Effect of Fermented Lactic Acid Bacteria on Antiallergic Effect of Artemisia princeps Pampanini

  • Artemisia princeps Pampanini, which is named as Sajabalssuk (SJ-1) in Korea, was fermented with lactic acid bacteria (LAB), and their antiallergic activities were investigated. When SJ-l was fermented with some LAB isolated from human feces, the inhibition of NO production in RAW264.7 cells and antioxidant activities of SJ-1 were not affected. However, the inhibitory activity of SJ-1 against degranulation of RBL-2H3 cells induced by IgE was increased by LAB fermentation. Among the LAB tested, Bifidobacterium infantis K-525 provided the most potent inhibitory effect of SJ-1 against degranulation of RBL-2H3 cells. SJ-1 extract fermented with B. infantis K-525 (F-SJ-1) potently inhibited the mouse passive cutaneous anaphylaxis reaction induced by IgE with antigen, skin dermatitis induced by 12-O-tetradecanoylphorbol-13-acetate, and scratching behaviors induced by compound 48/80. These inhibitory activities of F-SJ-1 were more potent than those of SJ-1. These findings suggest that the inhibition of SJ-1. extract against IgE-induced allergic diseases, such as rhinitis and asthma, can be enhanced by LAB fermentation.

Essential Oil Isolated from Iranian Yarrow as a Bio-rational Agent to the Management of Saw-toothed Grain Beetle, Oryzaephilus surinamensis (L.) (머리대장가는납작벌레의 합리적 방제 물질로 이란 서양가새풀 정유의 살충효과 평가)

  • Overuse of synthetic pesticides caused negative side-effects such as environmental contamination, development of insect pests' resistance, and effects on non-target organisms. Plant origin substances without/or with low mammalian toxicity have been considered as promising alternatives to the synthetic pesticides. Fumigant toxicity of the essential oil of Iranian Yarrow, Achillea millefolium L., was investigated against a cosmopolitan stored-product insect pest: saw-toothed grain beetle (Oryzaephilus surinamensis L.). Chemical profile of this essential oil was studied by Gas Chromatography-Mass Spectrometry. Tested concentrations were significantly effective to the mortality of insect pest. A positive correlation between essential oil concentrations and pest mortality were realized. LC50 value (lethal concentration needed to 50% mortality) was achieved as $17.977(16.195{\pm}20.433){\mu}l/l$ air. The main components were 1,8-Cineole (13.17%), nerolidol (12.87%), ${\alpha}$-cubebene (12.35%), artemisia ketone (6.69%), ${\alpha}$-terpineol (5.27%), alloaromadendrene oxide (4.71%) and borneol (3.99%). Terpenic compounds including monoterpene hydrocarbons (8.19%), monoterpenoids (44.23%), sesquiterpene hydrocarbons (21.69%) and sesquiterpenoids (22.24%) were 96.35% of the total identified compounds. Results indicated that the terpene-rich A. millefolium essential oil may be considered as a safe bio-agent in the O. surinamensis management.

Antioxidative and Antibacterial Activities of Artemisia princeps Pampanini Extracts (사자발쑥 추출물의 항산화 및 항균 활성)

  • In the present study, the antioxidative and antibacterial activities of Artemisia princeps Pampanini (A. princeps Pamp.) extract were investigated. The ethyl acetate fraction of A. princeps Pamp. showed the most prominent free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}=12.27{\mu}g/mL$). Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of A. princeps Pamp. extract on $Fe^{3+}-EDTA/H_2O_2$ systems were investigated using a luminol-dependent chemiluminescence assay. The ethyl acetate fraction of the extract ($OSC_{50}=0.33{\mu}g/mL$) had a 5 times greater ROS scavenging activity than L-ascorbic acid ($1.50{\mu}g/mL$), known as a water soluble antioxidant. The cellular protective effects of fractions of A. princeps Pamp. on the rose-bengal sensitized photohemolysis of human erythrocytes were examined. The aglycone fraction of extracts suppressed photohemolysis in a concentration dependent manner. The inhibitory effects of A. princeps Pamp. extract on tyrosinase were investigated to assess their whitening efficiency. The ethyl acetate fraction demonstrated a 7 times higher tyrosinase inhibitory effect ($IC_{50}=29.20{\mu}g/mL$) than albutin, known as a whitening agent. The antibacterial activity of ethyl acetate fractions against various normal skin flora were measured. The results showed that the antibacterial activity of the fraction was the highest on Staphylococcus aureus, Bacillus subtilis, and Propionibacterium acnes. Antioxidant substances were isolated and purified from the ethyl acetate fractions. Eupatilin and jaceosidin were identified. These results indicate that the extract/fractions of A. princeps Pamp. can function as antioxidant and/or antibacterial agents for the skin.

  • This study was conducted of Korean native plants to screen herbicidal activity which could be used for the development of new natural herbicides. Ninety-eight plants were collected from Wan Island, Chollanamdo in Korea and their methanol extracts were obtained. Herbicidal activities of the methanol extracts were determined by seed bioassay using canola (Brassica napus L.) seedlings. Among ninety-eight species, twenty plants were highly herbicidal ($GR_{50}<1,000\;{\mu}g\;g^{-1}$): Abies holophylla MAXIM., Ailanthus altissima (MILL.) SWINGLE, Anthemis nobilis L., Aralia elata SEEM., Artemisia iwayomogi KITAMURA, Asarum sieboldii MIQ., Brassica campestris subsp. napus var. nippo-oleifera MAKINO, Clematis terniflora DC., Crataegus scabrida SARG., Gnaphalium affine D. DON, Jasminum nudiflorum LINDL., Kalopanax pictus (THUNE.) NAKAI, Machilus japonica S. et Z., Myrica rubra S. et Z., Osmunda japonica THUNB., Phytolacca esculenta V. Houtte, Platanus occidentalis L., Quisqualis indica L., Rubus hirsutus THUNB., Yucca smalliana FERN. Fifty plants were shown moderate herbicidal activity $(1,000\;{\mu}g\;g^{-1}, however, twenty-eight plants were not shown any herbicidal activity.

In vitro Effects of Essential Oils from the Aerial Parts of Artemisia annua L. Against Antibiotic-Susceptible and -Resistant Strains of Salmenella typhimurium (항생제 내성 및 감수성 Salmonella typhimurium 균주에 대한 개똥쑥 지상부 정유와 Kanamycin의 병용효과)

  • The essential oil fraction from the aerial parts of A. annua was analyzed by GC-MS. As the results, caryophyllene oxide (11.7%), caryophyllene (7.54%), camphor (7.32%), 1,8-cineol (4.98%), and borneol (3.99%) were confirmed as the main components of the oil fraction. The effects of this oil and its main components on antibiotic-susceptible and -resistant strains of Salmonella enteritidis and S. typhimurium were assessed. A. annua oil fraction significantly inhibited all strains of the two Salmonella species examined, with minimum inhibiting concentrations (MICs) ranging from 2.0 mg/ml to 8.0 mg/ml. Among the main components of the oil, borneol and camphor showed relatively strong inhibiting activity with MICs between 1.0 mg/ml and 4.0 mg/ml. The MICs of caryophyllene and caryophyllene oxide were higher than 16 mg/ml. The combination effects of the oils with kanamycin were evaluated using a checkerboard microtiter assay. Against S. typhimurium KCCM11862 and CCARM8009 strains, the oil fraction of A. annua, camphor, and 1,8-cineol exhibited significant synergistic with kanamycin with fractional inhibitory concentration (FIC) indices in the range of 0.085 to 0.375. In conclusion, a combination of kanamycin and A. annua oil or its main component, camphor, and cineol, may be useful for reducing the minimum effective dose of antibiotic required for the treatment of resistant S. typhimurium infections.

Inhibitory Effect of Artemisiae Annuae Herba Extracts on Melanin Synthesis, Tyrosinase Activities and Production Levels of Tyrosinase, MMP-1 and MMP-9 in SK-MEL-2 Cells (SK-MEL-2 세포에서 청호(靑蒿) 추출물의 Melanin 생성, Tyrosinase 활성과 생성, MMP-1 및 MMP-9 생성 억제 효과)

  • Objectives: Artemisiae Annuae Herba is the dried aerial part of Artemisia annua L. (AAL). In Oriental medicine, Artemisiae Annuae Herba (AAH) is traditionally used to treat fever. AAH clears summerheat or damp-Heat, clears deficiency fevers, cools the blood and stops bleeding, stops malarial disorders and relieves heat, clears liver heat and brightens the eyes. Recently, there were many studies about effects of AAH on anti-oxidative, anti-inflammatory, anti-cancer, hair growth and plasma lipid composition. So, we expected AAH has an availability that can effect on skin whitening and elasticity. Methods: The present study was designed to investigate the effects of AAH on skin whitening and elasticity in SK-MEL-2 cells. In this experiment, the effects of AAH on proliferation rates, melanin synthesis, tyrosinase activities and production levels of tyrosinase, MMP-1 and MMP-9 in vitro were examined. Results: AAH did not affect viability of SK-MEL-2 cells and inhibited melanin synthesis induced by ${\alpha}$-Melanocyte-stimulating hormone (${\alpha}$-MSH) significantly. In addition, AAH also inhibited tyrosinase activity and lowered tyrosinase level in SK-MEL-2 cells. Finally, AAH inhibited productions of Matrix metalloproteinase-1 (MMP-1) and Matrix metalloproteinase-9 (MMP-9). Conclusions: These data suggest that AAH can be used to treat patients with skin diseases such as freckled face and also used as skin whitening agent.

Studies on Protective Effect of DA-9601\ulcorner an Artimisiae Herba Extract, against Ethanol-induced Gastric Mucosal Damage and its Mechanism (에탄올-유발 위점막손상에 대한 애엽추출물 (DA-9601)의 방어효과 및 기전에 관한 연구)

  • Protective effect of DA-9601, an extract of Artemisia Herb, against ethanol-induced gastric mucosal injury was evaluated in rats. In the prophylactic study, DA-9601 exhibited total protection (99.4%) against absolute ethanol-induced gastropathy, And the protective effect of DA-9601 lasted up to 2 hours, which was longer than those of other contemporary mucoprotectants. In the treatment study, DA-9601 significantly facilitated the healing of 70% ethanol-induced mucosal damage, which was superior to cetraxate, a commonly used anti-ulcer drug. The mechanisms of mucoprotection of DA-9601 were also assessed. DA-9601 increased the release of prostaglandin E$_2$ from murine neutrophils in a dose-dependent manner in vitro. The cytoprotective effect of DA-9601 against ethanol-induced mucosal damage was significantly diminished by the concommitant injection of N$\omega$-nitro-L-arginine methyl ester (L-NAME, 5 mg/kg, i.v.), a non-specific nitric oxide (NO) synthase inhibitor, while it was not affected by preinjection of indomethacin (5 mg/kg, s.c.), a prostaglandins-depletor. And it was found that DA-9601 significantly enhanced adaptive cytoprotective action of 10% ethanol against absolute ethanol (56.9$\pm$6.5 vs 23.0$\pm$3.3 mm$^2$, p<0.05, mean$\pm$SEM), though its exact underlying mechanism remains to be clarified. The present fin[lings demonstrate that DA-9601 exerts gastroprotecticv actions for the stomach against ethanol through several different underlying mechanisms, in which prostanglandins and NO are involved. In conclusion, the results obtained suggest that DA-9601 can be useful both in prevention and treatment of ethanol-induced gastric damage.

  • To detect the toxic relevant components, asarone, coumarin and thujone, in the 87 medicinal plants used in brewing a Korean traditional folk wine, their 20% ethanol extracts were prepared and purified by a SPE(solid phase extraction) method. The toxic components in the purified extracts were identified by GC-FID and GC/MS analysis. On analyses, asarone was detected in 6 species, Acorus gramineus Solander, Acorus asiaticus Nakai, Angelia gigus Nakai, Santalum album, etc.; coumarin in 22 species, Anethum graveolens, Foeniculum vulgare Gaertner, Lithosperum erythrorhizon Siebold et Zuccarinii, etc.; thujone in 24 species, Achyranthes japonica Nakai, Amomum xanthiodes Wallich, Artemisia asiatia Nakai, seed of Cannabis sativa L., Caragana sinica R., Chrysanthemum morifolium Ramat, Codonopsis lanceolata Bentham et flooker, Foeniculum vulgare Gaertner, etc.

Effects of Selenate Ion Concentration in Nutrient Solution on the Growth and Essential Oil Content of Wormwood( Artemisia absinthium L.) (배양액 내의 Selenate 이온농도가 웜우드(Artemisia absinthium L.)의 생육 및 정유함량에 미치는 영향)

  • This study was conducted to evaluate appropriate selenate ion concentration for the production of high functional vegetables. Sodium selenate was treated 0, 2, 4, 6 and 8mg/$\ell$ using herb nutrient solution developed by European R & D Vegetable Center in Belgium. Low level of Na2se04 concentration increased the growth of wormwood, but high selenate concentration decreased the growth. Total chlorophyll content was increased by sodium selenate. The higher selenate ion concentration in the nutrient solution, the more total chlorophyll content was. The vitamin C content in wormwood was high at 2 and 4mg/$\ell$ treatment, showing good growth, at higher concentration, however, the vitamin C content decreased. At 4mg/$\ell$ selenate ion concentration, essential oil content of wormwood was best. But higher selenate ion concentration decreased essential oil content. The uptake of Se by the plant increased with the increase of selenate ion concentration.
