Isolation and Purification of Novel Anti-Fungal Peptides from Hemolymph of Immunized Larvae of Housefly, Musca domestica (집파리유충 hemolymph 중신형의 anti-fungal peptides의 분리정제)

  • To isolate and purify anti-fungal active substances from immunized housefly (Musca domestica), low dose of Candida albicans was injected into the larvae of the housefly to induce the appearance of potent anti-fungal active substances in the hemolymph. This purification work was performed by the routine isolation and purification processes of protein, namely, solid phase extraction (SPE), SDS-PACE electrophoresis, HPLC purification. Three 4-16 kDa peptides which exhibited antifungal activity against Candida albican and other fungi were isolated from induced hemolymph. Consequently, further anti-fungal activity study showed that these three peptides were different either in molecular weight or in anti-fungal activity. All isolated substances were proved to be active and resistant to high-temperature. It was deduced that these peptides isolated from induced housefly were novel members of the insect defensin family and they were inducible.

Isolation and Selection of Actinomycetes Producing Anti-fungal Materials (항진균성 활성물질을 생성하는 토양방선균의 분리)

  • Anti-fungal materials producing bacteria were isolated from soil by bennett's agar and actinomycete isola-tion agar medium. The bacterla were identified as synonym of Actinomycetes. Based on the data obtained from its morphological and colony characteristics. The medium for production of anti-fungal materials was YEME (yeast extract 4 g, malt extract l0g, glucose 4 g, D.W 1ι, pH 7.0${\pm}$0.2). The culture conditions were 30$^{\circ}C$, 7 days and 200 rpm in shaking incubator. No. 13, No. 15 and No.28 strains were produced anti-fungal materials against fungal plant pathogens. Specially, The No. 28 strain showed a powerful biopesticide activity and broad spectrum effects of anti -fungal materials on Collectrichum coccodes, Botrytis cinerea, Cladosporium cucumerinum, Didymella bryoniae.

In vitro Anti-fungal Activity of Various Hydroxylated Fatty Acids Bioconverted by Pseudomonas aeruginosa PR3

  • The in vitro anti-fungal activity of hydroxylated fatty acids obtained from microbial conversion by Psuedomonas aeruginosa PR3 using ricinoleic acid(RA), eicosadienoic acid(EDA) and conjugated linoleic acid(CLA) as substrates, was investigated. Bioconverted hydroxylated fatty acids showed different anti-fungal activities potentials against the range of phytopathogenic fungi such as Botrytis cinerea, Rhizoctonia solani, Fusarium oxysporum, Sclerotonia sclerotiorum, Colletotricum capsici, Fusarium solani and Phytophthora capsici. RA and EDA showed up to 50% fungal mycelial inhibition at the concentration of $5{\mu}l\;ml^{-1}$. RA, EDA and CLA also exhibited anti-fungal activities with minimum inhibitory concentration(MIC), ranging from 500 to $1000{\mu}g\;ml^{-1}$. Screening was also carried out using varied concentrations of bioconverted RA and EDA for determining the anti-fungal effect on the spore germination of different fungi. Bioconverted RA and EDA showed a considerable degree of spore germination inhibition.

Anti-fungal materials Produced by Streptomyces albogriseus Isolated in Korean soil

  • An anti-fungal material producing actinomycete was isolated from domestic soil. This strain was identified as Streptomyces albogriseus by 16S rDNA sequence. YEME (yeast extract 4g, malt extract 10g, glucose 4g, D.W 1l , pH $7.0{\pm}0.2$) medium was used for production of anti-fungal materials. S. albogriseus was cultured in a shaking incubator for 2 weeks at 150 rpm and $25{\pm}1^{\circ}C$. An anti-fungal material produced by S. albogriseus was identified at 340nm by uv/vis- spectrometer and it showed powerful anti-fungal activity. This is the first report that secondary metabolite produced by S. albogriseus showed an activity against phytopathogenic fungi such as Collectrichum coccodes, Botrytis cinerea, Cladosporium cucumerinum, Didymella bryoniae.

  • The anti-fungal activity of plant extractives was investigated and the lacquer extractives were selected. The lacquer extractives were coated on anti-fungal paper and confirmed its effect. Water repellency, tensile index and brightness of anti-fungal paper treated with natural extractives were similar to commercial pesticide-treated paper. The incidence of Fusicaladium leuieri MAGNUS of commercial pesticide-treated paper and anti-fungal paper treated with lacquer were similar but, the incidence of Botryosphaeria dothidea (Moug.) Ces & De Not and Gloeodes pomigena of those were increased 10-60 % than commercial pesticide-treated paper.

  • To define diversity of domestic radish, we analysis genetic relationship of anti-fungal protein genes from several domestic radish (Raphanus sativus L.) seeds. We have isolated from domestic radish (Baekwoon) anti-fungal protein named RAP[12]. In this report, we isolate RNAs and raw protein from radish seeds then, RT-PCR analysis was done with another known anti-fungal sequences of radish from Gene Bank/EMBL and anti-fun- gal, anti-yeast activity were done against Bot교tis cenerea, Saccharomyces cerevisiaeι Candida albicans with it's raw proteins. The anti-fungal activity was shown used all seeds but anti-yeast activity was shown only two seeds (Myungsan, Baekwoon). RT-PCR products (about 0.2 Kb) were not shown only two seeds. To identify the sequencing relationship of the domestic radish, we have cloned and sequenced RAP genes of the radish and analysis the sequence relationship with clustalw program. Thus we report the result that there are some different relationship between domestic radish and known other radish's anti- fungal protein[15].

  • Generally, polymer materials are not air-permeable and hydrophilic. In addition, they do not possess anti-fungal property. Hydrophilicity, air-permeability, and anti-fungal properties of new composites consisting of polymer, ceramic nanoparticles, and silver ion were investigated by contact angle measurements, air permeation time, and cell culture. The hydrophilic, air-permeable, and anti-fungal composites can be used in health care industry.

Antifungal Activity of Korean Radish (Raphanus sativaus L) Extracts Against Pathogenic Plant (한국산 무 추출물의 곰팡이 병균에 대한 항진균성)

  • A study of the anti-fungal properties in Korean radish was conducted using a variety of purification procedures such as Extrelut column, RP(Reverse Phase) Cl8 Column Chromatography, HPLC etc. to separate anti-fungal substances from Korean radish juices to test them against a common gray mold called Botrytis cenerea. Dialysis tube operation showed that these substances were presumably thermostable compounds with low molecular mass (less than 3.5 kDa). Differences of anti-fungal activities depending upon types of radishes used did not show any noticeable variation. The antifungals were presumably composed of more than 5 compounds. Among these, the most anti-fungal fraction was analyzed by HPLC in which one peak was obtained. Disease-affected plants were inoculated with 10mg of Extrelut fraction and results showed similar anti-fungal activity to pesticides suggesting possible usage of these substances as environmentally friendly antibiotics.

Anti-fungal, anti-oxidant, and anti-inflammatory effects of supercritical fluid extracts from Ulmus davidiana (느릅나무 초임계 추출물의 항균, 항산화 및 항염증 활성)

  • The Ulmus davidiana have been used in traditional oriental medicine as remedies for inflammation, ulcers, cancers, bacterial infections and scabies. In this study, the anti-fungal, anti-oxidant, and anti-inflammatory activitiesof a supercritical extract of U. davidiana were investigated in vitro. To explore the anti-oxidant and anti-inflammatory characteristics of the supercritical extract, ABTS radical scavenging activity, and the inhibition of nitric oxide production in LPS-stimulated RAW 264.7 cells were examined, respectively. In addition, the anti-fungal activities of the extract were assessed. The results showed a concentration-dependent increase in ABTS radical scavenging activity. Cells stimulated with LPS produced more nitric oxide than normal control cells; however, cells treated with the supercritical fluid extract decreased this production in a concentration-dependent manner. Finally, the supercritical fluid extracts showed significant anti-fungal activity. These results suggest that extracts of the U. davidiana might be used to develop potent anti-fungal, anti-oxidant, and anti-inflammatory agents, and may be useful as ingredients for related new functional cosmetic materials.

Anti-tumor and immuno-stimulating activity of fungal polysaccharides

  • Low molecular weight molecules and high molecular weight substances were found to have anti-tumor and immuno-modulating activity. Previously polysaccharides have been received much attention because of adhesives, food additives or animal foods (Whistler et al., 1976). In effort of developing new anti-tumor substances with low toxicity, numerous polysaccharides from yeast, algae, bacteria, higher plants and especially fungi have been investigated for anti-tumor and immuno-modulating activities. Thus the high molecular weight molecule was reported to have anti-tumor activity through host mediated immunity. In this brief article, attention will be paid to polysaccharides which is especially fungal origin.
