Developmental toxicity and anti-inflammatory effect of the soft coral Dendronephthya gigantea collected from Jeju Island in zebrafish model

  • Recent in vitro studies have demonstrated that extract of soft coral Dendronephthya gigantea (SCDE) had strong anti-inflammatory activities. However, the direct effects of SCDE on anti-inflammatory activities in vivo model remained to be determined. Therefore, the present study was designed to assess in vivo anti-inflammatory effect of SCDE using lipopolysaccharide (LPS)-stimulated zebrafish model. We also investigated whether SCDE has toxic effects in zebrafish model. The survival, heart beat rate, and developmental abnormalities were no significant change in the zebrafish embryos exposed to at a concentration below $100{\mu}g/ml$ of SCDE. However, lethal toxicity was caused after exposure to 200 and $400{\mu}g/ml$ of SCDE. Treating zebrafish model with LPS treatment significantly increased the reactive oxygen species (ROS) and nitric oxide (NO) generation. However, SCDE inhibited this LPS-stimulated ROS and NO generation in a dose-dependent manner. These results show that SCDE alleviated inflammation by inhibiting the ROS and NO generation induced by LPS treatment. In addition, SCDE has a protective effect against the cell damage induced by LPS exposure in zebrafish embryos. This outcome could explain the profound anti-inflammatory effect of SCDE both in vitro as well as in vivo, suggesting that the SCDE might be a strong anti-inflammatory agent.

UHPLC/TOFHRMS analysis and anti-inflammatory effect of leaf extracts from Zizyphus jujuba in LPS-stimulated RAW264.7 cells

  • Zizyphus jujube is a plant in the buckthorn family (Rhamnaceae) that has been the subject of research into antibacterial, antifungal and anti-inflammatory properties of its fruit and seed. However, few studies have investigated its leaves. In this study, the anti-inflammatory activity of ZJL (an extract of Z. jujube leaf) was evaluated to verify its potential as an anti-inflammatory agent and SARS-CoV-2 medicine, using nitric oxide (NO) assay, RT-PCR, SDSPAGE, Western blotting, and UHPLC/TOFHRMS analysis. We found that ZJL suppresed pro-inflammatory mediators such as NO, inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), and tumor necrosis factor α (TNF-α) in lipopolysaccharide (LPS)-induced RAW264.7 cells. ZJL acted by inhibiting NF-KB and MAPK signaling pathway activity. We also confirmed that ZJL contains a phenol compound and flavonoids with anti-inflammatory activity such as trehalose, maleate, epigallocatechin, hyperoside, catechin, 3-O-coumaroylquinic acid, rhoifolin, gossypin, kaempferol 3-neohesperidoside, rutin, myricitrin, guaiaverin, quercitrin, quercetin, ursolic acid, and pheophorbide a. These findings suggest that ZJL may have great potential for the development of anti-inflammatory drugs and vaccines via inhibition of NF-κB and MAPK signaling in LPS-induced RAW264.7 cells.

A novel mechanism of Korean Red Ginseng-mediated anti-inflammatory action via targeting caspase-11 non-canonical inflammasome in macrophages

  • Background: Korean Red Ginseng (KRG) was reported to play an anti-inflammatory role, however, previous studies largely focused on the effects of KRG on priming step, the inflammation-preparing step, and the anti-inflammatory effect of KRG on triggering, the inflammation-activating step has been poorly understood. This study demonstrated anti-inflammatory role of KRG in caspase-11 non-canonical inflammasome activation in macrophages during triggering of inflammatory responses. Methods: Caspase-11 non-canonical inflammasome-activated J774A.1 macrophages were established by priming with Pam3CSK4 and triggering with lipopolysaccharide (LPS). Cell viability and pyroptosis were examined by MTT and lactate dehydrogenase (LDH) assays. Nitric oxide (NO)-inhibitory effect of KRG was assessed using a NO production assay. Expression and proteolytic cleavage of proteins were examined by Western blotting analysis. In vivo anti-inflammatory action of KRG was evaluated with the LPS-injected sepsis model in mice. Results: KRG reduced LPS-stimulated NO production in J774A.1 cells and suppressed pyroptosis and IL-1β secretion in caspase-11 non-canonical inflammasome-activated J774A.1 cells. Mechanistic studies demonstrated that KRG suppressed the direct interaction between LPS and caspase-11 and inhibited proteolytic processing of both caspase-11 and gasdermin D in caspase-11 non-canonical inflammasome-activated J774A.1 cells. Furthermore, KRG significantly ameliorated LPS-mediated lethal septic shock in mice. Conclusion: The results demonstrate a novel mechanism of KRG-mediated anti-inflammatory action that operates through targeting the caspase-11 non-canonical inflammasome at triggering step of macrophage-mediated inflammatory response.

오약순기산(烏藥順氣散)의 소염 진통작용에 관한 연구 (Studies on the Anti-inflammatory and Analgesic Activities of Ohyaksungisan)

  • Ohyaksungisan, combined preparation of crude drugs, has been used for hemiplegia, arthralgia and paralysis in traditional Korean medicine. The anti-inflammatory activity of the aqueous extract from Ohyaksungisan(OSSE) was investigated on acetic acid-induced edema and adjuvant arthritis in rats. Acute toxicity and analgesic action in mice were also examined. Its anti-inflammatory activity on 5% acetic acid-induced edema and adjuvant arthritis was observed with oral administration. The acute toxicity showed 10% mortality at 2400 mg/kg (p.o), but was not showed at 1200 mg/kg (i.p). OSSE was showed to have significant analgesic action (P<0.05) at 150 mg/kg and this action was strengthened at 300, 600 mg/kg. The anti-inflammatory effect was showed significant preventive effect on the hind paw edema from 90 min. and the adjuvant arthritis, when orally administered for 19 days. showed significant inhibitory effect on the hind paw edema from the 5th day.

봉독요법(蜂毒療法)의 근골격계질환(筋骨格界疾患) 치료기전(治療機轉)에 대한 문헌적(文獻的) 고찰(考察) (The Study of Literature Review on Mechanism of Bee Venom Therapy for Musculo-skeletal Disorder)

  • Objectives : There have been many studies of the effect of Bee Venom therapy about arthritis, but no one study was reported about its whole functional mechanism to musculo-skeletal system. This study was designed to investigate the effect, Indication, and side effect of Bee Venom therapy on musculo-skeletal disease by literature review of articles. Results : The effects of Bee Venom therapy to musculo-skeletal system are divided to Anti_inflammatory effect and Anti-nociceptive effect. Anti_inflammatory effect is achieved through competitive chemotaxis, immuno-regulation, increasing of cortisol secretion by stimulating hypothalamus-pituitary gland-adrenal cortex axis. Anti-nociceptive effect is achieved by Anti-inflammatory mechanism and it works similar to anti-nociceptive effect of the acupuncture acting on central and peripheral nociceptive transduction system. The Bee Venom therapy could cause severe side effect, for example, hypersensitivity and anaphylaxis, injury to central nerve system and cardiovascular system, peripheral blood system, and renal dysfunction. Conclusions : With its Anti-inflammatory and Anti-nociceptive mechanism, Bee Venom therapy is considered that has good effects to autoimmune disease, chronic inflammation of various musculo-skeletal disease and various pain syndrome. But the clinician must be careful for its side effects.

  • Experimental studies were conducted to investigate for the effect of Tongkyu-tang on analgesic, antipyretic, sedative, anti-inflammatory, isolated ileum and anti-histamine actions. Tongkyu-Tang was composed of sixteen crude drugs including Astragali Radix, which has been used for the treatment of common cold, headache, rhinitis, nasal obstruction, rhincospasm, etc. The following results have been obtained; analgesic and antipyretic actions were obtained. Prolonged action against the hypnotic duration of pentobarbital were significantly noted in mice. Anti-inflammatory effects in the paw edema induced by 1% carrageenin and 1% dextran were significantly shown in rats. Spontaneous motilities of isolated ileum of mice were strongly suppressed, and contractions of isolated ileum of mice and guinea-pigs induced by acetylcholine chloride, barium chloride and histamine were remarkably inhibited. Antihistamine actions were noted. Considering the above experimental results, it is suggested that effects of Tongkyu-tang based on the Oriental Medical References were similarly consistent with the actual experimental results.

  • Most inflammatory disorders are usually treated using anti-inflammatory drugs including non-steroidal anti-inflammatory drugs (NSAID) and steroidal anti-inflammatory drugs (SAID). In a prolonged use, however, they may frequently produce adverse side-effects. Thus, it is necessarily required to develop a new anti-inflammatory drug with little side-effects. Nephrite has been widely used by traditional oriental medicine to cure the various chronic diseases. In order to verify the anti-inflammatory activity of nephrite, the TPA (12-O-tetradecanoylphorbol-acetate) or the croton oil-induced edema was developed in the mouse ears and the nephrite powder suspension or the nephrite water was directly applied to the ear edema. It was found that nephrite could significantly reduce the ear swelling implying its strong potential as an active anti-inflammatory agent when comparing to indomethacin, a non-steroidal anti-inflammatory drug.

Binding Model of Fisetin and Human c-Jun NH2-Terminal Kinase 1 and Its Anti-inflammatory Activity

  • Fisetin is a naturally occurring flavonoid with some anti-cancer and anti-inflammation capabilities. In this study, we perform docking studies between human c-Jun N-terminal kinase 1 (JNK 1) and fisetin and proposed a binding model of fisetin and JNK 1, in which the hydroxyl groups of the B ring and oxygen at the 4-position of the C ring play key roles in binding interactions with JNK. Fluorescence quenching and saturation-transfer difference (STD) NMR experiments showed that fisetin exhibits good binding affinity to JNK, $1.32{\times}10^8M^{-1}$. The anti-inflammatory activity of fisetin was also investigated. Fisetin significantly suppressed tumor necrosis factor, the NO production, and macrophage inflammatory cytokine release in LPS-stimulated RAW264.7 mouse macrophages. We found that the anti-inflammatory cascade of fisetin was mediated through the JNK, and cyclooxygenase (COX)-2 pathways. Our findings suggest the potential of fisetin as an anti-inflammatory agent.

Recent Prospect of Compounds Derived from Marine Macroalgae for Medicinal Application of Anti-Inflammation for Chemoprevention of Cancer

  • Although marine living organism contains a numerious number of compounds, it is difficult to collect these compounds in a large scale for medicinal application. However, in recent years, several bioactive compounds isolated from marine macroalgae have been proved to be able to provide potential sources for development of medicinal products because they can be obtained in large amount from marine. A number of studies have reported a variety of effects of marine macroalgae but a few anti-inflammatory activity of marine macroalgae have recently been published. Herein, we reviewed novel anti-inflammatory compounds recently isolated from marine brown algae, green algae and red algae. From this survey, in particular, some compounds contained in edible macroalgae exert anti-inflammatory effects with inhibition on cyclooxygenase-2 (COX-2), inducible nitric oxide synthase(iNOS) and matrix metalloproteinases (MMPs) activity regulated by nuclear factor-kappa B transcription factor that play a key role in cancer as well as inflammation, demonstrating to be able to potentially apply to development of anti-inflammatory agent for chemoprevention of cancer. Furthermore, some macroalgae and their compounds with both excellent anti-inflammatory activity and very low toxicity can select a potential candidates capable of preventing or treating several chronic inflammation such as colitis, hepatitis and gastritis, leading to cancer.

  • Experimental studies were done to research the clinical effect of Bojeosodokeum on the Analgesic, Antipyretic, Anti-inflammatory, Antimicrobial and Anti-convulsive effects in mice and rats. The results obtained as follows : 1. The solid extract powders of Bojeosodokeum was revealed significant action on analgesic effect. 2. the solid extract powders of Bojeosodokeum was revealed significant action on Anti-inflammatory effect. 3. The solid extract powders of Bojeosodokeum was revealed significant action on antipyretic effect. 4. Antimicrobial effects of solid extract powders of Bojeosodokeum against Escherichia coli and staphylococcus aureus was observed. 5. The solid extract powders of Bojeosodokeum was revealed significant action on anticonvulsive effect. According to the above results, it is confirmed that analgestic, anti-inflammatory, antipyretic, antimicrobial and anticonvulsive effect were recognized in solid extract powders of Bojeosodokeum.
