Function of Blending Essential Oil in the Development of Anti-Dandruff Products

  • Purpose: In this paper, we show our blending ratio of 10 types of Essential Oils that survives beneficial bacteria and kills harmful bacteria in the scalp, and we investigate the possibility of application of our blending ratio to the development of anti-dandruff products and the possibility of being used as a raw material for clinical beauty and customized cosmetics. Methods: The scalp microorganisms used in our study were M. furfur, S. epidermidis, E. coli, and P. nitroreducens. There are a total of 10 Essential Oils such as True Lavender, Lime, Roman chamomile, Rosemary camphor, Cedarwood, Geranium, Clove, Tea tree, Palmalosa, and Peppermint. The antibacterial test of the blended Essential Oil was carried out according to the test method of the standardized evaluation methodology of "Food and Food Additives Code". Since M. furfur is related to the growth of sebum in the scalp, in this study we used the fnLNB and the fnLNA with 20 ㎖ of whole fat cow milk added. Results: The blending ratio of EO, which inhibits dandruff-causing bacteria such as M. furfur, S. epidermidis, E. coli, and does not inhibit P. nitroreducens showing dominant growth in a healthy scalp, was B8(Clove 0.2%, Roman chamomile 0.5%, Tea tree 0.3%), B9(Geranium 0.1%, Palmarosa 0.1%, Roman chamomile 0.5%, Tea tree 0.3%), B10(Clove 0.1%, Geranium 0.1%, Palmarosa 0.1%, Roman chamomile 0.5%, Tea tree 0.2%). Conclusion: It is thought that the blending ratio of BEO obtained as a result of this study can provide a basis for use as an alternative to antibiotics in developing anti-dandruff drugs and emerge as a new alternative to solve scalp microbial imbalance. In order for EO to be used as a useful raw material for anti-dandruff preparation, researches on 1) Standardization (the effects of products differ according to the types, regions, climate, extraction methods, etc.), 2) Antimicrobial effects, 3) Safety, etc., must be established.

  • Unstable cosmetic active ingredients could be degraded rapidly by chemical and photochemical process. Particularly, some of active ingredients like retinol are known to cause skin irritation when applied on the skin excessively. Therefore, it has become a very important issue to encapsulate cosmetic actives for the stabilization and skin protection. This study was performed in order to prepare a chitosan microcapsule containing liposoluble cosmetic actives and to investigate the stabilization effect of actives when chitosan microcapsule was applied in cosmetic formulation. Chitosan, deacetylated form of chitin, has been of interest in the industrial applications due to its biocompatibility, biodegradability, non-toxicity, antimicrobial activity and also used as a wall material of capsule. Retinol was used as a core material and was stabilized by a wall of chitosan and antioxidants. The chitosan microcapsule containing retinol(CMR) was prepared by using coacervation method and W$_1$/O/W$_2$ emulsification techniques. The CMR has 0.5~10.0 ${\mu}{\textrm}{m}$ size distribution and a long-term stability of more than an year inside the cosmetic formulation(O/W). Remaining retinol percentages at 45$^{\circ}C$ after 8 weeks in the CMR dispersion were 15.6%(pH 4.0), 59.8%(pH 6.0) and 65.0%(pH 6.0 with antioxidant) respectively. Retinol stability when added CMR inside a ONV emulsion was better than that of ONV emulsion added non-capsulated retinol. As a result, remaining retinol at 45$^{\circ}C$ after 8 weeks in O/W emulsion added non-capsulated retinol and O/W emulsion containing CMR was 12.7%, 70.5% respectively. It appeared that chitosan treated microcapsule may be used for a potential encapsulation method of unstable active ingredients.

쿼세틴 담지 나노캡슐을 함유한 알지네이트 스펀지의 제조 및 생리활성 특성 (Preparation and Study of Bioactive Characteristics of Alginate Sponge Containing Quercetin-encapsulated Nanocapsules)

  • Quercetin is one of flavonoids widely distributed in the plants and well known to have antioxidants, antiinflammatory, antimicrobial properties. In this study, alginate sponge containing quercetin-encapsulated nanocapsules was prepared by miniemulsion polymerization, dyring/crosslinking method and their bioactive characteristics were investigated. Alginate sponge containing quercetin-encapsulated nanocapsules were evaluated using a field emission scanning electron microscope(FE-SEM), a high performance liquid chromatography, cell viability, DPPH radical scavenging activity and antibacterial activity. The study indicates that alginate sponge containing quercetin-encapsulated nanocapsules had significant antioxidant, antiinflammatory and antibacterial activities. This study suggested that alginate sponge containing quercetin-encapsulated nanocapsules can be a potential candidate for medical materials.

Antibacterial Effect of Fructose Laurate Synthesized by Candida antarctica B Lipase-Mediated Transesterification

  • Sugar esters are valuable compounds composed of various sugars and fatty acids that can be used as antibacterial agents and emulsifiers in toothpaste and canned foods. For example, fructose fatty acid esters suppress growth of Streptococcus mutans, a typical pathogenic bacterium causing dental caries. In this study, fructose laurate ester was chosen as a target material and was synthesized by a transesterification reaction using Candida antarctica lipase B. We performed a solvent screening experiment and found that a t-butanol/dimethyl sulfoxide mixture was the best solvent to dissolve fructose and methyl laurate. Fructose laurate was synthesized by transesterification of fructose (100 mM) with methyl laurate (30 mM) in t-butanol containing 20% dimethyl sulfoxide. The conversion yield was about 90%, which was calculated based on the quantity of methyl laurate using high-performance liquid chromatography. Fructose monolaurate (Mr 361) was detected in the reaction mixture by high-resolution mass spectrometry. The inhibitory effect of fructose laurate on growth of oral or food spoilage microorganisms, including S. mutans, Bacillus coagulans, and Geobacillus stearothermophilus, was evaluated.

Anticaries Activity of Antimicrobial Material from Bacillus alkalophilshaggy JY-827

  • The present study was performed to investigate the excellent microbial anticaries substance, aminoglycoside antibiotic, which is more effective than chlorhexidine for the treatment of dental caries. The aminoglycoside antibiotic against Streptococcus mutans JC-2 from a novel alkaliphilic Bacillus alkalophilshaggy JY-827 exhibited no significant difference at the treatment concentration of $2.5{\times}10^{-7}M$, however, it inhibited the activity of the Streptococcus mutans glucosyltransferase by 70.2% and 99.8% at the concentrations of $2.5{\times}10^{-7}$M\;and\;2.5{\times}10^{-6}M$, respectively. Lineweaver-Burk plot of the inhibitory aminoglycoside antibiotic showed competitive inhibition, with $K_i$ value of $6.4{\times}10^{-6}$ M. The aminoglycoside antibiotic did not show any cytotoxicity against human gingival cells. To evaluate the industrial applicability of the aminoglycoside antibiotic, a toothpaste containing this substance was prepared and tested on the extracted human teeth. The inhibitory rate of tooth calcification and calcium ion elution by the aminoglycoside antibiotic were 50% and 2.5 times, respectively. These results suggested that the aminoglycoside antibiotic from Bacillus alkalophilshaggy JY-827 is an effective agent against dental caries.

Targeted Nanomedicine that Interacts with Host Biology

  • Nanotechnology is of great importance to molecular biology and medicine because life processes are maintained by the action of a series of molecular nanomachines in the cell machinery. Recent advances in nanoscale materials that possess emergent physical properties and molecular organization hold great promise to impact human health in the diagnostic and therapeutic arenas. In order to be effective, nanomaterials need to navigate the host biology and traffic to relevant biological structures, such as diseased or pathogenic cells. Moreover, nanoparticles intended for human administration must be designed to interact with, and ideally leverage, a living host environment. Inspired by nature, we use peptides to transfer biological trafficking properties to synthetic nanoparticles to achieve targeted delivery of payloads. In this talk, development of nanoscale materials will be presented with a particular focus on applications to three outstanding health problems: bacterial infection, cancer detection, and traumatic brain injury. A biodegradable nanoparticle carrying a peptide toxin trafficked to the bacterial surface has antimicrobial activity in a pneumonia model. Trafficking of a tumor-homing nanoprobes sensitively detects cancer via a high-contrast time-gated imaging system. A neuron-targeted nanoparticle carrying siRNA traffics to neuronal populations and silences genes in a model of traumatic brain injury. Unique combinations of material properties that can be achieved with nanomaterials provide new opportunities in translational nanomedicine. This framework for constructing nanomaterials that leverage bio-inspired molecules to traffic diagnostic and therapeutic payloads can contribute on better understanding of living systems to solve problems in human health.

Antimicrobial Activity of Vaccinium macrocarpon (Cranberry) Produced Proanthocyanidin (PAC) on the Growth and Adhesion Properties of Staphylococcus aureus

  • Cranberries have long been used by lay people to relieve the symptoms of urinary tract infections. Recent research has determined that the component of cranberry called proanthocyanidin (PAC) is the primary mechanism for inhibiting P-fimbriated E.coli adhesion to uroepithelial cells in vitro. A series of experiments were performed to determine the effects of PAC on growth and adhesion of uropathogenic E. coli and Staphylococcus aureus to urinary catheter material. The results showed that PAC-inhibited binding of Gram positive S. aureus to collagen coated surfaces and significantly decreased the growth of these bacteria. P-fimbriated E.coli did not bind well to the biomaterial and their growth was unaffected by the cranberry extract with the exception of some loss in viability at 1000 $\mu\textrm{g}$/mL after 5 to 18 hours of exposure. This is the first report of the potential for cranberries to interfere with the adhesion and growth of S. aureus, a multi-drug resistant organisms responsible for morbidity and mortality especially in hospitalized patients.

발효녹차액을 이용한 모발의 트리트먼트 효과 (A Study of Effects of Fermented Green Tea Extract-based Treatment on Hair)

  • This study attempted to determine the effects of fermented green tea extracts on hair by using them in a perm. For this, hair is bleached, and then the fermented green tea extracts and water were supplied to the hair using a mist sprayer. Then the hair was permed, and hair damage was tested. Regarding hair moisture levels and wave formation, specifically, the morphological changes of hair were investigated with the Scanning Electron Microscope(SEM). The study results found the following. In terms of wave formation, the fermented green tea extract-based perm was much better than the general perm. In addition, hair bleaching was more serious in the latter. According to analysis on hair moisture using SEM, hair damage was more prevalent in the general perm as well. Therefore, it was confirmed that green tea extract-based perms cause less damage to the cuticle. In other words, the potential of fermented green tea extracts as a hair cosmetics material was discovered. Furthermore, it appears that the study results contribute to the development of low-irritating scalp and hair care products using the anti-inflammatory and antimicrobial properties of the fermented green tea extracts.

Subacute Oral Toxicity of Chitosan Oligosaccharides on Sprague Dawley Rats

  • Chitosan is derived from chitin by deacetylation in the presence of alkali, which is a copolymer consisting of $\beta$-(1longrightarrow4)-2-acetamido-D-glucose and $\beta$-(1longrightarrow4)-2-amino-D-glucose units with the latter usually exceeding 80% (Arvanitoyannis et al., 1998). Chitosan has been developed as new physiological material since it possesses antibacterial activity, hypocholesterolemic activity and antihypertensive action. However, even though chitosan has very strong functional properties in many areas, its high molecular weight and high viscosity may restrict the use in vivo. In addition, there is little doubt that such properties will influence absorption in the human intestine. Recently, studies on chitosan have attracted interest for converted chitosan to oligosaccharide, because the oligosaccharide possesses not only water-soluble property but also versatile functional properties such as antitumor activity, immune-enhancing effects, enhancement of protective effects against infection with some pathogens in mice, antifungal activity, calcium absorption accelerating effect (Jeon et al., 1999) and antimicrobial activity. There is, however, little information on the toxicity of chitosan oligosaccharide. (omitted)

In Vitro Virucidal Effect of Mouthrinse Containing C31G on Seasonal Influenza Viruses

  • C31G is a potent antimicrobial agent and can disrupt the microbial membrane by the alkyl portion of the molecule. The objective of this study was to evaluate the virucidal effectiveness of C31G and mouthrinse containing C31G (Sense-Time) on seasonal influenza viruses. Evaluation of the virucidal activity against influenza viruses was performed with end-point titration in 10-day-old chicken embryos and Madin-Darby canine kidney cells. In vitro studies demonstrated that C31G and Sense-Time inhibited the growth of seasonal influenza viruses even in the presence of 5% organic material. Gargling with C31G or Sense-Time would enhance oropharyngeal hygiene, which would be helpful for reducing influenza transmission.
