Objectives: To experimentally assess the anti-depressant effects of Gammakdaejo-tang Complex Extracts (GMDJ-Tang) in rats. Methods: Twenty Wistar Hannover (160~170 g) stressed rats were treated with different concentrations of GMDJ-Tang extracts i.e., 0, 62.5 mg/kg, 125 mg/kg, and 250 mg/kg. Chronic mild stress was induced by food deprivation, empty bottles, forced swimming, flickered light, tilt cages, shaking cages, high density breeding, water deprivation, and by soaking the litter cover according to fixed schedule. Blood and brain tissue samples were collected for biochemical analysis. Tests included serotonin and ELISA assays. Results: GMDJ-tang increased the weight of treated rats as well as levels of serotonin, BDNF and TrkB; however, the differences were not significant. In contrast, the extracts significantly decreased blood glucose in stressed rats. GMDJ-tang extracts did not significantly impact Serum AST, ALT, leukocytes, erythrocytes and thrombocytes when comparing treatment groups to control rats. Likewise, hemoglobin, hematocrit and PLT increased in treatment groups following treatment with GMDJ-tang, but this change was without significance. Conclusions: These results suggest that GMDJ-tang can alleviate chronic mild stress in rats, possibly through anti-depressant activity.
The action of serotonin (5-Hydroxytryptamine) on the excretory function of the kidney was investigated in the dog, utilizing the clearance method and the stop-flow technique. It was shown that serotonin, $10{\mu}g/kg/min$, i. v., exerts a marked antidiuretic effects and elicits a marked hemodynamic changes in the kidney: a highly significant decrease of the glomerular filtration rate and a tendency of decrement in the renal plasma flow. Little change in the systemic blood pressure was noted, and the participation of the antidiuretic hormone in the antidiuretic action was ruled out by adding vasopressin to the infusion fluid. The stop-flow analysis showed that there is no evidence of altered activity in the tubules by serotonin. It was thus concluded that serotonin elicits anti diuresis in the dog by decreasing glomerular filtration rate, which results from the constriction of Vas afferens in the glomeruli.
효모 (Saccharomyces cerevisiae)는 인체에 무해한 Generally Recognized As Safe(GRAS)급으로 인정되며, 최근 효모 추출물이 정신적 긴장, 과민, 두통 같은 월경 전 증후군을 완화시키는 효과가 있음이 보고되어 있어, 효모추출물(Saccharomyces cerevisiae extract: SCE)과 그 분획(SCE-40)이 우울증과 불안증에서도 효과가 있는지 확인하고자 다음 실험을 실시하였다. 행동학적 검정으로 SCE의 항우울 효과를 확인하기 위해 SCE를 0, 1, 10, 100mg/kg의 농도로 웅성 ICR 생쥐에게 2주간 경구 투여한 후 강제수영검사(forced swimming test; FST)에서의 부동시간과, 또 다른 항우울 효과 검정 실험인 미현수검사(Tail Suspension Test: TST)에서의 부동시간을 측정하였다. 또한, SCE의 항불안 효과 검정을 위해서 SCE를 0, 1, 10, 100mg/kg의 농도로 웅성 ICR 생쥐에게 1회(1시간 전), 혹은 2주간 경구 투여한 후 고위십자미로검사(Elevated-plus-maze test : EPM)에서의 open arms에 머문 시간을 측정하였다 시험관내 실험으로는 웅성 백서의 대뇌피질에서의 SCE 및 SCE-40의 serotonin transporter (SERT), norepinephrine transporter(NET), 그리고 GABA의 ligand의 결합 억제능 측정과, serotonin (5-hydroxytryptamine: 5-HT), norepinephrine uptake 측정이 수행되었다. 행동적 실험 결과, FST에서 1회나 2주간 SCE 10mg/kg과 100mg/kg을 투여 받은 생쥐에서 생리 식염수만 투여 받은 생쥐보다 부동시간이 유의하게 감소됨을 보여 SCE에 의해 항우울 효과가 유발됨을 보였고, TST 실험에서도 유사한 결과가 나타났다. 또한, 항불안을 측정하는 EPM 실험에서도 1회나 2주간 SCE 10mg/kg과 100mg/kg을 투여 받은 생쥐에서 생리 식염수만 투여 받은 생쥐보다 open arms에서 머무는 시간이 유의하게 증가되어 SCE가 단기 또는 장기간의 항불안에 효과가 있음을 나타냈다. 시험관내 실험 결과에서는, SCE와 SCE-40이 SERT, NET, 그리고 GABA ligand의 결합 억제능이 있음이 확인되었고, SCE와 SCE-40은 serotonin(5-hydroxytryptamine: 5-HT)과, norepinephrine의 uptake를 억제하는 것으로 나타났다. 그러므로 본 연구는 효모 추출물(SCE)과 그 분획(SCE-40)이 행동적, 신경학적으로 항우울, 항불안에 효과가 있음을 확인하였으며, 효모 추출물(SCE)과 그 분획(SCE-40)이 안전한 천연식품으로서 우울증, 불안증 등의 관련 질환의 예방, 치료용 의약품 개발과 기능성 식품에 효과적으로 이용될 수 있음을 시사한다.
Objectives : This experiment was designed to investigate the effect of Gamisoyo-san(Jiaweixiaoyaosan, SYS) on serotonin activity of P815 Mast Cell. Methods : The effects of SYS on activation of TPH-1 mRNA and AAADC mRNA in P815 mast cell were investigated. The effect of SYS on content of serotonin in P815 mast cell was investigated. The effects of SYS on activation of DPPH and SOD in P815 mast cell were investigated. Results : 1. The SYS increased the activation of SOD and DPPH in P815 mast cell. 2. The SYS decreased the manifestation TPH-1 mRNA in P815 mast cell. 3. The SYS decreased the manifestation AAADC mRNA in P815 mast cell. Conclusions : This experiment shows that Gamisoyo-san might not be effective for treating depression in terms of biogenic amine theory. However, Gamisoyo-san showed significant anti-oxidative effect and it can not yet be ruled out for treating depression. Therefore, pathogenesis of depression and clinical research of Gamisoyo-san is suggested for future research.
Objectives: This study was perfomed to investigate the antioxidant activity and serotonin activity of Gami-guibi-tang on P815 Mast Cell. Methods: The effects of Gami-guibi-tang on activation of TPH-1 mRNA and AAADC mRNA in P815 mast cell were investigated. The effect of Gami-guibi-tang on content of serotonin in P815 mast cell was investigated. The effects of Gami-guibi-tang on activation of DPPH radical scavenging and SOD in P815 mast cell were investigated. Results: 1. The Gami-guibi-tang increased SOD activity and DPPH radical scavenging activity. 2. The Gami-guibi-tang increased the intracellular concentration of serotoninin 60 ${\mu}g/ml$, 80 ${\mu}g/ml$ experiment group. 3. The Gami-guibi-tang increased menaingful the manifestation TPH mRNA. 4. The manifestation of AAADC and MAO mRNA have not made menaingful changes on Gami-guibi-tang. Conclusions: This experiment shows that Gami-guibi-tang had significant anti-oxidative effect. And Gami-guibi-tang increased the intracellular concentration of serotonin. Therefore, Gami-guibi-tang can be used by the medication of major depression disorder. But Study on mechanism of increased serotonin and clinical research of Gami-guibi-tang is suggested for future research.
Rapid eye movement (REM) sleep has an essential role in the process of learning and memory in the hippocampus. It has been reported that linalool, a major component of Lavandula angustifolia, has antioxidant, anti-inflammatory, and neuroprotective effects, along with other effects. However, the effect of linalool on the cognitive impairment and behavioral alterations that are induced by REM-sleep deprivation has not yet been elucidated. Several studies have reported that REM-sleep deprivation-induced memory deficits provide a well-known model of behavioral alterations. In the present study, we examined whether linalool elicited an anti-stress effect, reversing the behavioral alterations observed following REM-sleep deprivation in mice. Furthermore, we investigated the underlying mechanism of the effect of linalool. Spatial memory and learning memory were assessed through Y maze and passive avoidance tests, respectively, and the forced swimming test was used to evaluate anti-stress activity. The mechanisms through which linalool improves memory loss and behavioral alterations in sleep-deprived mice appeared to be through an increase in the serotonin levels. Linalool significantly ameliorated the spatial and learning memory deficits, and stress activity observed in sleep-deprived animals. Moreover, linalool led to serotonin release, and cortisol level reduction. Our findings suggest that linalool has beneficial effects on the memory loss and behavioral alterations induced by REM-sleep deprivation through the regulation of serotonin levels.

The effect of alcoholic extracts of Costus specious (Family: Zingiberaceae) was evaluated in experimental models of pain and inflammation. Oral administration of 100, 200 and 300 mg/kg of C. specious extracts were used for the above study. Crude extracts of C. specious (300 mg/kg dose) showed maximum time needed for the response against thermal stimuli ($7.242\;{\pm}\;0.532\;s$) which is comparable to diclofenac sodium ($8.471\;{\pm}\;0.25\;s$) in the hot plate test. The MPH (Maximum Possible Analgesia) has been found to be 14.285 for 300 mg/kg dose of the crude extract while the MPH for diclofenac was 15.857 after 60 min of administration in the hot tail-flick method. The crude extract at 300 and 200 mg/kg doses showed significant reduction in acetic acid induced writhings in mice with a maximum effect of 59.661% reduction at 300 mg/kg dose which is comparable to standard diclofenac sodium (73.4%). Alcoholic extract of C. specious showed significant inhibition in serotonin and egg albumin induced hind paw oedema in rats at 100, 200 and 300 mg/kg of the crude extracts respectively (Serotonin induced edema 44.22; 53.75; 58.51%; egg albumin induced edema - 41.317; 53.892; 59.880% inhibition after 4 h respectively). The antiinflammatory effects showed by the extract were comparable to that of standard indomethacin 5 mg/kg (Serotonin induced edema 77.56%; egg albumin induced edema 77.844% inhibition after 4 h). These results suggest that the extract possesses both the anti-inflammatory and analgesic activity on mice and rat model.
Cedrus deodara (Pinaceae) has been used traditionally in Ayurveda for the treatment of central nervous system disorders. 3,4-bis(3,4-dimethoxyphenyl)furan-2,5-dione (BDFD) was isolated from heart wood of Cedrus deodara and was shown to have antiepileptic and anxiolytic activity. Thus, the present study was aimed to explore its anti-depressant effect and to correlate the effect with serotonin and nor adrenaline levels of brain. Albino mice were used as experimental animal. Animals were divided in to three groups; vehicle control, imipramine (30 mg/kg i.p.), BDFD (100 mg/kg i.p.). Tail suspension test (TST) and forced swim test (FST) was performed to evaluate antidepressant effect of BDFD. BDFD (100 mg/kg, i.p.) showed a significant decrease in immobility time when subjected to FST whereas immobility time was not significantly altered in TST. BDFD treatment increased serotonin and noradrenaline levels in the brain which is indicative of BDFD having possible atypical antidepressant action.

Safflower (Carthamus tinctorius) seeds are wellknown traditional oriental medicines that have long been used for the remedies of blood stasis and bone formation in east Asia. In this study, ethyl acetate (EtOAc) was used for extraction of the main chemical compounds from C. tinctorius seeds. Four major compounds were identified, acacetin, cosmosiin, N-feruloyl serotonin and N-(p-coumaroyl) serotonin. Each compound was evaluated for its inhibitory activity against the inflammatory process of macrophages. All compounds significantly inhibited production of lipopolysaccharide (LPS)-stimulated nitric oxide (NO) and pro-inflammatory cytokines. The protein levels of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) were dramatically decreased by serotonins in a dose-dependent manner in LPS-stimulated RAW 264.7 macrophages. These results suggest that serotonin derivatives from safflower seeds may reduce inflammation-related diseases.
This study aimed to investigate the anti-stress function of Aucklandiae Radix (AR) and Cyperi Rhizoma (CR). The essential oils used in the experiment were extracted from AR and CR using Steam Distillation Extraction and Super critical CO2 extraction. To observe the effects of sample administration, we measured feed intake, leukocytes, red blood cells, hemoglobin, platelets, serum serotonin content, immobility time, climbing time, and swimming time in mice subjected to chronic restraint stress as behavioral changes. The average body weight of all experimental groups increased than the average body weight of the control group. The immobility and climbing times of experimental groups A and B administered with supercritical extraction samples were shorter than those of the other experimental groups and the control group, and the swimming time was longer. The serotonin content in the blood of all experimental groups decreased compared to the normal group, and the serotonin content of the control group was increased. The authors suggest that Korean herbal medicines AR and CR may be utilized as anti-stress flavoring agents based on the above results.
