Superpixel-based Apple Leaf Disease Classification using Convolutional Neural Network (합성곱 신경망을 이용하는 수퍼픽셀 기반 사과잎 병충해의 분류)

  • The classification of plant diseases by images captured by a camera sensor has been studied over past decades. A method that has gained much interest is to use image segmentation, from which statistical features are derived and analyzed by machine learning. Recently, deep learning has been adopted in this area. However, image segmentation is still a difficult task to achieve stable performance due to a variety of environmental variations. The end-to-end learning in neural network has a demerit that train images may be different from real images acquired in outdoor fields. To solve these problems, we propose superpixel-based disease classification method using end-to-end CNN (convolutional neural network) learning. Based on experiments performed on PlantVillage apple images, the classification accuracy is 98.29% and 92.43% for full-image and superpixel. As well, the multivariate F1-score is (0.98, 0.93). Therefore we validate that the method of using superpixel is comparable to that of full-image.

Cross-tolerance of Alternaria mali to Various Fungicides (사과 반점낙엽병균(斑點落葉病菌)의 각종(各種) 살균제(殺菌劑)에 대한 교차내성(交差耐性))

  • Of the 1,200 single spore isolates of Alternaria mali causing apple leaf spot and fruit decay, the mycelial colonies showed 76, 96 and 15% growth at 100,000 ${\mu}g/ml$ of captafol, chlorothalonil and folpet, respectively, and five and three percent growth at 10,000 ${\mu}g/ml$ of iprodione and polyoxin. These isolates showed cross-tolerance of mycelial growth, spore formation, or spore germination to the above five fungicides plus captan, garbenda, and thiophanate methyl. Captafol, mancozeb, polydong, and propineb did not show any cross-tolerance of spore germination since no spore was germinated at 100 ${\mu}g/ml$ which was much lower than the concentration used by apple growers. Benomyl, fenarimol, oxidong, and triademefon showed medium degree of cross-tolerance of the fungal growth, sporulation, and spore germination.

Screening of Functional Materials from Solvent Fractions of Apple Flower Leaf Extract (사과꽃잎 추출물의 용매 분획으로부터 기능성 소재의 탐색)

  • Fractional solvent extraction by organic solvents such as hexane, chloroform, ethylacetate, and butanol was carried out using 70% ethanol extract of apple flower leaves. Biological activities including antioxidant, whitening, antimicrobial and anti-wrinkle activities were investigated and bio-active materials of the extracts were identified using GC/MSD. Among the tested solvent fractions, ethylacetate fraction showed the highest total polyphenol content (1218.94 ${\mu}g/mL$), and flavonoid (140 ${\mu}g/mL$). The DPPH radical scavenging activities was over 80% at a dry matterbased concentration of 200 ${\mu}g/{\mu}L$ and SOD-like activity was over 90% at 50 ${\mu}g/mL$ concentration in ethylacetate fraction that was slightly lower than of ascorbic aicd. Tyrosinase inhibition activity related to skin-whitening was over 60% by ethylacetate fraction of 100 ${\mu}g/mL$. As an anti-aging effect, elastase inhibitory activity was about 45% in ethylacetate fraction. Also, it showed a significantly antimicrobial activity against P. acenes. From GC/MSD analysis, a characteristic peak of high content in ethylacetate fraction was identified as kaempferol, which has been reported as a bioactive compound.

Carbamoyl Phosphate Synthase Subunit CgCPS1 Is Necessary for Virulence and to Regulate Stress Tolerance in Colletotrichum gloeosporioides

  • Glomerella leaf spot (GLS) is a severe infectious disease of apple whose infective area is growing gradually and thus poses a huge economic threat to the world. Different species of Colletotrichum including Colletotrichum gloeosporioides are responsible for GLS. For efficient GLS control, it is important to understand the mechanism by which the cruciferous crops and C. gloeosporioides interact. Arginine is among one of the several types of amino acids, which plays crucial role in biochemical and physiological functions of fungi. The arginine biosynthesis pathway involved in virulence among plant pathogenic fungi is poorly understood. In this study, CgCPS1 gene encoding carbamoyl phosphate synthase involved in arginine biosynthesis has been identified and inactivated experimentally. To assess the effects of CgCPS1, we knocked out CgCPS1 in C. gloeosporioides and evaluated its effects on virulence and stress tolerance. The results showed that deletion of CgCPS1 resulted in loss of pathogenicity. The ∆cgcps1 mutants showed slow growth rate, defects in appressorium formation and failed to develop lesions on apple leaves and fruits leading to loss of virulence while complementation strain (CgCPS1-C) fully restored its pathogenicity. Furthermore, mutant strains showed extreme sensitivity to high osmotic stress displaying that CgCPS1 plays a vital role in stress response. These findings suggest that CgCPS1 is major factor that mediates pathogenicity in C. gloeosporioides by encoding carbamoyl phosphate that is involved in arginine biosynthesis and conferring virulence in C. gloeosporioides.

Influence of Water Stress through Root Pruning on Tree Growth and Fruit Quality in 'Fuji'/M.9 Apple Tree at Full Bloom (만개기 단근처리를 통한 수분스트레스가 '후지'/M.9 사과나무의 수체반응 및 과실특성에 미치는 영향)

  • This study was conducted to find out the influence of water stress from May to June on photosynthesis, shoot growth and fruit quality of apple trees. Fuji'/M.9 apple trees were root pruned at full bloom (30th April), parallel to the row of 30 cm or 60 cm from the trunk, to one side or both sides of trunk. The average value of afternoon leaf water potential from May to June of the no root pruning was maintained over -1.80 MPa, but that of root-pruning treatments was maintained under -1.80 MPa. In the comparison of average value of leaf water potential from May to June by root-pruning degree, the root pruning both sides at 30 cm from trunk was lowest (-2.06 MPa), followed by the root pruning both sides at 60 cm (-2.02 MPa) and the root pruning one side at 30 cm (-1.91 MPa). Root pruning one side at 30 cm or both sides at 60 cm reduced photosynthetic rate from May to June by 80% or 65%, respectively, compared with no root pruning. So, the degree of decreasing total shoot length or average fruit weight compare to the no root pruning of the root pruning one side at 30 cm was 25% or 11%, respectively, and those of the root pruning both sides at 30 cm was 45% or 15%, respectively. However, the soluble solid content and fruit color of the root pruning was higher than those of the no root pruning, and those of the root pruning both sides was higher than root pruning one side. These results indicate that the fruit enlargement was decreased when the average value of average leaf water potential from May to June was under -1.80 MPa, and the shoot growth was decreased when that was under -2.00 MPa.

Effect of Artificial Light Sources on the Growth of Apple Rootstock M.9 Seedling (인공광원이 사과 대목 M.9 묘 생육에 미치는 영향)

  • To investigate the effect of light sources on the growth and photosynthesis of the dwarf apple rootstock M.9 for the production of standard seedlings, the plants were cultivated in a controlled environment for 6 weeks. The sources of light are six treatments [Red (R), Blue (B), White (W), RBUV (R7B3 containing UV-A), RBW (R3B1W1), SMF (high pressure sodium + metal halide + fluorescent lamp) under $154{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$. Growth characteristics of apple seedlings varied depending on artificial light source at 3 weeks and 6 weeks. The plant height of apple seedling was high in the R, RBUV, RBW, and SMF light sources at 3 weeks, and in the R light at 6 weeks. There was no significant difference on stem diameter among the treatments at 3 weeks, but showed high in RBUV and RBW light at 6 weeks. Leaf number was the highest in RBUV light at 3 and 6 weeks. The chlorophyll content (SPAD value) was high in the B and RBUV light at 3 weeks, but it was not significant at 6 weeks. The growth rate to height of the R light (1.12mm/day) was the highest among the treatments, followed by RBUV, RBW, SMF, W and then B. Leaf area was the highest in RBUV and RBW lowest in B. Specific leaf area was high in W and fresh and dry weight were high in RBUV. The photosynthetic rate at 6 weeks was highest in the B and lowest in the R. Stomatal conductivity and transpiration rate were higher in the B and W compared to the other light sources. Therefore, we are considered that light sources for growth of dwarf apple rootstock M.9 seedlings are suitable the R, RBUV, and RBW light sources with a high mixing ratio of Red and Red +Blue.

Studies on Mechanism and Damage Occurrence for Foliar Spray of NaCl Solutions in Fruit Trees (NaCl 엽면살포에 따른 과수의 피해양상과 기작에 관한 연구)

  • We investigated the mechanism on occurring of briny injury in four fruit species (apple, pear, grape, peach). Briny injury of apple, pear, peach, and grape was induced by the foliar splay of saline solutions. Browning symptom was initiated one hour after the spray of 3% NaCl in apple, pearl and peach trees. while two hour were needed for grape, but the latter developed the symptom faster than the formers. Foliar spray of 3% NaCl resulted in 4-fold increase in ethylene production 24 h after the spray, and the production was returned to early stage rate after 72 h. Ethylene production of control was nearly constant with slight decrease after 4 h. Browning and defoliation response to the spray of 3% NaCl on lower side were not different from the spray on both sides regardless of fruit species, while no injury symptoms occurred by the spiny on upper side. In saline damage for treated date, lateral bud was only germinated in apple at 30 July. A shoot apex was 100% grew in all fruit tree. In result for hormonal content of leaf and shoot apex, control was high t-zeatin content in leaf. However, IAA and ABA content was increasing in high saline concentration.

First Report of Apricot pseudo-chlorotic leaf spot virus Infecting Peach Trees in South Korea (복숭아나무에서 검출된 Apricot pseudo-chlorotic leaf spot virus의 국내 첫 보고)

  • In May 2016, 24 peach samples showing abnormal and virus like symptoms were collected in one of major peach producing area, Yeongcheon-si, Gyeongsangbuk-do, Korea. We performed RT-PCR diagnosis for confirmation of viral infection. The diagnostic targets are 17 species of viruses and viroids that quarantine and high risk pathogens when it occur. As a results, seven species of viruses and viroids, including an unreported (Apricot pseudo-chlorotic leaf spot virus, APCLSV) and a quarantine (Peach latent mosaic viroid, PLMVd) species in Korea, were detected. For the sequence analysis of unreported virus, APCLSV, the sequence of coat protein gene were amplified and cloned. The sequence showed 97% nucleotide identity with other APCLSV isolates and compared with other seven species of reported Trichoviruses. This virus was classified as APCLSV based on the sequence and phylogenetic analysis. This isolate was named Yeongcheon. As patterns of APCLSV occurrence, all samples that APCLSV detected were co-infected with Apple chlorotic leaf spot virus (ACLSV). As properties of ACLSV, APCLSV has high possibility of wide spread disease in fruit tree farms in Korea. Therefore, it is necessary to do related researches, such as infection route and influence of disease in commercial orchards.

A Study on Food Attitude and Dietary Habits of Pre-school Children (취학전(就學前) 아동(兒童)의 식품기호(食品嗜好)와 식습관(食習慣) 조사연구(調査硏究))

  • A group of 132 pre-school children with a mean age of six years and one month was the subjects of this investigation. These children were members of three nursery schools locating in Kwang-ju city. Each child's mother replied to a questionaire which included 42 food items and 4 questions. The results showed that the food preference of each child was variable and the group as a whole expressed more positive than negative feelings. The average number of foods liked per child was 22.9(range: $7{\sim}42$) among 42 food items. Peach, Apple, Strawberry, Laver, Egg, Tomato, Sweet Potato, Tangerine Orange and Beef were ranked in descending order of 'like' ratings. The average number of foods refused was 4.9 (range: $0{\sim}25$). Onion, Liver, Red Pepper Leaf, Root of Bellflower, Taro, Wild Seasame Leaf, Egg Plant, Cabbage, Water Cress and Bracken were ranked in descending order of 'refuse' ratings. The odor of hurting feelings, the hot taste and the flabby texture were the reasons why the children refused these foods. The average number of foods unexperienced was 4.4 (range: $0{\sim}14$). Kidney, Dried Small Whitebail, Tangle (DA SHI MA), Liver, SA RI Mushroom, Acorn Mook, Dried Fragrant Mushroom, Pine Agaric, Root of Bellflower and Ped Pepper Leaf were ranked in descending order of 'unexperience' ratings. Children's 'like' rating toward total foods was 57.3% and the 'refuse' rating was 11.7%. Among eight food groups, fruits showed the highest favority (91.4%) and mushroom showed the least (25.9%). The difference between male and female in the preference of total foods did not showed significant level. But the difference were significant in those of individual food groups; other vegetables, green and yellow leafy vegetables, mushrooms(above, p.<01), fishes and shellfish and sea weeds (above, p<.05). Children who had experienced meals missed rated 59.1% and 34.1% of these children missed meals once a week. The main reason for maels missed was due to the heavey snacks before meal time. Children who had snacks twice a day rated 45.6%. Main foods used as their snacks were starch foods (Cake, Biscuit, Sweet Potato) and citrous foods (Fruits, Apple, Tangerine).

Identification of Plant Viruses Infecting Pear Using RNA Sequencing

  • Asian pear (Pyrus pyrifolia) is a widely cultivated and commercially important fruit crop, which is occasionally subject to severe economic losses due to latent viral infections. Thus, the aim of the present study was to examine and provide a comprehensive overview of virus populations infecting a major pear cultivar ('Singo') in Korea. From June 2017 to October 2019, leaf samples (n = 110) of pear trees from 35 orchards in five major pear-producing regions were collected and subjected to RNA sequencing. Most virus-associated contigs matched the sequences of known viruses, including apple stem grooving virus (ASGV) and apple stem pitting virus (ASPV). However, some contigs matched the sequences of apple green crinkle-associated virus and cucumber mosaic virus. In addition, three complete or nearly complete genomes were constructed based on transcriptome data and subjected to phylogenetic analyses. Based on the number of virus-associated reads, ASGV and ASPV were identified as the dominant viruses of 'Singo.' The present study describes the virome of a major pear cultivar in Korea, and looks into the diversity of viral communities in this cultivar. This study can provide valuable information on the complexity of genetic variability of viruses infecting pear trees.
