애기땅빈대의 항산화 활성 성분 (Antioxidative Constituents from the Whole Plants of Euphorbia supina)

  • Eight compounds were isolated from the EtOAc soluble fraction of Euphorbia supina MeOH extract as the radical scavengers for antioxidant activity. Their structures were identified as kaempferol (1), quercetin (2), juglanin (3), avicularin (4), astragalin (5), isoquercitrin (6), hyperin (7), and nicotiflorin (8) by spectroscopic analysis. The antioxidant activity was evaluated by the ORAC (oxygen radical absorbance capacity) assay, which measures scavenging activity against peroxy radicals induced by 2,2'-azobis (2-methylpropionamidine) dihydrochloride, and the ORAC value is expressed as relative trolox equivalent. Compounds 4, 6, and 7 exhibited potent antioxidant activity, whereas the other compounds showed weaker activity than trolox.

Luteolin and Chicoric Acid, Two Major Constituents of Dandelion Leaf, Inhibit Nitric Oxide and Lipid Peroxide Formation in Lipopolysaccharide-Stimulated RAW 264.7 Cells

  • Luteolin and chicoric acid are the most abundant phytochemicals in dandelion (Taraxacum officinale) leaf. In this study, four kinds of extraction methods [hot water, ambient temperature (AT) water, ethanol, and methanol] were applied to analyze the contents of both phytochemicals and verify their anti-inflammatory and antioxidative activities. The methanol extract showed the most potent nitric oxide (NO) inhibitory effect. The luteolin and chicoric acid concentrations were 3.42 and $12.86\;{\mu}g/g$ dandelion leaf in the methanol extract. The NO-suppressive effect of luteolin and chicoric acid was identified in a dose-dependent manner with $IC_{50}$ values of $21.2\;{\mu}M$ and $283.6\;{\mu}M$, respectively, in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells without cytotoxicity. Malondialdehyde (MDA) concentration, as an index for free radical injury on cell membrane, was also dose-dependently inhibited by the two compounds. The suppressive effect was further examined using mRNA and protein expression levels, which were attributable to the inhibition of inducible nitric oxide synthase (iNOS). These results suggest that two phytochemicals in dandelion leaf, luteolin and chicoric acid, may play an important role in the amelioration of LPS-induced oxidative stress and inflammation.

The Promise of Dried Fruits in Cancer Chemoprevention

  • Chemoprevention is an attempt to use nontoxic natural and synthetic substances or their mixtures to intervene the relatively early stages of carcinogenesis, before invasive characteristics are manifested. The consumption of fruits is well known to reduce the risk of human cancers. Although most fruits are available only on a seasonal basis, recent advances in food processing technologies have made it possible to extend the shelf life of fruits and fruit-products. Fruits can be preserved by applying different drying processes to reduce the moisture content. Different varieties of dried fruits are now sold in supermarkets, thereby making them readily accessible to consumers. Since oxidative stress and chronic inflammation play important roles in cancer development, dried fruits with antioxidative and anti-inflammatory properties hold promise for cancer chemoprevention. The antioxidant, anti-inflammatory and chemopreventive activities of dried fruits are largely attributed to their polyphenols and vitamins. Dried fruits contain adequate amounts of bioactive principles, such as anthocyanins, acetogenins, catechins, coumarins, phenolic acids, terpenes, xanthones, and others. Since numerous health beneficial phytochemicals in fruits are conserved even after processing, regular intake of dried fruits can help prevent cancer. This review addresses the chemopreventive potential of representative dried fruits and their active constituents.

강황 지상부의 항산화 성분 (Antioxidant Constituents of the Aerial Parts of Curcuma longa)

  • As part of ongoing study focused on the discovery of natural antioxidants from Korean plants by measuring the DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging effect and superoxide quenching activity, ethanol extract of the aerial parts of Curcuma longa (Zingiberaceae) was found to show potent antioxidant activities. Subsequent activity-guided fractionation of the ethanol extract led to the isolation of two phenolic compounds, 1,2,3,4,6-penta-O-galloyl-${\beta}$-D-glucopyranoside (1) and gallic acid (2), as antioxidant compounds. Their structures were elucidated by spectroscopic studies. Compounds 1 and 2 were isolated for the first time from this plant. These compounds showed the significant antioxidative effects during the DPPH free radical scavenging test, and the riboflavin- and xanthine-originated superoxide quenching activity tests.

참깨의 품질평가 현황과 전망 (Current Status and Prospects of Quality Evaluation in Sesame)

  • Sesame (Sesamum indicum L.) is probably the most ancient oilseed crop known in the world. Sesame seed is known for its high nutritional value and for having oil (51%) and protein (20%) content. The fatty acid composition of sesame oil is palmitic acid (7.8%), stearic acid (3.6%), oleic acid (45.3%), and linoleic acid (37.7%). Sesame oil is characterized by a very high oxidative stability compared with other vegetable oils. Two lignan-type compounds, sesamin and sesamolin, are the major constituents of sesame oil unsaponifiables. Sesamol (a sesamolin derivative) can be present in sesame seeds and oils in very small amount. Other lignans and sesamol are also present in sesame seeds and oils in very small amount as aglycones. Lipid oxidation activity was significantly lower in the sesamolin-fed rats, which suggests that sesamolin and its metabolites contribute to the antioxidative properties of sesame seeds and oil and support that sesame lignans reduce susceptibility to oxidative stress. Sesaminols strongly inhibit lipid peroxidation related to their ability to scavenge free radical. The sesame seed lignan act synergistically with vitamin I in rats fed a low $\alpha$-tocopherol diet and cause a marked increase in a u-tocopherol concentration in the blood and tissue of rats fed an $\alpha$-tocopherol containing diet with sesame seed or its lignan. The authors are reviewed and discussed for present status and prospects of quality evaluation and researched in sesame seeds to provide and refers the condensed informations on their quality.

쑥부쟁이 추출물의 아세틸콜린에스테라제 저해 및 항산화 활성 (Acetylcholinesterase Inhibitory and Antioxidant Properties of Aster yomena Extract)

  • Objectives : To evaluate the radical scavenging and acetylcholinesterase (AChE) inhibitory activities of the ethylacetate (EtOAc)-soluble portion of a methanolic extract of Aster yomena, three different assay systems were performed. Methods : The antioxidant activity of A. yomena extract was tested as its capacity to scavenging free radicals of DPPH and $ABTS^+$, which has been widely used to evaluate the antioxidant activity of natural products from plant sources. AChE inhibitory activity was tested against mouse brain AChE by spectrophotometric method of Ellman using ELISA microplate reader. Results : The methanolic extract of A. yomena was fractionated and the EtOAc-soluble portion showed significant AChE inhibitory and free radical scavenging effects. Also the EtOAc-soluble portion revealed the highest phenolic contents as compared to the other extracts. Conclusions : These results indicate that phenolic compounds may be important constituents that give rise to the anti-AChE and antioxidative activities of A. yomena extract. Further phytochemical studies on this plant, for nutraceutical or pharmaceutical application, are warranted.

맥문동 열수추출액을 이용한 인삼정과의 품질 특성 (Physicochemical Characteristics of Ginseng JungKwa Produced with Hot-Water Extract from Maegmundong (Liriope platyphylla tubers))

  • 인삼정과 제조 시 이용되는 당 용액 대신에 올리고당 함량이 높은 맥문동의 열수추출액을 이용하여 인삼정과를 제조한 후 품질 특성을 조사하였다. 맥문동 추출액으로 제조한 인삼정과는 가용성 무질소물인 탄수화물의 함량이 76.40%로 설탕으로 제조한 인삼정과의 80.01%에 비하여 약 4% 정도 낮았고, 조단백질 및 조회분 함량은 각각 1.46% 및 3.49%로 일반인삼정과의 0.59% 및 0.96%에 비하여 약 3배 이상 높았다. 맥문동 열수추출액을 이용한 인삼정과에서 유리당 조성 및 함량은 fructose가 32.42%로 가장 많았고, 그 외 올리고당이 16.91%, 설탕 및 이당류가 13.91%, glucose가 13.16%이었으며, Rh2를 비롯한 진세노사이드 11종이 검출되어 총 함량이 740.1 mg%로 일반 인삼정과(675.6 mg%)보다 더 높았다. 총 페놀화합물과 플라보노이드 함량은 각각 5.46과 0.016%로 일반인삼정과의 5.02와 0.014%보다 높았다. 맥문동 열수추출액으로 제조한 인삼정과의 항산화활성은 DPPH $IC_{50}$값이 인삼정과 중량기준으로 약 34.5 mg/mL 이었으며, hydrogen peroxide 소거능은 $50{\mu}g$/mL의 농도에서 92%의 소거능을 보여주었다.

추출방법에 따른 백단향의 항산화 및 생리활성 (Antioxidative and Biological Activities of Santalum album Extracts by Extracting Methods)

  • 본 연구에서는 백단향의 다양한 추출방법에 따른 항산화활성, 미백 효과 및 세포 독성을 평가 하였다. DPPH 라디칼에 대한 소거능은 n-Hexane 추출물을 제외한 모든 추출물에서 양성대조군인 L-ascorbic acid ($IC_{50}$, $28.7\;{\mu}g/mL$)보다 강하거나 동등한 활성을 나타내었으며, 그중에서도 70% acetone추출물에는 강한 라디칼 소거능($IC_{50}$, $18.6\;{\mu}g/mL$)을 나타내었다. 또한 LDL 산화억제 실험에서도 활성이 인정되었고 총 페놀 함량이 높게 나타난 70% acetone 추출물에 가장 강한 활성($IC_{50}$, $58.3\;{\mu}g/mL$)이 확인되었으며, 다음으로 MeOH, EtOH, Hot water, n-Hexane추출물 순의 활성을 관찰할 수 있었다. Tyrosinase 저해활성을 평가한 결과 양성 대조군인 arbutin ($IC_{50}$, $13.7\;{\mu}g/mL$) 및 kojic acid ($IC_{50}$, $8.3\;{\mu}g/mL$)보다 다소 약한 활성을 나타내었으며 70% Acetone을 이용하여 백단향을 추출할 경우 미백 활성이 가장 강한 결과를 얻었다. 백단향의 열수 추출물은 마우스멜라노마세포(B16F10)에 대해서 1.0 mg/mL의 농도에서 24.7%의 성장억제효과를 나타내었다.

산마늘의 부위별 성분분석 및 생리활성 평가 (Analysis of the Various Constituents and Comparison of Biological Activities of Different Parts of Allium victorialis var. platyphyllum)

  • 산마늘의 부위별(뿌리, 줄기 및 잎) 무기성분 함량과 다양한 생리활성을 평가한 결과, 부위에 관계없이 K과 Ca이 다른 무기성분에 비해 그 함량이 월등히 높았다. 총 폴리페놀 함량(mg tannic acid eq./100 g dry wt.)은 잎($792.7{\pm}16.6$ mg) > 줄기($160.3{\pm}6.8$ mg) > 뿌리($101.5{\pm}2.7$ mg) 순이었으며, 총 플라보노이드 함량(mg quercetin eq./100 g dry wt.)은 잎($497.9{\pm}11.1$ mg) > 뿌리($35.7{\pm}0.4$ mg) > 줄기($18.5{\pm}1.4$ mg) 순이었다. DPPH 라디칼(최종농도 250 ${\mu}M$)을 이용한 항산화 활성을 검토한 결과, 뿌리 30 mg, 줄기 25 mg, 그리고 잎 4 mg에서 $SC_{50}$값을 보였고, CE-OOH 생성 억제 효과는 잎 > 줄기 > 뿌리 순이었다. 그리고 부위별 항균활성 검토 결과, 뿌리, 줄기, 잎 모두 C. perfringens, S. aureus, L. monocytogenes, V. vulnificus, V. parahaemolyticus에 항균활성을 보였으며, 그 중 특히 C. perfringens 균에 강한 항균활성을 보였다.

곽향의 성분 분석 및 주요 성분들의 in silico 항당뇨 타겟 단백질 탐색 (Analysis of Chemical Constituents of Agastachis Herba and in silico Investigation on Antidiabetic Target Proteins of its Major Compounds)

  • 곽향은 식욕부진, 메스꺼움 등의 치료에 사용될 뿐만 아니라 최근에는 항당뇨 효능도 알려졌다. 본 연구에서는 곽향의 항산화력과 주요 성분들을 조사한 다음, in-silico 방법론을 적용하여 타겟 단백질들을 예측하였다. 먼저 메탄올 추출물의 DPPH와 ABTS 라디칼 소거능의 EC50 값은 각각 78.6 ㎍/mL과 31.0 ㎍/mL이었다. 이것은 ascorbic acid의 값(9.9 ㎍/mL, 5.2 ㎍/mL)과 비교할 때 항산화력이 뛰어나다고 할 수 있다. HPLC-PDA-MS/MS를 이용하여 성분들을 정성 분석한 결과, 추출물의 주요 화합물로 rosmarinic acid, tilianin, agastachoside 그리고 acacetin을 확인하였다. 이들 성분들의 항당뇨 작용 타겟 단백질을 DIA-DB 서버를 사용하여 구조 유사도와 inverse doking 방법론을 적용하여 예측하였다. 본 가상 탐색 결과, 주요 타겟 단백질들은 PPAR-γ, DPP IV, glucokinase, α-glucosidase, SGLT2, aldose reductase, corticosteroid 11-beta-dehydrogenase로 예측되었다. 그리고 이들 단백질들 중 일부는 이미 실험적으로 검증된 타겟 단백질이었다. 따라서 in silico 검색 방법이 유효하다고 생각할 수 있다. 마지막으로 활성성분들의 최적의 추출 조건을 결정하기 위하여 여러 추출 용매들로 곽향을 추출하였다. 여러 유기 용매들 중에는 메탄올이 그리고 에탄올-물 혼합용매에서는 80% 에탄올이 가장 효과적인 추출 용매로 확인되었다.
