Virginiamycin 생합성 유도인자 Virginiae Butanolide C에 이한 Erythromycin 생산 유도 (Induction of Erythromycin by Virginiamycin Inducing Factor, Virginiae Butanolide C)

  • Streptomyces virginiae 유래의 virginiamycin 생합성 유도인자인 VB-C를 이용하여 다른 방선균에의 항생물질 생산 유도 기능을 검토하였다. Erythromycin 생산균인 Streptomyces erythraeus를 공시균주로 사용하였으며, VB-C 결합 receptor 유전자가 포함된 plasmid(pARS 701, 9 kb)를 공시균주에 도입하여 transformant로 사용하였다. Parent와 transformant 모두 유도인자인 VB류를 생산하였으며, VB-C의 signal 전달에 관여하는 결합 단백질의 존재가 확인되었다. Parent는 본배양 0, 20, 44시간째에 합성 VB-C 300 ng/ml를 첨가시 초기 항생물질 생산시기가 약 8시간 이상 단축되었다. Transformant는 본배양 44시간에 첨가시 6시간 이상 항생물질 생산시기가 단축되었고 항생물질 생산량도 증가되었으며, parent에 비해 B. subtilis에도 강한 항균력을 나타내었다. 또한 항균 spectrum은 parent보다는 transformant가 생산한 항생물질의 항균호과가 더 큰 것으로 나타났으며, VB-C를 첨가한 경우 미첨가 경우보다 넓은 항균 spectrum을 보였다. 이들 결과에서 공시균인 S. erythraeus에서 VB signal 전달에 따른 항생물질 생산유도가 입증되었으며, VB-C receptor gene의 도입에 따른 발현과 함께 VBs의 유도, 항생물질의 생산시기의 단축 및 생산량의 증가가 예상됨에 따라 다양한 Streptomyces sp. 균주를 대상으로 항생물질의 대량생산을 비롯한 새로운 2차 대사산물 생산에의 응용이 예상되었다.

시호가룡골모려탕(柴胡加龍骨牡蠣湯)을 이용한 발달장애 환자의 ADHD증상 관리 1례 (A case study on a patient of developmental disabilities with ADHD)

  • Objective : The purpose of this report is to submit a case study on a patient of developmental disabilities with ADHD by taking "Sihogayonggolmoryeo-tang". Methods : We gave herbal medicine to a patient and observe the progress as the way of Cornners Abbreviated Rating Scale(CARS), ADHD Rating Scale (K-ARS), The ADD-H Comprehensive Teacher's Rating Scale(ACTeRS), Child Attention Problem(CAP), IOWA Conners Rating Scale, Conners Teacher Rating Scale(CTRS), Conners Parents Rating Scale (CPRS), Home Situation Questionnaire(HSQ), School Situation Questionnaire(SSQ). Results : The total score of CARS, ACTeRS and other scales were decreased by taking herbal medicine, and especially hyperactive scores were significantly decreased. Conclusions : Patient's symptom was significantly improved by using "diagnostic system based on Shanghanlun six meridian patterns and provisions" and analyzing patient's psychological and physical conditions.

능동 후륜조타와 요우 모멘트의 협조제어에 관한 연구 (A Study on Integrated Control System Design of Active Rear Wheel Steering and Yaw-Moment Control Systems)

  • Conventionally, 2WS is used for vehicle steering, which can only steering front wheel. In case of trying to high speed lane change or cornering through this kind of vehicle equipped 2WS, it may occur much of Yaw moment. On the other hand, 4WS makes decreasing of Yawing Moment, outstandingly, so it is possible to support vehicle movement stable. And conventional ABS and TCS can only possible to control the longitudinal movement of braking equipment and drive which can only available to control of longitudinal direction. There after new braking system ESP was developed, which controls both of longitudinal and lateral, with adding of the function of controlling Active Yaw Moment. On this paper, we show about not only designing of improved braking and steering system through establishing of the integrated control system design of 4WS and ESP but also designing of the system contribute to precautious for advanced vehicle stability problem.

환경 안에서의 교육이 주의력 결핍 과잉 행동 장애(ADHD) 아동의 부적응 행동 변화에 미치는 영향 (Influence of Education in Environment on Maladjusted Behavior Changes among Children with ADHD)

  • This study investigated ADHD prevalence among the first to sixth graders in ordinary classes of H elementary school located in an area of special rural conditions and selected 4 sixth graders who received high scores in a self-diagnostic test on attention deficit and over 17 points from teachers in an ARS-IV test with a view to verifying the effects of the education in environment on the improvement of maladjusted behaviors among ADHD children when the education program for improvement of maladjusted behaviors is applied to the environment. The program was applied from March to mid-July for about 4.5 months in 13 periods. The program applied consisted of activities of experiencing the nature. Behavior changes of targets during the course of the program were recorded in the frequency of ADHD target behaviors card and the observation log to describe the results, which underwent a comparative analysis through pre-and post-tests of ADDES-SV and to graph the changes in maladjusted behaviors of target children. To sum up, the education in environment resulted in positive effects on the changes of maladjusted behaviors among ADHD children by reducing attention deficit impulsive behaviors and hyperactivity.

Plant-Parasitic Nematode Genera Associated with Turfgrass in Maryland Golf Courses and Athletic Fields

  • Field surveys were conducted to assess the occurrence and diversity of plant-parasitic nematodes (PPNs) in golf courses and athletic fields across Maryland, USA, during 2022 and 2023. A total of 28 golf courses and ten athletic fields were surveyed, revealing the prevalence and abundance of 13 PPNs taxa in the region. Criconemoides was identified as the most prevalent (94.9%) and Tylenchorhynchus as the most abundant (2.3) across all samples. Central golf courses (west side of the Chesapeake Bay) exhibited a high prevalence of Criconemoides and Tylenchorhynchus, while Eastern Shore golf courses and athletic fields displayed a higher prevalence of Helicotylenchus and Criconemoides. Further, Belonolaimus longicaudatus was reported for the first time from turfgrass in Maryland, raising concerns due to its potential to cause severe damage on both cool- and warm-season turfgrass. Biodiversity analysis indicated that richness (R2) was higher in athletic fields, while diversity (H') and evenness (J') were significantly greater in golf courses. This study provides baseline information for monitoring PPNs distribution in Maryland and also for the development of effective nematode management approaches in turfgrass ecosystems.

Use of δ-Aminolevulinic Acid in Swine Diet: Effect on Growth Performance, Behavioral Characteristics and Hematological/Immune Status in Nursery Pigs

  • Certain amino acids are essential precursors of a variety of important biomolecules in addition to their major function as protein building blocks. ${\delta}$-Aminolevulinic acid (ALA) is synthesized from the condensed form of succinyl-CoA with glycine after decarboxylation catalyzed by ALA synthase. The objective of the study was to determine the effects of ALA supplementation on growth performance, behavioral characteristics and hematological/immune status in nursery pigs. A total of 144 pigs weaned at 21 d of age were allotted to three dietary treatments representing (-) control (w/o antibiotics; NC), (+) control (w/carbadox at 50 mg/kg; PC), and the treatment group with ALA supplementation (0.05%; TA). Each treatment had 6 pens (replicates) with 8 pigs per pen. Pigs were fed phase 1 (21.9% CP, 1.40% Lys) and 2 (20.6% CP, 1.15% Lys) experimental diets for 3 and 2 wks, respectively. Feed intake and weight gain were measured weekly during phase 1 and at the end of phase 2. At the end of phase 2, blood samples were taken and analyzed using an automated hematology analyzer. Skin color and activity of pigs (48 h) from all pens in each treatment were measured at the second week of phase 2. Growth performance was not affected (p>0.05) by the dietary supplementation of ALA during the 5 wk nursery period. Pigs in the TA (6.46) and PC (6.68) had a higher (p<0.05) number of red blood cells ($10^6cell/{\mu}L$) than pigs in the NC (6.15). Pigs in PC (12.16) had a higher (p<0.05) hemoglobin level (g/dL) than pigs in the NC group (11.29) and the TA group (11.47). Pigs in the TA and PC had darker (p<0.05) and less (p<0.05) yellow skin color than pigs in the NC. Pigs in the PC tended (p = 0.081) to be less active than pigs in the other groups. There were no differences in behavioral characteristics between the NC and the TA. The data suggest that ALA supplementation has no adverse effects on growth performance of nursery pigs. Moreover, ALA supplementation increased red blood cell counts which may be beneficial to pigs.

효모 ABF1 단백질의 DNA Binding 부위에 대한 구조 기능 연구 (Structure-Function Analysis of DNA Binding Domain of the Yeast ABF1 Protein)

  • ABF1(Autonomously replicating sequence Binding Factor 1)은 효모 genome에서 $RTCRYN_5ACG$의 염기 서열을 가지고 있는 promoter, mating-type silencer, ARS에 결합하는 DNA binding 단백질이다. E. coli 에서 ABF1 유전자를 발현하기 위하여, ABF1 유전자를 pMAL-c2 벡터에 cloning하였다.(pMAHW). pMAHW를 E. coli에 형질전환하여, ABF1 융합단백질을 발현시키고, amylose resin affinity chromatography에 의하여 분리하였다. Factor Xa protease를 이용하여 분리된 융합단백질로부터 maltose binding protein을 잘라낸 후에 gel retardation analysis 방법으로 분리된 ABF1이 ARS1에 결합하는 능력을 지니고 있음을 확인하였다. DNA 결합에 관련된 부위를 찾기 위하여, 비전형적인 zinc finger motif가 위치하는 자리에서 pMAHW의 ABF1 유전자에 His-61을 다른 아미노산으로 치환하였다. DNA binding 부위로 추정되는 ABF1 단백질의 중간지역에 Leu-353, Leu-360를 다른 아미노산으로 치환하였다. Site-specific mutagenesis 를 통해 만들어진 mutant를 gel retardation analysis와 complementation test를 통해서 비전형적인 zinc finger motif이외에 다른 DNA binding motif가 있는 것을 알 수 있었다.

곤충기생성 성충, Steinernema feltiae와 Heterorhabditis heliothidis를 이용한 고자리파리 및 담배거세미나방의 생물적 방제 (Biological Control of Onion Maggot and Tobacco Cutworm with Insect-parasitic Nematodes, Steinernema feltiae and Heterorhabditis heliothidis)

  • 고자리파리와 담배거세미나방을 방제할 목적으로 곤충기생성 선충, S. feltiae와 H. heliothidis를 이용, 실내 실험한 결과는 다음과 같다. 고자리 파리에서 S. feltiae를 유충 한 마리당 30,60,120,240마리를 접종한 모든 구에서 80-100%의 높은 치사율을 얻었고, H. heliothidis 10,20,40,80마리 접종 구에서도 63.3-100%치사율을 얻었다. 담배거세미나방의 경우 S. feltiae는 유충한마리당 50,100,200,400마리를 접종하여 3령층은 전 선충 접종구에서 100%의 치사율을, 4령충은 76.7-100%, 5령충은 43.3-100%의 치사율을 나타내어 접종량과 령게에 따라 치아율에 차이를 보였다. H. heliothidis는 20,40,80,160마리를 접종하여 3령층에서는 67.7-100%의 높은 치사율을 보였으나 4령충과 5령충에서는 36.7-90%, 3.3-90%의 치사율을 각각 나타내었다. 먹이인 케일에 처리한 경우 S. feltiae는 비교적 효과가 있었으나 H. heliothidis는 효과가 현저히 검소하였다. 그러나 3령충에서는 효과가 있어 3령충이 가장 방제 적기이며 감수적 영기임이 확인되었다.

Implantation in Ruminants: Changes in Pre-Implantation, Maternal Recognition of Pregnancy, Control of Attachment and Invasion - Review -

  • As high as 50% of pregnancies are known to fail and the majority of such losses occur during the peri-implantation period. For the establishment of pregnancy in mammalian species, therefore, implantation of the conceptus to the maternal endometrium must be completed successfully. Physiological events associated with implantation differ among mammals. In ruminant ungulates, an elongation of the trophohlast in early conceptus development is required before the attachment of the conceptus to the uterine endometrium. Moreover, implantation sites are restricted to each uterine caruncula where tissue remodeling, feto-maternal cell fusion and placentation take place in a coordinated manner. These unique events occur under strict conditions and are regulated by numerous factors from the uterine endometrium and trophoblast in a spatial manner. Interferon-tau (IFN-${\tau}$), a conceptus-derived anti-Iuteolytic factor, which rescues corpus luteum from its regression in ruminants, is particularly apt to play an important role as a local regulator in coordination with other factors, such as TGF-${\beta}$, Cox-2 and MMPs at the attachment and placentation sites.

콩 Hypocotyl에서 Acetyl Soyasaponin $A_1$의 분리 및 구조 분석 (Isolation and Structural Analysis of Acetyl Soyasaponin $A_1$ from Hypocotyl of Soybean)

  • 본 연구는 국내육성 콩 품종 및 전통식품의 우수성을 입증하고, 생리활성이 우수한 고품질의 신품종 육성을 위한 기초기반기술 확립의 일환으로 group A soyasaponin의 분리 및 정제에 관한 연구를 수행한 결과를 요약하면 다음과 같다. 1. 콩 종실로부터 hypocotyl을 분리하고 탈지한 후 automated solvent extractor(ASE)를 이용하여 MeOH 추출 후 flash column($150mm{\times}40mm$ i.d.)을 이용하여 총 14개의 fraction을 분리하였다. 2. Flash column으로 얻어진 8 및 9번 fraction(Fraction-I) 을 Luna $C_{18}$ semipreparative reverse phase column($250cm{\times}50mm$ i.d.)을 이용하여 Fast PCLC로 정제하여 미지화합물(unknown compound : UKC)(Fr-I-2)을 분리하였다. 3. Compound 1을 NMR($^1H-NMR$, 600 MHz; $^{13}C-NMR$, 100 MHz; DEPT), IR, UV 및 ESI-MS 분석을 통하여 구조를 동정한 결과 분자량(MW)이 1436.6이며, 분자식이 $C_{67}H_{104}O_{33}$인 group A 계열의 acetyl soyasaponin $A_1$으로 확인되었다.
