This study was conducted to confirm the application as ingredients of cosmetics through an examination of the function for anti-oxidant activity of the fraction isolated from Pyrus ussuriensis leaves. The dried leaf of Pyrus ussuriensis were extracted with acetone-$H_2O$ (6:4, v/v), concentrated and fractionated with the upper layer of acetone on a separatory funnel. Each fraction was freeze dried, then a portion of acetone soluble powder was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol as eluents and also used the MIC-gel using a series of aqueous methanol as developing solvent. The isolated compounds were identified by silica-gel TLC. The concentration of total phenolic compound of Pyrus ussuriensis acetate soluble fraction was high, 914 mg/g. The results obtained from the analyses of the anti-oxidanat effects of Pyrus ussuriensis acetate fraction can be summarized as follows: In the result of DPPH scavenging radical activity, Pyrus ussuriensis acetate soluble fraction showed more than 80% at 100 ppm. SOD-like activity of one of Pyrus ussuriensis acetate soluble fractions was 77% at 1000 ppm. Xanthine oxidase inhibition of Pyrus ussuriensis acetate soluble fraction was 38% at 100 ppm. From these results, we confirmed that acetate fraction of Pyrus ussuriensis has a great potential as a natural ingredients with a natural antioxidant and antimicrobial source.

In this study. we evaluated anti-aging activity of medical plants that protect the skin cell damage induced by UV irradiation. We have investigated diverse biological activities of Selaginella tamariscina as an anti-aging ingredient of cosmetics. S. tamariscina was found to show scavenging activities of radicals and reactive oxygen species (ROS) with the $IC_{50}$ values of $65.1{\mu}g/mL$ against 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and $40.9 {\mu}g/mL$ against superoxide radicals in the xanthine/xanthine oxidase system, respectively. For testing intracellular ROS scavenging activity, the cultured human dermal fibroblasts were analyzed by increase in dichlorofluorescein (DCF) fluorescence upon exposure to UVB $20 mJ/cm^2$ after treatment of S. tamariscina. UVA-induced MMP-1 protein and mRNA expression in human dermal fibroblasts were reduced in a dose-dependent manner by S. tamariscina. Moreover, S. tamariscina inhibited MMP-2 (gelatinase) activity in UVA-irradiated human dermal fibroblasts assayed by zymography and semi-quantitative RT-PCR. Taken together, these results suggest that S. tamariscina may act as an anti-aging agent by Increasing collagen and preventing the skin cell damage induced by UV irradiation, and imply that S. tamariscina nay be useful as a new ingredient for anti-aging cosmetics.
The aim of the study was to assess the cosmeceutical activity of Pyrus ussuriensis leaves and it is possible that can be used as a cosmetic ingredient for application of cosmetic industries. P. ussurensis leaf was extracted with various solvents including water, 70% ethanol and 60% acetone. In the result of DPPH (1,1-diphenyl-2-picryl-hydrazyl) scavenging radical activity, water, ethanol and acetone extract of P. ussuriensis leaf were higher than 70%, 80% and 85% at 50 ppm concentration, respectively. Xanthine oxidase inhibition activity and superoxide dismutase (SOD)-like activity by P. ussuriensis extract were higher than 30%. In addition, SODlike activity of all extracts showed tendency of the significant increase with the increase of concentration. In the anti-inflammatory test, P. ussuriensis leaf extract inhibited generation of nitric oxide (NO) stimulated by LPS in the macrophage cell line (raw 264.7) after 12 to 24 hours. As above results, P. ussuriensis has a great potential as a cosmeceutical raw material as well as anti-oxidant and anti-inflammatory ability.
This study was designed to determine the biological activities of Chionanthus retusus flower extracts. Water and 90% ethanol extracts of C. retusus flower were prepared. The inhibitory activities of water and ethanol extracts with a phenolic content of $200{\mu}g/mL$ against xanthine oxidase were 25.60% and 15.92%, respectively. Further, the water extract did not show any inhibitory activity against ${\alpha}$-glucosidase whereas the ethanol extract showed 100.00% inhibition of ${\alpha}$-glucosidase. The inhibitory activities of the extracts against tyrosinase were 17.27% (water extract) and 36.13% (ethanol extract), which suggest that the extracts may have a whitening effect. The water extract did not inhibit elastase activity but showed a collagenase-inhibitory activity of 20.21%. On the contrary, the ethanol extract showed 96.26% and 35.93% inhibition of collagenase and elastase, respectively. These findings suggest that the extracts may have an anti-wrinkle effect. Lastly, the extracts showed a hyaluronidase inhibitory activity of 36.96% (water extract) and 88.70% (ethanol extract), suggesting that they may have an anti-inflammatory effect. The results indicate that C. retusus flower extracts containing phenolic compounds can be used as functional resources because they have anti-gout, carbohydrate degradation-inhibitory, whitening, anti-wrinkle, and anti-inflammatory effects.

The purpose of this study was to investigate anti-obesity and anti-hyperlipidemic effects of extracts from Geranium thunbergii (GT) in high-fat-diet-induced obese rats. Animals were randomly divided into four groups [normal diet, high fat diet, MeOH extract of GT (GTM), and BuOH fraction of GT (GTB)] and GT samples were treated with dose of 100 mg/kg for 8 weeks. It was observed that GTB-treated group significantly reduced body weight gain, food intake, epididymal fat weight, and triglyceride level in serum and liver compared to control group. The rats fed GTB also decreased contents of thiobarbituric acid-reactive substances (TBARS), hydroxyl radical, and xanthin oxidase (XO) increased by high fat diet. Furthermore, the anti-oxidant enzymes such as superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and catalase were increased by GTB treatment. The experimental results indicate that GTB has anti-obesity and anti-hyperlipidemic effects, as well as radical scavenging activity.

Objective : This study was performed to assess the effects of Aloe and Violae herba extracts on skin disease and skin beauty. Methods : Anti-oxidant effects were measured by the scavenging for DPPH radical, xanthine oxidase activity. Anti-inflammatory effects were examined by relations in NO synthesis, IL-$1{\beta}$, IL-6, TNF-$\alpha$, NF-kB, COX-2, MAP kinase. The skin wrinkle formation effects were measured by collagenase and elastase activities. The whitening effects were examined by tyrosinase activities, melanin synthesis in MNT-1 cell. Results : 1. In an anti-oxidant test, Aloe and Violae herba extracts showed high radical scavenging activity. 2. In an anti-inflammatory test, Aloe and Violae herba extracts strongly inhibited the lipopolysaccharide(LPS)-induced nitric oxide(NO) release from the RAW 246.7 macrophage cells. Aloe and Violae herba extracts also inhibited the LPS-induced IL-$1{\beta}$ and COX-2 expressions. The inhibitory effects of Aloe and Violae herba extracts on macrophage activation were via the inhibition of NF-kB, evidenced by transient transfection assay. Furthermore, Aloe and Violae herba extracts weakly inhibited the activation of Jun-N-terminal kinase(JNK) but they did not have any effects on p38 MAP kinase in RAW 264.7 cells. 3. In the skin wrinkle formation assay, Aloe extract strongly inhibited collagenase and elastase, whose activity are tightly related with the wrinkle formation. 4. In the skin whitening assay, Aloe and Viloae herba extracts weakly inhibited tyrosinase activity, however, it was not statistically significant. Besides they did not have any effects on melanin synthesis, indicating that they could not be applicable for skin whitening. Conclusion : This study show that Aloe and Violae herba extracts may play a significant role in skin disease and skin beauty.
1bis study investigated the antioxidative effect in kidney of senescence-accelerated prone SAMP8 mice with calorie restriction. 4-weeks-old SAMP8 female mice were divided into 4 groups according to the experimental feeding period: for 4, 8, 12 month, and at natural death. Each group was subdivided into 2 groups, with thirteen mice each one, as ad libitum group and as dietary restriction group (60% of ad libitum feeding amount). After feeding for a given period, the mice were sacrificed to get the following results: among the experimental groups, there wereno significant differences in xanthine oxidase (XOD) activity in their kidney tissues. The contents of cytochrome $P_{450}$ decreased in ad libitum group and dietary restriction group by age. The activity of NADPH-cytochrome $P_{450}$ reductase showed a trend similar to cytochrome $P_{450}$. Superoxide radical content increased with age. At the 4th, 8th and 12 months of the experimental period, the activity in the dietary restriction group was less than that of ad libitum group by as much as 17% 14% and 14% respectively. For hydrogen peroxide, the contents were increased in the ad libitum group with age, while no correlation between content and age was observed in the dietary restriction group. In the 8th and 12th months of the experimental period, the were in the dietary restriction group less than that of ad libitum group counterpart as much as 17% and 20o/c, respectively. For the cellular membrane stability of the kidney, no significant correlation with age was observed in either the dietary restriction group or the ad libitum group. However at the 12th month of the experiment, however, the stability in the dietary restriction group was 11 % higher than that in the ad libitum group. In conclusion, with these results obtained from the SAMP8 mouse model, we demonstrate that dietary restriction has the effects of anti-oxidation and anti-senescence in the kidney.
To study the effects of metal ions on the activities of antioxidative enzymes, the activities of superoxide dismutase(SOD), peroxidase(POD), catalase(CAT) of Persicaria vulgaris has been studied after treating with Cd, Cu, Zn and Al. 1. The activities of SOD in leaf and stem were decreased, but that in root was increased. Among the metal ions studied in this report, Al gave the highest increase in SOD activity in root. 2. The activities of POD after treating with Cd or Cu did not show any significant differences. POD activities after treating with Zn and Al has been decreased, however, that in root showed increased activities after treating with Zn 5,000 ppm or Al 500 ppm. 3. The activity of CAT in leaf was decreased with every metals studied. The CAT activity in root was increased with increased concentration. The root treated with Al showed highest activity. 4. The presence of isozymes after treated metal ions has been studied in gel electrophoresis. The POD treated plant did not show any new isozymes, but the intensity of one of pre-existent band was increased. The SOD treated plant showed the several new isozymes.
The aim of present study is to investigate antioxidative effect of the Rice Bran Extracts and Defatted Rice Bran Extracts. Rice Bran Extracts used Rice Bran Water Extract, Rice Bran Ethanol Extract, Defatted Rice Bran Water Extract, Defatted Rice Bran Ethanol Extract. This study was carried out to examine quenching effects of Rice bran extracts on DPPH-, Riboflavin-, and Xanthin oxidase- originated superoxide activities. In addition, in order to determine whether Rice Bran Extract can be safely applied to human skin, the cytotoxic effects of Rice Bran Extract in Human Dermal Fibroblast cells were determined using MTS Assay. These results demonstrated that RBE and DRBE had anti-oxidative properties and did not induce the cytotoxic effects in Human Dermal Fibroblast cells. Therefore, these findings suggest that anti-oxidative properties of RBE and DRBE may be considered convergence with skin care.
Curcumin (diferuloylmethane) is a major naturally-occurring polyphenol of Curcuma species, which is commonly used as a yellow coloring and flavoring agent in foods. Curcumin has shown anti-carcinogenic activity in animal models. Curcumin possesses anti-inflammatory activity and is a potent inhibitor of reactive oxygen-generating enzymes such as lipoxygenase/cyclooxygenase, xanthine dehydrogenase/oxidase and inducible nitric oxide synthase; and an effective inducer of heme oxygenase-1. Curcumin is also a potent inhibitor of protein kinase C(PKC), EGF(Epidermal growth factor)-receptor tyrosine kinase and LĸB kinase. Subsequently, curcumin inhibits the activation of NF(nucleor factor)KB and the expressions of oncogenes including c-jun, c-fos, c-myc, NIK, MAPKs, ERK, ELK, PI3K, Akt, CDKs and iNOS. It is proposed that curcumin may suppress tumor promotion through blocking signal transduction path-ways in the target cells. The oxidant tumor promoter TPA activates PKC by reacting with zinc thiolates present within the regulatory domain, while the oxidized form of cancer chemopreventive agent such as curcumin can inactivate PKC by oxidizing the vicinal thiols present within the catalytic domain. Recent studies indicated that proteasome-mediated degradation of cell proteins playa pivotal role in the regulation of several basic cellular processes including differentiation, proliferation, cell cycling, and apoptosis. It has been demonstrated that curcumin-induced apoptosis is mediated through the impairment of ubiquitin-proteasome pathway. Curcumin was first biotransformed to dihydrocurcumin and tetrahydrocurcumin and that these compounds subsequently were converted to monoglucuronide conjugates. These results suggest that curcumin-glucuronide, dihydrocurcumin-glucuronide, tetrahydrocurcumin-glucuronide and tetrahydrocurcumin are the major metabolites of curcumin in mice, rats and humans.
