The Therapeutic Role of Nanoparticle Shape in Traumatic Brain Injury : An in vitro Comparative Study

  • Objective : To perform a comparative analysis of therapeutic effects associated with two different shapes of ceria nanoparticles, ceria nanorods (Ceria NRs) and ceria nanospheres (Ceria NSs), in an in vitro model of traumatic brain injury (TBI). Methods : In vitro TBI was induced using six-well confluent plates by manually scratching with a sterile pipette tip in a 6×6-square grid. The cells were then incubated and classified into cells with scratch injury without nanoparticles and cells with scratch injury, which were treated separately with 1.16 mM of Ceria NSs and Ceria NRs. Antioxidant activities and anti-inflammatory effects were analyzed. Results : Ceria NRs and Ceria NSs significantly reduced the level of reactive oxygen species compared with the control group of SH-SY5Y cells treated with Dulbecco's phosphate-buffered saline. The mRNA expression of superoxide dismutases was also reduced in nanoparticle-treated SH-SY5Y cells, but apparently the degree of mRNA expression decrease was not dependent on the nanoparticle shape. Exposure to ceria nanoparticles also decreased the cyclooxygenase-2 expression, especially prominent in Ceria NR-treated group than that in Ceria NS-treated group. Conclusion : Ceria nanoparticles exhibit antioxidant and anti-inflammatory effects in TBI models in vitro. Ceria NRs had better anti-inflammatory effect than Ceria NSs, but showed similar antioxidant activity.

A Study on the Physiological Activity of Cosmetics in Bupleurum falcatum L. Extract (시호 추출물의 화장품 생리활성에 관한 연구)

  • Bupleurum falcatum L. is one of the medicinal crops and has excellent pharmacological activities such as reducing fever, sedation, inhibition of hepatopathy, anti-inflammatory, and anticancer. Currently, it is used for food or medicine, but the use as a cosmetic is insufficient, so the suitability and use value as a cosmetic material were reviewed. Using the Bupleurum falcatum L. extract, polyphenol, flavonoid, antioxidant, and anti-inflammatory tests were conducted. The total polyphenol content was 14.71±0.16 mg·TAE/g, and the total flavonoid content was 1.42±0.05 mg·QE/g. The antioxidant activity showed excellent activity in a concentration-dependent activative, and the NO expression inhibition ability was 61.21% at 1,000 ㎍/mL, confirming that the anti-inflammatory activity was excellent. Through this study, the antioxidant and anti-inflammatory effects of Bupleurum falcatum L. extract are excellent, so it is considered to be of sufficient value as a natural material for cosmetics.

Antioxidant and Anti-inflammatory Activities in Freeze-dried and Hot Air-dried Aronia (Aronia melanocarpa) Extracts (동결 및 열풍건조 아로니아 추출물의 항산화 및 항염증 활성)

  • This study examined the effects of drying methods of Aronia melanocarpa on contents of total polyphenols and flavonoids, antioxidant activity and anti-inflammatory activity. As a result, freeze-dried aronia extract (FDAE) significantly exhibited higher contents of total polyphenols and flavonoids (155.76 mg GAE/g and 105.70 mg QE/g) than hot-air dried aronia extract (HDAE) (134.93 mg GAE/g and 82.29 mg QE/g). Also, FDAE showed greater antioxidant activity than HDAE in both DPPH and ABTS. For anti-inflammatory activity, NO production from lipopolysaccharide activated RAW264.7 cell reduced at a dose-dependent manner in both FDAE and HDAE. However, reduction rate of NO production is higher in FDAE (62.7%) than in HDAE (33.5%). These results suggest that comparatively, freeze-drying is a better method for preserving the antioxidant and anti-inflammatory activities and of aronia.

  • Cudrania tricuspidata have been used for anti-inflammatory, anti-hepatotoxic, anti-hypertensive and anti-diabetic activities. In this study, in order to investigate the efficacy of antioxidant activity, the bio-activity guided fraction and isolation of physiologically active substance were performed. H$_2$O, 30%, 60%, 100% MeOH and acetone fractions were examined antioxidant activity by DPPH method. It was revealed that 30%, 60%, 100% MeOH frations have significantly antioxidant activity. Nutritional Components and Physiological Activity of Centella asiatica Cultured in Chungju by Drying Method (건조방법에 따른 충주산 병풀의 영양성분과 생리활성)

  • Centella asiatica (CA) has been used as a nutritional plant as well as a traditional herbal medicine around the world. This study, quality component (proximate composition, total polyphenol, and triterpenoid compound), and antioxidant and anti-inflammatory activities of CA dried using various methods were investigated. Proximate compositions of CA with different drying methods included a large amount of carbohydrates, crude protein, crude ash, and crude fiber. Among the drying methods, cold drying provided the highest total polyphenol content and antioxidant activity, while hot-air drying at 75℃ provided the lowest total polyphenol content and antioxidant activity. In addition, when the major triterpenoid compounds of CA were analyzed, the highest content of asiaticoside of triterpenoid glycoside was obtained with all drying methods. With respect to the total triterpenoid, the highest content was obtained with cold drying (68.8 mg/g) whereas natural drying (31.4 mg/g) provided the lowest content. In anti-inflammatory activity of LPS-stimulated RAW 264.7 cells, EtOH extract of cold drying showed a significantly higher inhibitory activity in comparison to the other drying methods. In conclusion, it is considered that the cold drying method is suitable for industrial preparation of functional materials with high physiological ingredients, and antioxidant and anti-inflammatory activities from CA.

Anti-oxidant Activities and Anti-inflammatory Effects on Artemisia scoparia (비쑥 추출물에 대한 항산화 활성 및 염증억제 효과)

  • We investigated the antioxidant activities such as DPPH radical scavenging capacity, xanthine oxidase inhibitory activity, and superoxide radical scavenging capacity of the aqueous EtOH extract and its solvent fractions of Artemisia scoparia. The ethyl acetate fraction showed high antioxidant activity, compared to positive controls such as ascorbic acid, butylated hydroxy anisole (BHA), trolox, and allopurinol in these assay systems. Moreover, we examined the inhibitory effect of solvent fractions of A. scoparia on the production of pro-inflammatory factors that the nitric oxide (NO), inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2) and prostaglandin E2 $(PGE_2)$ production activated with LPS $(1{\mu}g/ml)$ in murine macrophage cell line RAW264.7. The amounts of protein levels were determined by immunoblottting. Tn the sequential fractions of hexane and dichloromethane inhibited the NO and $PGE_2$ production and the protein level of iNOS and COX-2. These results suggest that A. scoparia may have anti-inflammatory activity through the antioxidant activity and inhibition of pro-inflammatory factors.

Antioxidant and Anti-inflammatory Effects of Atoberry in Atopic Dermatitis-like NC/Nga Mouse Model (아토피 피부염 유사 NC/Nga 마우스 모델에서 아토베리의 항산화 및 항염증효과)

  • Oxidative stress has been implicated in cutaneous damage in various inflammatory skin diseases, including atopic dermatitis (AD). Atoberry is the herb medicine extract which is composed with Spirodelae Herba, Xanthii Fructus, Houttuyniae Herba, Taraxaci Herba, Retinervus Luffae Fructus, Platycodi Radix, and Scutellariae Radix. In this study, we investigated the antioxidant and anti-inflammatory effects of Atoberry in AD-like skin lesion NC/Nga mice. Murine AD-like skin lesions were made by painting Dermatophagoides farinse (Df) extract. Atoberry significantly increased electron donating ability (DPPH), nitrite scavenging (NO) and superoxide dismutase (SOD) activities in dose dependant. Topically applied Atoberry significantly reduced clinical severity score, ear thickness and histological grade in AD-like skin lesion NC/Nga mice. In addition, the serum levels of IgE, NO and prostaglandin E2 were significantly reduced by Atoberry. Futhermore, skin tissue levels of SOD, catalase and glutathione peroxidase (GPx) were significantly reduced by Atoberry. These results demonstrate that topical application of Atoberry may be improve the AD-like skin lesion by antioxidant and anti-inflammatory effects.

Anti-inflammatory and Antioxidant Activity of Pomegranate Leaf Extract (석류 잎 추출물의 항염 및 항산화 활성)

  • This study aims to evaluate the antioxidant and anti-inflammatory effects of pomegranate (Punica granatum) leaves and to explore the potential of pomegranate leaf extract as an anti-aging cosmetic ingredient. The ethyl acetate fraction of ethanolic extract of pomegranate leaf (EFP) was assessed for its total polyphenol and flavonoid contents, as well as its antioxidant and anti-inflammatory activities. The beneficial components and skin improvement potential of pomegranate leaves were investigated. The results showed that the total polyphenol content of EFP was 871.6±16.3 mg gallic acid/g, and the flavonoid content was 36.6±0.3 mg quercetin/g. In the ABTS radical scavenging assay, EFP exhibited a dose-dependent antioxidant activity with an EC50 value of 24.62±0.48 ㎍/mL. In the skin cell cytotoxicity assay, EFP demonstrated high cell viability at concentrations below 50 ㎍/mL, indicating minimal cytotoxicity. In the nitric oxide (NO) production inhibition assay, EFP effectively inhibited NO production even at low concentrations, with near-complete inhibition at 6 ㎍/mL. These results suggest that EFP has the potential to be utilized as a natural cosmetic ingredient with antioxidant and anti-inflammatory properties.

The Effect of Rutin on Antioxidant and Anti-inflammation in Streptozotocin-induced Diabetic Rats

  • The study examined the antioxidant activities and anti-inflammatory effects of rutin from the streptozotocin (STZ)-induced diabetic rats. Results revealed that the levels of plasma glucose and serum glucose were remarkably higher in the STZ-treated group compared to other groups and were significantly reduced in the STZ+rutin treated group compared to the STZ-treated group. In terms of weight, it significantly increased in all experimental groups during the experiment period except for STZ-induced diabetic group. The weight of the STZ-treated group was remarkably reduced compared to other groups. Regarding the weight of each body organ, the STZ-treated group showed higher organ weight compared to the other groups while STZ+rutin-treated group showed significantly reduced kidney and liver weights compared to those of STZ-treated group. In the pancreas tissue of the STZ-treated group, ${\beta}$-cell destruction and vacuolization were observed. Inflammation in the heart, liver, kidney, and retina tissues were also vividly recorded. In the STZ+rutin administered group, the heart and retina tissues were shown to be preserved normally while the liver and kidney tissues showed reduced histopathology in general compared to the STZ-treated group. Conclusionally, the rutin has the effect on the antioxidant activities and anti-inflammation in the STZ-induced diabetic rats.

Evaluation of in vitro anti-oxidant and anti-inflammatory activities of Korean and Chinese Lonicera caerulea

  • BACKGROUND/OBJECTIVE: The honeysuckle berry (HB) contains ascorbic acid and phenolic components, especially anthocyanins, flavonoids, and low-molecular-weight phenolic acids. In order to examine the potential of HB as a hepatoprotective medicinal food, we evaluated the in vitro anti-oxidant and anti-inflammatory activities of Korean HB (HBK) and Chinese HB (HBC). MATERIALS/METHODS: Antioxidant and anti-inflammatory effects of the extracts were examined in HepG2 and RAW 264.7 cells, respectively. The anti-oxidant capacity was determined by DPPH, SOD, CAT, and ARE luciferase activities. The production of nitric oxide (NO) as an inflammatory marker was also evaluated. The Nrf2-mediated mRNA levels of heme oxygenase-1 (HO-1), NAD(P)H dehydrogenase [quinone] 1 (Nqo1), and glutamate-cysteine ligase catalytic subunit (Gclc) were measured. The concentrations of HB extracts used were 3, 10, 30, 100, and $300{\mu}g/mL$. RESULTS: The radical scavenging activity of all HB extracts increased in a concentration-dependent manner (P < 0.01 or P < 0.05). SOD (P < 0.05) and CAT (P < 0.01) activities were increased by treatment with $300{\mu}g/mL$ of each HB extract, when compared to those in the control. NO production was observed in cells pretreated with 100 or $300{\mu}g/mL$ of HBC and HBK (P < 0.01). Treatment with $300{\mu}g/mL$ of HBC significantly increased Nqo1 (P < 0.01) and Gclc (P < 0.05) mRNA levels compared to those in the control. Treatment with $300{\mu}g/mL$ of HBK (P < 0.05) and HBC (P < 0.01) also significantly increased the HO-1 mRNA level compared to that in the control. CONCLUSIONS: Thus, the Korean and Chinese HBs were found to possess favorable in vitro anti-oxidant and anti-inflammatory activities. Nrf2 and its related anti-oxidant genes were associated with both anti-oxidant and anti-inflammatory activities in HB-treated cells. Further studies are needed to confirm these in vivo effects.
