The Effect of Selaginella tamariscina on Inhibition of Pancreatic Lipase and Lipid Accumulation (부처손(Selaginella tamariscina) 추출물의 리파아제 저해 활성 및 지질 축적 억제 효과)

  • The objective of this study was to evaluate novel usability as natural anti-obesity supplement of Selaginella tamariscina extract. The total phenol contents and total flavonoid contents were $60.29{\pm}3.11GAE\;mg/g$ and $14.90{\pm}0.34QE\;mg/g$, respectively. To evaluate anti-obesity activity of Selaginella tamariscina extract, pancreatic lipase inhibition activity as well as its inhibition effect of lipid accumulation in adipocytes were conducted by Oil Red O staining and lipolysis assay. The result of pancreatic lipase inhibition activity of S. tamariscina extract showed a wide range between 40 and 73% dose dependently. While the incubation of 3T3-L1 cells with S. tamariscina extract inhibited differentiation of preadipocytes and reduced lipid accumulation, the level of released free glycerol into culturing medium was increased in multiple concentrations. These results showed that S. tamariscina extract inhibit adipogenesis and pancreatic lipase activity. Thus, S. tamariscina extract can be a candidate for regulating lipid accumulation in obesity.

Anti-obesity Effect of Sargassum confusum Ethanol Extract in Obese Rats (알쏭이모자반 주정추출물의 항비만 효과)

  • Effect of Sargassum confusum extract on the reduction of body weight gain and lipid contents in obese rats were evaluated to find natural materials with anti-obesity benefits. After inducing obesity by feeding 42.5% high-fat diet for 5 weeks, each 10 Sprague-Dawley rats were randomly assigned to high-fat diet control (HFD) group and high-fat diet group containing 3% Sargassum confusum extract (HFDSC). Weight gain of HFD group ($2.96{\pm}0.31g/day$) was significantly (p<0.05) higher as compared to that of normal diet (ND) group ($2.19{\pm}0.17g/day$). Weights of adipose tissues of HFD group were higher than those of ND group. Body weight gain of HFDSC group, however, was $2.36{\pm}0.24g/day$, which was significantly (p<0.05) lower by 21% than that of HFD group. In addition, weights of epididymal and perirenal adipose tissues were lower by 15% and 16%, respectively, as compared to those of HFD group. Biochemical analyses showed that concentration of triglyceride, total cholesterol, and fatty acids were significantly (p<0.05) lower in HFDSC group. These results suggest that Sargassum confusum extract has a high potential as an anti-obesity material by reducing weight gain and obesity-related factors in serum.

Anti-Obesity Effect of Fermented Detoxified Rhus verniciflua Vinegar Supplementation in Diet-Induced Obese Rats (무독화한 옻발효초가 고지방식이를 급여한 흰쥐에 미치는 항비만 효과)

  • The study aimed to investigate the anti-obesity effects of 1% Rhus verniciflua vinegar (RV) supplementation in high-fat-diet (60% fat)-induced obese rats. A total of 50 4-wk-old male Sprague-Dawley rats were fed normal chow diet or maintained on high-fat diet (HFD) for 12 weeks to induce obesity and were then randomized into five groups as follows: normal diet+ultra-pure water (CON), HFD+ultra-pure water (OB-DW), HFD+1% acetic acid (OBAA), HFD+1% RV (OB-RV), and HFD+0.1% caffeine (OB-CF). AA was used as a control for RV, and caffeine was used as a positive control due to its weight reducing effect. After 2 months, body weight, organ and adipose tissue weights, serum lipids, hepatic lipids, adipocyte size, and cell number per spot level were analyzed. As a result, food efficiency ratio, abdominal adipose tissue weight, serum levels of total cholesterol, triacylglycerol, free fatty acids, coronary artery index, and fecal lipid were significantly reduced in the RV treatment group. In this study, we found that dietary RV improved obesity by increasing lipid excretion and reducing lipogenesis. These results suggest that RV has potential as a functional anti-obesity food.

Anti-obesity effects of Glycyrrhiza uralensis ethanol extract on the inhibition of 3T3-L1 adipocyte differentiation in high-fat diet-induced C57BL/6J mice (감초 주정추출물의 3T3-L1 지방세포 분화 억제 및 고지방 식이로 유도된 C57BL/6J 마우스에 대한 항비만 효과)

  • The anti-adipogenic activity of Glycyrrhiza uralensis was investigated by examining the effects of its ethanol extract (GUE) on a mouse model with a high-fat diet (HFD) and 3T3-L1 preadipocytes during adipocyte differentiation. GUE administration for eight weeks significantly reduced weight gain in mice fed an HFD. GUE effectively inhibited 3T3-L1 preadipocyte differentiation and lipid droplet accumulation. This inhibitory effect is associated with the downregulation of key adipogenic regulators, including PPARγ and C/EBPα, and the modulation of adipose metabolism regulators, such as Fasn and Fabp4. LC-Q-TOF-MS analysis identified twelve phenolic and flavonoid compounds, including liquiritigenin and licorice saponin, in the GUE. These findings demonstrate that the anti-obesity effect of the GUE is attributed to the biological activity of its phenolic and flavonoid compounds. Therefore, the GUE has potential anti-obesity activity. Moreover, further studies on the isolation of bioactive components from the GUE and the investigation of the underlying molecular mechanisms of the GUE are required to establish its efficacy in metabolic disorders, including obesity.

In Vitro and In Vivo Physiological Characteristics of Dietary Fiber from By-product of Aloe vera Gel Processing (알로에 베라 유래 식이섬유의 In Vitro 및 In Vivo 생리기능 특성)

  • A fiber fraction (Aloe cellulose), the by-product obtained from Aloe vera gel processing was freeze dried and investigated for in vitro glucose/ bile acid retarding effects of powdered sample (100 mesh) comparing with commercial $\alpha$-cellulose as a reference sample. We also examined the effectiveness of physiological functionality such as the antiobesity and anti-constipation on Sprague-Dawley (SD) rat. The Aloe cellulose powders during in vitro dialysis experiment for 2 hours exhibited the glucose and bile acid retarding index of 20.32-35.2% and 53.13-28.30%, respectively. Especially, freeze dried aloe cellulose showed the 2.5 and 1.2-6 times higher effect on in vitro glucose and bile acid retardation than those of $\alpha$-cellulose. These relatively good retarding effects on glucose and bile acid diffusion suggest a potential of preventing from diabetes and arteriosclerosis of some extent. Also, the results from animal experiments on SD rats fed a high-fat diet for 4 weeks suggested that Aloe cellulose might be used as a novel dietary fiber showing an effective anti-obesity and anti-constipation effect.

The Anti-obesity Effect of 5 Herbal Formulas in High Fat Diet Induced Obese Mice (5종 방제의 항비만 효과 비교)

  • Objectives : Samhwangsasim-tang (SST), Hwangryeonhaedok-tang (HHT), Ukgan-san (UGS), Onjunghwadam-hwan (OHH) and Samul-tang (SMT) have been used for the treatment of various diseases. This study was performed to compare the anti-obesity effects of 5 herbal formulas in high fat diet-(HFD) induced obese mice. Methods : The mice were randomly divided into seven groups that were fed a normal diet (ND), a HFD, a HFD plus SST (HFD + SST), a HFD plus HHT (HFD + HHT), a HFD plus UGS (HFD + UGS), a HFD plus OHH (HFD + OHH), or HFD plus SMT (HFD + SMT) at 300 mg/kg/day for 7 weeks. All groups were assayed for body weights, food efficiency ratio (FER), final liver and fat weight and blood biochemical parameters. Results : The increased body weights, food efficiency ratio (FER), and serum total triglyceride were decreased in HFD + OHH group relative to the same measurements in HFD group. Furthermore, the HFD + SST group significantly reduced FER, liver and abdominal subcutaneous fat weight gains, and serum total triglyceride, whereas HDL-cholesterol level was increased compared to HFD group. Conclusions : These results suggested that HFD + OHH and HFD + SST exert anti-obesity effects in HFD-induced obese mice.

The Study on Anti-obesity Activity of the Wild Plants of Jeju Island (제주 자생 식물들에 대한 항비만 효능에 대한 연구)

  • In this study, we investigated the anti-obesity activity of extracts collected from wild plants in Jeju island. The inhibitory effect of plant extracts on the differentiation of preadipocyte 3T3-L1 was examined by oil red-O staining. We found that extracts collected from 6 plants among 31 plants, namely, Aralia elata(Miq.) Seem, Polygonum multiflorum Thunberg, Artemisia asiatica, platycodon grandiflorum(Jacq.) A. Dc., Polygonum cuspidatum S. et Z., Magnolia obovata Thunb, significantly inhibited preadipocyte differentiation. Additionally, 4 plant extracts were also found to have antioxidant activities in DPPH radical scavenging assay. Taken together, these results show that 6 plant extracts suppress the differentiation of preadipocytes, suggesting the potential use of 6 plant extracts as anti-obesity agents.

Inhibition of Lipase Activity and Preadipocyte Differentiation in 3T3-L1 Cells Treated with Sargassum horneri Extract (괭생이모자반 추출물의 리파아제 저해 활성 및 3T3-L1 지방전구세포 분화억제 효과)

  • In this study, in order to evaluate the anti-obesity effect of sargassum horneri extract, the effects of the extract on lipase activity and preadipocyte differentiation in 3T3-L1 cells were investigated. S. horneri extract between 0.0 and 1.0 mg/mL showed no cytotoxicity and inhibited lipase activity by 68.1%. When S. horneri extract was utilized at levels of 0.25, 0.5, and 1.0 mg/mL in 3T3-L1 cells, preadipocytes differentiation decreased by 11.4, 19.7, and 25.6%, respectively, showing anti-obesity effects. In addition, after treatment with 1.0 mg/mL S. horneri extract, the mRNA expression levels of sterol regulatory element binding proteins-1c (SREBP-1c), peroxisome proliferator activated receptor-γ (PPAR-γ), CCAAT enhancer binding protein-α (CEBP-α), fatty acid synthase (FAS), and stearoyl-CoA desaturase1 (SCD1) in 3T3-L1 cells were significantly decreased (p < 0.05) by 65.2, 54.9, 50.0, 33.8, and 33.8% respectively. These results showed that S. horneri extract suppresses lipase activity and prophylactic preadipocyte differentiation in 3T3-L1, and thus can be used as an anti-obesity agent in functional foods and medicines.

Anti-Obesity Effect of Pinus densiflora Leaf Extracts (솔잎(Pinus densiflora leaf) 추출물의 항비만효과)

  • In this study, we measured the inhibitory activity of Pinus densiflora leaf (PDL) against excessive lipid accumulation in mouse preadipocyte, 3T3-L1 cells to investigate whether PDL exerts anti-obesity activity. Lipid accumulation and the protein level were measured using Oil red O staining assay and Western blot analysis, respectively. We observed that PDL inhibited excessive lipid accumulation and decreased the expression of CEBPα, PPARγ and perilipin-1 related to lipid accumulation in 3T3-L1 cells. Therefore, considering these results, PDL can be used as a potential agent for anti-obesity.

Anti-obesity effects of an enzymatic extract of mandarin (Citrus unshiu) peel in 3T3-L1 adipocytes (감귤피 효소적 추출물의 지방세포에서의 항비만 효과)

  • Mandarin peel (MP) is a by-product of the processing of citrus juice or other products. This study aimed to investigate the potential anti-obesity effect of an enzymatic extract of MP on the inhibition of adipogenesis in 3T3-L1 adipocytes. The enzymatic extract (MPCE) was prepared using the commercial food-grade carbohydrase Celluclast. Lipid accumulation and triglyceride levels were significantly lower in MPCE-treated cells than in untreated cells. In addition, MPCE treatment reduced the protein expression levels of peroxisome proliferator-activated receptor-γ, CCAAT/enhancer-binding protein α, sterol regulatory element-binding protein 1, and fatty acid-binding protein 4. These results suggest that MPCE inhibits adipogenesis by downregulating the expression levels of adipogenesis-related proteins. Therefore, the current findings demonstrate that MPCE possesses potent anti-obesity properties and could be a potential ingredient in functional food industries.
