발아현미와 수종한약재 추출액의 스트레스 및 불안상태 개선 효과에 대한 임상연구 (A Clinical Trial about Anti-anxiety and Anti-stress Effect of a modified formula consisted with Several Herbs)

  • This double-blind randomized clinical trial was performed to evaluate the anti-anxiety, anti-stress effect of a modified formula WS-01 consisted with several herbs(Jujubae Fructus. Longanae Arillus. Poria Cocos) and germinated rice. The extract of the formula was administered (p.o) to 24 students and placebo was to 23 students for 4 weeks. STAI-KYZ(Hahn. Hakjisa), ABR2000(Meridian, Korea), CBC, LFT, U/A were checked before and after the administration period. There was no difference between the STAI score of WS-01 and that of placebo group in first test. But the STAI state anxiety score was 5.33±7.33 reduced in WS-01 group and it was 0.52±6.42 increased in placebo group in second test. this result was significant stastically. 10 students in WS-01 group showed improving result, but only 1 student in placebo group showed improving result of ABR2000 test. The result of ABR2000 was analyzed with curve 2, Regulation 2, and graph 1,2,3, because those reflect central nerve system and stressful condition of the body. There was no significant result in first and second CBC, LFT, Urine test. The results of this clinical study suggest that WS-01 has anti-anxiety, anti-stress effect, and it could be applied to various conditions or disorders related with anxiety and stress.

Anxiolytic Action of Taurine via Intranasal Administration in Mice

  • Taurine has a number of beneficial pharmacological actions in the brain such as anxiolytic and neuroprotective actions. We explored to test whether taurine could be transported to the central nervous system through the intranasal route. Following intranasal administration of taurine in mice, elevated plus maze test, activity cage test and rota rod test were carried out to verify taurine's effect on anxiety. For the characterization of potential mechanism of taurine's anti-anxiety action, mouse convulsion tests with strychnine, picrotoxin, yohimbine, and isoniazid were employed. A significant increase in the time spent in the open arms was observed when taurine was administered through the nasal route in the elevated plus maze test. In addition, vertical and horizontal activities of mice treated with taurine via intranasal route were considerably diminished. These results support the hypothesis that taurine can be transported to the brain through intranasal route, thereby inducing anti-anxiety activity. Taurine's anti-anxiety action may be mediated by the strychnine-sensitive glycine receptor as evidenced by the inhibition of strychnine-induced convulsion.

동물모델에서 시호가용골모려탕(柴胡加龍骨牡蠣湯)의 수면유도 및 항불안 효과 (Sleep-promoting and Anti-anxiety Effects of Shihogayonggolmoryo-tang in Mice)

  • Shihogayonggolmoryo-tang (ST) is a Korean medical herb cocktail which has been used to treat anxiety induced insomnia. In this study, we will examine sleep-promoting and anti-anxiety effects of ST, and investigate its mechanism. ICR mice were divided into three groups for the first examination : control group (n=11), ST50 group (50 mg/kg, po, n=11), ST200 group (200 mg/kg, po, n=11). Sleep-promoting effect was confirmed by measuring the sleeping duration time and sleeping onset time after thiopental sodium treatment (50 mg/kg, ip). ICR mice were divided into five groups for the second examination : control group (n=11), ST200 group (200 mg/kg, po, n=11), ST200+Flumazenil group (ST 200 mg/kg, po, flumazenil 0.3 mg/kg, ip, n=11), diazepam group (1 mg/kg, ip, n=11), diazepam+flumazenil group (diazepam 1 mg/kg, ip, Flumazenil 0.3 mg/kg, ip, n=11). Anxiety behavior and sleep-promoting effect was confirmed by open field test and measuring the sleeping duration time and sleeping onset time. Expression levels of c-fos in tuberomammillary nucleus (TMN) and ventrolateral preoptic nucleus (VLPO) were analyzed by immunohistochemistry. ST treated group showed significantly decreased anxiety behavior and enhanced sleeping duration time and sleeping onset time concentration dependently. The expression of c-fos was significantly upregulated in VLPO as sleep-inducing center and TMN as downregulated in arousal center by ST treatment. In addition, all effects of ST were reversed by flumazenil. Our results suggest that ST has sleep-promoting and anti-anxiety effects through regulating arousal center (TMN) and sleep-inducing center (VLPO).

소양인 형방도적산과 양격산화탕의 항 스트레스효과에 관한 실험적 연구 (An Experimental Study on the Anti-stress Effect by Soyangin Hyeongbangdojeok-san and Yanggyeoksanhwa-tang)

  • 1. Objective This study was done to identify the anti-stress effect of Hyeongbangdojeok-san (HDS), Yanggyeoksanhwa-tang(YST) in Soyangin. 2. Methods Experimental animals were composed of YST, HDS+stress groups which were administered each by 200mg/kg, 400mg/kg and the Saline+stress group. On the 1st day, making the rats forced swim and on the 2nd day, applying Forced swimming test to the rats. After FST, the levels of Corticosterone in the blood were measured. For the study of learning retardation, memory ability and anxiety reaction, experimental animals were composed of YST, HDS+restraint stress groups which were administered each by 400mg/kg, no stress group and the Saline+restraint stress group. Restraint stress were applied 2 hours a day for 3 weeks. On the last day of the 3rd week, Elevated Plus Maze(EPM) was applied to the groups and Morris Water Maze(MWM) was applied to the groups for 7 days. 3. Results 1. As the results of measuring FST which reflects depression, the YST+stress group and the HDS+stress group showed significant effect in comparison with the Saline+stress group. The levels of Corticosterone in the blood were decreased only in the 400mg/kg YST+stress group. 2. As the results of measuring how long EPM which reflects anxiety reaction stayed in the open arm, there was the trend which can suppress anxiety reaction in the HDS+restraint stress group bur no statistical significance. But there was any suppression of anxiety reaction in the YST+restraint stress group. 3. According to the result of MWM, the saline+restraint stress group showed the learning retardation which means increased time arriving at goal compared to the normal group at the second and third day of measurement. On the contrary, a learning retardation was significantly decreased in the YST+restraint stress group at the third day of measurement. 4. Among the Probe trial test a memory loss occurred in the saline+restraint stress group, but memory ability was significantly increased in the YST+restraint stress group. 4. Conclusion: As the results above, Soyangin Yanggyeoksanhwa-tang has significant influence to the antidepression effect, the learning retardation, the anxiety reaction and also in the Hormone level. Hyeongbangdojeok-san has significant influence to the antidepression effect, in the Hormone level, bur not to the learning retardation and anxiety reaction. prefer to drink cold water, and who are suffering from chronic gastritis.

아동청소년에서 운동의 항우울 효과와 BDNF와의 관련성에 대한 예비 연구 (Association between BDNF and Antidepressant Effects of Exercise in Youth: A Preliminary Study)

  • Objectives: The purpose of this study was to evaluate anti-depressive effects of exercise on child and adolescent and its association with brain derived neurotrophic factor (BDNF). Methods: Twenty nine middle school boys (age $13.3{\pm}0.7$) were divided into two groups, 15 boys for control group and 14 in the experimental group. The control group participated in a regular exercise program, 3 times a week for 15 weeks. During the same period, the experimental group participated in an aerobic exercise program specifically designed to enhance anti-depressive effect of exercise. Serum BDNF level and its performance of each group on the Beck Depression Index (BDI), Children's Depression Inventory (CDI), Screen for Child Anxiety Related Emotional Disorders (SCARED), Aggression Questionnaire (AK-Q), and Stroop task were compared before and after the exercise program. Results: Scores of BDI, CDI, SCARED, and AK-Q were significantly lower in both groups after the exercise programs compared to those before the programs. The Stroop task performances were significantly improved after the programs. However, there were no significant differences between two exercise programs, except SCARED separation anxiety, AK-Q physical, and verbal aggression scores. Also, no association was found between serum BDNF level and anti-depressive effects of exercise. Conclusion: Our preliminary results suggest a possible effect of exercise on depression, anxiety, aggression, and cognition of child and adolescents.

Phytochemical Standardization and Anti-Anxiety (Izterab-e-Nafsani) study of Aftimoon Hindi (Cuscuta reflexa Roxb.) on An Animal Model

  • Background: Cuscuta reflexa Roxb is a member of the Cuscutaceae family, and in Unani medicine, it is known as Aftimoon. It is a parasitic plant that can be found growing abundantly on various host plants in India up to 3000 metres in altitude during the rainy season. Unani physicians have been using it for years to cure a variety of illnesses, including psychiatric illnesses like melancholia, schizophrenia, and epilepsy. It has been used to cure hepatitis, palpitations, and skin disorders, among other things. Objective of the study: To evaluate anti-anxiety effect of Cuscuta reflexa Roxb in Swiss Albino mice of either sex. Materials and Methods: A total of 24 Swiss Albino mice weighing 25-35 g were used in this study. Animals were chosen at random and held in their cages for at least 7 days in a standard setting. Group A was given regular saline as a vehicle, Group B was given a hydro alcoholic extract of the lower dose of the test drug, Group C was given a hydro alcoholic extract of the higher dose of the test drug, and Group D was given the standard drug Diazepam 5 mg/kg orally. Aftimoon as hydro alcoholic extract (200 mg/kg and 400 mg/kg body wt.) was given in single and double doses and observed for 7 days. Results: For each parameter in each category, mean and standard deviations were computed. For multiple group comparisons, a one-way ANOVA was used, followed by Turkey's post hoc test. (p<0.05) was used as the significance standard. Conclusion: These results advocate that the Aftimoon as double dose (400 mg/kg body wt.) revealed anti-anxiety effect similar to standard drug.

Sogeunjung-tang improves depressive-like behavior decreased by forced-swimming test

  • Sogeunjung-tang (S-tang) is a traditional Korean medicine that has been used for the therapy of anxiety disorder in a variety of clinical situations. However, no study has investigated this effect. The aim of this study was to investigate the effectiveness of S-tang for reducing depression in animal model. The anti-depressant effect of S-tang was investigated in ICR mice by using forced swimming test (FST). After FST, we analyzed the blood biochemical parameters from blood. The immobility time was significantly decreased in the S-tang oral administration or pharmaceutical acupuncture group compared with the control group at 14 days (p < 0.05). The anti-depressant effect of S-tang was higher than that of fluoxetine (a classical anti-depressant). S-tang or fluoxetine significantly decreased the creatine kinase and lactate dehydrogenase level compared with the control group (p < 0.05) but not glucose, protein, and blood urea nitrogen level. Further investigation of S-tang as a potential pharmacologic agent in the management of depression related anxiety is warranted.

Anti-nociceptive effects of dual neuropeptide antagonist therapy in mouse model of neuropathic and inflammatory pain

  • Background: Neurokinin-1 (NK1) and calcitonin gene-related peptide (CGRP) play a vital role in pain pathogenesis, and these proteins' antagonists have attracted attention as promising pharmaceutical candidates. The authors investigated the anti-nociceptive effect of co-administration of the CGRP antagonist and an NK1 antagonist on pain models compared to conventional single regimens. Methods: C57Bl/6J mice underwent sciatic nerve ligation for the neuropathic pain model and were injected with 4% formalin into the hind paw for the inflammatory pain model. Each model was divided into four groups: vehicle, NK1 antagonist, CGRP antagonist, and combination treatment groups. The NK1 antagonist aprepitant (BIBN4096, 1 mg/kg) or the CGRP antagonist olcegepant (MK-0869, 10 mg/kg) was injected intraperitoneally. Mechanical allodynia, thermal hypersensitivity, and anxiety-related behaviors were assessed using the von Frey, hot plate, and elevated plus-maze tests. The flinching and licking responses were also evaluated after formalin injection. Results: Co-administration of aprepitant and olcegepant more significantly alleviated pain behaviors than administration of single agents or vehicle, increasing the mechanical threshold and improving the response latency. Anxiety-related behaviors were also markedly improved after dual treatment compared with either naive mice or the neuropathic pain model in the dual treatment group. Flinching frequency and licking response after formalin injection decreased significantly in the dual treatment group. Isobolographic analysis showed a meaningful additive effect between the two compounds. Conclusions: A combination pharmacological therapy comprised of multiple neuropeptide antagonists could be a more effective therapeutic strategy for alleviating neuropathic or inflammatory pain.

Intranasal administration of dexmedetomidine (DEX) as a premedication for pediatric patients undergoing general anesthesia for dental treatment

  • Background: The most important reason for pre-operative administration of medication is to reduce anxiety. Alleviation of fear and anxiety about surgery enables patients to remain comfortable during treatment. Dexmedetomidine (DEX) is a fast-acting drug that is used as a premedication in different circumstances because it has sedative and anti-anxiolytic effects, and stable hemodynamics. It also has the advantage of intranasal administration. The aim of this study was to investigate the effects and hemodynamic stability of DEX by retrospectively analyzing cases in which DEX was administered nasally as a premedication. Methods: Ten patients treated at Dankook University Dental Hospital, recruited between February and April 2015, received intranasal delivery of $2{\mu}g/kg$ DEX, 30 minutes prior to general anesthesia. Anesthesia records of anxiety, blood pressure, respiration, pulse, estimated arterial oxygen saturation ($SpO_2$), and partial pressure, or maximum concentration, of carbon dioxide ($ETCO_2$) were analyzed. Results: Administration of DEX prior to a general anesthetic effectively relieved anxiety. Respiratory depression, the most severe adverse effect of other sedatives, was not observed. Hemodynamic stability under general anesthesia was maintained during treatment and a reduction in emergence delirium was observed upon completion of treatment. Conclusions: Premedication administration of DEX is safe for pediatric patients undergoing dental treatment under general anesthesia.

Anti-stress Effects of Ethanol Extract of Ziziphus jujuba Against Corticosterone-Induced Apoptosis in PC12 Cells

  • The coronavirus disease 2019 (COVID-19) pandemic may be stressful for people. Public health actions, such as social distancing, can make people feel isolated and lonely and can increase stress and anxiety. As a result, there is a growing interest towards various materials to relieve stress. Thus, the present study aimed to investigate the anti-stress effects of ethanol extract of Ziziphus jujuba in PC12 cells treated with corticosterone and its underling mechanisms. Furthermore, the viability of the cells, the apoptosis of the cells, the level of phosphorylation of extracellular signal-regulated kinases (p-ERKs) expression were measured by MTT assay, LDH assay, Hoechst staining assay and western blotting. Our results showed that the extract of Ziziphus jujuba reversed corticosterone-induced damage in PC12 cells, which increased cell viability, decreased LDH release, and attenuated corticosterone-induced apoptosis as compared with the corticosterone-treated group. Therefore, these data suggest that the extract of Ziziphus jujuba could be a good candidate for development as a functional food supplement in the improve the anti-stress effect.
