  • The reactive oxygen species (ROS) caused the damage of macro molecules, many degenerative disease and cancer, which was produced in process of the aerotropic metabolic pathway as well as in response to the various genotoxic stresses. Recently, redox systems including the number of antioxidant proteins such as catalase, glutathione peroxidase, heam-containing peroxidase, peroxiredoxin and superoxide dismutase (SOD) has been reported to have chemopreventive effects. Antioxidant proteins has been known to have the activity directly removing ROS and affecting the protein-protein interaction and cell signaling to induce the cellular responses. We need to understand the mechanism of antioxidants prevent DNA damage from oxidative stresses for researching the cancer prevention and providing the development of cancer therapeutic drug.

Differential Antioxidant Mechanisms of Rice Plants in Response to Oxyfluorfen and Paraquat

  • The mechanisms of resistance to oxyfluorfen (OF) and paraquat (PQ) were investigated in rice plants. Examination of the concentration dependence of oxyfluorfen- or paraquat-induced increase in conductivity showed that conductivities in the OF- and PQ-treated leaf squares were increased with 0.1 ${\mu}M$ oxyfluorfen and 0.01 ${\mu}M$ paraquat and further increased with higher concentrations. The levels of conductivity were approximately 10-times higher in the PQ-treated plants than in the OF-treated plants, indicating that the PQ-treated plants suffered more severe photodynamic damage than the OF-treated plants. The photooxidative stress caused by foliar application of either 50 ${\mu}M$ oxyfluorfen or 100 ${\mu}M$ paraquat increased the enzyme activities of ascorbate peroxidase and peroxidase 1 day after the herbicide treatments and then further increased their enzyme activities 2 days after the treatments. The activities of catalase began to increase 2 days after the oxyfluorfen and paraquat treatments. These antioxidant enzymes appear to play an essential part of defense mechanisms against oxyfluorfen and paraquat. Our results demonstrate that paraquat caused more severe oxidative stress, as indicated by a greater change in conductivity, thereby resulting in greater increases in antioxidant responses in plants, compared with those of oxyfluorfen.

H2O2 Pretreatment Modulates Growth and the Antioxidant Defense System of Drought-stressed Zoysiagrass and Kentucky Bluegrass

  • This study investigated the effect of exogenous hydrogen peroxide ($H_2O_2$) on the antioxidant responses and growth of warm-season turfgrass (Zoysia japonica Steud.) and cool-season turfgrass (Poa pratensis L.) subjected to drought stress. Compared with control plants that were not pretreated with $H_2O_2$, plants pretreated with $H_2O_2$ had significantly greater fresh and dry weights of shoots and roots, and increased water content. $H_2O_2$ pretreatments before drought stress significantly decreased the concentrations of malondialdehyde and $H_2O_2$. DPPH radical scavenging and glutathione activities were significantly increased. The responsive activities of the antioxidant enzymes superoxide dismutase, ascorbate peroxidase, catalase, and peroxidase were also significantly enhanced. Our results suggest that exogenous $H_2O_2$ could improve the growth of warm-season and cool-season turfgrass under drought stress by increasing the activity of their antioxidant enzymes, while decreasing lipid peroxidation.

Explicating morphophysiological and biochemical responses of wheat grown under acidic medium: Insight into to the antioxidant defense and glyoxalase systems

  • Low soil pH causes from $H^+$ rhizotoxicity results in nutrients unavailability in the growing media, inhibits plant growth, development and reduces crop yields. The present study was carried out to reveal morpholophysiological and biochemical responses of wheat (Triticum aestivum L.) to acidity stress. Four wheat varieties viz. BARI Wheat-21, BARI Wheat-25, BARI Wheat-26 and BARI Wheat-30 were used in the study. Eight-day-old seedlings were exposed to different pH levels (3.5, 4.5, 5.5 and 6.5) of growing media. Acidity stress at any level reduced biomass, water, and chlorophyll contents in all the varieties; whereas BARI Wheat-26 showed the least damage. $H^+$ rhizotoxicity also caused oxidative stress through excess production of reactive oxygen species and methylglyoxal which increase lipid peroxidation in all the varieties but the lowest oxidative damage was observed in BARI Wheat-26 due to better performance of the antioxidant defense and glyoxalase systems. Considering the growth, physiological and biochemical attributes BARI Wheat-26 may be considered as acidity stress tolerant, among the variety examined.

Higher concentrations of folic acid reduced the dietary requirements of supplemental methionine for commercial broilers

  • Objective: An experiment was conducted to study the effect of supplementing DL methionine (DL Met) at graded concentrations on performance, carcass variables, immune responses and antioxidant variables in broiler chicken fed folic acid (FA) fortified (4 mg/kg) low-methionine diet. Methods: A basal diet (BD) without supplemental DL Met, but with higher level (4 mg/kg) of FA and a control diet (CD) with the recommended concentration of methionine (Met) were prepared. The BD was supplemented with DL Met at graded concentrations (0%, 10%, 20%, 30%, 40%, and 50% supplemental DL Met of CD). Each diet was fed ad libitum to 10 replicates of 5 broiler male chicks in each from 1 to 42 d of age. Results: Body weight gain (BWG) reduced, and feed conversion ratio (FCR) increased in broilers fed low-Met BD. At 30% and 20% inclusion of DL met, the BWG and FCR, respectively were similar to those fed the CD. Similarly, supplementation of 10% DL Met to the BD significantly increased ready to cook meat yield and breast meat weight, which were similar to those of the CD fed broilers. Lipid peroxidation reduced, the activity of antioxidant enzymes (GSHPx and GSHRx) in serum increased and lymphocyte proliferation increased with increased supplemental DL Met level in the BD. The concentrations of total protein and albumin in serum increased with DL Met supplementation to the BD. Conclusion: Based on the data, it can be concluded that supplemental Met can be reduced to less than 50% in broiler chicken diets (4.40, 3.94, and 3.39 g/kg, respectively in pre-starter, starter and finisher phases) containing 4 mg/kg FA.

Effects of Dietary Supplementation of a Meju, Fermented Soybean Meal, and Aspergillus oryzae for Juvenile Parrot Fish (Oplegnathus fasciatus)

  • In this study, dietary supplementations of Korean Meju, fermented soybean meal (F-SBM) by Aspergillus oryzae, and A. oryzae itself were evaluated on growth performance, feed utilization, immune responses and phosphorus availability in juvenile parrot fish, a marine aquaculture fish species. Four isonitrogenous and isocaloric diets were formulated to contain 8% soybean meal (control diet), 4% Meju (50% soybean meal was replaced by Meju), 4% F-SBM (50% soybean meal was replaced by F-SBM), or 0.08% A. oryzae itself. One of the four experimental diets was fed to triplicate groups of fish for 8 weeks. At the end of the feeding trial, no significant differences were found in growth performances and feed utilization. Red blood cell counts in the fish fed the A. oryzae diet were significantly higher than that of fish fed the control diet. The antioxidant activity in Meju diet was significantly higher than that of the control and A. oryzae diets. Fish fed Meju and F-SBM diets showed numerically higher antioxidant activity of serum compared to that of fish fed the control diet, even though it was not significant. Liver superoxide dismutase activity of fish fed the test diets was significantly higher than that of fish fed the control diet. The apparent digestibility coefficients of protein of fish fed all the diets were not significantly different. Phosphorus absorption was numerically increased in fish fed F-SBM and A. oryzae diets compared to that of fish fed the control diet. This study indicates that the fermentation process of soybean meal does not impair growth performance and feed utilization in parrot fish. The fermentation process could enhance the availability of phosphorus in soybean meal and non-specific immune responses of parrot fish.

Protective Effects of Acorn (Quercus acutissima CARR.) against IgE-mediated Allergic and Ovalbumin (OVA)-Induced Asthmatic Responses via Inhibition of Oxidative Stress

  • This work was performed to investigate the protective effect of ethanol extract (AEx) from acorn (Quercus acutissima CARR.) against allergic mediated responses in asthma model cells and mice. The AEx inhibited antigen-stimulated cytokine production such as interleukin (IL)-4, IL-13 and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and AEx also inhibited intracellular reactive oxygen species (ROS) generation against IgE-mediated allergic response in rat basophilic leukaemia RBL-2H3 cells. The ovalbumin (OVA)-sensitized mice were orally administered with AEx (100 or 300 mg/kg) and authentic tannic acid (75 mg/kg) every day for 15 days. Increased TNF-${\alpha}$ production by OVA-sensitization/challenge was significantly reduced by administration of AEx. The serum triglyceride levels of asthma mice were significantly reduced after feeding for 15 days with tannic acid or AEx. The mice fed with tannic acid or AEx also exhibited a significant reduction in body weights compared to those of asthma control group. The AEx increased the heme oxygenase (HO)-1 mRNA expression in the asthma model mice and showed DPPH radical scavenging activity. These results indicate that AEx protects against IgEmediated allergic and OVA-induced asthmatic responses via direct and indirect antioxidant activities. Reduced triglyceride and body weights may provide additional protective benefits of AEx on allergic asthma.

Tat-ATOX1 inhibits inflammatory responses via regulation of MAPK and NF-κB pathways

  • Antioxidant 1 (ATOX1) protein has been reported to exhibit various protective functions, including antioxidant and chaperone. However, the effects of ATOX1 on the inflammatory response has not been fully elucidated. Thus, we prepared cell permeable Tat-ATOX1 and studied the effects on lipopolysaccharide (LPS)- and 12-O-tetradecanoyl phorbol-13-acetate (TPA)-induced inflammation. Experimental results showed that transduced Tat-ATOX1 protein significantly suppressed LPS-induced intracellular reactive oxygen species (ROS). Also, Tat-ATOX1 protein markedly inhibited LPS- and TPA-induced inflammatory responses by decreasing cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) and further inhibited phosphorylation of mitogen activated protein kinases (MAPKs; JNK, ERK and p38) and the nuclear factor-kappaB ($NF-{\kappa}B$) signaling pathway. These results indicate that the Tat-ATOX1 protein has a pivotal role in inflammation via inhibition of inflammatory responses, suggesting Tat-ATOX1 protein may offer a therapeutic strategy for inflammation.

배수성이 다른 자생 버뮤다그래스의 휴면 전후 항산화 효소활성 및 세포막 안정성 변화 (Antioxidant Enzyme Activity and Cell Membrane Stability of Korean Bermudagrass Genotypes Different in Ploidy at Dormant Stage)

  • 기존 보고된 바에 의하면 한국 자생 버뮤다그래스는 군집 내에서 형태학, 생육 특성, 세포학적 특성에 대해 유전적으로 매우 다양한 변이를 보여주었다. 버뮤다그래스의 염색체 수와 핵 DNA 량에 따르면 배수성 수준의 범위가, 3배 체(2n=3x), 4배체(2n=4x), 5배체(2n=5x), 6배체(2n=6x)로 나타났었다. 본 연구에서는 한국에서 휴면이 유도되는저온과 짧은 일장에 대한 항산화효소(superoxide dismutase, catalase, peroxidase, ascorbate peroxidase)의 다양한 반응과 각 버뮤다그래스 세포형의 세포막 안정성을 조사하였다. 모든 항산화효소는 휴면 기간동안 높게 나타났으나, 과산화수소를 물과 산소 분자로 변환시키는 헴기를 함유한 카탈라제는 6배체 버뮤다그래스를 제외한 세 개의 세포형에서 휴면이 개시되기 전에 활성화되었다. 상대적으로 세엽이며 생육속도가 빠른3배체와 4배체는 superoxide dismutase와 peroxidase 효소의 활성이 증가됨을 확인하였다. 수산기를 가진 라디칼에 의해 손상을 받은 세포막에서 지질과산화의 산물인 말론디알데히드(MDA)는 온도가 감소함에 따라 모든 세포형에서 증가되었고, 방어적인 항산화효소를 더 갖고 있는 3배체와 4배체는 MDA 생산이 현저하게 더 낮게 나타났다. 전해질 유출은 5배체와 6배체에서 더 높았던 것과 유사하게, 저온이 적용될 때 외견상으로 세포막에 더 손상을 받는 것 같았다. 실험 결과, 서로 다른 세포형(cytotype)의 항산화 반응은 유전적으로 특이적이며, 이는 버뮤다그래스에서 저온 저항성과의 연관성을 분자 수준에서 더 연구하는 것이 필요하다.

Proline, Sugars, and Antioxidant Enzymes Respond to Drought Stress in the Leaves of Strawberry Plants

  • Drought is a severe abiotic stress that affects global crop production. A drought model was created for 'Toyonoka' Fragaria ${\times}$ ananassa, and the effects of drought stress on contents of proline, sugars, and antioxidant enzyme activities were investigated. Strawberry transplants with identical growth were chosen for the experiments and the randomized design included four replications (10 plants per block). The experimental sets differed in the moisture level of the culture medium relative to the range of moisture content as follows: control, 70-85%; mild drought stress, 50-60%; moderate drought stress, 40-50%; and severe drought stress, 30-40%. Drought stress was imposed by limiting irrigation. Plants were sampled and physiological parameters w ere measured on 0, 2, 4, 6, 8, and 10 days after the commencement of droughts tress. The water potential of strawberry leaves decreased in the plants under mild, moderate, and severe stress during the course of the water stress treatment and exhibited a significant difference from the control. Strawberry leaves subjected to drought stress had higher accumulation of proline, sugars, and malondialdehyde, and higher activities of superoxide dismutase, peroxidase, and catalase than leaves of control plants. Malondialdehyde levels increased in parallel with the severity and duration of drought stress. By contrast, antioxidant enzyme activity displayed dynamic responses to drought stress, first increasing and subsequently decreasing as the severity and duration of drought stress increased. These results suggest that strawberry plants respond to drought stress by altering the activities of antioxidant enzymes and the levels of osmotically active metabolites. These biochemical response changes may confer adaptation to drought stress and improve the capacity of plants to withstand water-deficit conditions.
