Effects of Rice Straw (RS) on Allergic Contact Dermatitis (ACD) Induced by DNCB in Mice (볏집 도초(稻草)이 DNCB로 유발된 생쥐의 알레르기성 접촉성 피부염에 미치는 영향)

  • Objectives In the theory of Korean medicine, rice straw (RS) has been used effectively as treatments for dyspepsia, diarrhea, enteritis, inflammatory epigastric diseases and various dermatitis. However, the theory has not been studied intensively yet about anti-inflammatory effects for human. This study was to investigate effects of RS for a treatment of allergic contact dermatitis (ACD) induced by 2,4-dinitrochlorobezene (DNCB) in mice. Methods In this experiment, effects of RS were investigated on changes in body weights, dorsum skin thickness, clinical aspects on the dorsum skin, spleen and body weight among these four groups; normal group (NOR), control group (CON), RS spread group (RSS) and RS spread and administer group (RSS+Adm). In addition, the effects on proliferations of splenocytes were also investigated in vitro and in vivo study. Results RSS group and RSS+Adm group showed increasing in body weights, diminished dorsum skin thickness and treated dermatitis on dorsum skin. In RSS+Adm group, spleen weights were lowered significantly compared to CON group. Conclusions In conclusion, these data suggest that RS can decrease symptoms of ACD significantly, and it shows the anti-inflammatory and immunosuppressant effect as well. Therefore, RS can be useful to treat patients with ACD.

Study on anti-allergic effects of Arctii Fructus herbal acupuncture (우방자약침(牛蒡子藥鍼)의 항(抗)알러지 효과(效果)에 대한 연구(硏究))

  • Objectives : We studied on anti-allergic effects of Arctii Fructus Herbal Acupuncture(AFHA) and Arctii Fructus Herbal Acupuncture Solution(AF). Methods : In vivo, Animals were herbal-acupunctured AFHA at both ST36 three times for 5 days. Then, we investigated compound 48/80-induced active systemic anaphylaxis(ASA) using ICR mice and anti-DNP IgE-induced passive cutaneous anaphylaxis(PCA) using Sprague Dawley rat. In vitro, we measured cell viability, b-hexosaminidase, IL-4 and TNF-a release from RBL-2H3 cells after treatment of AF of various concentrations. Results : In vivo, AFHA pretreatments at both ST36 inhibited compound 48/80-induced ASA. PCA was inhibited by AFHA pretreatments at both ST36. In vitro, AF treatments were not affect on cell viability and inhibited b-hexosaminidase, IL-4 and TNF-a release. Conclusions : These results suggest that AFHA and AF may be beneficial in the inhibition of allergic inflammatory response.

Anti-allergic effects of So-Cheong-Ryong-Tang in ovalbumin-induced allergic rhinitis model (난알부민 유도 알레르기 비염 마우스에 대한 소청룡탕(小靑龍湯)의 효능 실험연구)

  • Objective : Allergic rhinitis (AR) is an allergic inflammation of the nasal airways. Socheongryong-Tang traditionally has been used in Korea for treatment of allergy diseases. In the present study, we investigated whether SCRT suppresses the progression of AR in vivo and in vitro. Method : In this study, we investigated the effect of oral administration of SCRT on the Ovalbumin(OVA)-induced allergic rhinitis mouse models. Additionally, to find a possible explanation for the anti-inflammation effects of SCRT, we evaluated the inflammatory cytokine levels in LPS-stimulated mouse inflammation cell (RAW264.7) and mouse splenocyte. Results : The finding of this study demonstrated that SCRT reduced mast cells and inflammation cells infiltration in OVA-induced nasal cavity. Additionally, SCRT inhibited the production of inflammatory cytokines in splenocyte. However SCRT don't inhibited the production of inflammatory cytokines in RAW264.7 cells. Conclusion : Taken together, our results showed that oral administration of SCRT beneficial effects in allergic rhinitis, suggesting that SCRT might be a useful candidate for the treatment of allergic rhinitis.

Anti-Inflammatory and Anti-Allergic Effects of Adenophora triphylla var. japonica Extract (잔대 추출물의 항염 및 항천식 효과)

  • Asthma is an increasing global health problem, and novel strategies to prevent or ameliorate the condition are needed. The purpose of the present study was to evaluate the anti-inflammatory and anti-asthma effects of Adenophora triphylla var. japonica extracts. We investigated the molecular mechanism underlying the effects of 80% ethanol extracts (AE) of A. triphylla on lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. AE treatment inhibited pro-inflammatory cytokines such as TNF-${\alpha}$ and IL-6 as well as nitric oxide (NO) production in LPS-stimulated RAW 264.7 cells. In particular, NO and pro-inflammatory cytokine production was suppressed more effectively by aerial parts (AE-A) than roots (AE-R) of A. triphylla. Quantitative RT-PCR assay showed that AE reduced mRNA levels of iNOS and COX-2. We also evaluated the anti-asthmatic effects of AE-A in an ovalbumin (OVA)-induced BALB/c mouse model. AE-A supplementation significantly reduced the amounts of airway eosinophils, IL-4 and IL-13 levels in BALF, and IgE levels in serum as compared with untreated, OVA-induced mice. These results suggest that AE-A can be considered as a therapeutic agent to potentially relieve asthma.

Anti-inflammatroy Activity of YHJST in Activated Mast Cells (량혈제습탕(凉血除濕湯)의 비만세포 활성화 억제작용)

  • Activated mast cells play pivotal roles in allergic and non-allergic inflammatory responses through the release of inflammatory mediators such as histamin, cysteinyl leukotriens, pro-inflammatory cytokines as well as chemokine. We tested whether YHJST, which is clinically prescribed for the treatment of various inflammatory disease including allergic disease, modulate inflammatory reactions in activated mast cells. YHJST decreased the release of histamine and b-hexosamidase in pholbol-12-myristate 13-acetate and/or calcium ionophore A23187 stimulated HMC-1 and RBL-2H3 cells, respectively. Further, the gene expressions of tumor necrosis factor-a, IL-6 and IL-8 were significantly reduced by YHJST. YHJST suppressed powerful induction of NF-kB promoter-mediated luciferase activity. Taken together, these data suggested that YHJST showed it's anti-inflammatory effects through the down-regulation of mast cell activation.

Anti-inflammatory and Anti-allergic Effects of Lentinula edodes Extract by UVIrradiation (UV-B 조사에 따른 버섯 추출물의 항염증 및 항알레르기 활성)

  • In this study, the effects of UV irradiation-enhanced ergocalciferol (vitamin D2) content containing Lentinula edodes extract on inflammation and allergic responses were investigated in vitro. The anti-inflammatory and anti-allergic effects of the mushroom extract were tested by estimating the cytokine secretions, such as TNF-α, IL-6, and IL-1β in LPS-activated macrophages (RAW 264.7), or histamine release in PMA and A23187-activated mast cells (RBL-2H3). Under the condition of macrophage activation with LPS, mushroom extract significantly reduced the secretions of pro-inflammatory cytokines, TNF-α and IL-6, and their mRNA expression also matched the observation. The current mushroom extract also significantly reduced the amount of mast cell degranulation-induced histamine secretion from PMA- and A23187-treated mast cells as well as the reduced secretion of IL-4. These results suggest that mushroom extract, which has increased ergocalciferol content by UV irradiation, inhibits the expression of cytokines in inflammation and allergic reactions; therefore, it can be used effectively for the prevention and treatment of inflammatory and allergic diseases.

An Experimental Studies on the Anti-allergic Reaction, the Antipyretic Action, the Anti-inflammatory and the Analgesic Action of Yenhwagamchotang and Gamiyenhwagamchotang (銀花甘草湯과 加味方이 抗 알레르기, 解熱, 消炎, 鎭痛效果에 미치는 影響)

  • Yenhwagamchotang has been widely used in treatment of inflammatory disease which is based on Oriental Medical literatures. These studies were attempted experimental effects of Yenhwagamchotang(sample A), Yenhwagamchotang plus Forsythia koreana NAKAI.(sample B), and Yenhwa-gamchotang plus Taraxacum mongolicum HAND- MAZZ(sample C),on the Anti-allergic reaction, the antipyretic action,the anti-inflammatory and the analgesic action,in rats. THe results of the studies were as follow: 1. Vascular permeability responses to intradermal serotonin in rats were showed significant effect at all sample groups. 2. The homologous passive cutaneous anaphylaxis in rats provoked by the IgE-like antibody aganist egg white albumin showed the decreasing effect. 3. The delayed type hypersensitivity responses to picryl chloride in mice were showed significant effect at all sample groups. 4. The delayed type hypersensitivity response to sheep red blood cell in mice were showed significant effect at all sample groups. 5. In anti-pyretic effect by yeast method were showed significant effect at all sample groups. 6. The anti-inflammatory effect by carrageenin method were showed significant effect at all sample groups. 7. The analgesic action by acetic acid method in mice were showed significant effect at all sample groups. According to the above result, Yenhwagamchotang(sample A), Yenhwagamchotang plus Forsythia koreana NAKAI(sample B), AND yenhwagamchotang plus Taraxacum mongolicum HNAN- MAZZ.(sample C ) were concluded to have the anti-allergic reaction, the antipyretic action, the anti-inflammatory, the analgesic action.

Anti-Allergic Effect of Oroxylin A from Oroxylum indicum Using in vivo and in vitro Experiments

  • Oroxylum indicum has long been used in Asian traditional medicine to prevent and treat respiratory diseases, diabetes, diarrhea and other conditions. Oroxylin A is a flavone that is present in Oroxylum indicum and in Scutellaria baicalensis. Because the root extracts of both plants have been shown to have anti-allergic effects, the authors investigated whether oroxylin A is likely to have beneficial effects on allergic asthma using female Balb/c mice and rat RBL-2H3 mast cells. Antigen-induced degranulation was measured in vitro by measuring b-hexosaminidase activity. A murine ovalbumin-induced allergic asthma model was used to test the in vivo efficacy of oroxylin A. Sensitization and challenge of ovalbumin induced allergic asthma responses, the accumulations of eosinophils and Th2 cytokine levels in bronchoalveolar lavage fluid and lung tissues. Oroxylin A administration decreased numbers of inflammatory cells, especially eosinophils, and reduced the expression and secretion of Th2 cytokines, including IL-4 and IL-13, in lung tissues and bronchoalveolar lavage fluid. Histologic studies showed oroxylin A reduced inflammatory signs and mucin production in lungs. These findings provide evidence that oroxylin A has potential use as an anti-allergic therapeutic.

Effects of Amomum xanthiodes on the Mast Cell-Mediated Allergic Reaction (비만세포 유래의 알레르기 반응에 대한 사인의 효과)

  • The discovery of drugs for the treatment of mast cell-mediated allergic disease is a very important subject in human health. The Amomum xanthiodes (Zingiberaceae) has been used for centuries as a traditional medicine in Korea and is known to have an anti-inflammatory effect. However, its specific mechanism of action is still unknown. In this report, we investigated the effect of hot water extract from Amomum xanthiodes (EAX) on the mast cell-mediated allergic reaction and studied its possible mechanisms of action. EAX inhibited compound 48/80-induced systemic anaphylaxis and serum his­tamine release in mice. EAX decreased the passive cutaneous anaphylaxis reaction activated by anti-dinitrophenyl (DNP) IgE antibody. EAX dose-dependently reduced histamine release from rat peritoneal mast cells activated by compound 48/80 or anti-DNP IgE. EAX increased cAMP and decreased compound 48/80-induced intracellular $Ca^{2+}$ levels. Our findings provide evidence that EAX inhibits mast cell-derived allergic reactions, and also demonstrate the involvement of cAMP and intracellular $Ca^{2+}$ in these effects.

Anti-inflammatory effects of Hataedock with Coptidis Rhizoma and Glycyrrhiza Uralensis on Allergic Rhinitis through Regulating IL-4 Activation (알레르기성 비염에서 황련-감초 하태독법의 IL-4활성 조절을 통한 항염증효과)

  • The aim of this study is to evaluate the anti-inflammatory effect of Hataedock treatment using Coptidis Rhizome and Glycyrrhiza Uralensis (CG) mixed extract in allergic rhinitis induced NC/Nga mice. We divided NC/Nga mice into 3 groups as follows; allergic rhinitis-induced group after CG Hataedock treatment (CGT, n=10), no treatment group (Ctrl), allergic rhinitis elicited group (ARE). To induce allergic rhinitis, NC/Nga mice of 3 weeks age were sensitized on 7, 8 and 9week by Ovalbumin (OVA) antigen in intranasal space. Hataedock using CG extract was administered on week 3 in allergic rhinitis-induced group (CGT) after Hataedock treatment. To identify distribution of Interlukin (IL)-4, Cluster of differentiation 40 (CD40), high-affinity IgE receptor ($Fc{\varepsilon}RI$), substance P, Matrix metallopeptidase 9 (MMP-9), Nuclear $factor-{\kappa}B$ ($NF-{\kappa}B$) p65, Inducible nitric oxide synthase (iNOS) and Cycloxygenase-2 (COX-2), we used histological examination. CGT significantly inhibited IL-4 and CD40 response compared with ARE. The reduction of Th2 cytokine expression decreased inflammatory mediators such as $Fc{\varepsilon}RI$, substance P, MMP-9, $NF-{\kappa}B$ p65, iNOS and COX-2. Such immunological improvement induced reduction of respiratory epithelial damage and mucin secretion in goblet cell. These results indicate that Hataedock treatment suppresses allergic rhinitis through modulating of Th2 responses and diminishing various inflammatory mediators in nasal mucosal tissue. It might have potential applications for prevention and treatment of allergic rhinitis.
