Quantification of Quercetin in Different Parts of Onion and Its DPPH Radical Scavenging and Antibacterial Activity

  • Levels of quercetin in different parts of onion were investigated using high performance liquid chromatography (HPLC) and liquid chromatography mass spectrometry (LC/MS) suitable for use with functional food material. Two main peaks were observed on HPLC chromatograms from the extracts of the skin, and the outer, middle, and core parts of onion. Using LC/MS, peak 1 was tentatively identified as quercetin monoglucoside at m/z 466.4, and peak 2 as quercetin with [M]+ at m/z 303.3. The levels of quercetin in the skin, and the outer, middle and core parts of the plant were 16.83,2.67,0.95, and 0.35 mg/g, respectively. In the study of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, skin, the nonedible part, contained the highest amount of quercetin, compared to the other edible parts, and showed the highest DPPH radical scavenging activity. Levels of quercetin and DPPH radical scavenging activity increased from core to skin. All parts of onion exhibited the strongest antibacterial activity only against Staphylococcus aureus and Vibro parahaemolyticus. Antibacterial activities of onion exhibited that S. aureus was more sensitive than V. parahaemolyticus. Among the four onion extracts, the middle part showed the strongest inhibitory activity against S. aureus but all onion extracts showed similar antibacterial activities against V. parahaemolyticus.

Antibacterial Activities of Peptides Designed as Hybrids of Antimicrobial Peptides

  • CA(1-8)ME(1-12), the CA-ME hybrid peptide of the amino terminal segments of cecropin A (CA) and melittin (ME), has been reported to have a broad spectrum and improved potency without a hemolytic property. In order to obtain new synthetic peptides with powerful antibacterial activity without hemolytic activity, several hybrid peptides were designed from the sequences of CA, ME, magainin 2, bombinin and lactoferricin. All hybrid peptides were constructed to form an amphipathically basic-flexible-hydrophobic structure and synthesized by the solid phase method. Their hemolytic activities against human red blood cells and antibacterial activities against both Gram-positive and Gram-negative bacteria were detennined. CA(1-8)MA(1-12), CA(1-8)BO(1-12), MA(10-17)ME(1-12) and LF(20-29)ME(1-12) showed comparable activities with broad spectra against both Gram-positive and Gram-negative bacteria relative to CA(1-8)ME(1-12) but without hemolytic properties. These hybrid peptides, therefore, could be useful as model peptides to design a novel peptide with improved antibacterial activity and study on structure-activity relationships of antimicrobial peptides.

Antibacterial Activity of Ethanol Extract of Saussurea lappa (목향(木香) 에탄올추출물의 항균효과(抗菌效果))

  • Objective : The purpose of this study is to investigate the antibacterial activity of Saussurea lappa for the applications of herb-based extracts to both cosmetic and medicinal industries. Methods : Antibacterial activity of ethanol extract of Saussutes lappa (ESL) was assessed using agar diffusion and broth dilution methods, and determined by whether clear zone was formed around paper disc and in terms of the size (mm) of clear zone, Results : ESL provided activity against Staphylococcus aureus KCTC 1927 in concentration of 1 mg/ml with an clear zone of 16 mm, and showed an activity against Staphylococcus epidermids KCTC 1917 in concentration of 1 mg/ml with an clear zone of 18mm. Moreover, the minimum inhibitory concentration (MIC) of ESL against S. sureus and S. epidermidls were 1 mg/ml for both bacterial species. However, ESL showed no growth inhibition against Pseudomonas aeruginosa KCTC 12513, Enterobactet aerogenes KCTC 2190, Escherichia coli KCTC 2571, Salmonella typhimurium KCTC 1925 and Propionibacterium acnes KCCM 41747. Conclusions : Antibacterial activity of ESL against Staphylococcus sureus and Staphylococcus epidermids causing eye and skin diseases was proved. The result suggest that ESL may be useful as a natural preservative on behalf of synthetic preservatives.

Antibacterial and Nitric Oxide Production Inhibitory Activities of Prunus sargentii Branches Extract and Its Fractions against Pathogens of Acne (산벚나무 가지 추출물 및 용매 분획물의 Nitric Oxide 생성 억제 효과와 여드름 원인균에 대한 항균활성)

  • Background: In this study, we investigated the antibacterial and nitric oxide (NO) production inhibitory activities of 75% ethanol extract of Prunus sargentii branches and its fractions against acne pathogens. Methods and Results: The antibacterial activity against acne causing pathogens was determined using the disc diffusion assay. The ethyl acetate fraction showed higher activities against Propionibacterium acnes, Staphylococcus aureus and Staphylococcus epidermidis than those shown by other fractions. In the DPPH radical and NO scavenging assays, the butanol fraction showed strong DPPH radical and NO scavenging abilities. These activities were related to the total polyphenol and flavonoid contents of butanol fraction. On the other hand, the chloroform and ethyl acetate fractions exhibited the highest NO production inhibitory activity in Lipopolysaccharide (LPS)-stimulated Raw 264.7 cells compared to those exhibited by other fractions. Conclusions: The extract and its ethyl acetate fraction from the branches of P. sargentii exhibited antibacterial activity and could be used as functional materials in antimicrobial related fields. Moreover, the chloroform and ethyl acetate fractions are potential antiinflammatory agents and butanol fraction acts as an effective radical scavenger.

  • Over the last two decades, microbiologically influenced corrosion (MIC) of metallic materials has received considerable attention due to its serious effects on industrial field. In this context, it is important to devise control methods which inhibit biofilm formation on various metallic compounds and are compatible with environment. It was change of various conditions (duty cycle, current density, $AgNO_3$ concentration and pH) for injection of Ag particles in anodized Aluminum alloy pore using pulsed current. Optimal condition was obtained by means of FE-SEM, ICP analysis etc. The antibacterial metal's specimen were manufactured under optimal condition and this specimen were tested the antibacterial characterization and anticorrosion characterization. In result of test, we can confirmed that the antibacterial characterization and anticorrosion characterization of the specimens of injected Ag particles in anodized Aluminum alloy pore using pulsed current were better than the anodized Aluminum alloy specimens.

A Study on the Antibacterial and Deodorization of Silk Fabrics Dyed with Natural Dye (II) ― Sappan Wood― (천연염료를 이용한 염색물의 항균, 소취성에 관한 연구(I) ―소 목―)

  • Antibacterial and deodorization properties of silk fabrics dyed with Sappan wood are investigated by Halo, Shake flask, Bioassay and Detection column methods.The results are obtained as follows; 1. Although K/S values increase with Al mordant and concentrate concentrations, these are decreased in 10% Al mordant concentration in case of 4mlg dye concentration. 2. Reduction ratios of bacteria increase with K/S values in various tests. 3. When undyed silk fabrics are treated with Al mordant, reduction ratios of bacteria are not varied with Al mordant concentrations. 4. Deodorization property of Al pre-mordanted dyeing silk fabrics is better than one of unmordanted dyeing and raw silk fabrics. 5. Antibacterial properties of Cu and Cr mordanted dyeing fabrics are better than ones of the dyeing fabrics treated with the other mordants. 6. Natural dyes extracted from Sappan wood show the good antibacterial and deodorization properties, and colorant among various components contained in Sappan wood has a excellent antibacterial and deodorization properties.

  • Duck down is a feather of waterfowl and is used as an accessory for bedding products such as evil, pillow and outdoor sleeping bags due to its soft and bulky characteristics. Down is used mainly for winter outdoor such as padding and jumpers because of its excellent thermal insulation effect. Down wear is known as a product with high added value. Down is processed and sanitized because a large amount of oil and fat, bacteria, dust, insects, and soil are mixed. In particular, if the proper amount of oil and fat is not removed, it may cause odor or bacteria, and since the fishy smell peculiar to down and harmful VOCs (toluene, benzene, etc.) are released, there is a need to suppress the occurrence of bacteria through the provision of antibacterial function. In this study, we investigated the substances that cause the fishy odor of down, and confirmed the effect on the deodorization and antibacterial properties of down according to processing agents and processing conditions in order to impart deodorant and antibacterial properties to down.

Periplanetasin-2 Enhances the Antibacterial Properties of Vancomycin or Chloramphenicol in Escherichia coli

  • Periplanetasin-2 from cockroach exhibits broad-spectrum antimicrobial activity. The underlying antibacterial mechanisms rely on the stimulation of reactive oxygen species overproduction to induce apoptotic cell death. A promising strategy to increase the bioavailability of periplanetasin-2 involves reducing the dose through combination therapy with other antibacterials that show synergistic effects. Thus, the synergistic antibacterial activity of periplanetasin-2 with conventional antibacterial agents and its mechanisms was examined against Escherichia coli in this study. Among the agents tested, the combinations of periplanetasin-2 with vancomycin and chloramphenicol exhibited synergistic effects. Periplanetasin-2 in combination with vancomycin and chloramphenicol demonstrated antibacterial activity through the intracellular oxidative stress response. The combination with vancomycin resulted in the enhancement of bacterial apoptosis-like death, whereas the combination with chloramphenicol enhanced oxidative stress damage. These synergistic interactions of periplanetasin-2 can help broaden the spectrum of conventional antibiotics. The combination of antimicrobial peptides and conventional antibiotics is proposed as a novel perspective on treatments to combat severe bacterial infection.

Antibacterial activity and toxicity of Halymenia durvillei red seaweed from Kayangan island, South Sulawesi, Indonesia

  • This study aimed to determine the antibacterial activity and toxicity of methanol and hexane extracts of Halymenia durvillei red seaweed which were found abundantly in Kayangan island, South Sulawesi. The antibacterial activity of the crude extract was tested against five gram-negative bacteria, namely Escherichia coli, Salmonella typhi, Pseudomonas aeruginosa, Aeromonas hydrophila, and Vibrio harveyi at a dose of 200 g/disk. Extract toxicity was tested on Artemia salina larvae at concentrations of 1,000, 500, 250, 125, 62.5, and 31.25 ㎍/mL. The results showed that the methanol and hexane extracts of H. durvillei had the highest activity against S. thypi and A. hydrophila, respectively, with inhibition zones of 26.2 mm and 21.0 mm. On the other hand, the two extracts did not show activity against E. coli and P. aeruginosa, respectively. The toxicity of the methanol extract of H. durvillei was twice as high as that of the hexane extract with half-maximal inhibitory concentration of 98.24 and 184.21 ㎍/mL, respectively. Thus, the methanol and hexane extracts of red seaweed H. durvillei have the potential as new antibacterial agents respectively against the pathogenic bacteria S. typhi and A. hydrophila, but also have the opportunity to be developed into antitumor herbal compounds.

A Study on the Functional Improvement of Natural Dyed Fabrics (천연염색 직물의 기능성 향상에 관한 연구)

  • In this study, a dye bath was made using a mixture of distilled water, pine needle extract and bamboo vinegar. Analysis and comparison of various functions of cotton fabrics dyed using bamboo charcoal and gardenia seeds in distilled water and in the dye bath mixture were done. The summary of the results is as follows. Comparing bamboo vinegar and the pine needle extracts, the bamboo vinegar had a higher antibacterial function. In the case of natural dyeing cotton fabrics using bamboo charcoal and gardenia seeds, the additional amount of dyestuff for optimization is 10 g and 20 g respectively per 1 liter of distilled water. The suitable dye bath mixture ratio of distilled water, pine needle extract, and bamboo vinegar considering functionality and economical efficiency of dyed fabrics is 4:3:3. By using the mixture made in this study as a dye bath, improved antibacterial function, deodorization, and colorfastness can be obtained more than distilled water alone.
