  • This study was conducted from 1991 to 1993 to develop a fungicidal spray program for the apples exportable to the United States of America, in which quarantine and pesticide residue in agricultural products are strictly regulated. In 1991, 2 spray schedules were applied to an orchard, in one of which the 7 fungicides registered for apple both in Korea and U.S. were used, and in the other of which the Bordeaux mixture for which the tolerance was exempted in U.S. was used 2 times along with those 7 fungicides. The apple white rot and fruit infection by Alternaria mali were not effectively controlled by the 7 fungicides alone; however, the control efficacy was raised by adding the Bordeaux mixture to the spray schedule. In 1992, 4 spray schedules were applied in which the kinds of fungicides and spray intervals were different one another. The results suggested that an effective spray program can be developed by adopting the Bordeaux mixture for 3 times or adopting the imminoctadine-triacetate which has not tolerance level in U.S. for same times in the vulnerable stage of apple white rot and alternaria blotch. In spite of the high efficacies against major apple diseases, the Bordeaux mixture could not be recommended to the apple growers due to the various defects such as restrictions in compatibility with insecticides and acaricides, troublesomeness in preparation and spray, especially the harmful effect on the finish of Fuji apples. In 1993, a spray program adopting 3 times of imminoctadine-triacetate during the growing season of apple was developed, which not only can effectively control the major apple diseases but also avoid the pesticide residue problems if it was sprayed 2 or 3 times after bagging. On the basis of the 3 years results, a basic fungicide spray program was formulated in which 1~3 times of imminoctadine-triacetate and 1~2 times of bitertanol were adopted in addition of the 7 common fungicides registered in both countries. In the results of application of the spray schedule to the actual farming in the 4 areas of Kyungpook Province in 1994, no noticeable defects were detected at the first year trial. However, this spray program will be continuously evaluated and modified to obtain better control efficacies against major apple diseases.

  • The applicability of chitosan, a biodegradable natural polymer, as the coating material to prevent the dispersal of the spores of the apple white rot agent Botryosphaeria dothidea, was investigated. The physical properties of mixed chitosan/polyvinyl alcohol(PVA) film showed the increased physical properties for film formation, such as tensil strength, elongation, and viscosity, compared to either chitosan or PVA film. The FT-IR spectra of chitosan/PVA film indicated that the film was formed by simple blending not by any new synthetic bond. The chitosan and chitosan/PVA film showed effective antifungal activity on B. dothidea. The formed films were well decomposed by ASTM strains used for biodegradability test, on the other hand, the PVA film could not be decomposed by above standard strains. The field test at apple orchard showed that the dispersal of B. dothidea spores could be effectively reduced by coated film, especially by chitosan/PVA film. The spore dispersal was reduced upto 97.0% by 1.0% chitosan/5.0% PVA film during 4 months.

  • Cytochalasin E (CE) is a secondary metabolite secreted by Rosellinia necatrix, caused by white root rot, and has toxicity to apple as a toxin during disease progress. This study was conducted to demonstrate the relationship between the production of CE and its pathogenicity. CE producing isolates and non-producing isolates of R. nectatrix were isolated from the mycerial mat of diseased roots and was detected on that using a TLC and HPLC analysis and in vivo pathogenicity test. CE non-producing isolates were not pathogenic to apple roots and not detected CE by TLC and HPLC analysis. It was shown that the production of CE was related to the pathogenicity of R. nectatrix.

  • Two different pest control programs were applied on 8-year-old 'Ryoka'/M.26 apple trees (Malus domestica Borkh.). Lime sulfur or Bordeaux mixture with emulsified oil were applied 12 times from late March to mid-September as organic treatment, and synthetic chemicals were 7 times applied as control treatment. Over the entire apple-growing season, photosynthesis rates of apple trees were significantly lower in the organic treatment than in the control, and this photosynthetic differences were larger in July and August. Photosynthesis-related parameters such as stomatal conductance and transpiration behaved similarly to photosynthesis. The leaf area in the organic treatment was significantly smaller ($24.7cm^2$) than that in the control treatment ($30.7cm^2$). Organic leaves contained significantly less Chl. a ($15.5mg{\cdot}g^{-1}$) than did control leaves ($17.6mg{\cdot}g^{-1}$). Fruit yield per tree was significantly lower in the organic treatment (18.8 kg) than in the control (24.5 kg), because organic fruits experienced a higher rate of disease infection such as white rot (Botryosphaeria dothidae) and bitter rot (Glomerella cingulata) than did control fruits. Organic fruits had high flesh firmness but less color development (lower Hunter's a values). In this experiment, the pest control program with frequent applications of organic fungicides showed negative effects on photosynthesis and disease infection on leaves and fruits, and thus reduce the fruit quality and yield in 'Ryoka'/M.26 apple trees.

  • Airborne and waterborne ascospores and conidia of Botryosphaeria dothidea were collected in apple orchards at Suwon and Chunan in 1992 through 1994. Both waterborne and airborne spores were first detected in mid April to early May. Thereafter, spores were abundant in early June to late August and present until early December. Rainwater collections contained much more conidia than ascospores during the apple growing seasons. Airborne ascospores catches, which were also detected on humid days without measurable rainfall, were much more than airborne conidia catches. High amounts of ascospores were detected in early times of apple growing season, whereas most conidia catches occurred in later times of the season. The number of waterborne conidia and airborne ascospores was positively correlated with mean daily maximum, minimum, and average air temperatures during the trapping periods (p=0.01). However, no significant correlation was found between the number of spores and the total precipitation during the trapping periods.

  • Our study investigated the available chlorine content, contact time and difference among strains of each pathogen for sodium hypochlorite (NaOCl) to control chemically against soil-borne fungal pathogens, such as Phytophthora rot by Phytophthora cactorum, violet root rot by Helicobasidium mompa, and white root rot by Rosellinia necatrix, causing die-back symptom on apple trees. As a result, the colony growth of Phytophthora cactorum was inhibited completely by soaking over 5 s in 31.25 ml/l available chlorine content of NaOCl. Those of H. mompa and R. necatrix were inhibited entirely by soaking over 160 s in 62.5 and 125 ml/l available chlorine content in NaOCl, respectively. Also, inhibition effect on available chlorine in NaOCl among strains of each soil-borne pathogen showed no significant difference and was similar to or better than that of fungicides.

  • A basic spray program for apple in which fungicides are scheduled to spray at 15-day interval from petal fall to late August was formulated on the properties of several selected fungicides. In order to improve it, experimental plots, completely randomized block with 3 replications, were prepared in an orchard of 15 years old Fuji cultivar, and the spray programs in which only one chemical in the basic spray program was substituted with others were applied to each plot. It was revealed that only single substitution of the fungicide in the basic spray program makes a great differences in the control of white rot and bitter rot, and that the control property of the fungicides against the two diseases was quite variable even by the time of application. A simila! ! r trial was conducted in 2002 with a new basic spray program that was formulated with fungicides that have shown best control in each spraying time in the previous trial, similar results were obtained. Applying this method, the usefulness of certain fungicide in the spray program for apple could be properly assessed. Anthracnose of Robinia pseudo-acacia L. caused by Collectotrichum spp.

  • Laccase activity of Pleurotus ostreatus is significantly increased by the addition of apple pomace. Among various conditions, the best concentration of apple pomace and cultivation time for the production of laccase by P. ostreatus was 2.5% and 9 days, respectively. Reverse transcription polymerase chain reaction analyses of laccase isoenzyme genes, including pox1, pox3, pox4, poxc, poxa3, and poxa1b, revealed a clear effect of apple pomace on transcription induction. Our findings reveal that the use of apple pomace can be a model for the valuable addition of similar wastes and for the development of a solid-state fermenter and commercial production of oyster mushroom P. ostreatus.
