약용식물 물 추출물의 항산화 활성 및 α-Glucosidase 저해효과 (Antioxidant Activities and α-Glucosidase Inhibitory Effect of Water Extracts from Medicinal Plants)

  • We studied the total polyphenol content, DPPH radical scavenging activity, hydroxyl radical scavenging activity and ${\alpha}$-glucosidase inhibition of water extracts from 17 medicinal plants. Total polyphenol contents ranged from 10.0 (Coix lachryma-jobi L, CL) ~ 279.7 (Perilla sikokiana, PS)mg/g. The water extract from medicinal plants were evaluated for its free radical scavenging activities and compared with a commercial antioxidant, ascorbic acid. DPPH radical scavenging activity of Pyrus pyrifolia (PP), Chamaecyparis obtusa L. (COL), Chamaecyparis obtusa F. (COF), and PS were higher than positive control. Higher hydroxyl radical scavenging activity were shown in Acanthopanax senticosus (AS) and Cordyceps militaris (CM) than the other plants. The highest anti-${\alpha}$-glucosidase activity was observed in Cornus officinalis (CO) and Paeonia suffruticosa Andrews (PSA) water extracts. PSA showed not only the higher DPPH radical scavenging activity but also the anti-${\alpha}$-glucosidase activity. The results of our study that PP, COL, COF, PS, AS, CM, CO and PSA could be potential candidates for natural antioxidants.

남해산 고사리의 이화학적 특성 및 항산화 활성 (Physicochemical Characteristics and Antioxidant Activity of Bracken (Pteridium aquilinum Kuhn) in Namhae)

  • In this study, we investigate the physicochemical characteristics of dried bracken (Pteridium aquilinum Kuhn) harvested in Namhae. The moisture, ash, crude protein and crude lipid content was $10.79{\pm}0.31%$, $6.16{\pm}0.04%$, $33.20{\pm}0.40%$ and $2.45{\pm}0.27%$, respectively. The total mineral (Ca, Fe, K, Mg, Na, Mn) content was $36720.1{\pm}495.7mg/kg$, with K being the highest at $31890.0{\pm}503.8mg/kg$. The total free amino acid content was 704.41 mg/100 g, the amount of methionine, citrulline and sarcosine being higher than the others. The water and 50% ethanol extracts from the dried bracken were evaluated for the total poly phenolic compound content, antioxidant activity and xanthine oxidase and ${\alpha}$-glucosidase inhibitory activity. The total polyphenol content in the 50% ethanol extract ($1574.86{\pm}18.31mg/100g$) was higher than in the water extract ($1240.24{\pm}16.32mg/100g$). The DPPH and ABT radical scavenging activity was increased according to the concentration of bracken extract, and the activity in the 50% ethanol extract was higher than in the water extract. In addition, the xanthine oxidase and ${\alpha}$-glucosidase inhibitory activity was also higher in the 50% ethanol extract than in the water extract. In conclusion, bracken has great antioxidative and antidiabetic effects and can be used as a preventive agent for oxidation and diabetes.

택란 분획물이 당뇨유발 흰쥐에서의 혈당강하에 미치는 영향 (The antidiabetic properties of fractions of Lycopus lucidic Turcz in streptozotocin diabetic rats)

  • This study was carried out to evaluate the effects of fractions of methanol(MeOH) extracts of Lycopus lucidic Turcz on hyperglycemia and energy metabolites in streptozotocin(STZ) diabetic rats. Diabetes mellitus was induced in male Sprague-Dawley rats weighing 200-220 g by an injection of STZ dissolved in a citrate buffer into the tail vein at a dose of 45 mg/kg of body weight, and the rats were divided into 7 groups, that is, one normal group and 6 diabetic groups: STZ-control, hexane, chloroform(CHCl$\sub$3/). ethylacetate(EtOAc), butanol(BuOH) and H$\sub$2/O fraction-fed groups. All groups were fed an AIN-93 diet and the fractions of Lycopus lucidic Turcz were administered orally with 2 % Tween 80 for 14 days after the STZ injection. Body weight, diet intake and organ weights were monitored. The plasma levels of blood glucose, insulin and protein were determined. The plasma concentrations of cholesterol, HDL-cholesterol, triglycerides and free fatty acid were assayed. The plasma activities of aspartate aminotransferase (AST) and alanine aminotransferase(ALT) were also measured. Body weight losses were observed by feeding the fractions of Lycopus lucidic Turcz in STZ experimental groups, and the kidney weight was increased. The extent of blood glucose decrement was significantly greater in the hexane and BuOH fraction-fed groups than STZ-control group. The plasma protein level was significantly lower in the H$\sub$2/O fraction-fed group. The plasma cholesterol level was decreased in BuOH and H$\sub$2/O fraction-fed groups compared with the STZ-control group. The levels of free fatty acids in the CHC1$\sub$3/ and H$\sub$2/O fraction-fed groups were significantly decreased(p<0.05). ALT activitiy of BuOH fraction-fed group was lower than control but it was not significantly different. These results suggest that the fractions of Lycopus lucidic Turcz are capable of lowering blood glucose and fat metabolites concentrations when administered to STZ-treated rats, and AST/ALT activity and insulin levels show the possibility of therapeutic use to diabetes mellitus.

Effects of Epothilone A in Combination with the Antidiabetic Drugs Metformin and Sitagliptin in HepG2 Human Hepatocellular Cancer Cells: Role of Transcriptional Factors NF-κB and p53

  • Type 2 diabetes mellitus patients are at increased risk of many forms of malignancies, especially of the pancreas, colon and hepatocellular cancer. Unfortunately, little is known of the possible interaction between antidiabetic drugs and anticancer agents. The present study investigates the influence of metformin (MET) and sitagliptin (SITA) on the in vitro anticancer activity of the microtubule depolymerization inhibitor agent epothilone A (EpoA). Hepatocellular liver carcinoma cell line (HepG2) viability and apoptosis were determined by the MTT test and by double staining with PO-PRO-1 and 7-aminoactinomycin D, respectively, after treatment with EpoA, metformin or sitagliptin. The levels of nuclear factor NF-${\kappa}B$ and p53 were evaluated in the presence and absence of inhibitors. While EpoA and MET inhibited HepG2 cell proliferation, SITA did not. EpoA and SITA induced higher p53 levels than MET. All tested drugs increased the level of NF-${\kappa}B$. Only MET enhanced the proapoptotic effect of EpoA. The EpoA+MET combination evoked the highest cytotoxic effect on HepG2 cells and led to apoptosis independent of p53, decreasing the level of NF-${\kappa}B$. These findings support the link between NF-${\kappa}B$ and p53 in the modulation of apoptotic effects in HepG2 cells treated by EpoA. Our studies indicate that the combination of EpoA and MET applied in subtoxic doses has a stronger cytotoxic effect on liver cancer cells than each of the compounds alone. The therapeutic advantages of the combination of EpoA with MET may be valuable in the treatment of patients with diabetes mellitus type 2 (T2DM) and liver cancer.

포공영(浦公英) 물 추출물의 항당뇨 효과에 대한 연구 (Antidiabetic Effects of Water Extract from Taraxacum coreanum Nakai in type II Diabetic Mice)

  • Taraxacum coreanum Nakai (TM) has been investigated for its antidiabetic activity in streptozotocin induced diabetic animals, but few studies have been conducted to evaluate its efficacy for treatment of type II diabetic mice. This study was designed to evaluate the efficacy of the water extracts of TM for the treatment of type II diabetes. Twenty four mice(C57BLKS/J Iar $-+Lepr^{db}/+Lepr^{db}$) with type II diabetes were randomly assigned either to a experimental group A (leaves extraction of TM), experimental group B(roots extraction of TM), experimental group C(leaves, roots and flowers extraction of TM) or control group. Water extracts of TM were ingested for 10 weeks. The changes of body weight, level of blood glucose, numerical change of insulin-immunoreactive cells and level of blood insulin were measured after treatment of water extracts of TM. A reduction of the blood glucose level after treatment of TM was observed in all the experimental groups. Especially, experimental C group showed significant reduction of blood glucose level compared with control group. Experimental C group induced an increase the level of blood insulin and density of insulin immunoreactive cells after treatment of TM. These results show that water extracts of TM can improve the severity in type II diabetic mice.

Antidiabetic activity of Argyreia speciosa (sweet) (Burm.f.)Boj. in normoglycemic and Streptozotocin-induced diabetic rats

  • Effect of ethanol (ASE) and water (ASW) extracts of Argyreia speciosa on blood glucose and lipid profile was investigated in normoglycemic and Streptozotocin (STZ)-induced diabetic animals. In oral glucose and sucrose tolerance test, treatment with ASE and ASW (100 and 200 mg/kg) and Glidenclamide (10 mg/kg) significantly improved the glucose and sucrose tolerance in normal animals. In addition, respective treatment for fifteen-day resulted in significant percentage reduction in serum glucose (SG) ie., 30.39% (lower dose of ASE) and 33.21% (higher dose of ASW). In standardized STZ (50 mg/kg, iv)-induced diabetic rats, a single dose of ASE and ASW treatment exhibited reduction in SG levels at different time intervals compared to basal levels. Administration of both the doses of ASE and ASW for fifteen-day days exhibited greater percentage reduction in glycemia (24.6%, 24.7%, 23.9% and 21.9% respectively) and also ameliorated restored to near normal value of all tested lipid parameters. Further, treatment also exhibited significantly improved glucose tolerance over the period of 120 min compared to diabetic control group. Eventhough treatment failed to increase serum insulin levels significantly but peripheral utilization of insulin was increased as evident by insulin tolerance test. Taken together, present study supports the traditional usage of title plant in the treatment of diabetes mellitus.

Anti-hyperglycemic and Anti-hyperlipidemic Effects of the Triterpenoid-Rich Fractions from Rubus coreanus and Rubus crataegifolius and Their Main Component, Niga-ichigoside $F_1$, in Streptozotocin-induced Diabetic Rats

  • To find the antidiabetic activity of the tripterpenoid-rich fractions of Rubus coreanus (TRF-cor) and R. crataegifolius (TRF-cra) leaves or its main component niga-ichigoside $F_1$ (Niga-$F_1$), anti-hyperglycemic and antihyperlipidemic effects were investigated in the diabetic rat model induced by streptozotocin (STZ). Treatments of rats with 200 mg/kg of the TRF-cor, TRF-cra (each, p.o.) or 20 mg/kg of Niga-$F_1$ significantly inhibited the increase of blood glucose concentration about 44.8%, 28.7% or 20.6%, respectively, in the diabetic rats. In addition, treatments with those fractions inhibited the increase of serum concentrations of triglyceride, total cholesterol or LDL-cholesterol caused by STZ. The inhibitory rate on atherogenic index (AI) values of the TRFcor (200 mg/kg), TRF-cra (200 mg/kg) or Niga-$F_1$ (20 mg/kg)-treated groups were decreased about 55.7%, 36.3% or 22.6%, respectively, comparable to STZ-treated group. In the oral glucose tolerance test, treatment of TRF-cor or TRF-cra inhibited the increase of blood glucose concentration in the STZ-induced rats. Administration of 20 mg/kg of Niga-$F_1$ (p.o.) also exhibited similar effects with the effects of both TRFs at 200 mg/kg dose (p.o.). These results support that the triterpenoids, in particular Niga-$F_1$, are contributed to the antidiabetic effects of R. coreanus or R. crataegifolius.

돼지감자 부위별 메탄올 추출물의 항산화 및 항당뇨 활성 (Antioxidative and Antidiabetic Activities of Methanol Extracts from Different Parts of Jerusalem Artichoke (Helianthus tuberosus L.))

  • 본 연구에서는 돼지감자 꽃, 잎, 뿌리가 가진 기능성 식품 소재로서의 활용도를 높이고자 다양한 생리활성을 평가하였다. 돼지감자 꽃, 잎, 뿌리의 폴리페놀 함량은 32.80 mg GAE/extract g, 43.46 mg GAE/extract g, 24.53 mg GAE/extract g으로 나타났으며, 플라보노이드 함량은 각각 15.64, 18.67, 11.65 mg GAE/extract g으로 나타났다. 돼지감자 꽃, 잎, 뿌리의 메탄올추출물의 DPPH 라디칼 소거능 활성은 각각 77.78%, 66.07%, 26.93%로 나타났으며, hydroxyl radical 소거능 활성은 각각 55.33%, 38.30%, 34.89%로 나타났다. 돼지감자 꽃, 잎, 뿌리메탄올 추출물의 아질산염 소거능을 측정한 결과, 돼지감자 추출물의 농도가 증가함에 따라 그 활성 역시 증가하는 경향으로 나타났다. 돼지감자 부위별 메탄올 추출물(5 mg/mL)의 ${\alpha}-glucosidase$ 저해활성은 꽃, 잎, 뿌리에서 각각 41.08%, 55.67%, 60.76%로 유의적인 차이를 나타났으며, ${\alpha}-amylase$ 저해활성은 꽃, 잎, 뿌리에서 각각 49.95%, 59.88%, 66.25%로 유의적인 차이를 나타내었다.

참깨박 에탄올 추출물의 항산화 및 α-amylase및 α-glucosidase 저해 활성 (Antioxidant, α-amylase and α-glucosidase inhibitory ability effects of sesame meal ethanol extract)

  • 기름을 착유하고 얻어지는 부산물인 참깨박의 활용과 이용가치를 높이기 위해 일반성분(수분, 탄수화물, 조단백질, 조지방, 조회분), 무기질을 측정하였고 탈지 처리한 후 70% 에탄올 추출하여 항산화 활성 및 항당뇨 활성을 분석하였다. 참깨박의 일반성분 함량은 수분 6.51%, 탄수화물 16.22%, 조단백질 46.30%, 조회분 9.88%, 조지방 21.09%로 이었고, 무기질은 K 1128.08 mg/100 g, Ca 1356.27 mg/100 g, Fe 12.29 mg/100 g, P 2022.14 mg/100 g, Cu 2.08 mg/100 g, Mg 643.40 mg/100 g, Na 7.29 mg/100 g 함량을 나타내었다. 탈지 참깨박의 총 폴리페놀과 플라보노이드 함량은 각각 184.98 mg GAE/g, 27.63 mg QE/g였으며, DPPH 및 ABTS 라디칼 소거활성 IC50을 측정한 결과 각각 91.84, 340.09 ㎍/mL, 환원력은 0.87 OD으로 측정되었다. 탈지 참깨박의 α-amylase 및 α-glucosidase 저해 활성 IC50의 측정 결과는 각각 74.31, 16.73 mg/mL을 나타내었다. 따라서 70% 에탄올로 추출한 탈지 참깨박은 우수한 항산화 활성과 항당뇨 활성을 나타내 기능적 가치를 높일 수 있을 것으로 기대된다. 또한 참깨를 착유하고 남은 참깨박을 사용하게 되므로 환경보호와 자원순환 측면에 활용가치가 있을 뿐만 아니라 건강기능식품소재로 이용가치가 있을 것이라 판단된다.

산사 추출물의 항산화 및 항당뇨 활성 (Anti-diabetic and Anti-oxidative Activities of Extracts from Crataegus pinnatifida)

  • 장미과에 속한 낙엽교목인 산사나무의 열매인 산사의 항산화 및 항당뇨 관련 활성을 검토하고, 추출 용매에 따른 차이를 비교하기 위하여 총 페놀 함량, DPPH 라디칼 소거능, ABTS 라디칼 소거능, 최종당화산물 억제능, ${\alpha}$-glucosidase 억제능, aldose reductase 억제능을 측정하였다. 수율은 70% 에탄올 추출물이 33.16%로 가장 높았고, 50% 에탄올 추출물이 27.79%로 그 다음 높았으며, 물, 30% 에탄올, 100% 에탄올은 각각 21.71%, 21.88%, 19.03%를 나타내었다. 총 페놀함량은 50% 에탄올 추출물이 $41.83{\pm}0.07mg$ GAE/g으로 가장 높았고, DPPH와 ABTS 라디칼 소거능도 50% 에탄올 추출물이 각각 $80.79{\pm}0.83%$, $34.92{\pm}0.97%$로 가장 높았다. 최종 당화산물 억제능과 ${\alpha}$-glucosidase 억제능도 50% 에탄올 추출물이 $27.09{\pm}2.27%$$58.87{\pm}0.70%$로 가장 높게 나타난 반면에 AR은 물 추출물이 $30.68{\pm}1.41%$로 가장 높게 나타나면서 상이한 경향을 보였다. 이상의 결과를 종합해 보면, 알코올 농도에 따른 산사의 추출물 중 50% 에탄올 추출물이 천연 항산화제로서 가치가 있으며, 당뇨병 예방을 위한 천연물로서의 가능성도 보였으나 당뇨병 합병증 치료에 대한 효과는 미흡한 것으로 나타났다. 향후 좀 더 다양한 fraction에서의 연구와 in vivo 시험 등을 통한 추가적인 연구가 필요할 것으로 사료된다.
