Inhibitory Effect of Lycopus lucidus on Mast Cell-Mediated Immediate-Type Allergic Reactions (택란의 비만세포 매개 즉시형 알레르기 반응의 억제 효과)

  • The effect of aqueous extract of Lycopus lucidus Turcz. (Labiatae)(LLAE) on mast cell-mediated immediate-type allergic reactions was investigated. LLAE (0.01 to 1 mg/g) dose-dependently inhibited systemic anaphylaxis induced by compound 48/80. LLAE (0.001 to 1 mg/g) also dose-dependently inhibited local anaphylaxis activated by anti-dinitrophenyl (DNP) IgE. When LLAE was pretreated at the same concentration with systemic anaphylaxis, serum histamine levels were reduced in a dose-dependent manner. LLAE (0.001 to 1 mg/mι) dose-dependently inhibited histamine release from rat peritoneal mast cells (RPMC) activated by compound 48/80. The level of cAMP in human mast cell line (HMC-1) cells, when LLAE (1 mg/mι) was added, significantly was increased, compared with that of normal control. These results provide evidences that LLAE may be beneficial in the treatment of allergic diseases.

  • Immediate-type hypersensitivity is involved in many allergic diseases such as asthma, allergic rhinitis and anaphylaxis. The discovery of drugs for the treatment of allergic disease is an important subject in human health. Stimulation of mast cells releases inflammatory mediators, such as histamine and pro-inflammatory cytokines with immune regulatory properties. We investigated the effect of the aqueous extract of Schizonepeta tenuifolia (AEST) (Labiatae) on the immediate-type allergic reaction. AEST inhibited compound 48/80-induced systemic allergic reaction. AEST attenuated immunoglobulin E (IgE)-mediated skin allergic reaction and histamine release from human mast cell line (HMC-1) cells. In addition, AEST decreased the gene expression and secretion of pro-inflammatory cytokines in phorbol 12-myristate 13-acetate (PMA) plus calcium ionophore A23187 (A23187)-stimulated HMC-1 cells. Our results indicate that AEST inhibits the mast cell-derived allergic reactions and involvement of histamine and pro-inflammatory cytokines in these effects.

Cichorium Intybus inhibits mast cell-mediated immediate-type allergic reactions

  • We investigated the effect of aqueous extract of Cichorium intybus (CIAE) on mast cell-mediated immediate type allergic reactions. CIAE dose-dependently inhibited systemic anaphylactic reaction induced by compound 48/80 in mice. Especially, CIAE inhibited compound 48/80-induced anaphylactic reaction 100% with the dose of 1000 mg/kg. CIAE 1000 mg/kg also significantly inhibited local anaphylactic reaction activated by anti-dinitrophenyl (DNP) IgE. When mice were pretreated with CIAE at a concentration ranging from 0.1 to 1000 mg/kg, the plasma histamine levels were reduced in a dose-dependent manner. CIAE (1 to 1000 g/ml) dose-dependently inhibited histamine release from the rat peritoneal mast cells (RPMC) activated by compound 48/80 or anti-DNP IgE. These results indicate that CIAE inhibits mast cell-mediated immediate-type allergic reactions.

Inhibitory effect of Taemyeongcheong on allergic reactions

  • Taemyeongcheong (TMC) is Korean traditional extracted drink with various health ingredients. TMC has been used to treat hepatic damage, obesity, gastritis, and colitis. However, the role of TMC on allergic reaction has not been studied yet. In this study, we investigated the anti-allergic effects of TMC against a compound 48/80-induced systemic anaphylactic reaction and IgE-mediated passive cutaneous anaphylaxis (PCA). TMC significantly inhibited the compound 48/80-induced systemic anaphylactic reaction and IgE-mediated PCA reaction. Furthermore, TMC reduced the levels of interleukin (IL)-1β, IL-4, IL-13, and vascular endothelial growth factor in the serum of mice under PCA reaction. Taken together, these results suggest that TMC can play a useful role as an anti-allergic agent.

Anti-allergic Effects of Socheongyoug-tang on RBL-2H3 Mast Cell and Mice-mediated Allergy Model (RBL-2H3 비만세포와 동물병태모댈에서 소청룡탕(小靑龍湯)의 항알레르기 효과)

  • The discovery of drugs on the treatment of mast cell-mediated allergic disease is a very important subject in human heath. The Socheongyoug-tang(SCYT) has been used for centruries as a traditional medicine in Korea and is known to have an anti-inflammatory effect. However, its specific mechanism of action is still unknown. In this report, we investigated the effect of hot water extract from SCYT on RBL-2H3 mast cell-mediated allergic reaction and studied its possible mechanism of action. SCYT inhibited compound 48/80-induced systemic anaphylaxis and serum histamine release in mice. SCYT decreased the passive cutaneous anaphylaxis reaction activated by Anti-lgE antibody-HSA. SCYT dose-dependently reduced histamine release from mice peritoneal mast cells activated by Anti-lgE antibody-HSA. SCYT increased cAMP and decreased compound 48/80-induced intracellular $Ca^{2+}$ levels. Our findings provide evidence that SCYT inhibits mast-derived allergic reactions, and also demonstrate the involvement of cAMP and intracellular $Ca^{2+}$ in these effects.

The Effects of Herbal-acupuncture with Juglandis Semen on the Anti-allergic Reaction (호도약침(胡桃藥鍼)의 항(抗)알레르기 효과(效果)에 관한 연구(硏究))

  • Objectives : This experimental research has been done to study the effects of herbal-acupuncture with Juglandis Semen on the anti-allergic reaction. Methods : ICR mice were injected intraperioneally with compound 48/80 to induce the immediately type hypersensitivity (anaphylaxis). ICR mice were sensitized by PC and SRBC to induce the delayed type hypersensitivity. Sprage-Dawley rats were injected subcutaneously with egg albumin to measure the WBC. ICR mice and Sprage-Dawley rats were injected with Juglandis Semen herbal-acupuncture for the anti-allergic reaction test. Results : 1. In the restraint effect of systemic anaphylactic reaction, the mortality of mice decreased 20% each before 10 minutes and after 5 minutes. 2. In the contact dermatitis induced by PC, both Treat Ⅰand Treat Ⅱ group showed significant(p<0.01, p<0.05) decrease in comparison with the Control group, and also Treat Ⅲ group showed significance(p<0.005). 3. In the delayed type hypersensitivity in mice challenged by SRBC, Treat Ⅰ group showed significant(p<0.05) decrease in comparison with the Control group, and there was significance(p<0.05, p<0.05) in Treat Ⅲ and Treat Ⅳ group. 4. The change of the WBC in allergic rats induced by egg albumin showed significant(p<0.01, p<0.05, p<0.05) decrease in Treat Ⅰ, Treat Ⅱ and Treat Ⅲ group. Conclusions : The restraint effects of Juglandis Semen herbal-acupuncture on the anti-allergic reaction showed significance statistically. And also allergic group treated with $BL_{17}$ acupuncture (Treat Ⅲ group) showed same results.

4-CMTB Ameliorates Ovalbumin-Induced Allergic Asthma through FFA2 Activation in Mice

  • Free fatty acid receptor 2 (FFA2, also known as GPR43), a G-protein-coupled receptor, has been known to recognize short-chain fatty acids and regulate inflammatory responses. FFA2 gene deficiency exacerbated disease states in several models of inflammatory conditions including asthma. However, in vivo efficacy of FFA2 agonists has not been tested in allergic asthma. Thus, we investigated effect of 4-chloro-α-(1-methylethyl)-N-2-thiazoylylbenzeneacetanilide (4-CMTB), a FFA2 agonist, on antigen-induced degranulation in RBL-2H3 cells and ovalbumin-induced allergic asthma in BALB/c mice. Treatment of 4-CMTB inhibited the antigen-induced degranulation concentration-dependently. Administration of 4-CMTB decreased the immune cell numbers in the bronchoalveolar lavage fluid and suppressed the expression of inflammatory Th2 cytokines (IL-4, IL-5, and IL-13) in the lung tissues. Histological studies revealed that 4-CMTB suppressed mucin production and inflammation in the lungs. Thus, results proved that FFA2 functions to suppress allergic asthma, suggesting 4-CMTB activation of FFA2 as a therapeutic tool for allergic asthma.

Suppressive Effect of Arazyme on Neutrophil Apoptosis in Normal and Allergic Subjects

  • Arazyme is a metalloprotease secreted by Aranicola proteolyticus that was previously shown to suppress cytokine expression of keratinocytes and endothelial cells and inhibit histopathological features in an atopic dermatitis-like animal model. However, the regulatory effects of arazyme in other allergic diseases have yet to be elucidated. In this study, we investigated whether arazyme is effective against neutrophil apoptosis in allergic diseases such as allergic rhinitis and asthma. Arazyme inhibited neutrophil apoptosis of normal subjects in a dose-dependent manner. However, the antiapoptotic effect of arazyme was reversed by LY294002, an inhibitor of PI3K, AKTi, an inhibitor of Akt, PD98059, an inhibitor of MEK, and BAY-11-7085, an inhibitor of NF-${\kappa}B$. Arazyme induced activation of NF-${\kappa}B$ via PI3K/Akt/ERK pathway. The anti-apoptotic effect of arazyme is associated with inhibition of cleavage of caspase 3 and caspase 9. Arazyme inhibited constitutive apoptosis of neutrophil in a dose-dependent manner in allergic subjects, and its mechanism was shown to be associated with PI3K/Akt/ERK/NF-${\kappa}B$. The results presented here improve our understanding of neutrophil apoptosis regulation and will facilitate development of drugs for treatment of allergic diseases.

Effects of Seokgokamibokhapbang(SGBHB) on anti-CD40-and recombinant IL-4-induced cytokine production and immunoglobulin E and histanine release in highly purified mouse B cells (석고가미복합방이 Anti-CD40과 rIL-4로 유도된 생쥐의 B 세포에서 싸이토카인 생성 및 면역글로블린 E에 미치는 효과)

  • In the present study, we exarnined anti-allergic effect of SGBHB in cultured B cells. B cells were prepared from isolated murine splenocytes and activated by co-treatment of anti-CD40 monoclonal antibody and recombinant IL-4 allergens. Anti-allergic effects of SGBHB in activated B cells were determined by measuring B cell surface activated molecules (CD23+ and CD11a+), and expression levels of IL-$1{\beta}$, IL-6, IL-10, TNF-$\alpha$, IgE, and HRF. The major findings are summarized as follows. 1. SGBHB treatment did not produce significant cytotoxic effects on mouse lung fibroblast cells. 2. SGBHB produced significant inhibitory effect on the expression of B cell surface activated molecules (CD23+ and CD11a) in activated B cells. 3. SGBHB treatment significantly inhibited expression levels of IL-$1{\beta}$, IL-6, and TNF-$\alpha$ mRNAs in activated B cells.IL-6 protein levels were significantly decreased by $100{\mu}g/m{\ell}$ of SGBHB treatrrient, and TNF-$\alpha$ protein levels were decreased compared to the control group, but statistically insignificant. 4. SGBHB treatment significantly increased IL-10 at both mRNA and protein levels in activated B cells. 5. SGBHB treatment significantly inhibited levels of IgE production. Thus, the present data suggest that SGBHB has an anti-allergic effect on activated B cells by controlling irnmune responses, and further implicates the possibility on clinical application as a therapeutic agent.

Effect of Whitmania pigra whitman on the Allergic Inflammatory Response (수질(水蛭)이 비만세포의 알러지 염증 반응에 미치는 영향)

  • Objectives: Whitmania pigra whitman (WP) has been used in herbal medicine to treat various conditions, such as eczema, skin burns and frostbites in herbal medicine. The purpose of this study is was to investigate the effect of WP on anti-allergy mechanism. Methods: To clarify the mechanism, the effect of WP on vascular permeability of rat cutaneous tissue and histamine and cytokines (IL-6, IL-8, $TNF-{\alpha}$) released from mast cells were observed. Results: The results were 1. the pretreatment with WP significantly decreased the compound 48/80-induced degranulation and histamine release from RPMC 2. WP did not inhibit the anti-DNP IgE-induced increment of vascular permeability of rat cutaneous tissue 3. WP significantly reduced the PMA plus A23187-induced increment of expression of IL-6, IL-8, and $TNF-{\alpha}$ in HMC-1 cells. Conclusions: The present study providesevidence that WP acid inhibits mast cell-derived inflammatory allergic reactions by blocking histamine release and pro-inflammatory cytokine expression, and suggest the mechanisms of action. Furthermore, in vivo and in vitro anti-allergic effect of WP suggests a possible therapeutic application of this agent in inflammatory allergic diseases.
