  • The single cell gel electrophoressis(SCGE) assay, also known as the comet assay, is a rapid, simple, visual and sensitive technique for measuring and analysing DNA breakage in mammalian cells. The SCGE or comet assay is a promising test for the detection of DNA damage and repair in individnal cells. It has widespread potential applications in DNA damage and repair studies, genotoxicity testing and biomonitoring. In this microgel electrophoresis technique, cells are embedded in agarose gel on microscope slides, iysed and electrophoresed under alkaline conditions. Cells with increased DNA damage display increased migration of DNA from the nucleus towards the anode. The length of DNA migration indicates the amount of DNA breakage in the cell. The comet assay is also capable of identifying apoptotic cells which contain highly fragmented DNA. Here we review the development of the SCGE assay, existing protocols for the detection and analysis of comets, the relevant underlying principles determining the behaviour of DNA and the potential applications of the technique.

  • Apoptosis is now widely recognized as a common form of cell death and represents mechanism of cell clearance in many physiological situations where deletion of cells is required. In vivo administration of bacterial lipopolysaccharide (LPS) to Balb/c mice induced DNA fragmentation in the thymus. DNA fragmentation in the thymus was roughly dependent on the dose of LPS injected and reached the peak 18 hours after injection. This apoptosis in the thymus might be mediated due to LPS stimulant. DEN (diethylnitrosamine) has been shown to cause liver cancer in experimental animals and humans. The hepatic tumorigenesis was induced by ad libitum feeding of DEN only. It was suggested that DEN induced hepatic tumorgenesis in rat is a good reproducible model for studying biochemical and pathophysiological changes associated with the development of hepatic tumorigenesis and apoptosis.

  • The effect of silkworm hemolymph on insect cell apoptosis was investigated. The addition of silkworm hemolymph into the culture medium either before or during the baculovirus infection increased the host cell longevity. Silkworm hemolymph also inhibits apoptosis induced by actinomycin D, the RNA synthesis inhibitor. In this study, a flow cytometry was used for the quantitative analysis of apoptosis inhibition by silkworm hemolymph.

  • Samryungbaikchul-san(SRBCS) has been used in oriental medicine for the treatments of gastrointestinal and neurological disorders. Here, potential protective function of SRBCS was investigated in neural tissues in Alzheimer's disease(AD) mouse model. In primary cultured cells from the spinal cord of newborn rats, treatment of ${\beta}$-amyloid peptide elevated cell counts positive to glial fibrillary acidic protein(GFAP) or caspase 3 immunoreactivity, but the co-treatment of SRBCS reduced positive cell counts. In vivo administration of scopolamine, an inhibitor of muscarinic receptor, resulted in increases in the number of glial fibrillary acidic protein(GFAP) and caspase 3-positive cells in hippocampal subfields, which was then decreased by the treatment of SRBCS or acetylcholinesterase inhibitor galathamine. The present data suggest that SRBCS may play a protective role in damaged neural tissues caused by scopolamine treatments in mice.

  • Sulforaphane은 브로컬리나 십자화과 채소중의 glucoraphanin의 가수분해 산물인 isothiocyanate로서 이는 detoxification 효소의 phase II를 일으키는 것으로 나타났고 설치류에서 화학적으로 발생된 유선 종양을 억제하고 최근에는 대장암 세포에서 cell cycle arrest와 apoptosis를 일으킨다고 알려져 왔다. 여기서는 SUL이 Human MammaryMCF-7 adenocarcinoma 세포의 증폭을 억제하는 역할을 제시하였다. MCF-7 cell에 15umol/L SUL을 처리하였을 때 G2/M cell cycle이 arrest를 보였고 cyclin B1 protein이 24시간 이내에 증가하였다. 15umol/L의 SUL은 in vivo 상에서 histon Hl의 인산화를 유도하고, 초기 mitosis에서 cell을block하며 mitotic microtuble의 중합화를 방해하였다. In vitro 상에서 정제된 bovine braintubulin에 대한 SUL을 고농도로 투여했을 때, tubulin의 중합율과 총 tubulin 중합도의 억제를 보였다. 덧붙여서, isothiocyanate를 함유하는 SULanalog로 처리된 정제 tubulin도 비슷하게 저해를 받았다. 본 연구는 SUL이 mitotic cell cyclearrest를 포함한 mammary cancer 억제력을 가진 것과, 이러한 기작으로 정상적인 tubulin 중합화및 microtubule dynamic에 한층 효과적인 영향을 준다는 것을 제시하였다.

Effect of Injin Butanol Fraction with Thin Layer Chromatography on Fas-mediated Apoptosis (인진butanol 분획의 TLC추출성분이 Fas-mediated Apoptosis에 미치는 영향)

  • Objective: The aim of this study is to investigate the effect of Injin butanol fractions with Thin Layer Chromatography on Fas-mediated Apoptosis. Method: Injin-butanol fraction separated by TLC. MIT assay, cell cycle analysis, Caspase-3 protease assay, DNA fragmentation assay and quantitative RT-PCR were performed to evaluate the effects of TLC extraction of lnjin-butanol fraction on cell viability, cell cycle progression and apoptosis. Results: Scopoletin, luteolin, apigenin and unknown powder was isolated by TLC. Fas-mediated apoptosis analysis shows that scopoletin has inhibiting function on apoptosis. Caspase- 3 protease assay analysis shows that scopoletin inhibits activity of caspase-3. Quantitative RT-PCR analysis shows that no activity on caspase-3, but apoptosis inhibition cytokine -Bcl-2- is activated, and apoptosis activating cytokine -Bax- is unactivated. Conclusion: These results show that each fraction of Injin-butanol TLC extraction, especially scopoletin, acts as a protective function on liver cell viability, and inhibitory function on apoptosis.   • Xenoestrogens are chemicals with diverse structure that mimic estrogen. Bisphenol A, a monomer of polycarbonate and epoxy resins, has been detected in canned food and human saliva. Bisphenol A stimulate cell proliferation and induce expression of estrogen -response genes in vitro. The purpose of the this study was to evaluate cell proliferation of bisphenol A in the presence of a rat liver 59 mix contaning cytochrome P450 enzymes and Cu (II). The fragmentation of intact DNA, a parameter of apoptotic cell death, was evaluated quantitatively by diphenylamine reaction method. Bisphenol A induced apoptotic cell death in a dose-dependent manner The effect of radical scavenger on the apoptotic cell death induced bisphenol A was investigated. The DNA fragmentation induced by bisphenol A was significantly inhibited by addition of radical scavenger to the culture medium. This indicated that elevated oxidative stress caused by imbalance between the production and removal of free radicals occurred in cells. Taken together, these results suggest that free radical reacts with Cu (II) leading oxidative stress.

Effect of Glycyrrhizin on the Aoptosis of Melanoma Cells in Mel/ret Transgenic Mice (Glycyrrhizin이 Mel/ret transgenic mice에서의 melanoma 세포의 apoptosis에 미치는 영향)

  • The effect of glycyrrhizin on melanoma cells was investigated. DNA fragmentation in cultured melanoma cells was promoted by the addition of glycyrrhizin in a dose dependent manner. Administration(i.m.) of glycyrrhizin to Mel/ret transgenic mice resulted in apoptosis induction, reduction of mitochondrial transmembrane potential in melanoma cells. Decreased B220+ B cells were recovered by the treatment of glycyrrhizin in splenocytes and mesenteric lymph node cells, while Thy-1+ T cells were not influenced. Results suggested that glycyrrhizin acted as an inducer of apoptosis of melanoma cells and an immuno-potentiator via recovered B lymphocyte population in Mel/ret transgenic mice.
