Anti-Oxidant, Anti-Melanogenic, and Anti-Inflammatory Activities of Zanthoxylum schinifolium Extract and its Solvent Fractions (산초 추출물 및 분획물의 항산화, 미백 및 항염증 활성)

  • This study was designed to explore new nutraceutical and cosmetic resources possessing biological activities from the plant kingdom. To fulfill this purpose, we analyzed the anti-oxidative, anti-melanogenic, and anti-inflammatory activities of Zanthoxylum schinifolium extract (ZSE) and its solvent fractions using in vitro assays and cell culture model systems. Three kinds of ZSE treated with methanol, ethanol, and water exhibited potent anti-oxidative activities through DPPH radical scavenging capacity, and inhibited in vitro DOPA oxidation. Furthermore, Z. schinifolium methanol extract (ZSME) inhibited the ${\alpha}$-melanocyte stimulating hormone, which induces melanin contents in B16F10 cells. Its anti-melanogenic activity originates from the inhibition of tyrosinase enzyme activity and melanogenesis related protein expression. Moreover, lipopolysaccharide induced nitric oxide production in the RAW 264.7 cell line was also ameliorated by ZSME treatment in a dose dependent manner. Among the four solvent fractions of ZSME treated with dichloromethane, ethyl acetate, n-butanol, and water, three fractions, except water, showed significant anti-melanogenic and anti-inflammatory activities. Taken together, these results provide important new insights into Z. schinifolium, indicating that it possesses numerous biological activities such as anti-oxidative, anti-melanogenic, and anti-inflammatory activities. Therefore, it may well serve as a promising material in the field of nutraceuticals and cosmetics.

  • Natural essential oils showed anti-microbial activity on relatively broad spectrum of bacterial and fungal species. These materials had much more intensive anti-microbial activities than synthetic preservatives on C. albicans, A niger, and P. acnes, especially. In the experimental group, anti-microbial activity was order of tea tree oil (from Melaleuca alternifolia) > methylparaben > phytoncide (from Chamaecyparis obtusa). Also, natural essential oils had anti-oxidative and anti-inflammatory effects. These results suggest that natural essential oils can be useful as good cosmetic ingredients such as natural preservatives and anti-oxidants.

  • There is currently increased interest in the identification of antioxidant compounds that are pharmacologically potent and have low or no side effects. Plants produce significant amounts of antioxidants to prevent the oxidative stress caused by photons and oxygen, therefore they represent a potential source of new compounds with antioxidant activity. Moutan Root Sark (MRS) has been frequently used as analgesic. antispasmodic, anti-inflammatory and remedies for female diseases. In this study. the antioxidant activity of extract from MRS was studied in vitro methods by measuring the antioxidant activity by TEAC, measuring the scavenging effects on reactive oxygen species (ROS) [superoxide anion, hydroxyl radical] and on reactive nitrogen species (RNS) [nitric oxide and peroxynitrite] as well as measuring the inhibitory effect on $Cu^{2+}$ induced human LDL oxidation and the inhibitory effect on collagen induced platelet aggregation. The MRS extracts were found to have a potent scavenging activity, as well as an inhibitory effect on LDL oxidation and on platelet aggregation. In conclusion, the MRS extracts have anti-oxidative and anti-atherosclerotic effects in vitro system, which can be used for developing pharmaceutical drug against oxidative stress and atherosclerosis.

Study on Anti-Skin Aging Effect of Sanguisorba officinalis L. (지유(地楡)의 피부 노화(老化)에 대한 연구(硏究))

  • To develop a new anti-skin aging cosmetics or functional foods by using antioxidative activity and collagenase inhibitor, a potent collagenase or elastase inhibitor was screened from various extracts of medicinal plants and its optimal extraction condition was investigated. And antioxidative activity, antimicrobial activity and inhibitory of effect against collagenase activity were investigated. In the these results, we selected the Sanguisorba (Sanguisorba officinalis L.) that presents a potential biological activities. Sanguisorba which is very rich in triterpenoid saponin and tannins was recently reported its anti-oxidant activities and phytoestogenic activities in vivo test and many clinical studies. The experiments were carried out in vitro to determine anti-oxidant activities of Sanguisorba extracts on DPPH radical scavenging activity assay, Superoxidase scavenging activity assay, Elastase and collagenase activity assay. It show that the Sanguisorba extracts have the most significant anti-oxidant on free radical scavenging activity assay, and also inhibited significantly activities of elastase, collagenase. Further, Sanguisorba extracts are activated Type I collagen protein expression in CCD-986sk cells. These result suggest that the Sanguisorba extracts on DPPH radical scavenging activity assay, Superoxidase scavenging activity assay, elastase and collagenase activity assay, Type I collagen protein expression in CCD-986sk cells effected could be developed cosmetic ingredients for anti-aging.

Effects of BojungIkkiTang-Gamybang on Protective of Cell Death and Anti-Oxidative in C6 Glioma Cell (보중익기탕가미방(補中益氣湯加味方)이 신경교세포의 세포사멸보호 및 항산화에 미치는 영향)

  • This study was designed to investigate the effects of BojungIkkiTang-Gamybang freeze dried powder (BITG) on proliferauion, protective of cell death induced by chemicals such as paraquat, hydrogen peroxide etc and anti-oxidative effects in C6 glioma cells. In our results, BITC accelerated proliferation rates of C6 cells in vitro. In addition, protective effects on cell death induced by paraquat and hydrogen peroxide. And, BITC did not have effects on SOD and total glutathione activities, but decresed malone dialdehyde activity. In conclusion, these results suggest the possibility of BojungIkkiTang-Gamybang to protect brain cell or neuronal cell from damage induced by oxidative stress. And also suggest that related mechanisms are involved in malone dialdehyde activity.

Quantification of γ-Oryzanol Components and Comparison Its Biological Activity in Brown Rice (현미(추청벼, 흑진주벼, 신동진벼)에 함유된 γ-Oryzanol의 함량 분석 및 생리활성 효과 비교)

  • We analyzed the contents of ${\gamma}$-oryzanol, which is contained in brown rice of the nation rice varieties Chucheong, Heukjinju and Sindongjin, by HPLC. Furthermore we also performed experiments on its biological activity, to prove the effectiveness of rice bran. The contents of ${\gamma}$-oryzanol contained in hulled rice showed 1,587 ppm for Heukjinju, followed by Chucheong(1,038 ppm), and by Sindongjin(472 ppm). In anti-oxidative activity, we performed an experiment, by measuring the radical scavenging activity of DPPH. Heukjinju showed the best effect, and Chucheong showed the worst effect. In cholesterol lowering activity, Heukjinju showed the best activity and Sindongjin showed the worst effect. In anti-bacterial activity as well, Heukjinju showed the best activity, and Sindongjin showed the worst effect. Through these experiments, we compared the contents of ${\gamma}$-oryzanol, which is contained in hulled rice(Chucheong, Heukjinju, Sindongjin). Also, we found the anti-oxidation effect, cholesterol lowering effect, and anti-bacterial activity of the ${\gamma}$-oryzanol extracts. Based on our research, we expect that ${\gamma}$-oryzanol can work as a new drug, or nutritional supplement.

Salt-water Processing-dependent Change in Anti-oxidative and Anti-inflammatory Effects of Cortex Eucommiae (염수초 포제법에 따른 두충의 항산화 및 항염증 활성 변화 비교연구)

  • Objectives The present study aimed to investigate the change in marker compounds, anti-oxidative and anti-inflammatory effects of salt-water processed Cortex Eucommiae. Methods To evaluate the influence of processing on anti-oxidant effect of Cortex Eucommiae, changes in total phenol, total flavonoid, 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) free radical scavenging, and ferric reducing antioxidant power (FRAP) between processed and raw Cortex Eucommiae were assessed. In addition, nitrite assay was conducted to determine the influence of processing on anti-inflammatory effect of Cortex Eucommiae. Cell viability was also examined as to elucidate whether processing affects cytotoxicity of Cortex Eucommiae. Finally, high-performance liquid chromatography (HPLC) analysis was conducted to monitor changes in pinoresinol diglucoside amount of processed and raw Cortex Eucommiae. Results Salt-water processed Cortex Eucommiae showed higher total phenol and flavonoid amount, compared to raw Cortex Eucommiae. Furthermore, anti-oxidative activity of processed Cortex Eucommiae was improved as discovered in DPPH, ABTS, and FRAP assays. Anti-inflammatory effect of Cortex Eucommiae was also enhanced following salt-water processing, as evidenced in nitrite assay. HPLC analysis found that the amount of pinoresinol diglucoside, widely known as the marker compound of Cortex Eucommiae, increases through salt-water processing. All experiments were performed with non-toxic concentration of Cortex Eucommiae; processing did not affect the cytotoxicity of Cortex Eucommiae up to the currently adopted concentration. Conclusions The present results support that salt-water processing of Cortex Eucommiae is beneficial in terms of marker compound amount, anti-oxidative, and anti-inflammatory activities. Additional investigations are needed to standardize the processing method of Cortex Eucommiae.

Antioxidant and Anti-inflammatory Effects of Solvent Fractions from the Peel of the Native Jeju Citrus ‘Hongkyool’ and ‘Pyunkyool’ (제주 재래종 감귤 ‘홍귤’, ‘편귤’ 과피 분획물의 항산화, 항염증 효과)

  • This study investigated the anti-oxidative and anti-inflammatory activity of unripe fruit peel solvent fractions of the native jeju citrus ‘Hongkyool’ (Citrus tachibana Tanaka) and ‘Pyunkyool’ (C. tangerina Hort. ex Tanaka). The total polyphenol content and total flavonoid content were highest in the butanol fraction of both ‘Hongkyool’ (534.4 mg/g, 431.8 mg/g) and ‘Pyunkyool’ (342.9 mg/g, 415.7 mg/g). In both cultivars, the butanol fraction showed the strong antioxidant activity by DPPH radical scavenging and ABTS radical scavenging. The DPPH radical scavenging of the butanol fraction from ‘Hongkyool’ and ‘Pyunkyool’ was 89% and 64% at a concentration of 1 mg/ml, respectively. The ABTS radical scavenging of the butanol fraction from ‘Hongkyool’ and ‘Pyunkyool‘ was 94% and 85% at a concentration of 1 mg/ml, respectively. We investigated the effect of the anti-inflammatory activity of the ethyl acetate fraction from ‘Hongkyool’ and ‘Pyunkyool’ on LPS-induced NO production, IL-6, iNOS, and COX-2 protein expression in Raw 264.7 cells. At concentrations of 50 and 100 μg/ml of the ‘Hongkyool’ ethyl acetate fraction, the anti-inflammatory effect was excellent. These results suggest that ethyl acetate and butanol fractions from ‘Hongkyool’ and ‘Pyunkyool’ could be useful functional materials, with anti-oxidative and anti-inflammatory properties.

Anti-Oxidant and Anti-Inflammatory Complex Health Functional Analysis of Chinese Resource Plant Boehmeria, Carlemannia and Clausena (중국 자원식물 보메리아, 칼라마니아, 클로세나의 항노화 항염 복합 건강관리 기능성 연구)

  • Ageing has been known to be deeply related to oxidative activities. Furthermore, inflammation is a response initiated by conditions such as infection and injury. It has been known that complex connections occur between oxidative stress and the inflammatory response, however underlying mechanism needs to be investigated. Chinese resource plants have been widely used as functional food and medicine for a long time, however it is not studied well how many of these resource plants work. We first decided testing anti-oxidant and anti-inflammatory activities with 95% ethanol extracts of the chinese resource plants, Boehmeria pilosinscula (Blume) Hassk., Carlemannia tetragona Hook f. and Clausena emarginata C.C.Huang. To measure anti-oxidant activity, we performed DPPH assay in Raw264.7 cells with 95% ethanol extracts. Clausena extracts showed significantly higher anti-oxidant activity than those of Boehmeria and Carlemannia. We then performed Nitric Oxide assay to measure the inflammation suppression levels with 95% ethanol extracts of these plants. Clausena extracts showed significantly higher suppression of nitric oxide production than those of Boehmeria and Carlemannia. This indicates that inflammation levels are significantly reduced by Clausena. After measuring anti-oxidant and anti-inflammatory activities, we then performed MTT assay to measure the cell survival rate of Raw264.7 cells treated with these extracts. Boehmeria showed much more cell survival rate than Carlemannia and Clausena. Taken together, this result suggests that Clausena extracts have more anti-oxidant and antiinflammatory activities than Boehmeria and Carlemannia, while Boehmeria extracts have more cell survival rate than Carlemannia and Clausena.

Anti-oxidative Properties of Lipids Extracted from Rainbow Trout (Oncorhynchus mykiss) Fed with Carotenoids and Conjugated Linoleic Acid

  • A commercial diet supplemented with carotenoids and conjugated linoleic acid was fed to rainbow trout (Oncorhynchus mykiss) for 8 weeks. To investigate the anti-oxidative properties of these compounds, lipids from the muscle and viscera of the fish were subjected to different assays. At $10\;{\mu}g/mL$, L-ascorbic acid exhibited 95% 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging activity, while the tissue lipids showed little radical scavenging activity. At 50 and $100\;{\mu}g/mL$, the lipids of the muscles and viscera showed 11.7-22.6% and 11.3-24.9% DPPH radical-scavenging activity, respectively. A lipid peroxidation inhibitory assay using the ferric thiocyanate method was also performed in comparison with $\alpha$-tocopherol at a concentration of 6.0 mg/mL. Our results indicate that the anti-oxidative property of the lipids in fish muscle, which was 85.2% compared to 85.3% for the visceral lipids, was stronger than that of $\alpha$-tocopherol (74.3%) following 3 days of storage at $40^{\circ}C$.
