Alteration of hepatic anti-oxidant systems by 4-nonylphenol, a metabolite of alkylphenol polyethoxylate detergents, in Far Eastern catfish Silurus asotus

  • Objectives This study aimed to estimate the effects of 4-nonylphenol (NP), a ubiquitously present surfactant in aquatic environments, on the anti-oxidant systems of the liver in the Far Eastern catfish Silurus asotus. Methods Changes in biochemical parameters involved in glutathione (GSH)-related and other anti-oxidant systems were analyzed following 4 weeks of 4-NP administration (0.1 and 1.0 mg/kg diet) via a formulated diet to catfish. Results 4-NP exposure induced an elevation in hepatic lipid peroxide levels and an accompanying decrease in reduced state GSH after 2 weeks, suggesting pro-oxidant effects of the chemical in catfish. This oxidative stress was associated with an inhibition of the GSH-utilizing enzyme glutathione peroxidase at the same time point. This inhibition was restored after 4 weeks. The activities of other anti-oxidant enzymes, i.e., glutathione reductase, superoxide dismutase and catalase were increased after 4 weeks. These enzyme increases occurred more strongly at the higher 4-NP concentration (1.0 mg/kg diet). Conclusions 4-NP given to catfish at 0.1 to 1.0 mg/kg diet, concentrations relevant to environmental levels, depletes the endogenous anti-oxidant molecule GSH and temporarily inhibits GSH-related anti-oxidant enzymes. Such declines in anti-oxidant capacity and elevated oxidative stress seem to be compensated eventually by subsequent activation of various anti-oxidant enzyme systems.

Comparison of Properties of Polyclonal Anti-N-Acetylchitooligosaccharides and Anti-Chitooligosaccharides Antibodies Produced for ELISA

  • To develop the enzyme-linked immunosorbent assay (ELISA) for the analysis of N-acetylehitooligosaccharides (NACOS) and chitooligosaccharides (COS), specific antibodies (Abs) were produced, and their properties were compared. N-acetylehitohexaose (NACOS6), chitohexaose (COS6), and COS mixture (COSM) conjugated to bovine serum albumin (BSA) were used to immunize rabbits. By the use of specific Abs and NACOS6-horseradish peroxidase (HRP), COS6-HRP, and COSM-HRP conjugates, competitive direct ELISA (cdELISA) was developed. The detection limits of NACOS6 by the anti-NACOS6 Ab and COS6 by the anti-COS6 and the anti-COSM Abs in the cdELISAs were about 0.2, 2, ana 2 ng/ml (ppb), respectively. In the cdELISA, the anti-NACOS6 Ab was found to recognize NACOS3-NACOS6, but not N-acetyl-D-glucosamine (GlcNAc), NACOS2, and COSs; the anti-COS6 Ab recognized COS2-COS6 and COSM, but not glucosamine (GlcN) and NACOSs. The recognition pattern of the anti-COSM Ab was almost the same as that of the anti- COS6 Ab, except that the former recognized COS2 and COS3 slightly better than the latter.

  • Anti-G 밸브는 Anti-G Suit에 연결되어 급격한 기동 시 조종사에게 적절한 여압기능을 제공한다. 또한 Anti-G 밸브는 연결된 레귤레이터를 조종하여 기동 간 조종사의 생리학적 산소요구량 증가에 따라 적절한 PBG(Pressure Breathing for G)가 제공되도록 조절한다. FA-50과 같이 고기동 임무가 요구되는 항공기에서 Anti-G 밸브는 조종사의 생존 및 항공기의 안전을 위해 매우 중요한 요소이다. 본 논문에서는 Anti-G 밸브 개발 중 발생된 문제점을 바탕으로 개발 요구도 및 설계개선 내용을 기술하였으며, 개선된 연구에 대해 구성품 성능시험 및 비행시험검증 결과를 함께 소개한다.

Anti-Cyclic Citrullinated Peptide(Anti-CCP) 측정에 대한 연구 (A Study on the Measurement of Anti-Cyclic Citrullinated Peptide (Anti-CCP))

  • This study was carried out to review and evaluate anti-CCP ELISA assay for diagnostic in RA patients from an early arthritis clinic (EAC). The subjects were obtained from patients visiting the outpatient clinic of the Dept. of Rhumatology med.of P hospital in Daegu, during 6 months from July 1, 2006 to December 31, 2006. The subjects were 140 cases : 80 cases from RA patients (60 women and 20 men; mean age 58 years; range, 32-68 years) confirmed by clinical diagnostic. 39 cases of these RA patients were classified as having early RA (EAC). 50 cases (non-RA) did not fulfill the criteria for RA, and 10 cases were from healthy individuals. We performed the analysis with solid phase-ELISA method (ETI-max3000, Diasorin; Italy) for anti-CCP and Nephelometry assay (Roche/Hitachi 902 analyzer; USA) for RF. The results obtained were summarized as follows ; anti-CCP ELISA is more specific than RF Nephelometry assay (specificity 94% vs 90%) to diagnose RA patients with suspected EAC (early arthritis clinic). The combination test "anti-CCP and RF" had a very high specificity (specificity 98.3%, PPV; RA group 96%, EAC 95%), the difference was statistically significant (p<0.05). Anti-CCP ELISA had more sensitivity in EAC (Early arthritis clinic) patients than chronic RA patients (sensitivity 64% vs 24%, respectively), anti-CCP of RA group and EAC group was more specific than RF (anti-CCP PPV; 92%, 89% vs 89%, 81% respectively), the difference was statistically significant (p<0.05). The difference of antibody concetration between anti-CCP and RF for RA and the control group is statistically significant (p<0.05). In conclusion, anti-CCP ELISA testing may be useful if performed concomitantly with RF Nephelometry assay to diagnose RA patients with suspected EAC (early arthritis clinic).

항진균성 활성물질을 생성하는 토양방선균의 분리 (Isolation and Selection of Actinomycetes Producing Anti-fungal Materials)

  • Anti-fungal materials producing bacteria were isolated from soil by bennett's agar and actinomycete isola-tion agar medium. The bacterla were identified as synonym of Actinomycetes. Based on the data obtained from its morphological and colony characteristics. The medium for production of anti-fungal materials was YEME (yeast extract 4 g, malt extract l0g, glucose 4 g, D.W 1ι, pH 7.0${\pm}$0.2). The culture conditions were 30$^{\circ}C$, 7 days and 200 rpm in shaking incubator. No. 13, No. 15 and No.28 strains were produced anti-fungal materials against fungal plant pathogens. Specially, The No. 28 strain showed a powerful biopesticide activity and broad spectrum effects of anti -fungal materials on Collectrichum coccodes, Botrytis cinerea, Cladosporium cucumerinum, Didymella bryoniae.

수중폭발충격에 대한 수중음향 트랜스듀서의 내충격 특성 해석 (Analysis for Anti-shock Characteristics of Underwater Acoustic Transducers to the Explosive Shock)

  • Underwater acoustic transducers can be exposed to a underwater explosive shock caused by various types of underwater weapon. So, a robust anti-shock design is required for transducers to endure the underwater explosive shock. To check the anti-shock characteristics of a transducer, underwater explosive shock test is needed. The conditions of underwater explosive shock test are set up referring to various oversea explosive shock test specifications, and the explosive shock pressure values are calculated according to those conditions. Transient analyses art: carried out for two kinds of underwater acoustic transducer model to verify the anti-shock characteristics. The applied model has robust anti-shock characteristics enough to endure the explosive shock up to 2300 psi. In the future, the transducer design should be certified through the fields test, and modified if needed.

나노 측정시스템을 위한 방진 구조물의 동적 설계에 관한 연구 (A Study on the Dynamic Design of Anti-Vibration Structures for Nano-class Measuring System)

  • It is necessary to design the anti-vibration system of precision machinery for a quality assurance. However, in general, the allowable vibration limit is not well known. In this paper, the vibration criteria for foundation of sensitive machinery is proposed and anti-vibration system is designed by using vibration measurement results of foundation. Also, the finite element analysis is performed to verify the effectiveness of the designed anti-vibration system and to determine the allowable dynamic loads of precision measuring system. The results of this study will be helpful for the future design of anti-vibration structures with nano scale measuring system.

유기농 콩 추출물의 항염증 및 항알레르기 효과 (Anti-inflammatory and Anti-allergic Effect of Extractsfrom Organic Soybean)

  • 유기농재배 콩을 다양한 용매에서 추출하여 그 추출물에 대한 항산화 효과, 항염증 효과, 대식세포의 세포사멸 보호 효과, 그리고 알레르기 저해효과를 비교하였다. DPPH 법과 SOD 효소의 활성 측정으로 항산화 능력을 평가하였으며, 항염증 효과는 LPS로 유도시킨 Raw 264.7 세포주에서 염증 매개 단백질인 Cox-2와 iNOS의 발현량을 Western blotting으로 조사하여 분석하였다. 또한 대식세포 보호 효과를 세포사멸 조절 단백질인 Bcl-xl의 발현량조사로 평가하였고, 1mM $H_2O_2$ 처리로 유도되는 세포사멸의 보호효과를 세포 수 측정법으로 조사하였다. 항알레르기 효과는 흰쥐의 비만세포인 RBL-2H3 세포주를 사용하여 IgE로 매개되는 비만세포의 탈과립화를 OPT assay로 측정하여 분석하였다. 그 결과, 유기 재배 콩의 다양한 추출물에서 항산화, 세포사멸 보호, 항염증 효과를 나타내었다. 특히 유기농 콩의 열수 추출물은 LPS로 유도시킨 염증반응을 효과적으로 억제하는 것으로 나타났다. 그러나 항알레르기 효과는 모든 추출물에서 히스타민 유리량이 약간 감소하는 것으로 나타나 유기농 콩의 특별한 효과를 확인할 수 없었다. 콩에는 많은 유용한 생리활성물질 뿐 만 아니라 알레르기 유발 물질도 존재하는 것으로 알려져 있지만 조추출물 상태에서는 특이적인 알레르기 유발보다 오히려 항알레르기 효과가 나타났다. 결론적으로, 만성 염증 및 천식, 그리고 아토피피부염과 같은 염증관련 질환에는 유기 재배된 콩이 좋은 억제 효과를 가지고 있다고 판단된다.

야전 운용자료를 이용한 대함유도탄의 신뢰도 분석 연구 (A Study on the Reliability Analysis of Anti-ship missile using Field Data)

  • 미국을 비롯한 선진국들은 배치 운용중인 개발 무기체계의 야전 운용자료를 수집 분석하여 신뢰도 유지여부를 확인하고 설계 재반영, 군수지원 요소 최적화를 위한 기초 경험자료 등으로 잘 활용하고 있으나 국내에서는 적용이 미흡한 실정이다. 따라서 본 논문에서는 국내 연구개발후 함정에 배치 운용 중인 ${\bigcirc}{\bigcirc}$대함유도탄을 대상으로 최근 6년간 ${\bigcirc}{\bigcirc}{\bigcirc}$발에 대한 구성품별 고장현황 등이 포함된 야전 운용자료를 수집하여 Weibull++ v7 소프트웨어에 입력후 모수적 추정방법을 이용하여 신뢰도를 분석하였다. 분석결과를 개발단계에서 분석된 유도탄과 구성품별 MTBF와 비교하여 초기신뢰도 유지여부를 분석하였고, 고장패턴과 검사주기 적절성도 확인하였다. 도출결과를 개발기관 및 품질보증기관, 제작사에 환류하여 RCM 분석을 통한 초기 신뢰도 회복 조치 및 총수명주기간 유지보수비가 절감되도록 정비정책에 대한 개선방안을 제시하였다.
