고혈압에 사용되는 한약처방 5종의 항비만 효과 (Anti-obesity Effect of Five Formulas Which can be used to Treat Hypertension in High Fat Diet Induced Obese Mice)

  • Daesiho-tang (DST), Yijin-tang (YJT), Gyeoneumjisil-hwan (GJH), Ukdam-hwan (UDH), and Sojojung-tang (SJT) have been used to treat hypertension. The aim of this study was to investigate whether five formulas were effective for improving obesity in high fat diet (HFD) induced obese mice. The mice were divided into seven groups: (1) a normal diet (ND), (2) a high fat diet (HFD) (3) a HFD plus DST (HFD + DST), (4) a HFD plus YJT (HFD + YJT), (5) a HFD plus GJH (HFD + GJH), (6) a HFD plus UDH (HFD + UDH), or (7) HFD plus SJT (HFD + SJT) at 150 mg/kg/day for 7 weeks. All five formulas treatments significantly lowered blood pressure, final liver weights, and serum total triglyceride levels. The four formulas (HFD+DST, HFD+YJT, HFD+UDH, and HFD+SJT) decreased body weights. Also, HFD+DST, HFD+YJT, and HFD+UDH groups reduced abdominal and epididymal fat weights. The serum LDL-cholesterol levels were decreased in HFD+YJT, HFD+UDH, and HFD+SJT groups compared to the HFD group. These results demonstrate that five formulas ameliorated obesity.

Impact of Cassia acutifolia Infusion on Glucose Levels in Obesity and Diabetes Rat Model

  • Objectives: The aim of this study was to investigate the effects of Cassia acutifolia on the obesity and the glucose levels in a rat model of obesity and diabetes. Methods: By random selection, 36 Wistar male rats were divided in two control groups, the positive and the negative control groups, and into four experimental groups receiving different infusions of Cassia acutifolia in water ad libitum. Results: The results revealed a statistically significant anti-obesogenic effect (P = 0.02), although this was not considered clinically significant. Additionally, Cassia acutifolia lowered the glucose levels by 30 mg/dL to 90 mg/dL (P = 0.05). However, we observed adverse effects in the liver, a two-fold increase in transaminase levels (P = 0.002), and in the kidneys, decreased creatinine levels (P = 0.001), and these adverse effects had no viable explanation. Conclusion: Cassia acutifolia has anti-hyperglycemic effects in obese diabetic rats. However, Cassia acutifolia also has adverse effects, so it should not be administered to patients.

Anti-obesity Effect of Fermented Whey Beverage using Lactic Acid Bacteria in Diet-induced Obese Rats

  • High-protein fermented whey beverage (FWB) was manufactured using whey protein concentrate (WPC) and Lactobacillus plantarum DK211 isolated from kimchi. This study was designed to evaluate the anti-obesity activity of FWB in male rats fed a high-fat diet. Male Sprague-Dawley rats were randomly assigned to three groups (n=8 per group). The three groups differed in their diet; one group received a normal diet (ND), another, a high-fat diet (HD), and the third, a HD plus fermented whey beverage (HDFWB), for 4 wk. Supplementation with FWB (the HDFWB group) prevented weight gain and body fat accumulation. The food intake in the HDWFB group was significantly lower (p<0.05) than that of the HD group. The HDWFB group also showed a significant decrease in organ weights (p<0.05), except for the weight of the testis. There was a significant decrease in total cholesterol, LDL-cholesterol, and triglycerides in the HDFWB group compared with the HD group (p<0.05), but there was no significant difference in serum HDL-cholesterol levels among the experimental groups. Rats ingesting FWB (the HDFWB group) also showed a significant decrease in blood glucose levels, and plasma levels of insulin, leptin, and ghrelin compared to HD group (p<0.05). These results indicate that FWB has beneficial effects on dietary control, weight control, and reduction in fat composition and serum lipid level; consequently, it may provide antiobesity and hypolipidemic activity against high fat diet-induced obesity in rats.

  • Objectives: The aim of this study was to examine the effect of Asparagus cochinchinensis (AC) and fermented AC (fAC) on microorganisms and efficacies. Methods: AC was fermented for four weeks without using any bacterial strains. Then we investigated fermentation characteristics including potential of hydrogen (pH), total sugar, microbial profiling and antioxidant compound contents such as total polyphenol and total flavonoid. The anti-obesity effects of AC and fAC were evaluated by using Oil Red O staining in 3T3-L1 adipocyte. Also anti-diabetic effects of them were evaluated by using 2-[N-(7-nitrobenz-2-oxa-1,3-diazol-4-yl)amino]-2-deoxy-d-glucose (2-NBDG) uptake in C2C12 skeletal muscle cell. Results: Both pH and total sugar of fAC were decreased significantly compared to unfermented AC. And the abundance of total bacteria and lactic acid bacteria increased during fermentation, especially Lactobacillus plantarum. Also fermentation of AC increased the content of total polyphenol. On the metabolic aspects, we found that AC and fAC suppressed fat accumulation. Conclusions: After four weeks of fermentation, AC increased concentrations of active compounds, altered microbial composition, and inhibited fat accumulation such as triglyceride. These results indicate that fermentation of AC might be a beneficial therapeutic approach for obesity.

Single-Cell Hemoprotein Diet Changes Adipose Tissue Distributions and Re-Shapes Gut Microbiota in High-Fat Diet-Induced Obese Mice

  • We have previously observed that feeding with single-cell hemoprotein (heme-SCP) in dogs (1 g/day for 6 days) and broiler chickens (1 ppm for 32 days) increased the proportion of lactic acid bacteria in the gut while reducing their body weights by approximately 1~2%. To define the roles of heme-SCP in modulating body weight and gut microbiota, obese C57BL/6N mice were administered varied heme-SCP concentrations (0, 0.05, and 0.5% heme-SCP in high fat diet) for 28 days. The heme-SCP diet seemed to restrain weight gain till day 14, but the mice gained weight again later, showing no significant differences in weight. However, the heme-SCP-fed mice had stiffer and oilier bodies compared with those of the control mice, which had flabby bodies and dull coats. When mice were dissected at day 10, the obese mice fed with heme-SCP exhibited a reduction in subcutaneous fat with an increase in muscle mass. The effect of heme-SCP on the obesity-associated dyslipidemia tended to be corroborated by the blood parameters (triglyceride, total cholesterol, and C-reactive protein) at day 10, though the correlation was not clear at day 28. Notably, the heme-SCP diet altered gut microbiota, leading to the proliferation of known anti-obesity biomarkers such as Akkermansia, Alistipes, Oscillibacter, Ruminococcus, Roseburia, and Faecalibacterium. This study suggests the potential of heme-SCP as an anti-obesity supplement, which modulates serum biochemistry and gut microbiota in high-fat diet-induced obese mice.

  • Obesity is one of the most serious health problem because it induced numerous metabolic syndrome and increases the incidence of various disease, including diabetes, hypertension, dyslipidemia, atherosclerosis, and cancer. In 3T3-L1 adipocytes, increases in reactive oxygens species (ROS) occur with lipid accumulation. NADPH oxidase, producing superoxide anion, may contribute to the development of obesity-associated insulin resistance and type 2 diabetes. In this study, we elucidated the effect of Chaenomeles sinensis koehne extract (CSE) against the development of obesity and the inhibition mechanisms in 3T3-L1 preadiocytes. CSE decreased triglyceride content and inhibited the expression of adipogenic transcription factors including peroxisome proliferator-activated receptor $(PPAR){\gamma}$, CCAT/enhancer binding protein $(C/EBP){\alpha}$ and sterol regulatory element-binding protein (SREBP-1). In addition, CSE highly increased antioxidant activity in a dose-dependent manner. CSE remarkably reduced intracellular ROS increase and NAD(P)H oxidase activity, NOX1, NOX4, Rac1 protein expression, and phosphorylation of p47phox and p67phox We also studied the effect of CSE on weight gain induced by high-fat diet. The oral treatment of CSE (500 mg/kg, body weight) in diet-induced obese (DIO) mice showed decrease in triglyceride and adipocyte size. Therefore, these results indicate that the effect of CSE on anti-obese effects, adipocyte differentiation and reducing triglyceride contents as well as adipocyte size in obese mice, may be associated with inhibition of NAD(P)H oxidase-induced ROS production and adipose transcription factors. These results showed the potential to inhibit the obesity by CSE treatment through controlling the activation of NAD(P)H oxidase in vitro and in vivo obese model.

고지방 식이로 유도된 비만 쥐에서 실리빈 (Silybin)이 체중 및 내당 능에 미치는 영향 (Effect of Silybin on Body Weight and Glucose Tolerance in High-fat-diet Induced Obese Mice)

  • In this study, we investigated the effects of silybin on body weight and glucose tolerance in mice fed high fat diet mice. We found that body weight, plasma TG contents, fat size, glycerol, 3-hydroxybutyrate and total cholesterol were significantly decreased in silybin (500 mg/kg) supplemented groups compared to high fat diet group. Whereas, total food intake was not changed between high fat diet group and high fat diet plus silybin group. Futhermore, supplement of high fat elevated the glucose intolerance and was improved in silybin supplement group. Finally, we examined the effect of silybin on circulating adipocytokine level to explore the possible mechanism by which silybin improves high fat diet-induced obesity and diabetes. The silybin supplement significantly reduced the level of adipocytokine, such as leptin, resistin, IL-6, and MCP-1 induced by high fat diet. These results suggest that silybin can be used to improve obesity and diabetes.

Dietary Aloe Improves Insulin Sensitivity via the Suppression of Obesity-induced Inflammation in Obese Mice

  • Background: Insulin resistance is an integral feature of metabolic syndromes, including obesity, hyperglycemia, and hyperlipidemia. In this study, we evaluated whether the aloe component could reduce obesity-induced inflammation and the occurrence of metabolic disorders such as blood glucose and insulin resistance. Methods: Male C57BL/6 obese mice fed a high-fat diet for 54 days received a supplement of aloe formula (PAG, ALS, Aloe QDM, and Aloe QDM complex) or pioglitazone (PGZ) and were compared with unsupplemented controls (high-fat diet; HFD) or mice fed a regular diet (RD). RT-PCR and western blot analysis were used to quantify the expression of obesity-induced inflammation. Results: Aloe QDM lowered fasting blood glucose and plasma insulin compared with HFD. Obesity-induced inflammatory cytokine (IL-$1{\beta}$, -6, -12, TNF-${\alpha}$) and chemokine (CX3CL1, CCL5) mRNA and protein were decreased markedly, as was macrophage infiltration and hepatic triglycerides by Aloe QDM. At the same time, Aloe QDM decreased the mRNA and protein of $PPAR{\gamma}/LXR{\alpha}$ and $11{\beta}$-HSD1 both in the liver and WAT. Conclusion: Dietary aloe formula reduces obesity-induced glucose tolerance not only by suppressing inflammatory responses but also by inducing anti-inflammatory cytokines in the WAT and liver, both of which are important peripheral tissues affecting insulin resistance. The effect of Aloe QDM complex in the WAT and liver are related to its dual action on $PPAR{\gamma}$ and $11{\beta}$-HSD1 ression and its use as a nutritional intervention against T2D and obesity-related inflammation is suggested.

원유에서 분리한 Enterococcus faecalis MD366의 생리적 특성 및 비만 억제 효과 (Physiological Characteristics and Anti-Obesity Effect of Enterococcus faecalis MD366 isolated from Raw Milk)

  • 본 연구는 원유로부터 비만 억제 능력이 있는 젖산균을 분리 및 동정하고, 이 균주의 생리적 특성을 규명하여 상업적으로의 이용가능성을 검토하고자 실시하였다. 이를 위해 Modified MRS 분별배지를 사용하여 노란색 집락을 형성하는 균주를 대상으로 각각 Anti-lipase activity와 Anti-adipogenic activity가 우수한 균주를 선발한 결과, MD366 균주가 최종 선발되었다. MD366 균주는 lipase 억제활성 $65.03{\pm}0.94%$, 지방분화억제활성 $27.4{\pm}1.4%$로나타났으며, 동정결과Enterococcus faecalis로 판명되었고, Enterococcus faecalis MD366으로 명명하였다. E. faecalis MD366의 최적 생장 온도는 $37^{\circ}C$ 이었으며, 담즙산과 산성의 pH에서 모두 우수한 생존력을 나타내었다. 효소활성은 전반적으로 낮았으나 acid phosphatase에 대해 비교적 높은 효소 활성을 나타내었다. 항생제 내성 실험 결과 neomycin, kanamycin, vancomycin에 내성이 있는 반면, novobiocin에 감수성을 나타냈으며, Escherichia coli, Salmonella Typhimurium과 Staphyloccous aureus에 대해 각각 80.4%, 60.2%와 65.4%의 억제 효과를 지니고 있는 것으로 나타났다.

제2형 당뇨 동물모델을 이용한 방풍통성산의 고혈당 개선효과 (Anti-hyperglycemic Effect of Bangpungtongseong-san on Type II Diabetes Animal Models)

  • Background: Type II diabetes is considered as one of the common diseases. Bangpungtongseongsan (BPS) has been used as a traditional medicine for treating obesity and hypertension in Korea. According to previous reports, it has anti-obesity, anti-chronic asthma, anti-oxidant, and anti-inflammatory properties. However, the effects of BPS on type II diabetes have not yet been elucidated. Thus, in this sutudy, we evaluated the water extracts of BPS using type II diabetes animal models. Methods and Results: Each group was orally administered with BPS (170, 850 and 1,700 mg/kg) for approximately 13 weeks. A mixture of 150 mg/kg metformin and 10 mg/kg sitagliptin (MS) was used as a positive control. The glycated hemoglobin (HbA1c) and glucose levels, and hematological parameters including blood urine nitrogen, creatinine, low density lipoprotein and total cholesterol, were measured using blood samples. Treatment with 170 mg/kg BPS decreased the HbA1c and glucose levels in blood without affecting the weights of the animals. However, threatment with 1,700 mg/kg BPS reduced the weights and fatty liver, and increased the blood glucose level in type II diabetes animal models Conclusions: These results indicate that a low dose of BPS for 13 weeks, which reduces HbA1c and blood glucose levels, could be used for the treatment of type II diabetes. However, further studies are required to elucidate how active ingredients of BPS influence HbA1c and glucose levels in blood.
