Experimental Effects of Taklihwangki-Tang on the Anti-Cancer And Immuno-Action (托裏黃기湯이 抗癌 및 免疫作用에 미치는 實驗的 效果)

  • Taklihwangki-Tang was a drug that treated carbuncle and cellulitis. So, the purpose of this Study was to investigate effect of Taklihwangki-Tang on the anti-cancer and proliferation of immunocytes, nitric oxide(NO) production of peritoneal macrophages. We used Taklihwangki-Tang extract(THT) with freeze-dried, 8wks-old male mice and cancer cell lines(L1210, S-180) for this Study. The proliferation of cells was tested using a colorimetric tetrazoliun assay(MTT assay). The results of this Study were obtained as follow ; THT was showed cytotoxicity on the L1210 and S-180 cell lines, increased proliferation of thymocytes. And the combined effects of THT and vincristine were became cytotoxicity of cancer cell lines and increased significantly proliferation of thymocytes. THT accelerated proliferation of thymocytes in normal mice, and decreased significantly proliferation of L1210 cells and accelerated significantly NO production of peritoneal macrophages in L1210 cells transplanted mice. This results suggest that THT inhibit proliferation of cancer cells by becoming immunocytes activity(NO production, proliferation of T-cell).

Anti-cancer and Anti-microbial Effect of the Fraction Isolated from Pyrus ussuriensis Leaves (산돌배나무(Pyrus ussuriensis) 잎 분획물의 항암 및 항균활성에 관한 연구)

  • This study was conducted to confirm the application as ingredients of cosmetics through an examination of the function for anti-cancer and anti-microbial of the fraction isolated from Pyrus ussuriensis leaves. The dried leaf of P. ussuriensis were extracted with acetone-$H_{2}O$ (6:4, v/v), concentrated and fractionated with the upper layer of acetone on a separatory funnel. Each fraction was freeze dried, then a portion of acetone soluble powder was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol as eluents and also used the MIC-gel using a series of aqueous methanol as developing solvent. The isolated compounds were identified by silica-gel TLC. The growth inhibition activity was measured using the MTT assay by the mouse meltioma (B16F10) cell. The cancer cell growth inhibition rate of fractions isolated from P. ussuriensis leaf was 80%. In anti-microbial activity test, the fraction of P. ussuriensis with 0.25 mg/disc resulted in the clear zone of 1.3 cm and 2 cm for Staphylococcus aureus and S. epidermidis of gram positive bacillus, respectively. In Escherichia coli of gram negative bacillus, the fraction with 0.5 mg/disc resulted in the clear zone of 1.1 cm~1.5 cm each fraction. From these results, we confirmed that acetate fraction of P. ussuriensis has a great potential as a natural ingredients with a anti-cancer and anti-microbial source.

  • This study was designed to compare the quality characteristics of freeze and spray dried red ginseng extract powders(RGEPs) by drying methods, which have been required to maintain the stability for the quality. Chemical compositions, major ginsenoside contents and color intensities of these Products were compared by drying conditions. The moisture absorption rates and optical densities also were compared during storage under maltreatment conditions of a various relative humidities (75, 86and 92 RH) and two different temperatures (37 and $50^{\circ}C$). It was found that decreases of total major ginsenosides contents in freeze and spray dried RGEPs were 5.4 % to 6.7 % during storage for 6 months at $37^{\circ}C$, 75 % RH. When these products packaged with inner seal of Al-foil laminate paper (Al-foil; 9 ${\mu}m$) were stored for 6 months at $37^{\circ}C$, 75 % RH. the moisture absorption rates of freeze and spray dried RGEPs were ranged 42 % to 82 %, 8 % to 16 %, respectively. In storage for 6 months at $37^{\circ}C$, 86 % RH, spray dried RGEP was higher in brown pigment($400{\sim}490nm$) than freeze dried RGEP while freeze dried was higher in pyrazine (278 nm), HMF and furfural (285 nm) than spray dried RGEP. It was found that RGEPs showed a strong anti-oxidative activity by electron donating ability to DPPPH, but there was no significant difference between freeze and spray dried RGEPs.

  • In this study, effect of freezing and cryogenic crushing on physico-chemical characteristics of sardine, pollack and sqiud representative for domestic frozen fishery products was investigated and some product using them was tried to be prepared. Dehead and viscerated, washed fishes were subjection to freezing without air circulation and liquid N2 gas at -20$\^{C}$,-40$\^{C}$ and -80$\^{C}$, and then frozen fishes were crushed by hammermill, masscolloider and the product was stored added with anti-freeze such as sorbitol, phosphates, starch and egg Powder, qualify of frozen squid surimi was not changes during 70 days at below -20$\^{C}$ . The results of quality characteristics and sensory evaluation of patties and nugget which made from shattered squid and pollack were similar to commercial products in flavor, color and texture, but sardine meat was inferior to commercial products in flavor and color.

  • This paper investigates insulating effects and strength properties of concrete under cold climate carrying out the experiment with insulating forms on the slab member. According to test results, under $-10^{\circ}C$ of outer temperature, the surface of concrete exposed to outer temperature goes below zero after 10hours since exposed. And, the surface of concrete covered with vinyl and blanket goes below zero after 42hours. However, the surface of concrete covered with insulating materials such as polystyrene foam does not drop below zero until 55hours. And, according to coring sample strength test, compressive strength cured with insulation shows much higher strength than no insulation curing condition and vinyl and blanket curing condition.

Anti-inflammation and Anti-aging Effect of Lactic Acid Bacteria Fermented Lycium chinense Extracts (젖산균 발효 구기자(Lycium chinense ) 추출물의 항염증 및 항노화 효능 연구)

  • In this study, fermented Lycium chinense (L. chinense) extract was used to analyze the efficacy of nitric oxide (NO) and anti-aging inhibition to verify its usefulness as a functional cosmetic material. L. chinense was extracted with hydrothermal and 70% ethanol as a solvent, fermented, concentrated, and freeze-dried to prepare a sample, followed by MTT assay, NO inhibition assay, western blot assay, and UPLC analysis.As a result of the analysis of NO inhibitory efficacy, fermented L. chinense water extract (FLW) and fermented L. chinense 70% ethanol extract (FLE) showed excellent NO inhibitory efficacy of 47.96% and 56.71%, respectively. As a result of measuring the expression patterns of the wrinkle-related proteins MMP-1, and TRPV-1 through Western blot, both FLW and FLE confirmed the inhibitory efficacy of MMP-1 and TRPV-1. Based on the results of the experiment, the fermented L. chinense extract is expected to have a high application value as a functional cosmetic material related anti-inflammatory, and anti-aging.

  • Objectives : Sam-Myo-Whan(SMW) has been known traditional prescription with anti- anthritis activities. We investigated inhibitory effects of SMW on lipopolysaccharide (LPS)-induced nitric oxide(NO), $TNF-{\alpha}$ and inducible nitric oxide synthase(iNOS) production from RAW264.7 cells and BV-2 Microglia cells. Methods : SMW, which had been extracted with 70% MeOH, concentrated and freeze-dried was used for this experiment. After BV2 mouse brain macrophages and RAW264.7 mouse peritoneal macrophages were pretreated with increasing concentrations of SMW extract for 30min, and then activated with LPS. To investigate cytotoxicity of SMW extract, cell viability was measured by MTT assay. NO production was measured in each culture supernatant by Griess reaction. mRNA expression of iNOS in two type cells was investigated by RT-PCR. $TNF-{\alpha}$ production was measured in each culture supernatant by ELISA. Results : SMW extract significantly inhibited LPS-induced NO and $TNF-{\alpha}$ production in BV2 cells and RAW264.7 cells dose-dependently. SMW extract also greatly suppressed mRNA expression of iNOS in both type cells activated with LPS. Conclusion : These data suggests that SMW extract may have an anti-inflammatory effect through the inhibition of iNOS expression.

Bioactivities of the Herb Extracts Used for Gamhongroju, a Korean Liqueur

  • In this study, antioxidative and anti-inflammatory activities of the herb (cinnamon, clove, glehnia root, ginger, violet-root cromwell, licorice, citrus peel and longan) extracts used for gamhongroju, one of the popular liqueurs in Korea, were investigated. Twenty grams of individual herbs were extracted in 60% purified ethanol and freeze-dried. A mixture of the individual herb extracts (HEM) was separately prepared. Cytotoxicity of the individual extracts and HEM on murine RAW264.7 macrophage cells were examined along with their recovering activity on $H_2O_2$-treated RAW264.7 cells. Antioxidant and anti-inflammatory activities of the extract-treated cells were determined by measuring superoxide dismutase (SOD) and glutathione peroxidase (GPx) activities, and Trolox equivalent antioxidant capacity (TEAC), nitric oxide (NO) and prostaglandin E2 (PGE2) levels. Violet-root cromwell extract showed the least cytotoxicity in terms of treated concentration. Most of the extracts, below levels of cytotoxicity, recovered the $H_2O_2$-treated cells. Treatment with some of the extracts increased SOD and GPx activities and TEAC levels while a majority inhibited the production of NO and PGE2 in lipopolysaccharide (LPS)-treated cells.

Comparison of the Effects of Straight and Twisted Heat Trace Installations Based on Three-dimensional Unsteady Heat Transfer (열선의 직선시공과 감기시공의 동파방지 효과 비교를 위한 3차원 비정상 수치해석)

  • This paper numerically examines, straight and twisted electrical heat trace installations for their anti-freezing effects on water inside a pipe. The unsteady incompressible Navier-Stokes equations coupled with an energy equation were solved to compare the two installation methods. The heat conduction of the pipe with a heat source interacts with the natural convection of the water, and the conjugate heat transfer was considered using a commercial code (ANSYS-FLUENT) based on a SIMPLE-type algorithm. Numerical experiments, were done to investigate the isotherms and the vector fields in the water region to extract the evolutions of the minimum and maximum temperatures of the water inside the pipe. There was no substantial difference in the anti-freezing effects between the straight and twisted. Therefore, the straight installation is recommended after considering the damage and short circuit behavior of the electrical heat trace.

Screening of a Natural Feed Additive Having Anti-viral Activity against Influenza A/H5N1 (안전한 닭고기 생산을 위한 고병원성 조류인플루엔자 A/H5N1에 항바이러스 효과를 가진 천연 사료첨가제의 탐색)

  • To search for anit-H5N1 influenza virus agent, the anti-viral activity of methanol and aqueous extracts from thirty medicinal plants were examined in this study. The plant material (30 g) was extracted with methanol (300 mL) for 24 hr at room temperature. Methanol extracts were filtered and evaporated, then freeze-dried. Aqueous extracts were prepared with dried plant material (30 g) and hot distilled water (300 mL). After 3 hr, the aqueous extracts were filtered and evaporated, then lyophilized. Extracts prepared from different plants were tested the antiviral activity against influenza viruses [A/vietnam/1194/04 (H5N1)-NIBRG-14] using the hemagglutination (HA) assay. Among the test plants, Asarum sieboldii was found to be a potent inhibitor of H5N1 influenza virus in MDCK cell culture. Virus titers were 7 log, whereas with methanol extract of Asarum sieboldii for 48 hr titers were 3 log, indicating that methanol extract of Asarum sieboldii inhibited the H5N1 influenza viruses from the infected cells.
