The heavy use of petroleum products in modern livings has brought ubiquitous environmental contaminants of aromatic compounds, which persist in aquatic and geo-environment without the substantial degradation. The persistence and accumulation of the aromatic compounds, which include xylene, phenol, toluene, phthalate, and so on are known to cause serious problems in our environments. Some of soil and aquatic microorganisms facilitate their growth by degrading aromatic compounds and utilizing degrading products as growth substrates, the biodegradation helps the reentry of carbons of aromatic compounds, preventing their accumulation in our environments. The metabolic studies on the degradation of aromatic compounds by microoganisms were extensively carried out along with their genetic studies. A Rhodococcus sp. EL-GT isolated in activated sludges has shown the excellent ability to grow on phenol as a sole carbon source. In the present study investigated a gene encoding phenol-degrading enzymes from a Rhodococcus sp. EL-GT.

To understand the effects of marine environmental and meteorological parameters on laver Porphyra yezoensis production at Nakdong River Estuary, we analyzed marine environmental (water temperature, salinity, nutrients, etc.) and meteorological properties (air temperature, wind speed, precipitation, sunshine hours) with yearly and monthly variations in laver production over 10 years (2003-2013). Air and water temperature, wind speed, sunshine hours and precipitation were major factors affecting yearly variability in laver production at the Nakdong River Estuary. Lower air and water temperatures together with higher levels of nutrients and sunshine and stronger wind speeds resulted in higher laver harvests. Salinity and nitrogen did not show clear correlations with laver production, mainly due to the plentiful supply of nitrogen from river discharge and the low frequency of environmental measurements, which resulted in low statistical confidence. However, environmental factors affecting monthly laver production were related to the life cycle (culturing stage) of Porphyra yezoensis and were somewhat different from factors affecting annual laver production. In November, a young laver needs lower water temperatures for rapid growth, while a mature laver needs much stronger winds and more sunshine, as well as lower temperatures for massive production and effective photosynthesis, mostly in December and January. However, in spring (March), more stable environments with fewer fluctuations in air temperature are needed to sustain the production of newly deployed culture-nets ($2^{nd}$ time culture). These results indicate that rapid changes in weather and marine environments caused by global climate change will negatively affect laver production and, thus, to sustain the yield of and predict future variability in laver production at the Nakdong River estuary, environmental variation around laver culturing farms needs to be monitored with high resolution in space and time.
Biomass and community composition of microphytobenthos in the Saemankeum tidal flat were studied by HPLC analysis of the photosynthetic pigments from November 2001 to November 2002. The environmental factors of sediment were also investigated to examine the relationship between microphytobenthos biomass and sedimentary environments. The detected photosynthetic pigments of microphytobenthos were chlorophyll a, b, c, fucoxanthin, 19'-hexanoyloxyfucoxanthin, violaxanthin, diadinoxanthin, alloxanthin, diatoxanthin, zeaxanthin+lutein, peridinin and beta-carotene. Pheophytin a, the degradation product of chlorophyll a, was also detected. The results of pigmen analysis suggest the presence of diatom (fucoxanthin), euglenophytes (chlorophyll b), chlorophytes (chlorophyll b + lutein), cyanobacteria (zeaxanthin), cryptophytes (alloxanthin), chrysophytes (fucoxanthin + violaxanthin), prymnesiophytes (19'-hexanoyloxyfucoxanthin) and dinoflagellates (peridinin). Chlorophyll a concentration in the top 0.5 cm of sediment was in the range of $0.24\;mg{\cdot}m\^{-2}\;-32.11\;mg{\cdot}m\^{-2}$ in the study area. The increase of chlorophyll a concentration in the spring indicates the occurrence of a microphytobenthic bloom. In the summer, there was a sharp decrease of the chlorophyll a concentration which was probably due to high grazing activity by macrobenthos. The annual mean chlorophyll a concentration in the study area was low compared to that in most of other tidal flat areas probably due to active resuspension of microphytobenthos and high grazing activity by macrobenthos. There was no clear relationship between microphytobenthos biomass and sedimentary environments because of a large variety of physical, chemical and biological factors, Pigment analysis indicated that while diatoms were dominated in the microphytobenthic community of the Geojon tidal flat, euglenophytes and/or chlorophytes coexisted with diatoms in the Mangyung River tidal flat.
Dominant species of macrobenthos were analyzed based on differentiation of three distinct methods: the density based method, the biomass based method and LeBris method, by considering the frequency of occurrence using quantitative data collected over 5 years (1993-1998) at 21 stations in Chonsu Bay. Sedimentary environments as well as species composition and diversity showed the spatial and temporal distribution patterns. The ranks of dominant species as determined by the density based method were more similar to the results by the LeBris method than to those from the biomass based method. Considering the temporal variation, LeBris method were more efficient than any other methods for the determination of dominant species in Chonsu Bay. Lumbrineris longifolia, Theora fragilis, and Moerella jedoensis were recognized by all three methods. A one-way analysis of variance indicated spatial distributions patterns among most of the dominant species. These species showed positive correlations to sedimentary parameters such as mean grain size. However, T. fragilis and Paraprinospio pinnata showed the temporal patterns in their distribution, and were also correlated to the benthic environment, organic content and dissolved oxygen. Some dominant species, e.g., T. fragilis, S. scutata, G. gurjanovae proved to be useful benthic indicators based on the environmental variations determinated by long-term benthic ecological monitoring in Chonsu Bay.
Environmental monitoring was conducted to identify the characteristics and patterns of sedimentary environments of tidal flats in 12 areas on the west coast of Korea. The habitat of the mud shrimp Upogebia major contained higher organic and mud contents compared to the habitat of the manila clam Ruditapes philippinarum. In particular, the mud content increased from 84.38% to 89.18% in the Seongam-4 area, where the mud shrimp and manila clam coexist, and the mean grain size exhibited a finer particle size, from 5.48 Φ to 5.80 Φ. In the mud shrimp habitat, the sediment mud content was > 60% and the mean grain size was > 5 Φ. Additionally, the mud shrimp only inhabited open coast tidal flat areas. The management of shellfish aquaculture farms by physical methods should be continued based on comparison of the sedimentary environments in the Boryeong and Seongam areas in response to the damage to the mud shrimp habitat.
The embryos of marine bivalves have been commonly used in bioassays for quality assessments of marine environments. Although several standard protocols for the developmental bioassay of bivalves have been proposed, only a few trials for application of these protocols in environmental assessments or for the development of a new protocol with Korean species have been conducted. As such, there is a strong need to establish standard bioassay protocols with bivalves commonly found in Korean waters. To determine the sensitivity of Mytilus galloprovincialis to establish a standard bioassay, their fertilized eggs were exposed to six metals (Ag, Cd, Cr, Cu, Ni, and Zn). The order of biological impact was Ag > Cu > Ni > Zn > Cr > Cd and their lowest observed effective concentration were 5, 16.4, 25.4, 142, 187 and 1,500${\mu}g/l$, respectively. The proportion of normal larvae appeared to decrease linearly with the logarithm of each toxicant concentration within the tested range. The average values of median effective concentrations $(EC_{50})$ from the triplicate experiments for Ag, Cd, Cr, Cu, Ni, and Zn were 6.8, 1,797, 786, 16.6, 68.1, and 139.2${\mu}g/l$, respectively. There was a more than 100-fold difference in $EC_{50}$ values of Cu and Cd. The value of $EC_{50}$ or median lethal concentration of Cu was within the range observed for other bivalve developmental bioassays. The overall sensitivity of M. galloprovincialis in the present developmental bioassay was also similar to that of other marine organisms commonly used in aquatic bioassays (e.g. sea urchins, oysters). Hence, the bioassay using the embryo of M. galloprovincialis is considered to be a useful tool to monitor and evaluate the quality of marine aquatic environments.
It is very important to differentiate of DNA derived from live or dead bacteria within mixed microbial communities in aquatic environments. Ethidium monoazide (EMA) is a DNA intercalating agent and the treatment of EMA with strong visible light cleaves the genomic DNA of bacteria. In dead bacterial cells, EMA intercalates into the genomic DNA, induces the cleavage of DNA, and inhibits the PCR amplification. In this study, we developed the EMA-PCR and EMA real-time PCR to detect the DNA derived from viable Escherichia coli (E.coli) in mixed cultures of live and dead E.coli. The treatment of EMA, $50{\mu}g/mL$, and 650 W visible halogen light exposure for 2 minutes cleaved the genomic DNA derived from heat killed E.coli but did not those of live E.coli. EMA-PCR could detect the DNA from live E.coli in mixed culture samples of live and dead E.coli at various ratio and there was no DNA amplification in only dead E.coli cultures. Similar results were observed in EMA real-time PCR. Further studies are needed to develop various EMA-PCR methods to detect viable waterborne pathogens such as Helicobacter pylori, Giardia lamblia, and so on.
Concentrations of perfluoroalkyl substances (PFASs) were measured in seawater, sediment, and biota collected from Gunsan coast, Korea to investigate their occurrence, distribution, and risk of exposure to humans through seafood consumption. The total concentrations of PFASs in seawater, sediment, and biota ranged from 5.97 to 74.9 ng/L, 0.01 to 13.3 ng/g dry weight, and 0.02 to 5.73 ng/g wet weight, respectively. Predominant PFAS compounds differed across matrices, indicating that the distribution of PFASs in multiple environmental samples is governed by their carbon-chain length. The concentrations of PFASs in seawater were significantly negatively correlated with salinity (P<0.01), suggesting terrestrial input (including land-used PFASs) as the major source of PFAS contamination in coastal environments. The estimated daily intakes (EDIs) of perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) through seafood consumption were 0.05 and 0.06 ng/kg body weight/day, respectively. The EDIs of PFOA and PFOS measured in this study were lower those the proposed by the United States Environmental Protection Agency and Canada guidelines, indicating limited health risk for Korean population from PFASs through consumption of seafood from Gunsan coastal environment.
We investigated the physio-chemical and geochemical parameters in the spraying shellfish aquacultures (Yeoja and Gangjin Bay) to establish the systematic strategy for effective environmental management. Spatial variation of each parameter showed partially significant difference (P<0.05) between Yeoja and Ganjin Bay, inferring the discriminative progress (i.e., accumulation and degradation) of the autochthonous organic matter within the aquaculture environments. We additionally integrated various properties (e.g., water/sediment quality, natural hazard, and biological health) which may affect the biological growth within the aquaculture habitats based on the biogeochemical cycles related to environmental components and aquaculture species. We used a screening approach (i.e., one out-all out; OOAO) which can permit the assessment of the health levels of aquaculture species, the scoring for other parameters (seawater, sediment, and natural hazard) as three levels (excellent, moderate and poor) depending on the complex interactive properties occurring in the aquaculture environments. Actual, discriminative scores obtained via our case studies may confirm that these stepwise processes are effectively evaluated for optimal health conditions within the aquaculture habitats. Thus, this approach may provide valuable insights for effective environmental management and sustainable growth of aquaculture operation.
Plesiomonas shigelloides distributed in the aquatic systems was isolated and identified in this study and compared with the c1inica] isolates in view of their physiological characteristics, Biochemica] charactristics of the isolates of P. shigelloides one sample taken from Gupo and two samples taken from Mu]gum, were studied. However, none was isolated in Haeundae, Dadaepo, Kangdong and Nakdong estuary. The isolated bacteria had an optimum growth condition in peptone water of $25~35^{\circ}C$, pH 7.5-8.0 and 1% NaCI concentration. The cell grew most properly on the selective enrichment media which were made from adding inositol to peptone water. DNase was s]owly produced and the results were different from those of other studies. The components of the fatty acid were 3% of 3-hydroxy]ated fatty acid containing $C_{12}~C_{18}$. 0-10% cyclopropane ($C_{17:0}$), 25~30% hexadecanoic acid ($C_{16:0}$), 32~43% hexadecenoic acid ($C_{16:1}$), 1~2% octadecanoic acid ($C_{16:0}$), and 9~14% octadecenoic acid ($C_{18:1}$). Bacterio]ogica] characteristics, susceptibility of antibiotics, and the components of fatty acid of the c1inica] isolates were similar to those of the strains isolated from the aquatic systems. The strains isolated from c1inica] sources degraded lactose more fast than those isolated from the aquatic systems. There existed resistant bacteria to chlorampenicol in the strains from patients, but there were no resistant bacteria in the strains from the aquatic systems. The components of fatty acid of the clinical isolates were 0~2% $C_{17:0 cyclo}$ and 2~3% $C_{18:0}$, but those of the strains from the aquatic systems were 2~10% and 1~2%, respectvely, which showed the quantitative difference between both components.
