Effects of Lycopene Alone or Combined with Melatonin on Methotrexate-Induced Nephrotoxicity in Rats

  • Methotrexate (Mtx), used for its anticancer and immunsuppresive properties, is known to be a nephrotoxic agent. We aimed to investigate the effects of lycopene (Lyc) alone or combined with melatonin (Mel) on Mtxinduced nephrotoxicity since both of these agents have antioxidant and anti-inflammatory effects. Nephrotoxicity was induced by intraperitoneal administration of methotrexate at a dose of 20 mg/kg. Treatment both with Lyc alone and Lyc combined with Mel provided significant reduction in tumor necrosis factor-alpha, interleukin 1-beta and ceruloplasmin levels in Mtx administered rats. Hovewer, Lyc combined with Mel provided a significant reduction also in NO levels. Hstopathological examination showed that there was an obvious improvement in the degenerative changes compared to Mtx administrated group with the Lyc combined Mel group giving best protection. In conclusion Lyc alone and combined with Mel provided significant improvement against renal damage caused by Mtx, preseumably via antioxidant and anti-inflammatory activities.

Oxidative Stress and Skin Diseases: Possible Role of Physical Activity

  • Background: The skin is the largest body organ that regulates excretion of metabolic waste products, temperature, and plays an important role in body protection against environmental physical and chemical, as well as biological factors. These include agents that may act as oxidants or catalysts of reactions producing reactive oxygen species (ROS), reactive nitrogen species (RNS), and other oxidants in skin cells. An increased amount of the oxidants, exceeding the antioxidant defense system capacity is called oxidative stress, leading to chronic inflammation, which, in turn, can cause collagen fragmentation and disorganization of collagen fibers and skin cell functions, and thus contribute to skin diseases including cancer. Moreover, research suggests that oxidative stress participates in all stages of carcinogenesis. We report here a summary of the present state of knowledge on the role of oxidative stress in pathogenesis of dermatologic diseases, defensive systems against ROS/RNS, and discuss how physical activity may modulate skin diseases through effects on oxidative stress. The data show duality of physical activity actions: regular moderate activity protects against ROS/RNS damage, and endurance exercise with a lack of training mediates oxidative stress. These findings indicate that the redox balance should be considered in the development of new antioxidant strategies linked to the prevention and therapy of skin diseases.

Curcumin-induced Growth Inhibitory Effects on HeLa Cells Altered by Antioxidant Modulators

  • Curcumin (diferuloyl methane), originated rhizomes of Curcuma longa L. has been suggested as an anti-inflammatory and anti-carcinogenic agent. In the present study, modulation of cytotoxic effects of curcumin on HeLa cells by different types of antioxidants was investigated. Cytotoxic effects of curcumin were significantly enhanced in the presence of superoxide dismutase (SOD) by decreasing $IC_{50}$ to 15.4 from $26.0\;{\mu}M$ after 24 hr incubation; the activity was not altered by catalase. The effect of curcumin was significantly less pronounced in the presence of 4 mM N-acetylcysteine (NAC). Low concentration (<1 mM) of NAC, however, increased the efficacy of curcumin. Cysteine and ${\beta}$-mercaptoethanol that have a thiol group, showed the similar biphasic patterns as NAC for modulating curcumin cytotoxicity, which was, however, constantly enhanced by ascorbic acid, a non-thiol antioxidant. In the presence of SOD, ascorbic acid, and 0.5 mM NAC, cellular levels of curcumin were significantly increased by 31-66%, whereas 4 mM NAC decreased the level. The present results indicate that thiol reducing agents showed a biphasic effect in modulating cytotoxicity of curcumin; it is likely that their thiol group is reactive with curcumin especially at high concentrations.

Induction of DNA Breakage by the Hot-water Extracts of Fructus Chaenomelis (Chaenomeles sinensis Koehne)

  • The possible mechanism of the DNA strand breaking activity of the hot-water extract of Fructus Chaenomelis (dried fruit of Chaenomeles sinensis) in a closed circular duplex replica form DNA (RFI DNA) was studied through agarose gel electrophresis under various conditions. Induction of DNA strand scission by the hot-water extract of C. sinensis occurred in dose and time-dependent manners. $Cu^{2+}$ was indispensable for the induction of DNA strand breakage. Exogeneous chelating agents inhibited the DNA breaking activity, conforming the catalytic action of $Cu^{2+}$ on generation of free radicals responsible for oxidative damage. Antioxidant enzymes and some radical scavengers were used to investigate the major radical species triggering the DNA strand scission, demonstrating that a highest inhibitory activity was found in the presence of catalase, while less in the presence of tiron (a scavenger for superoxide radical), 2-aminoethyl-isothiuroniumbromide-HBr, cysteamine (scavengers for hydroxyl radical), and 1,4-diazabicyclo [2,2,2] octane (a scavenger for singlet oxygen) in decreasing order. The findings implied that oxygen radical species generated in presence of transition divalent cation during the oxidation of some compounds contained in the hot-water extract of C. sinensis is mainly responsible for inducing genotoxicity.

Sea Buckthorn (Hippophae rhamnoides L.) Leaf Extracts Protect Neuronal PC-12 Cells from Oxidative Stress

  • The present study was carried out to investigate the antioxidative and neuroprotective effects of sea buckthorn (Hippophae rhamnoides L.) leaves (SBL) harvested at different times. Reversed-phase high-performance liquid chromatography analysis revealed five major phenolic compounds: ellagic acid, gallic acid, isorhamnetin, kaempferol, and quercetin. SBL harvested in August had the highest total phenolic and flavonoid contents and antioxidant capacity. Treatment of neuronal PC-12 cells with the ethyl acetate fraction of SBL harvested in August increased their viability and membrane integrity and reduced intracellular oxidative stress in a dose-dependent manner. The relative populations of both early and late apoptotic PC-12 cells were decreased by treatment with the SBL ethyl acetate fraction, based on flow cytometry analysis using annexin V-FITC/PI staining. These findings suggest that SBL can serve as a good source of antioxidants and medicinal agents that attenuate oxidative stress.

Changes in Antioxidant and Whitening Activities of Buckwheat Seeds with Germination Time

  • The purpose of this study was to evaluate the antioxidant and whitening action of Fagopyrum esculentum and F. tataricum buckwheat seeds depending on their germination time. In a previous study, we reported significant changes in sprout yield and phytochemical content in ethanol extracts from F. esculentum and F. tataricum seeds with increase in germination times. DPPH radical scavenging activities of F. tataricum increased with increasing germination time, whereas that of F. esculentum decreased. Next, we investigated anti-melanogenic activities of these species by estimation of melanin content and tyrosinase activity. Inhibition of melanin production in ${\alpha}-MSH$ (${\alpha}$-melanocyte stimulating hormone)-induced B16F10 cells by extracts from these seeds were analyzed. Among the two, F. tataricum extracts were characterized by higher inhibitory activity against melanin production. In addition, when B16F10 cells were incubated with L-DOPA for detection of in situ tyrosinase activity, F. tataricum and F. esculentum extracts were observed to reduce melanin production in cells. Taken together, these results indicate that extracts from buckwheat seeds could influence cellular processes via modulation of tyrosinase activity. Hence, buckwheat seeds could be utilized as whitening agents in the cosmetic industries and as therapeutics for hyperpigmentation disorders in a clinical setting.

Evaluation of the Antioxidant and Antiproliferative Properties of a Hot-water Extract from Gulfweed, Sargassum fulvellum

  • Sargassum fulvellum (gulfweed) is a widespread seaweed in the coastal areas of northeast Asia. In the present study, we identified the phenolic compounds present in aqueous and ethanolic extracts of S. fulvellum and evaluated their antioxidative properties and their abilities to block cell proliferation using in vitro assays: antioxidant activity was assessed by using a DPPH assay and superoxide anion scavenging activity, anti-tyrosinase activity, and anti-proliferative activity were assessed using MTT and lactate dehydrogenase [LDH] assays in vascular smooth muscle cells. The hot-water ($65^{\circ}C$) extract had a higher phenol content than the ethanolic extract. The hot-water extract showed a statistically significant increase in free radical scavenging activity and a greater ability to reduce proliferation of vascular smooth muscle cells stimulated with platelet-derived growth factor-BB. Taken together, hot-water extracts of S. fulvellum may be an important source of antioxidative and antiproliferative agents.

약용곤충추출물 라이브러리를 이용한 항산화 활성의 초고속 검색 (High Throughput-compatible Screening of Anti-oxidative Substances by Insect Extract Library)

  • 산화적인 스트레스(Oxidative stress)는 신경염증의 발병요인 중의 하나로 알려져 있다. 이에 본 연구는 약용곤충추출물을 이용하여 항산화 물질을 찾고자 초고속 적용가능한 스크리닝 방법을 적용하였다. 우선, 분자염증은 활성산소 관련의 물질과 밀접한 관련이 있으므로 이들의 억제를 동반하는 추출물을 먼저 선별하였다. 항산화 실험과 관련하여 DPPH (1,1-Diphenyl-2-picrylhydrazyl), FRAP(Ferric ion reducing antioxidant power), HO (Hydroxyl radical) 소거, linoleic acid에 대한 항산화 활성 등을 assay하였고 hydrogen peroxide(H2O2)에 의한 세포사멸 억제 활성을 보기 위해 MTT assay를 실시하였다. 실험 결과, 사마귀, 늦반딧불이, 무당벌레에서 다른 library에 비교하여 항산화 활성이 높게 나타났다.

녹차 추출물을 함유한 옥피 단백질 필름의 특성 (Characterization of a Corn Fiber Protein Film Containing Green Tea Extract)

  • 옥수수의 습식 분쇄 공정 중 나오는 부산물인 옥피로부터 옥피 단백질(CFP)을 추출하였다. 다양한 가소제와 가교제를 첨가하여 CFP 필름을 제조한 후, 필름의 물성을 측정하였다. 본 연구에서 사용된 가소제와 가교제 중, 2 g fructose와 0.03% cinnamaldehyde를 첨가한 필름이 가장 좋은 물성을 가졌다. 또한 여러 농도(0, 0.5, 1.0, 1.5%)의 녹차 추출물(GTE)을 첨가한 CFP 필름을 제조하였다. GTE의 농도가 증가함에 따라 CFP 필름의 인장강도, 수분 용해도, 불투명도는 증가하였고, 반면에 신장률과 필름의 수분투과도는 대조구에 비해 감소하였다. GTE 첨가한 CFP 필름의 항산화성은 DPPH 소거능으로 측정하였는데, GTE 농도가 증가함에 따라 증가하였다. 그리고 E. coli O157:H7, L. monocytogenes, S. aureus 에 대한 항균성 실험을 한 결과 또한 GTE 농도가 증가함에 따라 증가하였다. 따라서 본 연구 결과, GTE 첨가는 CFP 필름의 항산화, 항균효과를 가져다 준다고 판단된다.

추황배 청징배즙의 제조 및 항산화 활성 (Easy production techniques for clear pear juice and its antioxidant activities of 'Chuwhangbae' pear)

  • 본 연구는 배 과즙의 가공조건을 최적화하기 위해 여과, 청징 및 갈변 방지 방법 및 가공 조건에 따른 항산화능을 조사하였다. 배를 분쇄 착즙 후 면포여과, 감압여과 및 원심분리를 실시한 결과 원심분리(3000 rpm, 10분간)를 할 경우 colorimetry value가 높은 'L'값(34.31)과, 낮은 'a'값(-0.13)을 보여 가장 좋은 청징 효과를 나타냈으며, 그 다음으로 감압여과(31.8%) 및 면포여과(11.0%) 순이었다. 청징제 종류에 따른 청징 효과는 1%의 gelatin이나 bentonite가 우수하였으며, tannin과 egg albumin은 효과가 없었다. 갈변저해제 처리에 따른 갈색도 측정결과 0.1% L-ascorbic acid 처리 시 11.5%의 갈변억제 효과를 보였다. 유리당은 fructose, sucrose, glucose, sorbitol이 검출되었으며, 가열배즙에서 fructose와 glucose는 증가되는 반면 sucrose는 감소되었다. 가공 조건에 따른 항산화능 조사 결과 총폴리페놀 함량은 가열배즙이 청징배즙에 비해 유의하게 많아졌으나 총플라보노이드 함량은 청징배즙과 가열배즙이 유사하게 증가하였다. DPPH radical 소거활성과 아질산염 소거능은 가열배즙과 청징배즙에서 착즙액보다 유의적으로 높은 것을 확인할 수 있었다.
