Anti-bacteria effect of green tea, mulberry, and mate leaves extracts on S. mutans (녹차잎, 뽕잎, 마테잎 추출물이 S. mutans에 미치는 항균효과)

  • This study aims to figure out how green tea leaves, mulberry leaves, and mate leaves that are often taken around in drinking tea influence on S. mutans known as dental caries causative bacteria. After vaccinating 1% of S. mutans on the extract added badge and incubating 6 hours and 10 hours in $37^{\circ}C$, it calculated absorbance and bacterial colony number. Extract of green tea leaves and mate leaves were added with 0, 0.5, 1.0, 2.0, and 4.0% of concentration. As a result of growth suppressive effect of S. mutans, it may be observed that colony number significantly decreases in higher concentration. When it calculated bacterial colony number with adding 2% extract after 10 hours, 99.0% for green tea leaves, 97.1% for mulberry leaves, and 89.6% for mate leaves appeared.

Effect of antibacterial effects of myrrh, rhatany, chamomomilla against to oral microorganisms (몰약, 라타니아, 카모밀레 등의 구강 내 병원균에 대한 항균작용)

  • Even though there exist a lot of study about antibacterial effects and reactions of extracted materials from plant, few study exist about oral pathogenic bacteria. Therefore we tried to recognize about the suppression effect to the periodontal pathogenic bacteria and halitosis, when add some kinds of plant extracted materials, myrrh, rhatany, chamomolilla to saliva. We used Crude drug : Myrrh tincture (100mg/ml), Ratanhia tincture (100mg/ml), Chamomile tincture(100mg/ml). We inspected about the cariogenic bateriae, S. mutans GS5 and S. sobrinus 6715, periodontal pathogenic bacteria, P. gingivalis 2561, P. intermedia ATCC 25611, Candida albicans ATCC 18804, and E. feacalis ATCC 4083, then the result follow. The plant extracted material, myrrh, rhatany, chamomomilla, which have convergence effect, bacteriocidal effect and anti-inflammation effect, show an antibacterial effect and reaction to the oral pathogenic bacteria. And with treating rhatany that have the most strong antibacterial effect, through transmission electron microscopy we could see a severe morphologic change of bacteria. This means with the plant extracted material, we can suppress the oral harmful bacteria and prevent periodontal diseases, caries, halitosis and oral inflammations. And within the future studies for the improvement of oral hygiene, our result might be a clinical evidence.

Antibacterial properties of composite resins incorporating silver and zinc oxide nanoparticles on Streptococcus mutans and Lactobacillus

  • Objectives: Recurrent caries was partly ascribed to lack of antibacterial properties in composite resin. Silver and zinc nanoparticles are considered to be broad-spectrum antibacterial agents. The aim of the present study was to evaluate the antibacterial properties of composite resins containing 1% silver and zinc-oxide nanoparticles on Streptococcus mutans and Lactobacillus. Materials and Methods: Ninety discoid tablets containing 0%, 1% nano-silver and 1% nano zinc-oxide particles were prepared from flowable composite resin (n = 30). The antibacterial properties of composite resin discs were evaluated by direct contact test. Diluted solutions of Streptococcus mutans (PTCC 1683) and Lactobacillus (PTCC 1643) were prepared. 0.01 mL of each bacterial species was separately placed on the discs. The discs were transferred to liquid culture media and were incubated at $37^{\circ}C$ for 8 hr. 0.01 mL of each solution was cultured on blood agar and the colonies were counted. Data was analyzed with Kruskall-Wallis and Mann-Whitney U tests. Results: Composites containing nano zinc-oxide particles or silver nanoparticles exhibited higher antibacterial activity against Streptococcus mutans and Lactobacillus compared to the control group (p < 0.05). The effect of zinc-oxide on Streptococcus mutans was significantly higher than that of silver (p < 0.05). There were no significant differences in the antibacterial activity against Lactobacillus between composites containing silver nanoparticles and those containing zinc-oxide nanoparticles. Conclusions: Composite resins containing silver or zinc-oxide nanoparticles exhibited antibacterial activity against Streptococcus mutans and Lactobacillus.

  • Cow's milk and dairy products are elements of the human diet that could play an important role in improving human health. The macronutrients and micronutrients found in milk could supply the nutrients required to maintain human health. Among them, milk-derived bioactive peptides have been identified as potential ingredients found in health promoting functional foods. These bioactive peptides target diet-related chronic diseases, particularly non-communicable ones such as cardiovascular disease, diabetes and obesity. Additionally probiotics such as lactic acid bacteria (LAB) are can be considered live microorganisms that confer health benefits for the host-, when administered in adequate amounts. Further, the calcium, vitamin D, and protein content of milk and dairy products could play a role in proving bone health. The effect of milk and calcium on bone mineral density could prevent against fracture, osteoporosis and rickets. Furthermore, milk and dairy products also contain which factors that, which protect against dental caries (anti-cariogenic properties). This paper reviews the various nutritional functions of milk and dairy products in improving human health.

  • Grapefruit seed extract has been reported to have antimicrobial effect. The purpose of this study was to evaluate the antimicrobial and anti-gingivitis effect of chewing gum containing grapefruit seed extract and xylitol. 40 healthy subjects with gingivitis or early periodontitis were divided into two groups. Subjects in the experimental group chewed gum containing grapefruit seed extract and xylitol while subjects in the control group chewed gum containing only xylitol. All subjects received scaling and tooth brushing instruction. 1 week after scaling was set as baseline. Gingival index and plaque index were scored at baseline, 1 week, 2 week, 3 week and 4 week. Bleeding index, probing pocket depth and clinical attachment level were scored at baseline, 2 week and 4 week. The number of total bacteria and Streptococcus mutans in unstimulated saliva of experimental group were counted at 1 week, 2 week, 3 week and 4 week. Gingival indices of experimental group and control group at baseline, 1 week, 2 week, 3 week and 4 week were 0.850${\pm}$0.298, 0.575${\pm}$0.345, 0.533${\pm}$0.332, 0.459${\pm}$0.311, 0.408${\pm}$0.224 and 0.758${\pm}$0.379, 0.592${\pm}$0.276, 0.563${\pm}$0.281, 0.454${\pm}$0.194, 0.413${\pm}$0.199 (mean${\pm}$SD), respectively. Plaque indices of experimental group and control group at baseline, 1 week. 2 week, 3 week and 4 week were 0.497${\pm}$0.500, 0.375${\pm}$0.484, 0.332${\pm}$0.471, 0,286${\pm}$0.452, 0.210${\pm}$0.407 and 0.411${\pm}$0.492, 0.375${\pm}$0.484, 0.354${\pm}$0.479, 0.313${\pm}$0.463, 0.193${\pm}$0.395, respectively. Bleeding indices of experimental group and control group at baseline, 2 week and 4 week were 0.377${\pm}$0.177, 0.298${\pm}$0.152, 0.192${\pm}$0.108 and 0.383${\pm}$0.124, 0.318${\pm}$0.153, 0.225${\pm}$0.126, respectively. Probing pocket depth of experimental group and control group at baseline, 2 week and 4 week were 2.56${\pm}$1.00, 2.40${\pm}$0.65, 2.23${\pm}$0.64 and 2.45${\pm}$0.682.37${\pm}$0.57, 2.19${\pm}$0.57, respectively. Clinical attachment level of experimental group and control group at baseline, 2 week and 4 week were 2.58${\pm}$1.01, 2.43${\pm}$0.67, 2.26${\pm}$0.65 and 2.49${\pm}$0.70, 2.40${\pm}$0.59, 2.22${\pm}$0.62, respectively. The % of reduction of total bacteria in saliva of experimental group at 2 week, 3 week and 4 week were 46 ${\pm}$ 53%, 53 ${\pm}$ 5% and 69 ${\pm}$ 33%. The % of reduction of Streptococcus mutans count in saliva of experimental group at 2 week, 3 week and 4 week were 52${\pm}$69%, 88${\pm}$30% and 89${\pm}$17%. From these findings, it can be concluded that regular use of grapefruit seed extract /xylitol chewing gum may be effective to control and prevent gingivitis and may have caries-preventive effect.

Effects of Foreign Plant Extracts on Cell Growth and Biofilm Formation of Streptococcus Mutans (해외 자생식물추출물이 Streptococcus mutans의 세포 성장 및 생물막 형성에 미치는 영향)

  • Chemically synthesized compounds are widely used in oral hygiene products. However, excessively long-term use of these chemicals can cause undesirable side effects such as bacterial tolerance, allergy, and tooth discoloration. To solve these issues, significant effort is put into the search for natural antibacterial agents. The aim of this study was to assess the extracts of foreign native plants that inhibit the growth and biofilm formation of Streptococcus mutans. Among the 300 foreign plant extracts used in this study, Chesneya nubigena (D. Don) Ali extract had the highest antimicrobial activity relatively against S. mutans with a clear zone of 9 mm when compared to others. This plant extract also showed anti-biofilm activity and bacteriostatic effect (minimal bactericidal concentration [MBC], 1.5 mg/ml). In addition, the plant extracts of 19 species decreased the ability of S. mutans to form biofilm at least a 6-fold in proportion to the tested concentrations. Of particular note, C. nubigena (D. Don) Ali extract was found to inhibit biofilm formation at the lowest concentration tested effectively. Therefore, our results reveal that C. nubigena (D. Don) Ali extract is a potential candidate for the development of antimicrobial substitutes, which might be effective for caries control as well, as demonstrated by its inhibitory effect on the persistence and pathogenesis of S. mutans.

Effects of Antibacteria and Adhesive Inhibition of Scutellaria baicalensis Extract on Streptococcus mutans (황금(Scutellaria baicalensis) 추출물에 의한 Streptococcus mutans의 항균 및 부착억제 효과)

  • The natural products are used to be development of new antibacterial substances against human pathogenic bacteria. Adherence to the tooth surface by S. mutans is an important step in initiation of dental caries. This study was to examine antibacterial activity and anti-adhesive effect of Scutellaria baicalensis extract against S. mutans. Extracts of S. baicalensis were tested for antimicrobial activities by paper disc methods and radial diffusion assay methods, and bacterial adherence assay using 3 type of hydroxyapatite. The antibacterial level of ethyl acetate extract, IPK-3 on the growth of S. mutans was 125 mg/ml of minimum inhibitory concentration (MIC). The maximum growth of S. mutans in medium added with IPK-3 extract (50 mg/ml) was delayed to 30 hr, while the highest at 24 hr in control medium. The pH values of the control medium was 5.63 at 18 hr, but the media supplemented with IPK-3 extract was pH 6.50 at 12 hr. In adhesive inhibition assay, S. mutans was labelled with the fluorescent indicator DAPI and measured with fluorescence microscope. Adhesion of S. mutans on hydroxyapatite beads was inhibited by IPK-3 extracts. These results suggest that S. baicalensis extract can be used as an effective material for antibacterial activity and adhesive inhibition against S. mutans.

Fluorine-releasing of Dental Restoration Materials in which the Fluorine is Contained (불소 유리로 본 불소 함유 수복재)

  • Restoration materials used to investigate effects of fluorine such as enamel strengthening and anti-caries effects in several types of dental restoration materials were five kinds including Ionoseal(VOCO GmbH, Cuxhaven, Germany), Fuji Filling LC(GC Co. Tokyo, Japan), Quadrant Universal LC(CAVEX Holland BV, Netherlands), PermaCem$^{(R)}$(DMG, Hamburg, Germany) and Dyract$^{(R)}$ AP(Dentsply GmbH, Germany), and the amount of fluorine-releasing was measured with ICS-5000 Reagent-FreeTM Ion Chromatography(RFICTM, Dionex, U.S.A.). The results of this study are as follows. 1. In all types of restoration materials, the amount of fluoride-releasing was reduced with time passage and it was declined sharply to show significance in four weeks. Fuji Filling LC(12.445PPM) or resin-reinforced glass ionomer and PermaCem$^{(R)}$(16.121PPM) or compomer were found to release fluorine for a long term(P<.001). 2. Ionoseal(0.887PPM) or glass ionomer and Quadrant Universal LC(0.957PPM) or composite resin released a few fluorine of 1PPM or less than 1PPM after one week, and Dyract$^{(R)}$ AP or compomer released fluorine of 8.631PPM in one week and its amount of releasing decreased dramatically in two and four week by recording 0.175PPM and 0.116PPM, respectively. Therefore, the effect of releasing fluorine in four weeks was observed to be poor (P<.001). 3. Fuji Filling LC or resin-reinforced glass ionomer and PermaCem$^{(R)}$ or compomer released fluorine of 33.372 and 1.902PPM, respectively in one week and their amount of releasing increased to be 36.371 and 18.223PPM, respectively in two weeks. So, their amount of fluorine-releasing recorded the highest levels in two weeks(P<.001).
