Antimicrobial Effect, Antioxidant and Tyrosinase Inhibitory Activity of the Extract from Different Parts of Phytolacca americana L.

  • This study was conducted to clarify the antimicrobial effect, antioxidant and tyrosinase inhibitory activities of the biological composition having the Phytolacca americana, and to enhance the natural materials utilization of foods and cosmetics. The antimicrobial activities of the different parts of P. americana were evaluated using the agar diffusion test. The antimicrobial activity of P. americana was relatively high in Malassezia furfur known as a skin fungi and Vibrio parahaemolyticus compared to Escherichia coli and Staphy-lococcus epidermidis. However, the antimicrobial activity in Vibrio parahaemolyticus did not show at all parts of P. americana. Both the DPPH radical scavenging activity and ABTS radical scavenging activity have been increased with the higher concentration of methanol extract. In particular, leaf extract of P. americana exhibited the highest activity both ABTS radical scavenging activity and DPPH radical scavenging activity. The nitrite scavenging activity was decreased when the pH was changed from pH 1.2 to pH 6.0. The highest nitrite scavenging activity was exhibited from the methanol extract of fruit, followed by root, stem, and leaf at pH 1.2. However, the nitrite scavenging activity at pH of 6.0 was not almost detected. All plant parts of P. americana showed tyrosinase inhibitory activity. The highest activity was found in the stem, and followed by root, leaf, and fruit in order. These tyrosinase inhibitory activity was progressively increased in a concentration-dependent manner. In this experiment on the methanol extracts of different organ from P. americana, we confirmed that the extract of P. americana showed potent tyrosinase inhibitory activity. Taken together, we conjectured that the P. americana had the potent biological activities, therefore this plant having various functional components could be a good material for development into source of natural food additives and cosmetics.

감귤류 종자 추출물의 향균활성 (Antimicrobial Activity of Extracts from Citrus Seeds)

  • To develope natural food preservatives antimicrobial effect of the natural products against food-related bacteria and yeast The purpose of this study was evaluate antimicrobial effect of the citrus seeds. antimicrobial activities of methanol extracts from the citrus seeds investigate against Escherichia coli O26, Staphylococcus aureus 6358, Saccharomyces cerevisiae IBM 4274, Bacillus licheuiformis 9945a and Alcoligenes faecalis. Citrus seeds is containing to moisture 4∼6.5%, curd protein 11∼15%, curd lipid 32∼46%, curd carbohydrate 22∼45 % and ash 2∼4 %, that is containing to flavornoid 12∼48mg% and phenolic compound 22∼53mg%. Solidity content of the methanol extract from the citrus seeds was 0.8∼1.2%. Almost all of the methanol extracts from citrus seeds exhibited growth inhibiting activities for most of microorganisms tested. The methanol extracts from Citrus grandis, C. sunki, C. sulcata showed the growth inhibitory effects against Escherichia coli O26. The methanol extracts from C. obovoidea, C. sulcata, C. aurantium showed the growth inhibitory effects against Staphylococcus aureus 6358. The methanol extracts from C. obovoidea, C. sulcata, C. tangerina showed the growth inhibitory effects against Saccharomyces cerevisiae IBM 4274. The methanol extracts from C. obovoidea, C sunki, C. sulcata, C. tangerinan, C. natsudaidai, C. iyo, C. aurantium showed the growth inhibitory effects against Bacillus licheuiformis 9945a. The methanol extracts from C. obovoidea, C sunki, C. sulcata, C. aurantium showed the growth inhibitory effects against Alcoligenes faecalis. Among this especially, Showed growth inhibiting activity of the methanol extracts from Citrus sulcata that about microorganisms investigated. If apply searching suitable application method about such the citrus seeds antimicrobial activity, role as good antimicorbial material in storage or cooking of food, processing is expected.

식품위해성균, 피부사상균 및 식물성 병원균에 대한 산초유의 항균 활성 및 항산화 활성 (Antimicrobial Activity against Food-hazardous Microorganisms, Dermatophytes, and Pytopathogens and Antioxidative Activity of Sancho Oil)

  • Background: Although Sancho (Zanthoxylum schinifolium Siebold & Zucc) oil has traditionally been used for its antibiotics properties, there is currently a lack of scientific evidence regarding its biological activities. In this study, we investigated the antimicrobial and antioxidant activities of Sancho oil against food-hazardous microorganisms, phytopathogens, and dermatophytes. Methods and Results: We investiated the antimicrobial activity of Sancho oil against 11 food-hazardous microorganisms, nine phytopathogens, and six dermatophytes. The Sancho oil was found to show the strongest antibacterial activity against Shigella flexneri and Listeria spp. Sancho oil also showed high antifungal activity against plant pathogens, particularly Fusarium oxysporum, and showed antimicrobial activity against dermatophytes such as Trichophyton rubrum, Microsporum canis and Candida albicans. The antioxidant activity of Sancho oil was measured using the DPPH method, and was found to be stronger than that of unrefined oil. Moreover, this activity increased with increasing oil concentration. Conclusions: We found that Sancho oil showed differing antimicrobial activities against food-hazardous microorganisms, dermatophytes, and plant pathogens. The antimicrobial activity spectrum of Sancho oil was not broad and varied among microbial strains. On the basis of our findings, we consider that Sancho oil could be used an antibacterial material for food-borne S. flexneri and Listeria spp., a biopesticide for Fusarium spp., and a treatment for dermatophytes such as T. rubrum.

붉나무(Rhus chinensis Mill) 에탄올 추출물의 항균 및 항산화 활성에 대한 연구 (A Study on Antimicrobial and Antioxidant Activity of Ethanol Extract of Rhus chinensis Mill)

  • In this study, the Rhus chinensis Mill was divided into bark and inner bark. The antimicrobial activity and the antioxidative activity of the extracts were investigated by using the organic solvent fractions after the extraction and concentration with ethanol. This study showed the possibility of functional materials such as raw materials for cosmetics and food supplements. This study was the antimicrobial activity and antioxidant activity of five microorganisms (S. aureus, E. coli, B. subtilis, P. aeruginosa, and P. ovale). The following conclusions were obtained. First, the antimicrobial activity of B. subtilis was found to be high in the ethanol extract of. Second, DPPH scavenging activity was 86.4% free radical scavenging activity at 2.5 mg/mL bark part and 61.9% free radical scavenging activity at 2.5 mg/mL in inner Rhus chinensis Mill bark part. The ABTS scavenging activity was 79.2% free radical scavenging at 1 mg/mL bark fraction and 63% free radical scavenging activity at 1 mg/mL in inner bark, and bark showed higher antioxidant activity than inner bark. These results suggested that the antimicrobial activity and antioxidant activity of Rhus chinensis Mill extract can be used as a natural material. Specific and diverse physiological activity studies are expected in the future.

방사선 이용 항균성 천연추출물 함유 하이드로겔 제조 및 특성 연구 (Radiation Preparation and Characterization of Antimicrobial Hydrogels)

  • Taraxacum platycarpum (TP) has been used for years without restriction on usual dose for its non-toxic nature and nonexistence of the side effects. To develop antimicrobial hydrogel, poly (vinylalcohol) (PVA) hydrogels containing the aqueous extracted TP as an antimicrobial agent were prepared by using gamma-rays irradiation. The antimicrobial activities of the TP hydrogels were tested against Staphylococcus aureus and Staphylococcus epidermidis by disc diffusion method. The inhibition zones (IZ) of the TP extracts and TP hydrogels against S. aureus were 16 mm and 20 mm and against S. epidermidis was 10 mm and 13 mm, respectively. In conclusion, the TP hydrogel that has an excellent antimicrobial activity was proved to be a valuable material for functional skin care.

Antimicrobial activity of fermented Maillard reaction products, novel milk-derived material, made by whey protein and Lactobacillus rhamnosus and Lactobacillus gasseri on Clostridium perfringens

  • Objective: The objective of this study was to evaluate the antimicrobial effects of fermented Maillard reaction products made by milk proteins (FMRPs) on Clostridium perfringens (C. perfringens), and to elucidate antimicrobial modes of FMRPs on the bacteria, using physiological and morphological analyses. Methods: Antimicrobial effects of FMRPs (whey protein plus galactose fermented by Lactobacillus rhamnosus [L. rhamnosus] 4B15 [Gal-4B15] or Lactobacillus gasseri 4M13 [Gal-4M13], and whey protein plus glucose fermented by L. rhamnosus 4B15 [Glc-4B15] or L. gasseri 4M13 [Glc-4M13]) on C. perfringens were tested by examining growth responses of the pathogen. Iron chelation activity analysis, propidium iodide uptake assay, and morphological analysis with field emission scanning electron microscope (FE-SEM) were conducted to elucidate the modes of antimicrobial activities of FMRPs. Results: When C. perfringens were exposed to the FMRPs, C. perfringens cell counts were decreased (p<0.05) by the all tested FMRPs; iron chelation activities by FMRPs, except for Glc-4M13. Propidium iodide uptake assay indicate that bacterial cellular damage increased in all FMRPs-treated C. perfringens, and it was observed by FE-SEM. Conclusion: These results indicate that the FMRPs can destroy C. perfringens by iron chelation and cell membrane damage. Thus, it could be used in dairy products, and controlling intestinal C. perfringens.

나노 금속복합체의 박과 작물 종자 분리균에 대한 항균효과 (Antimicrobial Activities of Nano Metal Hybrid Materials against the Microorganisms Isolated from Cucurbit Seeds)

  • 이 연구는 플라즈마 기술 (radio frequency-thermal plasma system과 direct current sputtering system)을 이용하여 제작된 나노 금속복합체를 이용하여 박과 작물(수박, 호박, 박)의 종자에서 분리한 미생물의 항균 활성 효과를 검정하기 위하여 실험을 진행하였다. 8종의 나노 금속복합체와 4가지의 담체를 이용하여 5종의 곰팡이와 10종의 세균을 대상으로 기내 실험을 수행하였다. 그 결과, 곰팡이를 대상으로 한 항균실험에서 나노 금속복합체 Brass/CaCO3의 경우 1,000 ppm 농도에서 5종의 곰팡이에 대하여 100%의 항균효과를 나타내었다. 세균을 대상으로 한 항균실험의 경우 나노 금속복합체 Brass/CaCO3(1,000 ppm)은 Weissella sp., Rhodotorula mucilaginosa, Burkholderia sp. 그리고 Enterococcus sp. 4가지 세균을 100% 억제하는 것으로 확인되었다. 나노 금속복합체 G-Ni은 Rhizopus stolonifer에 대하여 100% 항균효과를 나타냈으며, 4가지 곰팡이에 대해서는 52.94-71.76% 정도의 항균효과를 나타내었다. 하지만 나노 금속복합체 G-Ni은 10종의 세균에 대해서는 효과가 없는 것으로 나타났다. 요약하면, 8가지 나노 금속복합체와 4가지 담체 중에서 Brass/CaCO3가 5종의 곰팡이와 4종의 세균 대하여 항균효과가 있었으며, G-Ni는 5종의 곰팡이에 대해서 52.94-100% 효과를 보였으나 세균에 대해서는 항균효과가 없는 것으로 확인되었다.

Casein 효소 가수분해물의 항균 활성과 그 응용 (Application and Antimicrobial Activities of Casein Hydrolysates Treated with Asp.oryzae Protease)

  • 단백 효소 가수분해물의 천연 항균제로서의 응용성을 검토하기 위하여 casein에 5종의 단백질 가수분해 효소를 작용시켜 얻어진 가수 분해물의 항균활성을 측정하고 활성이 가장 높은 가수분해물을 부분 정제하여 그 응용성을 검토하였다. Casein에 5종 단백질 분해효소를 작용시켜 얻은 가수분해물의 항균활성은 Aspergillus oryzae protease에 의한 것이 가장 높았다. 효소처리에 의하여 얻어진 가수분해 물을 30,000, 10,000, 3,000 membrane filter로 한외여과 하였을 때 항균활성은 3,000이하 분획물에 대부분 함유되어 있었으며 공시균주에 대한 최소저해농도는 $1.0\~1.5\;mg/mL$이었다. Aspergillus oryzae pretense로 작용시킨 casein 가수분해물은 $121^{\circ}C$에서 10분간 가열했을 때도 그 항균 활성을 유지하는 것으로 보아 열에 안정하였다. 가수분해물을 HPLC로 220 nm 280 nm에서 검출된 peak 별로 수집하여 항균활성을 측정 한 결과 retention time 12.6, 13.2 분에서 분취된 peptide가 활성이 있었다. 가수분해 동결건조물을 된장에 첨가하였을 때 미생물의 생육이 저해되었으며 실용성이 있었다.

니켈 코팅된 탄소나노튜브/산화아연 나노복합소재의 제조와 항균 및 기계적 특성 분석 (Preparation of Nickel Coated-carbon Nanotube/Zinc Oxide Nanocomposites and Their Antimicrobial and Mechanical Properties)

  • 본 연구에서는 탄소나노튜브, 니켈 및 산화아연의 항균특성 활용을 목적으로 새로운 항균 나노 복합 재료를 개발하였다. 니켈 코팅된 탄소나노튜브의 제조 및 그들의 동시발생 집중 및 비활성 세균에 대한 항균소재의 잠재적 응용성을 평가하였다. 제조된 니켈 도금 탄소나노튜브의 형상 및 니켈 도금은 전계 방출 주사전자현미경(FE-SEM) 및 X선 에너지 분산 분광기(EDS)를 사용하여 분석하였다. 나노복합소재의 항균 특성을 확인할 수 있는 타깃 세균은 황색포도상구균과 대장균으로 지정하였다. 니켈 도금된 탄소나노튜브와 산 처리된 탄소나노튜브로 각각 강화된 나노복합소재의 황색포도상구균 및 대장균에 대한 항균특성을 비교하였을 때, 니켈 도금된 탄소나노튜브/산화아연으로 강화된 나노복합소재의 항균 특성이 더 우수한 결과를 보이는 것을 확인할 수 있었다. 이러한 결과에 따라서, 탄소나노튜브 표면에 도금된 니켈이 복합소재 내에서 세균의 살균 작용에 중요한 역할을 하는 것으로 판단할 수 있다. 또한, 첨가된 산화아연은 나노복합소재의 항균특성 향상을 목적으로 제안되었다.

잘피(Zostera marina) 추출물의 항균효과에 대한 연구 (A Study of Antimicrobial Effect of Zostera marina Extracts)

  • 본 연구는 잘피 추출물의 화장품 소재로서 활용가능성을 평가하기 위해 건조된 잘피 전체, 뿌리, 잎 줄기를 70% 에탄올로 추출하여 항균활성을 확인하였다. 잘피 추출물의 항균활성은 Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Pseudomonas aeruginosa, Candida albicans, Propionibacterium acnes 균주로 disc diffusion 방법을 통해 생육저해환(clear zone)을 측정하였다. 그 결과 잘피 뿌리 추출물처리군에서 Staphylococcus epidermidis의 clear zone이 $13.00{\pm}0.50mm$로 확인되었고, Staphylococcus aureus, Propionibacterium acnes, Pseudomonas aeruginosa의 clear zone은 각각 $11.75{\pm}0.25mm$, $12.00{\pm}0.50mm$, $12.25{\pm}0.25mm$로 확인되어 잘피 추출물의 항균 효능을 확인할 수 있었다.
