%) in June, 6 times (0.61%) in July, and 4 times (0.41%) in August. Birds using the artificial nest boxes were observed in 57 (32.95%) of the 173 installed artificial nest boxes, and they included Great tit (Parus major) using 12 boxes (21.05%), Varied Tit (Parus varius) using 7 boxes (12.28%), and unidentified birds using 38 boxes (66.67%). This study is the first to consider citizen participation in the monitoring of artificial nest boxes, a survey method for the reproductive ecology of the Paridae family, including Great tits, and it can be utilized as basic data for the design of ecological monitoring combined with citizen science in the future.

  • The purpose of this study was to investigate the inhabiting ecology of wild birds using artificial nests in Gyeongnam province. The sort of birds, the utilization of artificial nests by the materials, by the sites and by the forest type, and other inhabiting ecology of birds were investigated from 1984 to 1986. The results obtained were as follows; 1. Wild birds in surveyed areas were classified into 20 families and 35 species. Among them the birds inhabiting the artificial nests were 6 species and those using them most frequently were Parus ater (17.2%), P. major (16.7%) and P. palustris (12.2%). 2. The artificial nests installed on the top and at the toot of the mountain were chiefly utilized by P. ater and P. major, but the artificial nests in park area were mostly used by Passer montanus and Sturnus cineraceus. 3. Sixty-one percent of the artificial nests were utilized by the birds; 45.5% were used for breeding and 15.6% were inhabitation only. 4. When different nest materials and shapes were tested, mixed nests (sawdust and cement) were used most frequently, and C type. (Diamond shape) of wood nests was used more frequently compared with the A (Roof shape) and B (Box shape) types of wood nests. Meanwhile, the birds prefered the 3cm hole size (81.1% of utilization) to 4cm of hole (57.8%) and 5cm of hole (24.4%). 5. The artificial nests constructed in mixed forest were more frequently inhabited (73.3%) by the birds than coniferous forest (68.3%) and broad-leaved forest (63.3%). 6. The period of egg-laying in Parus species was from late April to early May, while Eophona migratoria, Lanius bucephalus, Motacilla alba and Passer montanus laid eggs generally in the middle of May. 7. Parus species, Passer montanus and Motacilla alba laid one egg every day for 5-10days, but Eopphona migratoria laied 4 eggs in 7 days and Lanius bucephalus laid 5 eggs in 4 days. The incubation period of Parus species was 16-18 days, while the others were about 11 to 14 days. 8. Thirteen days after the hatch of Parus major, chicks reached 13.9g of body weight, 72.0mm of wing-length and 20.9mm of tarsus length. P. montanus chicks reached 20.3g of weight, 66.2mm of wing-length and 20.2mm of tarsus length during the same period. 9. Food item of feeding chicks almost consisted of creatures (98.2%) with following composition; 55.2% of insect larvae, 37.2% adult insects, 2.8% of pupae and 2.8% of spiders. Vegetable items were 2% only. Among those creature food items, 95.2% were insect pests to forest.

The effect of Equilume light masks on the timing of seasonal ovulation of Thoroughbred mares in South Korea

  • Advancing the estrous cycle of mares is an essential breeding strategy that is routinely conducted by Thoroughbred breeders to improve economic outcomes. For this purpose, Equilume light masks have been developed as an alternative to existing technologies such as artificial lighting or hormonal treatments because they are considered as valid as existing methods with additional animal welfare advantages. For example, with the Equilume light masks, horses can be let out into the pasture, whereas they have to be kept indoors during lighting treatment. Because the function of Equilume light mask on the estrous cycle of mares is influenced by environmental factors such as nutrition condition and temperature, Equilume light mask should be studied in various environments. The objective of the present study was thus to verify the effect of Equilume light masks on the onset of the estrous cycle in Thoroughbred mares in South Korea. Mares were randomly selected and separated into two groups at two Thoroughbred horse breeding farms. The mares in the treatment group were equipped with Equilume light masks from November 18 to February 10 the following year. The body condition, the number and size (> 35 mm) of uterine follicles, and the uterine horn score of the mares were assessed on January 6 and February 10. The body condition scores were not different between the two groups. The treatment with the Equilume light mask had no positive effects on developing follicles and the reproductive tract. In conclusion, the use of Equilume light masks did not influence the seasonal resumption of the estrus cycles in Thoroughbred mares in South Korea.

GENOTYPIC AND PHENOTYPIC CORRELATIONS IN A SOYBEAN CROSS

  • In a plant breeding program, an efficient selection of desired characters in a population is important. Generally, many agronomic characters in a given population are determined by polygenes and quantitatively inherited. In practice, the genetic relationship between two observed characters which are undoubtedly subjected to the environmental influence is difficult to identify. In recent years, many workers have attempted to understant the genetic relationship between characters in terms of genotypic correlation, and the knowledge thus gained should furnish many important and useful information for the planning of breeding, selection, and interpretation of the result. The genotypic correlation is the result of pleiotropy, linkage of genes(2, 3, 5, 6, 8) and natural or artificial selection(4). The purposes of this study were to estimate genotyric and phenotypic correlations between all possible pairs of nine characters. and to seek certain characters which may be useful as indicators of certain important agronomic characters. Weber and Moorthy(10), Johnson et al. (5) and Sheth(7) found that in general, the genotypic correlations were higher than the phenotypic correlations. Weiss et al. (11) obtained significant positive correlations between maturity and oil content, maturity and low protein content, and high protein content and low oil content. Weber and Moorthy(10) reported the positive genotypic correlations between flowering and maturity, yield and maturity, yield and plant height, yield and seed weight, and negative genotypic correlations between maturity and oil content, and oil content and seed weight. Johnson et al. (5) studied the genotypic and phenotypic correlations among 24 characters and concluded that selection based entirely on a long fruiting period, lateness, heavy seed, low protein, high oil and resistance to lodging would be effective in increasing yield. Sheth(7) found the following positive associations among characters; height and maturity, yield and lodging, low protein content and high oil content, and yield and low protein content. Hanson et al.(1) also reported high negative correlation between seed yield and protein content.

Breeding of Good Quality Leaves High Yielding Mulberry Cultivar "Suilppong" (Morus alba L.) (양질다수성 뽕품종 "수일뽕" 육성)

  • New triploid mulberry cultivar, Suilppong was bred among selected lines from F$_1$ seedlings obtained by crossing the female, artificial tetraploid No. 411 which obtained from Chungilppong seedlings, and the male, Kugsang 21. Local adaptability test has been carried out at four places (Suwon, Kongju, Jeonju and Jangsung) for five years since 1997. Leaf quality was tested by silkworm rearing for two silkworm rearing seasons (spring and autumn) at Suwon in 2000. Suilppong was recognized to be superior in productivity to control cultivar, Kaeryangppong. The new cultivar, Suilppong, has following agronomic characteristics as compared with Kaeryangnppong, which is leading cultivar widely cultivating in Korea. It is belonging to Morus alba L. with mixed shapes from the ovate to 5 lobated leaf shape, sprouting in spring nearly the same to the Kaeryangppong (Morus alba L.), high leaf yielding of 20% compared to Kaeryangppong (Morus alba L.). It is adaptable to everywhere except the places where cold damage occurs frequently in Korea. Suilppong is recommended to rear silkworms not only in spring but also in autumn besides for young silkworms.

Comparison of Embryo Production Performance and Conception Rate after Embryo Transfer between Mongolian Cattle and Korean Native Cattle

  • This study is to compare the effect of estrus synchronization and embryo transfer between Korean and Mongolian cattle. Embryos were collected from 9 donors housed in Asan city in South Chungcheong Province, South Korea. Embryos were collected 9 donors from Khushaat sum, Selenge province and Bayanchandmani sum, Tov province in Mongolia. Follicle Stimulating Hormone (FSH), Controlled Internal Drug Release (CIDR) and Prostaglandin (PG) were used for superovulation. Subsequently, Artificial Insemination (AI) was done for donor cow and embryo was collected after 7 and 8 days. Collected embryos were compared between Mongolian and Korean cattle. Finally, good quality and fresh embryos were transferred to 50 and 22 recipients of cows in Korea and Mongolia respectively. The findings show that Korean native cattle each donor cow produced on an average 16.9 embryos and, 10.9 embryos were found transferable. But in case of Mongolia the average production of embryos per donor cow was 8.6 embryos and, 6.2 embryos were found transferable. Embryo collection after 7 and 8 days was not difference in embryo production in Korea. But, in Mongolia embryo production after 8 days was found more efficient than the 7 days. Korean native recipient's cows (74.6%) and Mongolian recipient's cows (71.0%) respectively were found transferable ovarian stage. The result suggested that efficiency of embryo production from the superovulation method treated of Korean cow were higher than the Mongolian cow. The pregnancy rate of Korea native cattle was 72%, which was about 10% higher than that of Mongolia cattle.

  • A combined technology of transvaginal ovum pick-up(OPU) system with in vitro-oocyte manipulation technique can be used for improving reproductive efficiency in the cattle. The objective of this study was to establish a newly-conceived breeding program using OPU in the pregnant cows. The OPU trial was performed in pregnant cows every 10 days from 40 through 90 days of artificial insemination (Al), and number of follicles in ovary, number of retrieved oocytes and embryo development following in vitro-fertilization, were evaluated. Reduced number of follicles in the ovaries of pregnant cows was firstly detected from 70 days after A' and a significant (P<0.05) decrease in the follicle number (5.4 follicles /donor) was found at 90 days than at 40, 50, 60 and 80 days after Al (8.0~9.2). A similar pattern was also observed in the number of oocytes retrieved by OPU apparatus during experimental period. When retrieved oocytes were matured and inseminated in vitro with frozen bull semen, development of the oocytes to the blastocyst stage was not significantly affected by the retrieval time. Four embryos (morula or blastocyst stage) derived from oocytes retrieved from pregnant cows were nonsurgically transferred to four recipient cows on day 7 of estrus cycle. For the first time in Korea, three of four transferred embryos developed to live calves with normal physiological parameters. In conclusion, an effective breeding program employing pregnant cow can be developed by use of OPU trial and in vitro culture techniques of oocytes ; OPU system could be repeated in pregnant cows with no risk of abortion and viable offsprings were borne after transfer to the recipients.

Population Change of Each Ardeidae Species in Damaged Habitats of Development Area (개발지역의 서식지 훼손에 따른 백로류 종별 개체군 변화)

  • This study aims to provide the biological and geographical materials to identify the changes in population of Family Ardeidae included Egretta alba modesta, Egretta garzetta, Bubulcus ibis, and Nycticorax nycticorax and establish their conservation measurement in terms of the fragmented and deteriorated habitat by human development such as housing and park projects. Wonjucheon Stream was measured the highest species diversity since the survey site was relatively less interfered by the surrounding human beings showing lower disturbance elements. However, the closer the stream comes downstream, the more the artificial disturbance elements generate, confirming that the species better adapted to contamination grow in more density so that the species composition becomes simplified. It implies that feeding place as well as breeding-growing places is also more closely related to the changes in population structure and species composition of Family Ardeidae. The results of Family Ardeidae immigration in Taejang-dong, Wonju, among the other survey sites, revealed that a total of 231 came to the site in 2006; and 210 in 2011, showing more or less reduction in the population. In the mean while, Namyang-dong in Hwaseong City showed the continuous decrease in population number of Family Ardeidae with 135 before development and 60 during development, resulting in the number never growing but continuously narrowing over and over after development. As a result of surveying the number of Family Ardeidae that bred following the findings above, Taejang-dong in Wonju City had 4 species of Family Ardeidae being bred, however, showing the decrease in number of population from 998 in 2006 to 589 in 2011. Namyang-dong in Hwaseong was confirmed to have as many as 352 of the population in 2006 and 230 in 2008; after the deforestation on their habitat, they had not lived in the site any longer, suggesting that the development would significantly affect Family Ardeidae.

Breeding for New Evergreen Dianthus Cultivar 'URI 2010-6' with Bright Red Flower Color (밝은 적색계의 상록성 패랭이꽃 신품종 'URI 2010-6' 육성)

  • A new Dianthus cultivar 'URI 2010-6' was selected among F1 progenies from a cross between interspecific hybrid 'Evergreen Emerald' as the seed parent and D. chinensis ' Ruby' as the pollen parent. In 2007, the cross w as made, and the seedlings of $F_1$ progenies were obtained and investigated. After three rounds of characteristic evaluation from 2008 to 2010, a clone was selected and registered as 'URI 2010-6' at the Korea Seed and Variety Service (plant variety protection number: 4431) in 2011. The flower of 'URI 2010-6' is caryophyllaceous, single type, and mainly bright red (Red, N79C) with secondary white (White, N155C) on the edge or as spots on the petal. The flower has diameter of 2.7 cm and length of 0.8 cm. This cultivar blooms continuously about 150 d on average and may be used as evergreen plant during winter months.
