Miconazole과 Amphotericin B의 항진균 활성에 대한 Anethole의 선택적 병용 효과 (Selective Combination Effect of Anethole to the Antifungal Activities of Miconazole and Amphotericin B)

  • The combination effect of anethole with amphotericin B, fluconazole, miconazole, or 5-fluorocytosine was investigated against Saccharomyces cerevisiae. When combined with $\frac{1}{2}$ minimum inhibitory concentration (MIC) or $\frac{1}{2}$ minimum fungicidal concentration (MFC) of anethole, the antifungal activities of fluconazole and 5-fluorocytosine were not changed, but the fungistatic and the fungicidal activities of miconazole were increased 64-fold, respectively. In the case of amphotericin B, the fungistatic activity was increased 2-fold, while the fungicidal activity was decreased 2-fold. The combination effect of anethole with miconazole or amphotericin B was also investigated at the various concentrations using the macrobroth dilution checkerboard method. The fractional inhibitory concentration (FIC) and the fractional fungicidal concentration (FFC) index between B exhibited the FIC index of 8.25 and the FFC of 32.06, respectively. Thus, it is analyzed that the combination of anethole with miconazole or amphotericin B on the antifungal action shows synergism and antagonism, respectively.

천연물 Polygodial과 Imidazole계 화합물의 병용에 의한 항진균 활성의 증진 (Synergistic Effect of Polygodial with Imidazole Drugs on the Antifungal Activity)

  • The fungistatic and fungicidal activities of amphotericin B, fluconazole, miconazole, econazole, and 5-fluorocytosine against Saccharomyces cerevisiae were estimated in the presence of 1/2 minimum inhibitory concentration (MIC) and 1/2 minimum fungicidal concentration (MFC) of polygodial, respectively. Among them, the antifungal activities of miconazoles by polygodial was still shown against several yeast-like fungi including Candida albicans, Candida utilis, Cryptococcus neoformans, except for Candida krusei. The combination of polygodial with imidazole drugs against Saccharomyces cerevisiae was further examined using the macrobroth dilution checkerboard method. The fractional inhibitory concentration (FIC) and the fractional fungicidal concentration (FFC) index between polygodial and miconazole were 0.16 and 0.19, while the combination of polygodial with econazole exhibited the FIC index of 0.19 and the FFC of 0.25, respectively. These results suggest that polygodial and the imidazoles on the fungistatic and fungicidal action are highly synergistic.

수종 허브정유의 Candida속 진균에 대한 억제 활성 및 Ketoconazole과의 병용효과 (Antifungal Activities of Herbal Essential Oils and Combination Effects with Ketoconazole against Candide spp.)

  • The antifungal activities of the essential oils from Anthemis nobilis, Ciderus atlantica, Juniperus communis, Lavandula angustifolia, Pelargonium graveolens, Pogestemon patchouli, Rosmarinus officinalis, and Styrax tonkinensis which are recommended for the treatment of microbial infections in aromatherapy and complementary medicines were tested against Candida spp. The activities were measured by broth dilution method and disk diffusion assay. Most of the test oils inhibited growth of Candida albicans, C. utilis and C. tropicalis. Especially, the essential oil from Pelargonium graveolens and its main component, citronellol showed the strongest activity among the herbs except benzoic acid from Styrax tonkinensis which is well-known antimicrobial compound. As a result of checkerboard microtiter test. synergistic effect of citronellol, was shown when the component was combinated with ketoconazole, displaying a fractional inhibiting concentration (FIC) index of 0.37 against C. albicans.

Screening of nanoemulgels for physicochemical stability and antifungal efficacy

  • The nanoemulgel was prepared to induce a synergistic effect along with higher efficacy. Nine sets of macroemulsion were made in which liquid paraffin was stabilized by the two non-ionic surfactants, Tween® 80 and Span® 80. Comparative stability analysis of the macroemulsions was used to determine the effective surfactant concentrations that gave the most stable systems (NE 2, NE3, NE4, NE5). High-speed homogenization was then applied. The final formulation was evaluated for globule size and polydispersablity index, physical properties (color, homogeneity, consistency, syneresis), pH, viscosity, spreadability with 200 g and 500 g weight, conductivity, drug content, stability, skin irritation, antifungal efficacy. Zeta size analysis confirmed the nanosize of the droplets in NE2 (284.8 nm), NE3 (79.89 nm), NE4 (194 nm) but not NE5 (632.8 nm), which was outside the nanoemulsion range. The antifungal assay exhibited zone of inhibition for NE3 (43±1.0 mm) and NE4 (42±1.7 mm), a marketed cream (33±1 mm), fluconazole alone (35±1 mm) and terbinafine alone (35.0±1.7 mm). The zone of inhibition of nanoemulgels increased compared with the drugs when used individually and when compared a placebo.

수목추출물의 생리활성에 관한 연구(VII) - 산벚나무 심재 추출성분의 항균 및 항산화활성 - (Studies on Biological Activity of Wood Extractives(VII) - Antimicrobial and Antioxidation Activities of Extractives from the Heartwood of Prunus sargentii -)

  • 천연물 유래의 보다 안전하고 활성이 우수한 항균 및 항산화물질을 탐색할 목적으로 산벚나무 심재로부터 분리한 pinostrobin, eriodictyol, naringenin. pinocembrin, taxifolin, verecundin의 항균 및 항산화활성을 검정 하였다. 배지점적법을 이용하여 항진균활성을 조사한 결과, pinocembrin(5,7-dihydroxyflavanone)이 공시균주 모두에 대해 80% 이상의 균사생장억제율을 나타내어 가장 활성이 우수한 것으로 나타났으며 그 다음이 naringenin(4',5,7-trihydroxyflavanone), 그리고 verecundin(7-hydroxyflavanone 5-O-${\beta}$-D-glucopyranoside)은 활성이 없는 것으로 나타났다. 특히, pinocembrin의 경우 현재 천연물로부터 분리되어 실용화되고 있는 항균물질인 히노키치올에 비견될 수 있는 항진균활성을 나타내었다. 한천배지확산법을 이용한 항세균활성 조사에서는 단리물질 모두 생육저지환을 형성하지 않아 활성이 없는 것으로 나타났다. 또한, 항산화활성 검정을 위해 프리라디칼소거능 및 과산화물가를 조사한 결과, taxifolin(3,3',4',5,7-pentahydroxyflavanone)과 eriodictyol(3',4',5,7-tetrahydroxyflavanone)이 천연항산화제인 ${\alpha}$-tocopherol의 2배의 프리라디칼소거능을 나타내었으며, 항산화지수도 천연항산화제보다 높은 것으로 나타나 항산화활성이 우수한 것으로 판단되었다. 이상의 결과 산벚나무 심재 에탄올 추출물의 높은 항균활성은 pinocembrin에, 항산화활성은 taxifolin과 eriodictyol에 기인하는 것으로 사료되었다.

Diversity and Antifungal Activity of Endophytic Fungi Associated with Camellia oleifera

  • Endophytic fungi strains (n = 81) were isolated from the leaves, barks, and fruits of Camellia oleifera from Hunan province (China) to delineate their species composition and potential as biological control agents of C. oleifera anthracnose. The fungi were identified by morphological and phylogenetic analyses. Fungal colonization rates of the leaves, barks, and fruits were 58.02, 27.16, and 14.81%, respectively. The isolates were identified as 14 genera, belonging to two subdivisions, Deuteromycotina and Ascomycotina; 87.65% of all isolates belonged to Deuteromycotina. The dominant species, occurring with a high relative frequency, were Pestalotiopsis sp. (14.81%), Penicillium sp. (14.81%), and Fusarium sp. (12.35%). The Simpson's and Shannon's diversity indices revealed the highest species diversity in the leaves, followed by the barks and fruits. The similarity index for the leaves versus barks comparison was the highest, indicating that the number of endophytic fungal species shared by the leaves and barks was higher than barks and fruits or leaves and fruits. Based on the results of dual culture experiments, only five strains exhibited antifungal activity against C. oleifera anthracnose pathogen, with isolate ty-64 (Oidium sp.) generating the broadest inhibition zones. Our results indicate that the endophytes associated with C. oleifera could be employed as natural agents controlling C. oleifera anthracnose.

Evaluation of Ciclopirox as a Virulence-modifying Agent Against Multidrug Resistant Pseudomonas aeruginosa Clinical Isolates from Egypt

  • Targeting the pathogen viability using drugs is associated with development of drug resistance due to selective pressure. Hence, there is an increased interest in developing agents that target bacterial virulence. In this study, the inhibitory effect of ciclopirox, an antifungal agent with iron chelation potential, on the microbial virulence factors was evaluated in 26 clinical MDR Pseudomonas aeruginosa isolates collected from Alexandria Main University Hospital, a tertiary hospital in Egypt. Treatment with 9 ㎍/ml ciclopirox inhibited the hemolytic activity in 70% isolates, reduced pyocyanin production, decreased protease secretion in 46% isolates, lowered twitching and swarming motility, and decreased biofilm formation by 1.5- to 4.5-fold. The quantitative real-time PCR analysis revealed that treatment with ciclopirox downregulated the expression levels of alkaline protease (aprA) and pyocyanin (phzA1). Ciclopirox is used to treat hematological malignancies and the systemic administration of ciclopirox is reported to have adequate oral absorption with a satisfactory drug safety profile. It is important to calculate the appropriate clinical dose and therapeutic index to reposition ciclopirox from a topical antifungal agent to a promising virulence-modifying agent agent against P. aeruginosa, a problematic Gram-negative pathogen.

Effect of Acaromyces Ingoldii Secondary Metabolites on the Growth of Brown-Rot (Gloeophyllum Trabeum) and White-Rot (Trametes Versicolor) Fungi

  • We investigated the antifungal activities of an endophytic fungus identified as Acaromyces ingoldii, found on a loblolly (Pinus taeda L.) pine bolt in Louisiana during routine laboratory microbial isolations. The specific objectives were to determine the inhibitory properties of A. ingoldii secondary metabolites (crude extract) on the mycelial growth of a brown-rot fungus Gloeophyllum trabeum and a white-rot fungus Trametes versicolor, and to determine the effective concentration of A. ingoldii crude preparation against the two decay fungi in vitro. Results show the crude preparation of A. ingoldii from liquid culture possesses significant mycelial growth inhibitory properties that are concentration dependent against the brownrot and white-rot fungi evaluated. An increase in the concentration of A. ingoldii secondary metabolites significantly decreased the mycelial growth of both wood decay fungi. G. trabeum was more sensitive to the inhibitory effect of the secondary metabolites than T. versicolor. Identification of specific A. ingoldii secondary metabolites, and analysis of their efficacy/specificity warrants further study. Findings from this work may provide the first indication of useful roles for Acaromyces species in a forest environment, and perhaps a future potential in the development of biocontrol-based wood preservation systems.

Isolation and Characterization of an Antifungal and Plant Growth-Promoting Microbe

  • Fungal diseases including anthracnose, stem rot, blight, wilting, and root rot of crops are caused by phytopathogens such as Colletotrichum species, Sclerotinia sclerotiorum, Phytophthora species, and Fusarium oxysporum and F. solani which threaten the production of chili pepper. In this study, to identify biological control agents (BCAs) of phytopathogenic fungi, potentially useful Bacillus species were isolated from the field soils. We screened out five Bacillus strains with antagonistic capacity that are efficiently inhibiting the growth of phytopathogenic fungi. Bacillus species were characterized by the production of extracellular enzymes, siderophores, and indole-3-acetic acid (IAA). Furthermore, the influence of bacterial strains on the plant growth promoting activity and seedling vigor index were assessed using Brassica juncea as a model plant. Inoculation with Bacillus subtilis SRCM 121379 significantly increased the length of B. juncea shoots and roots by 45.6% and 52.0%, respectively. Among the bacterial isolates, Bacillus subtilis SRCM 121379 showed the superior enzyme activities, antagonistic capacity and plant growth promoting effects. Based on the experimental results, Bacillus subtilis SRCM 121379 (GenBank accession no. NR027552) was finally selected as a BCA candidate.

Leptospermum scoparium 추출물중 케톤체 분획물의 항균력 및 항생제와의 병용효과 (Activities of Ketonic Fraction from Leptospermum scoparium alone and Synergism in Combination with Some Antibiotics Against Various Bacterial Strains and Fungi)

  • Whole oil and ketonic fraction (KF) of Leptospermum scoparium have been tested for their antimicrobial activity and combination effect with several antibiotics against various bacterial strains and fungi by using microbiological assay methods. Antibacterial activities of KF against a number of test strains were 2-3 fold stronger than those of whole oil. MICs of the KF were $65~125{\;}{\mu\textrm{g}}/ml$ against seven gram positive bacterial strains, $65~250{\;}{\mu\textrm{g}}/ml$ against 19 methicillin resistance Staphylococcus aureus strains, and $65~50{\;}{\mu\textrm{g}}/ml$ against 14 quinolone resistance strains. However, KF showed little or no activity against gram negative bacteria. MICs of the KF were $16~250{\;}{\mu\textrm{g}}/ml$ against more than 50% of the anaerobic bacterial strains tested. KF showed the higher antibacterial activity than bacitracin against 10 strains of Bacteroids thetaiotaomicron, or three strains of Bacteroides ovatus, and the more active than ciprofloxacin against one strain of Bacteroides thetaiotaomicron and three strains of Bacteroids ovatus. The MICs of KF was 63 and $250{\;}{\mu\textrm{g}}/ml$ against Aspergillus niger and Candida albicans, respectively. Antibacterial activities of KF in combination with 19 antibiotics against 14 strains and with four antifungal agents against one fungal strain were determined by paper strip diffusion method. While most of combination showed additivity, KF showed synergism with bacitracin, exfadroxil, cephradin, and meropenem for 29~57% of the strains tested. However, ofloxacin, enoxacin, sparfloxacin showed antagonism with KF for 43~71% of the strains. KF alone and in combination with bacitracin, gentamycin, neomycin, itraconazole, fluconazole, terfinafine and ketoconazole against five bacterial strains or one fungus strain synergistic effect was demonstrated against 33% of strains examined with FIC index value below 0.5 by checkerboard study. Synergistic effect of KF with gentamicin against Staphylococcus epidermidis 329 (QRS) was found by time-kill study.
