Antitumor Components of Korean Basidiomycetes

  • To find antitumor components in the shake-cultured mycelia of Volvariella bombycina, the mycelia were extracted with hot water. After the extract was dialyzed and freeze-dried, a protein-polysaccharide fraction was obtained and examined for antitumor activity against the solid form of sarcoma 180 in ICR mice. It showed 60.3% inhibition ratio at a dose of 20mg/kg/day for 10 days. It was found to consist of a polysaccharide moiety and a protein moiety. After gel filtration on Sepharose 4B, Fraction B was obtained and showed the highest inhibition ratio of 71.1%. When the antitumor component was examined for immunopotentiating activity, it was found to increase the macrophage accumulation in the peritoneal cavity as well as the antibody production of the spleen cells of the mice.

  • The carpophores of three Korean mushrooms, Coriolus versicolor, Pleurotus ostreatus and Lentinus edodes were respectively extracted with hot water and the extracts were dialyzed through Visking tube. They were found to exert an antitumor activity against sarcoma-180 implanted in mice. Especially, the inhibition ratio of the extract of Coriolus versicolor (100 mg/ Kg.i.p) was almost 100% . But all the extracts did not affect the growth of leukemia L517Y cells in vitro. Therefore these facts indicate that the extracts appear to stimulate cell-mediated immunity.

유산균의 배양물로부터 분리된 물질에 대한 암 유발 ICR Mice의 항암 효과 (Antitumor Activity of CBT-AK5 Purified from Lactobacillus casei against Sarcoma-180 Infected ICR Mice)

  • 최근 많은 관심을 가지는 유산균은 사람이나 동물의 장내 방어 장벽인 위산, 담즙산염, 췌장에 효과가 있고, 이들 생균은 장 점막에 쉽게 집락화하는 특성을 가지면서도 인체에는 무해한 균이기 때문에 식품첨가물 부분에서 GRAS(Generally Recognized As Safe)로 분류되어 있고, 이런 안전성을 기반으로 수많은 제품이 제조되어 유통되고 있다. 유산균의 다양한 연구는 유산균 생균제, 사균체, 유산균 파쇄액을 이용하여 항암 효과, 유당 단백질의 흡수 증진, 콜레스테롤의 저하 류머티스 관절염 치료 및 정장작용 개선에 효과적인 기능을 가진다고 보고하고 있다. 이런 다양한 효과를 가지는 유산균의 기능을 이용하기 위하여 금번 연구에서는 항암 활성이 우수한 Lactobacillus casei의 2차 산물인 배양여액에서 순수 분리한 것으로 UItrafiltration membrane(3, 30 KDa)으로 분리 농축한 단백질 물질인 CBT-AK5을 실험에 사용하였다. 이전 연구로 CBT-AK5가 정상세포와 암세포주를 이용한 세포 독성 실험 및 암세포 생육 억제 활성을 측정한 결과, 정상세포에 대한 세포 독성은 20% 이내로 낮은 세포 독성 효과를 나타난 반면, 각 장기별 암세포주에 대한 암세포 생육 억제 활성을 측정한 결과 70% 이상의 높은 암세포 생육 억제 활성을 나타내어 항암 활성 물질임을 확인하였다. Lactobacillus casei의 2차 산물인 배양여액에서 순수 분리한 CBT-AK5의 항암 효과에 대한 in vivo 실험을 실시하였다. In vivo 실험으로 6~8주령된 ICR mice를 사용하였고, 복수암 유발 및 고형암 유발을 위해 Sarcoma-180 cell line이용하였다. 투여 방법으로는 경구투여와 복강내 투여로 나누어 진행하였고, 각 투여 방법에 따른 치료군과 예방군으로 나누어 실험하였다. 또한 치료군과 예방군마다 다른 농도별($25\;{\mu}g/m{\ell}$, $250\;{\mu}g/m{\ell}$, $2.5\;{\mu}g/m{\ell}$ 차이도 관찰하였다. 위 실험에서 경구투여에 따른 항암 활성 효과는 양성대조군보다 체중 증가와 생존율 비교에 있어서 정상군보다 다소 높게 관찰되었으나, In vitro 실험시 높은 항암 활성 효과에 비해 유의적이지 않았다. 하지만, 복강내 투여에 따른 항암 활성 효과는 양성대조군보다 체중 증가와 생존율 비교에 있어서 높은 활성 효과가 관찰되었고, 치료군과 예방군 모두에서 항암 활성이 우수한 물질임을 확인하였다. 이상의 실험에서 Lactobacillus casei의 2차 산물인 배양여액에서 순수 분리한 CBT-AK5가 항암 활성에 우수한 물질임을 확인하였다.

  • To develop potent anitumor polysac-charides from higher hungi of Korea the alcohol precipitated polysaccharides, which were obtained from hot water extracts of shake-cultured mycelia of eight species of basidiomycetes, were subjected to antitumor screening tests, using sarcoma 180 as a tumor cell. When administered i. p, at the dose level of 20mg/kg/day for ten days into the mice which were implanted with $1{times}10^{8}$ cells of sarcoma 180 twenty-four hour before the start of polysaccharide injection, the polysaccharides of Plutcus cervinus ISA-Pc-1004, Pleurotus pulmonarius, Hydnum repandum ISA-Hr-1006, and Laccaria laccata ISA-LI-1008 respectively showed inhibition ratios of 62.0%, 51.7%, 64.8%, and 57.8%. They were selected for further study, while those of Pancillus serotina ISA-Ps-1012, Laccaria amethystina ISA-La 1001, Pholiota adiposa ISA-Pa-1010, and Lepista spISA-Ls-1007 respectively showed inhibition ratios of -9.3%, 25.3%, 27.3% and 33.0%.

매미눈꽃동충하초(Paecilomyces sinclairii)로부터 추출한 조다당류의 면역 활성과 항암 효과에 관한 연구 (Studies on Immuno-Modulatory and Antitumor Effects of Crude Polysaccharides Extracted from Paecilomyces sinclairii)

  • 매미눈꽃동충하초로부터 중성염용액, 열수 및 메탄올 추출물을 분리하였다. 세포독성 실험 결과, 열수 추출물은 $100{\mu}g/ml$의 농도에서 HT-29에 대해서 세포독성을 나타냈으나, NIH3T3, HepG2, Sarcoma 180에 대해서는 $0{\sim}2,000{\mu}g/ml$의 농도에서 세포독성을 나타내지 않았다. 중성염용액 추출물과 메탄을 추출물에서는 약간의 독성을 나타내었다. Sarcoma 180 복수암에 대한 항암 효과는 중성염용액 추출물과 메탄을 추출물을 투여한 실험군에서 32.3%의 생명 연장 효과를 나타내었다. 매미눈꽃동충하초의 중성염용액 추출물은 대조군에 비해 비장세포를 $2.4{\sim}2.6$배 증가시켰고, B 임파구의 alkaline phosphatase 활성을 $2.7{\sim}3.9$배 증가시킴으로써 비장세포의 증식능과 B 임파구의 면역 활성 효과를 증가시켰다. 대식세포주 RAW 264.7에 $50{\mu}g/ml$의 농도로 중성염용액 추출물을 처리하였을 경우, 양성 대조군이 $79{\mu}M$의 nitric oxide(NO)를 발생시킨 것에 비해 다소 높은 $89{\mu}M$의 NO가 발생되었다. 매미눈꽃동충하초의 중성염용액 추출물이 가장 높은 항암 효과와 B 임파구와 대식세포 활성을 나타내었으며, 따라서 매미눈꽃동충하초 중성염용액 추출물의 항암 효과는 숙주의 면역 기능을 활성화 시킨 것에 기인된 것으로 사료된다.

항암제와 Lactobacillus casei HY2782의 병용투여에 의한 항암효과의 증강 (Augmentation of antitumor activity of antitumor drugs in combination with Lactobacillus casei HY2782)

  • Augmentation of antitumor activity of antitumor drugs in combination with Lactobacillus casei HY2782 (LC2782) was studied against Sarcoma-180 (S-180) and Lewis lung carcinoma (3LL). Antitumor drugs used in this study were 5-fluorouracil (5-fu) and cyclophosphamide (CP). The prolongation effect of LC2872 on the life span of mouse intraperitoneally implanted with S-180 was stronger than that of OK-432 and BCG, while the inhibitory effect of OK-432 and BCG on the growth of 3LL solid tumor was a little stronger than that of LC2782. Average survival rates of mice administrated LC2782, OK-432 and BCG were 192%, 141%, and 112%, respectively, when that of the control was 100%, Intralesional administration of 5-Fu, CP, 5-Fu+LC2782 and CP+LC2782 resulted in 93%, 69%, 99% and 73% inhibition rates against 3LL solid tumor proliferation. The combination therapy of 5-Fu or CP with LC2782 significantly prolonged the life span of S-180-inoculated ICR mice. Average survival rates of mice administrated 5-Fu and CP alone were 115% and 99%. Furthermore, survival rates of mice administrated 5-Fu and CP in combination with LC2782 were 226% and 244%, respectively.

비늘버섯의 성분(成分)에 관한 연구(硏究)(제 2 보) (Studies on the Components of Pholiota squarrosa(II))

  • To investigate antitumor components in Korean higher fungi, the carpophores of Pholiota squarrosa belonging to the family Strophariaceae were collected and extracted with hot water. A protein-bound polysaccharide fraction was obtained by adding ethanol to the extract and by dialyzing through Visking tube. The fraction was examined for antitumor activity against sarcoma 180 implanted in mice. It showed an inhibition ratio of 78.7% at the dose of 20mg/kg/day. The tumor in two of the ten mice was completely regressed. The chemical analysis of the antitumor fraction by Anthrone and Lowry-Folin methods showed that it consisted of 42% polysaccharide and 55% protein. The enzyme fraction of the carpophores showed no proteolytic activity on casein.

  • To elucidate action mechanism of lyophyllan A, an antitumor polysaccharide of Lyophyllum decastes, its immunological activities were examined. Lyophyllan A increased significantly the weights of spleen and liver of mice. Lyophyllan A also restored the decreased thymic weight in tumor-bearing mice. It did not show any direct cytotoxicity against tumor cells, but showed immunopotentiating activities by increasing the number of the plaques in hemolytic plaque assays. Lyophyllan A increased the number of peritoneal exudate cells (PEC) and inhibited the growth of sarcoma 180 mixed with PEC. Moreover the macrophages from lyophyllan A-treated mice exhibited a strong cytotoxic activity towards L5178Y target cells.

  • A protein-polysaccharide fraction was isolated from the mycelial culture of a Korean basidiomycetous fungus, Hebeloma crustuliniforme, and was named HCA (Hebeloma crustulineforme fraction A). When intraperitoneally injected at 200 mg/kg/day once daily for 10 days, HCA inhibited the growth of sarcoma 180 solid tumor in ICR mice by 65.1% (p<0.05). HCA also exerted protective effect against leukopenia induced by cyclophosphamide with the protection ratio of 39.7% (p<0.01).

Comparison of Lectin from Pseudixus japonicus and Concanavalin a on Lymphocytes Proliferation and Cytotoxicity

  • Pseudixus japonicus agglutinin (PJA) was isolated. And its characteristics were compared with those of concanavalin A (Con A). PJA is a glycopritein composed of 49.3% carbohydrate and 50.7% protein which had relatively high percentages of glutamic acid, aspartic acid and phenylalanine residues. The hemagglutinating activity of PJA was approximately one-eighth of that of Con A when tested with mouse crythrocytes. PJA failed to simulate the proliferation or transformation of human and mouse lymphocytes in contratst to Con A. PJA and Con A showed cytotoxicities against SNU-1 (human stomach cancer cells), SNU-CI (human colon cancer cells) and mouse Sarcoma 180 cells when tested by 3-(4, 5-dimethyl thiazol-2-yl)2. 5-diphenyl tetrazolium bromide (MIT) colorimetric assay. The antitumor activity of the lectin in vivo was also tested in Sarcoma 180 bearing mice. There was no significant difference in prologation of lifc span of the mice after the treatment with PJA and Con A for 10 consecutive days.
