Synthesis and Antihypertensive Activity of Certain Mannish Bases of 2- Ethoxycarbonylindoles and 5H-Pyridazino [4,5-b] indoles

  • This manuscript reports the synthesis of two series of Mannich Bases 3-12 and 21-40 obtained respectively by the reaction of either 2- ethoxycarbonylindoles 1-2 or 5H-pyridazino [4,5-b]indoles 17-20 as a substrate with formalin and the appropriate $2^{\circ}C$amines under the suitable Mannich conditions. fourteen of the synthesized Mannich basese were screened as antihypetensive agents in normotensive anesthetized rats. The effect of compound 4 in normotensive anesthetized dogs was also studied.

Hypertensive Encephalopathy with Reversible Brainstem Edema

  • Presented here is a 36-year-old male with arterial hypertension who developed brainstem edema and intracranial hemorrhage. Magnetic resonance scan revealed diffuse brainstem hyperintensity in T2-weighted and fluid-attenuated inversion-recovery images, with an increase in apparent diffusion coefficient values. After a reduction in blood pressure, rapid resolution of the brainstem edema was observed on follow-up. The patient's condition was thus interpreted as hypertensive brainstem encephalopathy. While many consider this a vasogenic phenomenon, induced by sudden, severe hypertension, the precise mechanism remains unclear. Prompt recognition and aggressive antihypertensive treatment in such patients are essential to prevent permanent or life-threatening neurologic injury.

  • To investigate the effects of N-4-salicyloamide-2-amino-6-piperidinopyrimidine 3-oxide (SALMI) on the blood pressure, this study was carried out in rats, using ethanol as vehicle. When Salmi and its salt were administered into the rat by oral administration or i.v. injection, both of them decreased blood pressure. But when Salmi was administered not more than 200 mg/kg(oral) and 30 mg/kg(i.v.) and Salmi·HC1 was administered not more than 100 mg/kg(oral), they did not significantly influence on the depressor effects of blood pressure.

  • It was previously indicated that 'Samulanshintang' had been examined for effects of the anticonvulsive, antipyretic, analgesic, sedative and antispasmodic actions. In order to investigate the effect on the cardiovascular system and anemic rabbits induced by phenlhydrazine HCl, pharmacological studies have been carried out with extract. The results of this investigation were summarized as follows: The relaxation of bronchial smooth muscle and antihistamine action were recognized. Antihypertensive and vasodilating actions due to vascular smooth muscle relaxation were noted in frogs and rabbits. 'Samulanshintang' lowered the rate of anemia induced by phenylhydrazine and rapidly recovered from the anemia state.

  • These studies were conducted to investigate the effect of 'Inchungbaek' water extract on analgesic and sedative actions, the relaxing action of isolated ileums, vasodilating action and blood pressure. The results of these studies were summarized as follows: Analgesic and sedative actions of 'Inchungbaek' were recognized in mice and rats. The relaxing action of isolated ileums were seen in mice and rabbits, antagonist action was seen acetylcholine and $BaCl_2-induced$ contraction of isolated ileums, and then recognized direct action in the ileum smooth muscle. Hypotensive and vasodilating actions due to vascular smooth muscle relaxation were noted in rabbits and cats.

  • Chitin, a poly $\beta$-(1longrightarrow14)-N-acetyl-D-glucosamine, is best known as a cell wall component of fungi and as a skeletal materials of invertebrates. Chitosan is derived from chitin by deacetylation in the presence of alkali. Chitosan has been developed as new physiological materials since it possesses antibacterial activity, hypocholesterolemic activity and antihypertensive action. However, the actions of chitosan in vivo still remain ambiguous as the physiological functional properties because most animal intestines, especially the human gastrointestinal tract, do not possess enzyme such as chitosanase which directly degrade the $\beta$-glucosidic linkage in chitosan, and consequently the unbroken polymers may be poorly absorbed into the human intestine. Therefore, recent studies as chitosan have attracted interest for chitosan oligosaccharides, because the oligosaccharides process not only water-soluble property but also versatile functional properties such as antitumor activity, immune-enhancing effects, enhancement of protective effects against infection with some pathogens in mice and antimicrobial activity (Kingsnorth et al., 1983, Mori et al., 1997). Effects of Siegesbeckia Glabrescens on the Vascular Relaxation and Antioxidative Status (희렴의 혈관이완 효능과 항산화 동태에 관한 연구)

  • This study investigated the effects of Siegesbeckia glabrescens, an antihypertensive remedy, on the contraction evoked by phenylephrine and KCl in isolated rat thoracic arata, and also analyzed antioxidative status in vitro. Siegesbeckia glabrescens revealed dose-dependent relaxation on phenylephrine(PE)/KCl-induced arterial contraction and more markedly on PE-induced contraction. Siegesbeckia glabrescens reduced malondialdehyde(MDA)levels, Phosphatidyl choline-liposome(PC-OOH) contents, linoleic acid-induced lipid peroxidation and exerted 1,1-diphenyl-2- picryl-hydrazyl(DPPH) radical scavenging effect, in vitro. These results indicated that Siegesbeckia glabrescens doesn't relaxe artery through a blocking α-adrenergic receptor and calcium channel mediated by voltage-operated calcium channel, and it s antioxidative effects may be involved in endothelium-dependent relaxation of arteries via vascular protective properites. Growth Characteristics and Physiological Functionality of Yeasts in Pear Marc Extracts

  • Kluyveromyces fragilis KCTC 7260 and Saccharomyces cerevisiae KCTC 7904, which both grew well in pear marc extract, were selected and their growth profiles and physiological functionalities were determined. Both of the selected yeasts established maximal growth by 20 hr of cultivation at $30^{\circ}C$ in pear marc extract. The cell-free extracts showed high antihypertensive angiotensin I-converting enzyme inhibitory activity of 68.9% and 52.1%, respectively. The extracts also displayed 9.2 U/mL and 12.0 U/mL of protease activity, respectively.

  • The present study examined the effect of alcohol extract of Isaria sinclairii on blood pressure in spontaneously hypertensive rats (SHR). The blood pressure and heart rate were measured after treatment of alcohol extract of I. sinclairii by indirect tail cuff method and direct tn vivo model. Male SHR were treated with extracts for 2 or 4 weeks starting at 12 weeks of age. (omitted)

  • In cardiovascular disease the flow adaptation of erythrocytes can be affected by reduced shear stresses and metabolic influences on red cell fluidity as a consequence of tissue hypoxia. In addition there are indications that risk factors of cardiovascular diseases are able to decrease the intrinsic red cell deformability. Erythrocyte deformability was studied by the filtration technique of Reid et al. to investigate the relationship between blood pressure chances and erythrocyte deformability. In this experiment normotensive rats, spontaneously and DOCA-salt treated hypertensive rats were used. Erythrocyte deformability was significantly reduced by blood pressure elevation in hypertensive rats but was not fully recovered by normalization of blood pressure after antihypertensive drug treatment. Therefore other factors than blood pressure may be involved in erythrocyte deformability reduction during blood pressure elevation.
