Antigastritic Effect of Carbenoxolone Disodium from Glycyrrhizae Radix (감초성분 Carbenoxolone Disodium의 항위염 효과)

  • Glycyrrhizae Radix, the dried roots of Glycyrrhiza glabra or Glycyrrhiza uralensis Fischer(Legumino-sae), has been used as a medicine for treatment of imflammation, arthritis, respiratory ailment, skin diseases and liver problems. The purpose of this study was to examine the effect of 70% ethanol extract, 18-${\beta}$-glycyrrhetinic acid, glycyrol and carbenoxolone disodium from Glycyrrhizae Radix on gastritis and gastric cancer. Using these materials, we tested antibacterial activity against Helicobacter pylori, antigastritic activity for HCI-ethanol-induced gastric lesion and the pylorus ligated gastric secretion with rats, and cell viability in gastric cancer cell. 18-${\beta}$-glycyrrhetinic acid and carbenoxolone disodium decreased the volume of gastric secretion and acid output in pylorus ligated rats. Also, carbenoxolone disodium had a strong effect of antibacterial activity on H. pylori. In addition 18-${\beta}$-glycyrrhetinic acid and glycyrol reduced cell viability in human gastric cancer cells(AGS and SNU638 cell) in dose-dependent manner. The reduction of total acid output and gastric secretion as well as the anti-bacterial activity against H. pylori might account for the antigastritic effects of carbenoxolone disodium.

  • Scutellaria Radix, the dried roots of Scutellaria baicalensis Georgi (Labiatae), has been used in oriental traditional medicine for treatment of fever urine disorder, diarrhea, inflammation. Present study was carried out for the gastroprotective effect of chrysin, baicalein, baicalin, wogonin from Scutellaria baicalensis. This reports evaluated antioxidant effect, antibacterial activity against Helicobacter pylori and HCI-ethanol-induced gastric lesion in rats and showed the significant effectiveness. It may be regarded that the antigastritic effects and antibacterial activity of baicalin, baicalein from Scutellaria baicalensis are originated from acid-neutralizing capacity, free radical scavenging effects and the antibacterial activity against Helicobacter pylori.

Antigastritic and Antiulcerative Action of the Extract of Zingiberis Rhizoma (생강추출물의 항위염 . 항궤양 작용)

  • The rhizoma of Zingiber officinale has been used as antiemetic, expectorants, stomachache relieving drugs and digestive accelerators. From the observation of antigastritic action of the methanol extract of the rhizoma, it was fractionated with hexane, chloroform, ethyl acetate and butanol, followed by bioassay on antigastritic and antiulcerative activity. The hexane and the chloroform fraction reduced significantly HCl ethanol induced gastric lesion at the dose of 370 and 210 mg/kg, p.o., respectively. On the gastric ulceration and gastric secretion in pylorus-ligated rats, the hexane fraction decreased the volume of gastric secretion and acid output, and also increased pH at the dose of 370 mg/kg, i.d.. It showed considerable curative ratio of acetic acid induced ulcer without inhibition of indomethacin induced gastric lesion. The methanol extract showed low acute toxicity with minimum lethal dose of more than 3000 mg/kg, p.o. in mice. In conclusion, Zingiberis rhizoma exhibited antigastric and antiulcerative activity which might be attributable to inhibition of gastric secretion. It is revealed that the active component may be present in the hexane fraction.

Antigastritic and Antiulcer Actions of the Extract of Head of Panax ginseng Radix (인삼노두 추출물의 위염 및 위궤양에 대한 효과)

  • In a preliminary screening of plant extracts for the antigastritic and antiulcer actions in rats, the extracts of head of Panax ginseng Radix showed positive activity in HCl ethanol-induced gastric lesion. Among the systematic fractions of hexane, chloroform, butanol and water, the most potent butanol fraction reduced significantly HCl ethanol-induced gastric lesion at the oral dose of 500 mg/kg. In pylorus ligated rats, hexane and butanol fraction showed decreases in the volume of gastric secretion and acid output, of which effects were stronger in butanol fraction. Further assays with butanol fraction disclosed that it significantly suppressed the aspirin-induced and Shay ulcer. The butanol fraction at the intraduodenal dose of 500 mg/kg showed significant stimulation of mucus secretion.

  • In a preliminary screening of the plant extracts for the antigastritic action in rats, the extract of Aralia elata(Araliaceae) showed positive activity in HCI plus ethanol induced gastric lesion. Systematic fractions with hexane, chloroform, ethyl acetate and butanol resulted in the most patent activity with the butanol fraction: This butanol fraction at the oral dose of 200 mg/kg exhibited significant inhibition of absolute alcohol induced gastric lesion which was more potent than 100 mg/kg of cimetidine and had significant stimulation of mucus secretion. The butanol fraction showed significant decreases in the ulcer indices of Shay ulcers and inhibition of gastric juice secretion with acid output in pylorus-ligated stomachs of rats. It also suppressed the acetic acid induced gastric ulcer. These results might suggest that the butanol fraction had inhibitory action in gastric lesion and ulceration through inhibition of gastric acid secretion and stimulation of mucin secretion in the stomachs of rats.

  • The methanol extract of Taraxaci Herba was found to have inhibitory effect on the gastric lesion induced by HCl-ethanol. The systematic fractionation of the methanol extract resulted in positive action with water fraction in the gastric lesion. It also showed significant inhibition of gastric lesion induced by HCl-aspirin and absolute ethanol, but did not prevent indomethacin induced gastric lesion. This fraction did not affect basal gastric acid secretion but showed a decrease of pepsin activity in pylorus-ligated rats.

  • Eupatilin is a major active component of Stillen\ulcorner(Artemisia Herba Extract) having a potent antigastritic effect. We investigated the physical properties of eupatilin using high performance liquid chromatography. Solubility, stability & partition coefficient of eupatilin were investigated. pH-stability of eupatilin was examined over the broad range through pH1-9 at 37$^{\circ}C$ & it has good stability above the broad range pH. The solubility of eupatilin was extremely low but the value of logP was more than 2. Also, a high performance liquid chromatographic method was developed for the determination of eupatilin in rat plasma. (omitted)

  • Sophorae Radix, the dried roots of Sophora flavescens Aiton (Leguminosae), has been used in oriental traditional medicine for treatment of skin and mucosal ulcers, sores, gastrointestinal hemorrhage, diarrhea, inflammation and arrhythmia. Present study was carried out for the gastroprotective effect of trifolirhizin from Sophora flavescens. This reports were evaluated antibacterial activity against Helicobacter pylori and HCl ethanol-induced gastric lesion in rats and showed the significant effectiveness. In pylorus ligated rats, the treatments of trifolirhizin showed decrease in the volume of gastric secretion and acid output. Also we evaluated the antibacterial activity against H. pylori treated with methanol extract and trifolirhizin from Sophora flavescens had a equivalent antibacterial activity with ampicillin against H. pylori at the dose of $100{\mu}g/mL$. It may be regarded that the antigastritic effects and antibacterial activity of trifolirhizin from Sophora flavescens are originated from reduction of total acid output identified by gastric secretion reduce, free radical scavenging effects and the antibacterial activity against H. pylori.
