Combination of Curcumin and Paclitaxel-loaded Solid Lipid Nanoparticles to Overcome Multidrug Resistance

  • Multi-drug resistance (MDR) has been known as a major hurdle in cancer chemotherapy. One of the most clinically significant causes of MDR was the efflux of anticancer agents mediated by p-glycoprotein (p-gp) over-expressed in MDR cancer cells. To overcome MDR, there have been several strategies such as co-administration with p-gp inhibitors and encapsulation of anticancer drugs into drug delivery systems. In the present study, curcumin was evaluated for its potential as p-gp inhibitor and MDR reversal activity when combined with paclitaxel incorporated into lipid nanoparticles (PTX/LN). Western blot assay showed curcumin did not modulate the level of p-gp expression in MCF-7/ADR which is a MDR variant of human breast cancer cell line, MCF-7, and over-expresses p-gp. However, curcumin inhibited p-gp-mediated efflux of calcein in a dose-dependent manner even though it showed lower activity compared to verapamil, a well-known p-gp inhibitor. Incorporation of paclitaxel into lipid nanoparticles partially recovered the anticancer activity of paclitaxel in MCF-7/ADR. The combined use of curcumin and PTX/LN exhibited further full reversal of MDR, suggesting susceptibility of PTX/LN to the efflux system. In conclusion, combined approach of using p-gp inhibitors and incorporation of the anticancer agents into nano-delivery systems would be an efficient strategy to overcome MDR.

  • The present investigation was undertaken to examine the anticancer activity of the methanol extract from Houttuynia cordata on ICR mouse with induced abdominal cancer and L1210 cancer cells. When the methanol extract of Houttuynia cordata (10∼200 $\mu\textrm{g}$/$m\ell$) was administered orally to ICR mouse with abdominal cancer, 47.8% of the best life prolonging effect was obtained. In case of cytotoxicity study (inhibition of cell proliferation) of Houttuynia cordata extract against L1210 cells, $IC_{50}$/ was found to be 62.8 $\mu\textrm{g}$/$m\ell$. In contrast to such considerable toxicity against cancer cell line, the toxicity demonstrated by the identical extract against normal lymphocytes was very meagre as shown to be < 5% compared with 86.5% in case of L1210 cells at the same condition. To get an insight into the reaction mechanism undelying the anticancer activity, $O_2$ion quantity and antioxidant enzyme activities such as superoxide dismiutase (SOD) and glutathione peroxidase (GPx) of L1210 cells in the presence of Houttuynia cordata extract were measured. The increased values of SOD and GPx enzyme activities in addition to the augmented generation of $O_2$ ion in L1210 cells implied that the reactive oxygen species induding $O_2$ion which were presumably induced by Houttuynia cordata extract might have participated in the process of L1210 cells cytotoxicity.

Monoterpenes의 항암작용과 활성산소 전환 효소의 활성 변화 (Anticancer Activity of Monoterpenes and the Changes of Enzymes Activities Responsible for the Conversion of Reactive Oxygen Species)

  • The present study was undertaken to investigate the anticancer activity of monoterepenes in the animal and the cancer cell line tests. Both of the noncyclic and cyclic monoterpenes showed significant life prolonging effects on ICR mouse with abdominal cancer induced by Sarcoma 180 cells up to 67.4% and 63.5% in case of linalool and geraniol, respectively. Linalool and geraniol also exhibited very excellent cytotoxicity against L1210 leukemic cells with $IC_{50}$/ value of 0.32 $\mu\textrm{g}$/mι in 5 days culture condition. In the presence of linalool and geraniol, the generation of $O_2$$^{[-10]}$ ion were found to be increased proportionally to the cytotoxicity arisen from these monoterpenes. Furthermore, the antioxidant enzymes activities such as superoxide dismutase (SOD) and glutathione peroxidase (GPx) responsible for the conversion of $O_2$$^{[-10]}$ ion to $H_2O$$_2$ and then to $H_2O$ augmented remarkably by linalool and geraniol. All data put together it can be postulated that monoterpenes may kill abdominal cancer cells of ICR mouse probably by activating anticancer system of the body, whereas the death of L1210 cells may be due to the detrimental attacks of reactive oxygen species (ROS) including $O_2$$^{[-10]}$ in spite of antioxidant enzymes activities to overcome the ROS attacks.

세신 비극성 분획의 항암 활성 및 성분 분석 (Anticancer Activity and Chemical Composition of a Non-Polar Fraction from Asiasari Radix et Rhizoma)

  • The study aimed to characterize chemical composition and anticancer property of the n-hexane fraction derived from Asiasari Radix et Rhizoma. The anticancer activity was evaluated on a panel of cancer cell lines including HN22, HSC2, HSC3, and HSC4 cells (human oral cancer), HCC827 and HCC827GR cells (human lung cancer), and KYSE30 and KYSE450 (human esophageal cancer) by MTS assay. As a result, The least polar subfraction from n-hexane-soluble layer displayed notable cytotoxicity on the tumor cell lines with IC50 ranging from 1.20 to 17.0 ㎍/ml. The chemical composition of constituents in the active subfraction was determined by gas chromatography-mass spectrometry (GC-MS). The essential oils comprised of sesquiterpenes including β-gurjunene (7.45%), γ-amorphene (6.61%), guaia-6,9-diene (6.40%), δ-guaiene (5.21%) and a phenylpropanoid, safrole (0.49%) were mainly identified in addition to long-chain hydrocarbons including n-heptadecane (24.60%), 7-hexadecene (4.44%) and a diterpenoid, ent-kaur-16-ene (6.57%).

순무가루를 첨가한 순무김치의 항산화 활성 및 대장암세포(HT-29) 항암효과 (Antioxidant Activity and Anticancer Effects of Turnip Kimchi with Turnip Powder on Colorectal Cancer Cells (HT-29))

  • In this study, the quality characteristics of kimchi, such as its salinity, pH, and acidity, were measured and compared, and the HT-29 human colon cancer cells were used to show the anticancer effects of kimchi. The kimchi samples used herein included standard kimchi (SK), turnip kimchi (TK), and turnip-powder-added kimchi (TPK). The measured pH and acidity of TK and TPK showed no significant differences with those of SK. Compared to SK and TK, TPK had higher DPPH scavenging activity and higher total flavonoid content, confirming its antioxidant activity. The cancer cell growth inhibition rates of TK and TPK were significantly higher than that of SK. In HT-29 cells treated with TPK, the mRNA expression of Bcl-xL, an anti-apoptosis-related gene, was lower, and the mRNA expressions of the apoptosis-related genes Bax, Bad, and caspase-9 were higher. TPK showed significantly higher levels of mRNA expressions for the cell-cycle-related genes p53 and p21 than the other samples, in addition to suppression effects on cancer cell proliferation. Compared to SK, TK and TPK suppressed the growth of colon cancer cells and showed higher anticancer effects. Therefore, it is shown that kimchi with added turnip powder had high anticancer effects.

Gold Nanoparticles Enhance the Anticancer Activity of Gallic Acid against Cholangiocarcinoma Cell Lines

  • Gold nanoparticles (GNPs) were conjugated with gallic acid (GA) at various concentrations between 30 and $150{\mu}M$ and characterized using transmission electron microscopy (TEM) and UV-Vis spectroscopy (UV-VIS). The anticancer activities of the gallic acid-stabilized gold nanoparticles against well-differentiated (M213) and moderately differentiated (M214) adenocarcinomas were then determined using a neutral red assay. The GA mechanism of action was evaluated using Fourier transform infrared (FTIR) microspectroscopy. Distinctive features of the FTIR spectra between the control and GA-treated cells were confirmed by principal component analysis (PCA). The surface plasmon resonance spectra of the GNPs had a maximum absorption at 520 nm, whereas GNPs-GA shifted the maximum absorption values. In an in vitro study, the complexed GNPs-GA had an increased ability to inhibit the proliferation of cancer cells that was statistically significant (P<0.0001) in both M213 and M214 cells compared to GA alone, indicating that the anticancer activity of GA can be improved by conjugation with GNPs. Moreover, PCA revealed that exposure of the tested cells to GA resulted in significant changes in their cell membrane lipids and fatty acids, which may enhance the efficacy of this anticancer activity regarding apoptosis pathways.

Differential anticancer effect of fermented squid jeotgal due to varying concentrations of soymilk additive

  • Fermentation plays a vital role in the nutritional enrichment of food. Korea has a long tradition of adding fermented food to the daily diet and jeotgal is one of the common fermented and salted foods in Korean cuisine. In our study, we added soymilk as an additive to squid jeotgal to improve its functionality. We mixed different concentrations of soymilk (2, 5, and 10 mg/g) with squid jeotgal samples, fermented them for one week, and then tested their antioxidant and anticancer activities to compare with those of squid jeotgal samples without soymilk additive. To investigate the anticancer characteristics, glutathione-S-transferase (GST)-pi enzyme assay was used. To test the antioxidant activities, various assays were performed, including 2,2-diphenyl-1-picryl hydrazyl free radical scavenging activity, 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium saltradical cation scavenging assay, and reducing power assay. Samples fermented with a small amount of soymilk showed excellent anticancer activity. The addition of only 2 mg/g of soymilk to squid jeotgal inhibited the activity of GST-pi by almost 50% when compared with the sample with no addition. Moreover, no undesirable bitterness or astringency was noticed. Our results could help to improve the current food status of squid jeotgal and it could be used to reduce the risk of chronic disease along with its basic nutritional function.

유근피(楡根皮)로부터 단백다당체의 분리 및 항암 면역활성 연구 (Separation of Glycoprotein and its Anticancer Immunostimulating Activity from Dried Barks of Slippery Elm [Ulmus parvifolia])

  • 본 연구에서는 민방이나 한방에서 천연 약재로 많이 사용하고 있는 느릅나무 뿌리껍질(유근피)로부터 수용액 추출물을 분리하여 이 중 단백다당체를 분리하여 성분분석 및 단백다당체가 갖고 있는 면역활성에 의한 항암효과를 실험적으로 제시하였다. 느릅나무 추출물 당단백질의 화학적 조성을 보면 총당함량은 55.8∼72.1%였고, 총산성당 함량은 30.0∼30.5%, 총단백질 함량은 5.0∼6.1%으로 수용성 고분자이며, 50 내지 500 kDa 범위의 분자량을 나타내었다. 그리고, 단백다당체를 구성하는 주요 당성분으로는 글루크론산, 람노스아라비노스, 글루코스, 갈락토스 등이었다. 단백다당체의 면역활성 검정에 따르면 면역세포의 증식을 자극하는 mitogen 역할을 하며, 특히 T세포에 의한 면역증강 효과를 나타내었다 B16 흑색종 이식 마우스를 이용한 생체내 면역활성에 의한 항암효과를 조사한 결과 유의성 있는 항암효과를 나타내었으며, 느릅나무 추출물인 단백다당체가 3mg/kg 군에서 가장 강한 항암효과를 나타내어 용매대조군에 대해 약 140% 생존연장효과를 나타내었다.

한방생약재를 이용한 약선차의 항산화 및 항암효과 (Antioxidant and Anticancer Activities of Yak-Sun Tea Prepared by Oriental Medicinal Herbs)

  • Various types of tea are consumed as a popular beverage worldwide particularly in Asian countries such as Korea, China, and Japan. The purpose of this study was to investigate the mineral contents, antioxidant properties and anticancer activity of Yak-Sun tea that is prepared by six oriental medicinal herbs. The results of the mineral contents were as follows; Ca, Mg, and Na contents were higher than those of green tea, whereas Fe, P, and K contents were lower than those of green tea. The total phenolics and flavonoid content of the Yak-Sun tea were higher than those of green and black teas. The $IC_{50}$ values of DPPH radical, hydroxyl radical, hydrogen peroxide radical scavenging of Yak-Sun tea were 0.78, 1.58, and 2.04 $mg/m{\ell}$, respectively, whereas the radical scavenging values of ${\alpha}$-tocopherol was 0.06, 0.05, and 0.09 $mg/m{\ell}$, respectively. When cancer cells were treated with Yak-Sun tea, the anticancer activity increased in a dose dependent manner. HCT116 colon cancer cell lines were dramatically increased, as compared to other cancer cell lines, such as MCF-7, H460, and MKN45 cell lines. The results of this study demonstrated that Yak-Sun tea could function as a tea to enhance health conditions for antioxidant and anticancer activity.

한국산 길조류에서 추출한 Fucoidan의 황산기에 따른 항암작용 (Increased Anticancer Activity by the Surfated Funcoidan from Korean Brown Seaweeds)

  • Fucoidan은 갈조류에 다량 함유되어있는 다당류로서 항암작용, 항혈응고작용, 항혈전작용, 항염증작용, 항virus작용과 같은 생리활성작용력을 가지고 있어 최근 많은 연구가 수행되어지고 있다. 본 논문은 갈조류에서 fucoidan을 추출 하여 그 항암활성을 SV40 DNA replication assay, RPA-ssDNA binding assay, 그리고 MCF7 cell growth inhibition assay를 이용하여 알아보았으며 이 항암활성은 fucoidan의 황산기가 중요한 역할을 하고 있음을 알아내었다. 화학적으로 황산기의 함유량을 증가시킨 fucoidan이 황산기의 함유량을 제거시킨 fucoidan보다 항암활성이 뛰어났고, 이는 RPA의 ssDNA 결함력을 떨어 EM리기 때문이라 예측되어 진다. 본 연구는 한국산 갈조류에서 추출한 황산기를 함유한 fucoidan의 항암활성능력을 보여주었다.
