Cuticular waxes on tea (Camellia sinensis L.) loaves consisted mainly of alkanes, fatty acids, primary alcohols, triterpenes, and a group of unknown compounds, dominated by primary alcohols and triterpenes. Tea tree accessions used in this study were M-1, M-2, Sakimidori, and Yabukita. For all accessions, the alkane, fatty acid, and primary alcohol constituents consisted of a homologues series, and the major constituents of primary alcohol class were the C28 and C30 homologues. Triterpenes consisted of friedelin, $\beta-amyrin$, and three unidentified ones and friedelin was the most abundant. Leaf area and the total amounts of cuticular waxes per leaf increased with lower leaf position from the apical bud in Yabukita variety. With different leaf position, total wax amount per unit leaf area on the youngest leaves of P1 (the uppermost leaf position) showed the largest amount $(12.80{\mu}g/cm^2)$, and on mature loaves of P2 to P6 ranged from 7.08 to $7.77{\mu}g/cm^2$, and then on the oldest loaves of P7 (the lowest leaf position) remained at an increased level $(17.53{\mu}g/cm^2)$. During leaf development (lower leaf position), the amount of primary alcohols decreased from P1 to P6 and increased at P7, whereas that of triterpenes increased from P1 to P7. The percentage of each wax class in the total wax amount occurred a decrease in primary alcohol and an increase in triterpene, with leaf age.
This experiment was carried in order to compare some propagation methods and the effective components in the root of Paeonia albiflora pallas, Paeonia moutan Sims and Paeonia albiflora pallas root grafted with Paeonia mouton sims The optimum seeding time of August 20 to september 10 for increasing germination ratio. The growing and developing of Paeonia albiflora Pall. for root dividing methods was advantageous to practice at Autum than at spring season. The shoot cutting method during the flowering time was proper to increase the rooting ratio with the aid of Ruton, the plant growth regulator but not emerged of apical bud. The grafting stages between the Paeonia albiflora pall. and Paeonia moutan Sims was available to practice during the September for increasing the thickness growth of root. Among the effective components of the paeony the paeoniflorin was shown high, while paeonol in paeony moutan. The paeoniflorin of herbaceous paeony / paeony moutan was increased about 75.9% than herbaceous paeony alone, and the other compoents are similar to those of herbaceous paeony root.
This study was conducted to determine the effect of a light-emitting diode (LED) on in vitro shoot growth and rooting in teak (Tectona grandis L.). In the experiments with apical bud explants, the greatest shoot elongation (3.2 cm) occurred when they were cultured on DKW medium under 50% blue and 50% red LED mixture (BR), whereas no differences in growth were observed in different light sources (florescent light [F] or BR) or media (MS or DKW). The highest number of shoot multiplication (2.4/explant) or elongation (4.94 cm) was achieved with 0.5 or 1.0 mg/L 6-Benzyladenine (BA) treatment under BR. In addition, the best rooting rate (93.8%) or root length (1.3 cm) was recorded with 0.5 mg/L indole-3-butyric acid (IBA) treatment under BR, and the highest root induction (3.1/explant) was observed in 0.2 mg/L IBA under BR. The in vitro rooted plantlets were hardened and survived well on soil.
Somatic embryos or adventitious buds were formed from the segments of zygotic embryo of Lycium chinense Mill. on Murashige and Skoog (MS) medium supplemented with auxins (2,4-D, NAA, IAA) and / or cytokinins (zeatin, kinetin, BAP). Embryogenic callus formed on MS medium containing 0.5 mg/L 2,4-D and then differentiated into somatic embryos without transfer to hormone free medium. On the other hand, adventitious buds were formed on medium with 0.01 mg/L 2,4-D and 0.5 mg/L zeatin. Among various parts of zygotic embryos, the morphogenic potential was higher in the cotyledonary region, and the most organogenic potential was found in cotyledon followed by radicle, hypocotyl, and whole embryo. Histologically, bipolar structure of the heart-shaped embryos were confirmed and in adventitious buds only shoot apical meristems were found to exist.
This study aimed to know difference in freezing resistance among different clonal seedlings or different seed source seedlings of Cryptomeria japonica which has been selected where extreme cold prevails in Korea and Japan. The freezing resistance of three 12-50 year old trees was also measured in the experiment. The freezing resistance was measured in different tissue parts: mainly leaf, cambiam and xylem, at three different collection dates in two different collection places during the winter of 1977-1978. The following results and discussions were made: 1. The clonal difference in freezing resistance of Cryptomeria japonica was $9^{\circ}$ to $15^{\circ}C$ in maximum according to the collection place. However, the clonal difference in freezing resistance was not related to the difference in climatic conditions where the parent tree have been growing. This impiled that the natural selection of cold resistant genes in Cryptomeria japonica has not reached its evolutional equilibrium yet since most of the Cryptomeria forest has been established by artificial regeneration. 2. The difference in freezing resistance among leaf, cambium and xylem was not apparent except that leaf of several clones showed higher freezing resistance than cambium or xylem when they collected at mid-winter. The least freezing resistant tissue part, thought its freezing resistance was not measure in all clones and all temperatures were appeared in the apical buds. The new shoot growth was observed in the next spring with being replaced by its dormant or adventitious bud growth when the apical bud was injured dy cold during winter. 3. The freezing resistance of leaf, cambium and xylem was shown high enough so that freezing resistance Cryptomeria clones in this experiment were supposed to be able to survive in cold winter conditions at the middle part of Korea. However, it was reported that the most susceptible tissue part to winter injury was the basal stem, but of which freezing resistance was not-measured in this experiment. Several silvicultural methods for prevention of Cryptomeria seedlings from cold damage were discussed in literature.
This study was aimed to mutant induction in Gentiana scabra BUNGE through the tissue culture techniques and comparison of gentiopicroside content in each mutant plants derived from tissue culture. In order to induce the mutants of Gentiam scabra, seed, apical and lateral bud were soaked in $NaN_3$, EMS and MNU solutions and cultured on MS basal medium containing 0.5mg/l NAA, BA $NaN_3$, MNU-treated buds survived about 50% but not survived in EMS treatment. Seed germination rate was extremely low in EMS and MNU treatments but various types of mutant plantlets were obtained by those treatments. 63% of elongated type and 37% of dwarf type were obtained from tissue culture treated by various mutagen. Gentiopicroside contents of regenerated plantlets was analyzed. Root of dwarf type contained more gentiopicroside(1.383%) than that of elongation type (0.083%).
To clarify some ambiguous diagnostic characters of Callicarpa japonica Thunb. and Callicarpa dichotoma (Lour.) K. Koch in Korea (Verbenaceae), the external morphology and micromorphology (leaf, anther, pollen, fruit, in particular endocarp surface and structure) of two taxa are studied and described in detail. It is confirmed that the following characteristics (e.g., the stem outline in a cross-section, the bud shape and length, the corolla tube length and the lobe length, the pattern of the anther dehiscence and length, and the endocarp morphology) are useful for distinguishing these two taxa. In particular, following three characteristics are most useful for an identification: (1) the corolla tube length of C. japonica (2.5-4.6 mm) is longer than that of C. dichotoma (0.7-1.0 mm); (2) the anther is dehiscing by an apical pore at the anthesis of C. japonica, while it is opening by a long fissure from the apex to the base in C. dichotoma; and (3) the edge of the endocarp (lateral view) in C. japonica is concave, while in C. dichotoma it is shown to be flat. On the other hand, the teeth state on the leaf margin and the position of inflorescence are not good diagnostic characteristics for identification. Additionally, the keys for all known Callicarpa taxa in Korea are provided.
As an endeavor to establish a micropropagation system for Epimedium koreanum Nakai., this study was carried out to define methods to disinfect its explants and media for callus induction, proliferation and plant regeneration. The lowest infection rates by fungi or bacteria on apical and axillary bud explants of rhizome were observed when they were immerged in 0.3% NaOCl solution for 20 min after soaked in 0.1% $AgNO_3$ solution for 30 min, but leaf explants were seldom infected with fungi or bacteria by this disinfectant method. The highest rate of plantlet formation was obtained from the explants disinfected in 0.3% NaOCl solution for 20 min after soaked in 0.1% $AgNO_3$ solution for 60 min for tip buds and in 0.1 % $AgNO_3$ solution for 30 min for axillary buds of rhizome. Induction rate of callus was the highest from the explants disinfectd in 0.3% NaOCl solution for 20 min after soaked in 0.2% $AgNO_3$ solution for 15 min. Callus growth was proper in a modified 1/2 MS medium including half strength of $NH_4NO_3$ with $0.02-0.2mg{\cdot}L^{-1}$ BA and $2.0mg{\cdot}L^{-1}$ NAA. Low rate of plantlet regeneration was obtained in 1/2 UM or 1/2 White medium with $2.0mg{\cdot}L^{-1}$ BA and $0.2mg{\cdot}L^{-1}$ AA.

Mammary epithelial cells contain a subpopulation of cells with a large proliferativ potential which are responsible for the maintenance of glandular cellularity and are the progenitor cells of mammary cancer. These clonogens give rise to multicellular clonal alveolar or ductal units(AU or DU) on transplantation and hormonal stimulation. To isolate putative mammary clonogens, enzymatically monodispersed rat mammary epithelial cells from organoid cultures and from intact glands are sorted by flow cytometry according to their affinity for FITC labeled peanut lectin(PNA) and PE labeled anti-Thy-1.1 antibody(Thy-1.1) into four subpopulations : cells negative to both PNA and Thy-1.1(B-), PNA+cells, Thy-1.1+cells, and cells positive to both reagents(B+). The in vivo transplantation assays indicate that the clonogenic fractions of PNA+cells from out-growths of organoids in primary cultures for three days in complete hormone medium(CHM) are significantly higher than those of cells from other subpopulations derived from cultrues or from intact glands. Extracellular matrix(ECM) is a complex of several proteins that regulated cell function ; its role in cell growth and differentiation and tissue-specific gene expression. It can act as a positive as well as a negative regulator of cellular differentiation depending on the cell type and the genes studied. Regulation by ECM is closely interrelated with the action of other regulators of cellular function, such as growth factors and hormones. Matrigel supports the growth and development of several different multicellular colonies from mammary organoids and from monodispersed epithelial cells in culture. Several types of colonies are observed including stellate colonies, duct-like structures, two- and three-dimensional web structures, squamous organoids, and lobulo-duct colonies. Organoids have the greatest proliferative potential and formation of multi-cellular structures. Phase contrast micrographs demonstrate extensive intracellular lipid accumulation within the web structures and some of duct-like colonies. At the immunocytochemical and electron micrograph level, casein proteins are predominantly localized near the apical surface of the cells or in the lumen of duct-like or lobulo-duct colonies. Squamous colonies are comprised of several layers of squamous epithelium surrounding keratin pearls as is typical fo squamous metaplasia(SM). All-trans retinoic acid(RA) inhibits the growth of SM. The frequency of lobulo-ductal colony formation increased with the augmentation of RA concentration in these culture conditions. The current study models could provide powerful tools not only for understanding cell growth and differentiation of epithelial cells, but also for the isolation and characterization of mammary clonogenic stem cells.
