Staphylococcus aureus의 항생제 내성 plasmid에 관한 연구 (R-plasmids in staphylococcus aureus)

  • Small size antibiotic resistance plasmids having molecular weights less than 10 Mdal were isolated and characterized from ten clinically isolated multiple resistant Staphylococcus aureus. Agarose gel electrophoresis profiles and antibiotic resistance patterns divided these strains into four groups. Strain 2-23-6, the representative strain of a group of five strains conferred two plasmids of molecular weights $1.6{\times}10^6\;dal\;and\;2.0{\times}10^6$ dal. The small plasmid (pSBK 112) specified macrolides, lincosamides and streptogramin type B (MLS) resistance gene which are expressed constitutively. Lage plasmid (pSBK 125) specified chloramphenicol resistance gene which is inducible. Strain 10-5 conferred a $3.0{\times}10^6$ dal plasmid (pSBK 141) which carry an inducible ampicillin resistance gene and strain P-H-2 conferred and $1.6{\times}10^6$ dal plasmid (pSBK 190) which carry a constitutive MLS resistance gene. Strain D-H-1 conferred four plasmids of molecular weights $0.8{\times}10^6$ dal (pSBK 201), $1.6{\times}10^6$ dal (pSBK 202), $2.5{\times}10^6$ dal (pSBK 203), and $1.2{\times}10^7$ dal (pDBK 204), respectively. Among those four plasmids, only pSBK 203 specified chloramphenicol resistance gene. Curing of constitutive MLS resistance using acriding orange or ethidium bromide in 2-23-6 and P-H-2 strains produced 'inducible' MLS resistance strains which are less resistant to MLS than the wild type strains, suggesting that there are two resistance genes in both strains; one is constitutive and the other is inducible.

Prevalence of methicillin-resistant Staphylococcus aureus in Iranian children: a systematic review and meta-analysis

  • Background: Antibiotic resistance is associated with longer hospitalizations, higher treatment costs, and increased morbidity and mortality rates. Purpose: This study aimed to determine the prevalence of methicillin-resistant Staphylococcus aureus (MRSA) in Iranian children. Methods: International databases, including Web of Science, PubMed, Embase, and Scopus, and Iranian databases, including Scientific Information Database (www.sid.ir), Magiran, and Iranian Database for Medical Literature (idml.research.ac.ir), were systematically searched for articles published between January 2000 and August 2019. Sources of heterogeneity were determined using subgroup analysis and meta-regression. Results: Overall, 343 studies were identified; of them, 20 were included in the meta-analysis to estimate the pooled prevalence. The pooled prevalence of MRSA was 42% (95% confidence interval [CI], 29-55) among culture-positive cases of S. aureus, 51% (95% CI, 39-62) in hospitalized children, and 14% (95% CI, 0.05-27) in healthy children. Conclusion: The overall pooled prevalence of MRSA in children was 42%. Appropriate infection control measures and effective antibiotic therapy are needed.

  • 메로페넴에 내성을 갖는 Pseudomonas peli CJ30 균주가 대한민국의 한강에서 분리되었다. CJ30 균주의 유전체는 크기가 4,919,106 bp이고 G + C 함량이 60.0%인 아홉 개의 contig로 조립되었다. CJ30 균주의 유전체 서열은 5,411개의 단백질 코딩 유전자, 18개의 rRNA 유전자 및 70개의 tRNA 유전자를 포함하였다. 균주 CJ30에는 blaSFC-3 및 ampC β-락타마아제 유전자가 포함되어 있습니다.

의료기관관련 폐렴 (Healthcare-Associated Pneumonia)

  • Pneumonia is frequently encountered in the clinical fields, both as a cause for admission and as a complication of the underlying disorder or as the course of treatment. Pneumonia is the second most common hospital-acquired infection and is associated with the highest morbidity and mortality rates among hospital-acquired infections. The guidelines for the management of hospital-acquired pneumonia by the American Thoracic Society include identifying individuals who have recently received antibiotics therapy or have been in medical facilities; these individuals are at higher risk for infection with multiple drug resistant organisms. Individuals, who have acquired pneumonia according to this clinical scenario, have what is known as healthcare-associated pneumonia (HCAP). Patients with HCAP should be considered to have potentially drug-resistant pathogens and should receive broad spectrum empiric antibiotic therapy directed at the potentially resistant organisms. In this paper, the diagnosis, risk factors, and treatment of HCAP are discussed.

An Antimicrobial Activity of a Peptidic Molecule from the Centipede, Scolopendra subspinipes mutilans L. Koch

  • An antimicrobial molecule was purified from centipede, Scolopendra subspinipes mutilans L. Koch, by reverse phase-HPLC. Its molecular weight was determined to be 1208.5493 by using matrix-assisted laser desorption ionization-time of flight (MALDI-TOF) mass spectrometry. Total amino acid composition analysis revealed that it consists of E, G, P, V, L, F, and W. It exhibited a broad antimicrobial spectrum against not only Gram-negative, but also Gram-positive bacteria. Furthermore, it was found to have an antimicrobial activity against vancomycin resistant enterococci (VRE). It may be a useful molecule for a new antibiotic development, especially against drug-resistant bacteria. We suggest that it may playa role in the defense system of this animal. This is the first report of a peptidic antimicrobial substance from centipede.

Antibacterial Activity of the Phaeophyta Ecklonia stolonifera on Methicillin-resistant Staphylococcus aureus

  • In an effort to discover an alternative therapeutic agent against methicillin-resistant Staphylococcus aureus (MRSA), several medicinal plants and seaweeds were evaluated for its antibacterial activity against MRSA. A methanolic extract of the Phaeophyta Ecklonia stolonifera exhibited significant antibacterial activity against MRSA. To perform more detailed investigation on antibacterial activity, the methanol extract of E. stolonifera was further fractionated with organic solvents such as hexane, dimethylchloride, ethyl acetate, and n-butanol. Among them, the hexane fraction showed the strongest antibacterial activity against MRSA strains with MIC from 500 to $600 {\mu}g/mL$. The fraction also exhibited a bactericidal activity against MRSA, indicating that E. stolonifera contains a bactericidal substance against MRSA.

Multidrug-resistant Staphylococci Found on Book Surfaces in East London Libraries

  • There is an increase in the presence of drug-resistant staphylococci outside of the nosocomial and healthcare setting. Although the presence of staphylococci has been studied in several public spaces, nothing is known on the presence of staphylococci in public libraries. Book surfaces from public libraries in the East London area, United Kingdom were swabbed and cultured and identity of the isolates determined by matrix-assisted laser desorption/ionization time of flight (MALDI-TOF) mass spectrometry (MS). Seven different staphylococcal species were identified by MALDI-TOF-MS analysis. This short study provides evidence of the presence of multidrug-resistant staphylococci in public libraries in the East London area.

  • Staphylococcus aureus KH13 and Staphylococcus aureus KH28 were resistant to chloramphenicol, ampicillin, clindamycin, erythromycin, gentamicin, kanamycin, streptomycin, tobramycin, and norfloxacin. A plasmid (pKH13) and two plasmids (pKH14, pKH15) were isolated from Staphylococcus aureus KH13 and Staphylococcus aureus KH28, respectively and complete nucleotide sequences of three plasmids were determined. It was found that pKH13 and pKH15 mediated chloramphenicol resistance and pKH14 was a cryptic plasmid.

해수에서 분리한 장염비브리오의 항생제 내성 및 암피실린 내성 유전자의 동정 (Antibiotic-Resistance Profiles and the Identification of the Ampicillin-Resistance Gene of Vibrio parahaemolyticus Isolated from Seawater)

  • The antibiotics-resistance profiles of 28 strains of Vibrio parahaemolyticus isolated from seawater were investigated. All of the strains studied were resistant to ampicillin (100%), but susceptible to 12 other antibiotics. The minimum inhibitory concentration (MIC) of V. parahaemolyticus to ampicillin was as high as $1,024-2,048\;{\mu}g{\cdot}mL^{-1}$. The phenotype of strain 8 changed from ampicillin-resistant to susceptible with an in-frame deletion mutant of VPA0477, a putative ${\beta}$-lactamase gene, and the MIC for ampicillin of the mutant strain was $1{\mu}g{\cdot}mL^{-1}$. In conclusion, our findings suggest that the VPA0477 gene acts as a ${\beta}$-lactamase in ampicillin-resistant V. parahaemolyticus strains.

동물에서 분리한 Proteus mirabilis의 항생제 감수성에 관한 연구 (Antibiotics susceptibility of Proteus mirabilis isolated from domestic animals in Chonbuk province)

  • Isolates of 70 Proteus mirabilis were tested against 10 different antibiotics by a disc diffusion method as recommended by National Committee for Clinical Laboratory Standards (NCCLS). The isolates were resistant in order of tetracycline(100.0%), enrofloxacin(95.7%), ampicillin(74.3%), choramphenicol(62.9%), cephalothin(58.6%), streptomycin(50%), kanamycin(47.2%), neomycin(35.8%), gentamicin(15.7%), and amikacin(2.9%). In the antibiotic resistant patterns, 37 kinds of multiple resistance patterns of P mirabilis isolates were detected. The highest resistant pattern was ampicillin-cephalothin-chloramphenicol-enrofloxacin-tetracycline(11.6%).
