Molecular Events on Experimental Skin Inflammation and Modulation by Topical Anti-inflammatory Flavonoids

  • There have been various animal models of skin inflammation. These models have been used for establishing anti-inflammatory activity of the topical agents including cosmetics. Here, the molecular mechanisms of most widely-used animal models of skin inflammation including contact irritation, acute and chronic inflammation, and delayed-type hypersensitivity are summarized. Against these animal models, varieties of plant flavonoids showed anti-inflammatory activity. The action mechanisms of anti-inflammation by topical flavonoids are presented. A therapeutic potential of flavonoids is discussed.

Reduced Anti-inflammatory Activity of Acetylsalicylic Acid Maltol Ester, Aspalatone

  • The anti-inflammatory activity of acetylsalicylic acid maltol ester (aspalatone), a potential anti-thombotic agent, was studied using the several experimental animal models of inflammation. By oral administration, aspalatone was found to possess the weak anti-inflammatory activity in models of an acute inflammation, in which aspalatone showed approximately one-third to one-fourth of the anti-inflammatory activity of aspirin. Aspalatone (200 mg/kg/day) and aspirin (50 mg/kg/day), however, did not show the inhibitory activity against granuloma fomation and adjuvant-induced arthritis.

추출용매에 따른 산초 (Zanthoxylum schnifolium)열매 추출물의 라디칼소거능과 항염증 효과 (Radical Scavenging and Anti-inflammation Activities from Different Extracts of Zanthoxylum schnifolium Fruits)

  • This study was presented to the physiological activities such as metal chelating capacity, superoxide dismutase-like activity, anti-inflammation, cytotoxicity from Zanthoxylum schnifolium fruit. The values for metal chelating capacity of 1 mg/mL of 70% EtOH and 70% MeOH extracts were 68% and 67%, respectively, it was shown that metal chelating capacity in a dose-dependent manner. The 100% EtOH and 100% MeOH extract exhibited strong superoxide dismutase-like activity. The highest SOD-like activity obtained from 100% EtOH was 51% at a concentration of 4 mg/mL. The all extracts of Zanthoxylum schnifolium fruit except $H_2O$ extract significantly decreased the concentration of LPS-induced NO in Raw 264.7 cells. The 100% EtOH, 100% MeOH and 70% EtOH extract represented the highest activity in the anti-inflammation properties in vitro. However, the 100% EtOH and 100% MeOH extracts has cytotoxicity in Raw 264.7 cells and it may affect cell viability. Conclude that 70% EtOH extract was most suitable for anti-inflammation. Our results revealed that the Zanthoxylum schnifolium fruit expected to physiological activities.

Rhamnazin inhibits LPS-induced inflammation and ROS/RNS in raw macrophages

  • Purpose: The aim of this work was to investigate the beneficial effects of rhamnazin against inflammation, reactive oxygen species (ROS)/reactive nitrogen species (RNS), and anti-oxidative activity in murine macrophage RAW264.7 cells. Methods: To examine the beneficial properties of rhamnazin on inflammation, ROS/ RNS, and anti-oxidative activity in the murine macrophage RAW264.7 cell model, several key markers, including COX and 5-LO activities, $NO^{\cdot}$, $ONOO^-$, total reactive species formation, lipid peroxidation, $^{\cdot}O_2$ levels, and catalase activity were estimated. Results: Results show that rhamnazin was protective against LPS-induced cytotoxicity in macrophage cells. The underlying action of rhamnazin might be through modulation of ROS/RNS and anti-oxidative activity through regulation of total reactive species production, lipid peroxidation, catalase activity, and $^{\cdot}O_2$, $NO^{\cdot}$, and $ONOO^{\cdot}$ levels. In addition, rhamnazin down-regulated the activities of pro-inflammatory COX and 5-LO. Conclusion: The plausible action by which rhamnazin renders its protective effects in macrophage cells is likely due to its capability to regulate LPS-induced inflammation, ROS/ RNS, and anti-oxidative activity.

Anti-Inflammatory Activity of Elsholtzia splendens

  • Elsholtzia splendens Nakai has been used in North-East Asia as an ingredient of folk medicines for treating cough, headache and inflammation. The present investigation was carried out to establish its in vivo anti-inflammatory activity using several animal models of inflammation and pain. The 75% ethanol extract of the aerial part of E. splendens significantly inhibited mouse croton oil-induced, as well as arachidonic acid-induced, ear edema by oral administration (44.6% inhibition of croton oil-induced edema at 400 mg/kg). This plant material also showed significant inhibitory activity against the mouse ear edema induced by multiple treatment of phorbol ester for 3 days, which is an animal model of subchronic inflammation. In addition, E. splendens exhibited significant analgesic activity against mouse acetic acid-induced writhing (50% inhibition at 400 mg/kg), while indomethacin (5 mg/kg) demonstrated 95% inhibition. E. splendens ($5-100{\;}{\mu}g/mL$) significantly inhibited $PGE_2$ production by pre-induced cyclooxygenase-2 of lipopolysaccharide-treated RAW 264.7 cells, suggesting that cyclooxygenase-2 inhibition might be one of the cellular mechanisms of anti-inflammation.

도화(桃花, Prunus persica Flos)의 항산화 및 항염증 활성 (Anti-oxidant and Anti-inflammation Activities of Prunus persica Flos)

  • 도화 추출물의 항산화능 및 항염증 효과를 검증하여 기능성 화장품 소재로서의 가능성을 검증하였다. 도화의 항산화능 측정으로 전자공여능을 측정한 결과 도화 에탄올 및 아세톤 추출물 500ppm에서 90.0% 이상의 효과를 나타내어, 합성 항산화제로 알려진 BHT와 유사한 효과를 나타내었으며, SOD 유사활성능 측정 결과 도화 에탄올 및 아세톤 추출물 1,000ppm에서 40.0% 이상의 활성을 나타내었다. Xanthine oxidase 저해활성능 측정 결과 도화 에탄올 및 아세톤 추출물 1,000 ppm에서 vitamin C와 유사한 30.0%의 효과를 나타내어, 도화 열수 추출물에 비해 도화 에탄올 및 아세톤 추출물의 효과가 높게 나타났다. 항염증 효과 측정으로 hyaluronidase 저해활성을 측정한 결과 도화 열수, 에탄올 및 아세톤 추출물에서 35.0%의 저해활성을 나타내었으며, macrophage 세포를 이용한 nitric oxide (NO) 저해활성을 측정한 결과 도화 추출물 모든 농도에서 12시간 이후부터 NO 생성량이 억제됨을 확인할 수 있었다. 또한 피부염증 유발균인 Staphylococcus epidermidis와 Propionibacterium acnes의 항균효과를 측정한 결과 도화 에탄올 추출물에서 항균 효과를 나타내었다.

Inhibitory effects of resveratrol analogs on lipopolysaccharide-induced cyclooxygenase-2 activity in RAW264.7 cells

  • It has been known that resveratrol, a phytoalexin present in grapes mainly, has antioxidant. anti-inflammatory, and cancer chemopreventive activity. One mechanism of its anti-inflammation and cancer prevention is considered to modulate cyclooxygense-2 (COX-2) activity. Since COX-2 plays an important role in inflammation and carcinogenesis, the potential COX-2 inhibitors have been considered as anti-inflammatory or cancer chemopreventive agents. (omitted)

성유탕가감방(聖愈湯加減方)의 항염증(抗炎症) 및 항산화(抗酸化) (The Experimental Study on Anti-oxidant and Anti-inflammatory Effect of Sungyoutanggagambang(SYTG))

  • Purpose: This study was performed to evaluate anti-oxidant activities and anti-inflammatory effects of Sungyoutanggagambang(SYTG). Methods: In the study of anti-oxidant activities. SYTG was investigated by DPPH radical scavenger activity. superoxide dismutase activity and superoxide anion radical scavenger activity. In the study of anti-inflammatory effects. SYTG was investigated using cultured cells and murine models. As for the parameters of inflammation. levels of several inflammatory cytokines and chemical mediators which are known to be related to inflammation were measured in mouse lung fibroblast cells(mLFCs) and RAW264.7 cells. Results: Prior to the experiment. we investigated the security of SYTG by measuring GOT and GPT in serum. 1. SYTG showed high antioxidant activity in a concentration-dependent degree by measured scavenging activity of DPPH free radical, superoxide dismutase and superoxide anion radical. 2. SYTG inhibited IL-1$\beta$, IL-6. TNF-$\alpha$, COX-2 and NOS-II mRNA expression as compared with the control group in a concentration-dependent degree in RAW264.7 cell line. 3. SYTG inhibited IL-1$\beta$, IL-6 production significantly at 100 ${\mu}g/ml$ and TNF-$\alpha$ production significantly at 50, 100 ${\mu}g/ml$ as compared with the control group in RA W264.7 cell line. 4. SYTG inhibited IL-1$\beta$, and IL-6 production significantly as compared with the control group in serum of acute inflammation-induced mice. and decreased IL-1$\beta$, IL-6 production in spleen tissue. and also decreased IL-1$\beta$, IL-6 production in liver tissue. Conclusion: These results suggest that SYTG can be useful in treating diverse female diseases caused by inflammation such as endometrosis, myoma, pelvic congestion. chronic cervicitis, chronic pelvic inflammatory disease and so on.

화피의 충치균과 치주질환균에 대한 항균활성 및 항염효과 (Antibacterial Activity against S. mutans or P. gingivalis and Anti-inflammatory Effect of Betulae Cortex)

  • Betulae Cortex of Betula platyphylla Suk. var. japonica Hara (Betulaceae) has long been used for treatment of various inflammation, fever and cough in Eastern Asia. In order to investigate antibacterial activity of the Betulae Cortex against Streptococcus mutans or Porphyromonas gingivalis, MIC (minimal inhibitory concentration) and pH were checked, and for anti-inflammation activity, the experiments about trypsin-induced paw edema, vascular permeability and myeloperoxidase activity in rat's hind-paw tissue, were carried out with various extracts of Betulae Cortex (BCXs) respectively. The BCXs showed significant antibacterial activity, and at the dose of over 50 mg/kg, BCX showed significant inhibition on the paw edema, vascular permeability and myeloperoxidase activity. These results indicate that BCXs have antibacterial activity against oral cariogenic bacteria and anti-inflammatory effect.

Recent Prospect of Compounds Derived from Marine Macroalgae for Medicinal Application of Anti-Inflammation for Chemoprevention of Cancer

  • Although marine living organism contains a numerious number of compounds, it is difficult to collect these compounds in a large scale for medicinal application. However, in recent years, several bioactive compounds isolated from marine macroalgae have been proved to be able to provide potential sources for development of medicinal products because they can be obtained in large amount from marine. A number of studies have reported a variety of effects of marine macroalgae but a few anti-inflammatory activity of marine macroalgae have recently been published. Herein, we reviewed novel anti-inflammatory compounds recently isolated from marine brown algae, green algae and red algae. From this survey, in particular, some compounds contained in edible macroalgae exert anti-inflammatory effects with inhibition on cyclooxygenase-2 (COX-2), inducible nitric oxide synthase(iNOS) and matrix metalloproteinases (MMPs) activity regulated by nuclear factor-kappa B transcription factor that play a key role in cancer as well as inflammation, demonstrating to be able to potentially apply to development of anti-inflammatory agent for chemoprevention of cancer. Furthermore, some macroalgae and their compounds with both excellent anti-inflammatory activity and very low toxicity can select a potential candidates capable of preventing or treating several chronic inflammation such as colitis, hepatitis and gastritis, leading to cancer.
