Effects of Combination Therapy with Anti-Obesity Herbal Medicine Including Ephedra Herba and Lorcaserin in Obese Patients: Two Case Reports (마황이 포함된 한방비만 처방과 Lorcaserin 병용 투여 후 호전된 비만 환자 치험 2예)

  • Although there has been many studies about the co-administration of herb formula and western medicine, there is none about obesity. The aim of this study was to report the effects of co-administration of herb formula containing ephedra sinica and lorcaserin on two obese patients for about 2~4 months. During the treatment, we checked body weight and body composition, and collected blood samples to evaluate liver, kidney, thyroid function and lipid profile. This treatment not only decreased body weight but also improved body composition and lipid profile in both patients. There was no hepatic or renal toxicity, nor any serious side effects of treatment. This study demonstrated that co-administration of herb formula containing ephedra sinica and lorcaserin is a safe and effective therapy to lose weight and improve metabolic parameters for obese patients. Further large-scale clinical trials are needed to evaluate the anti-obesity effect of the combination of herbal and western medicines.

Anti-Obesity Effect of a New Dietary Supplement Consisting of Hydroxycitrate, Carnitine and Red Pepper (3D-Relax Diet) in High-Fat Fed Rats

  • Anti-obesity effect of a new dietary supplement (3D-relax) in high-fat fed rats. The aim of this study was to assess the effects of 3D-relax; a proprietary formulation containing hydroxycitrate (233 mg/g), carnitine (150 mg/g) and red pepper (150 mg/g); on body weight, body fat, and serum lipids levels in rats fed a high-fat diet. Male SD 7-wk-old rats (n=8) were fed a high fat diet [52% total dietary energy (E%) from fat, 15.4 E% protein, 32.6E% carbohydrate] with or without 3D-relax administration (1 g/kg body weight/day) for 3 weeks. Administration of 3D-relax significantly reduced the increase in body weight compared to the group fed high fat without 3D-relax. Food efficiency ratio (FER) tended to be decreased with administration of 3D-relax, but was not significant. The perirenal and epididymal fat pad weights of vats administered 3D-relax were significantly lower than those of the high fat group that did not ingest 3D-relax during the 3 weeks. The oral administration of 3D-relax significantly increased HDL-cholesterol level and lowered total cholesterol level compared to those of high fat alone group. These results suggest that 3D-relax reduced body weight and fat gains, and those effects are presumably linked to its inhibitory effects on lipogenesis.

Anti-obesity Effect of Rhizoma Atractylodis Herbal Acupuncture in High Fat Diet-induced Obese ICR Mouse (고지방 식이로 유도된 비만 생쥐에서 창출약침의 항비만 효과)

  • Objectives : The aim of this study was to investigate the anti-obesity potential and mechanisms of action of Rhizoma Atractylodis(RA) herbal acupuncture in high fat diet- induced obese ICR mice. Methods : Sample solutions for herbal acupuncture were prepared from the Rhizoma Atractylodis water extract powder at concentration of 150mg/kg and 300mg/kg with distilled water. Five week-old ICR mice acclimatized to the laboratory environment for 1 week were allocated into four groups: regular diet group (RD), high fat diet group(HFD), groups fed HFD with 150mg/kg RA herbal acupuncture treatment (RAE 150) and with 300mg/kg RA herbal acupuncture treatment(RAE 300). Herbal acupuncture groups were injected with either 150mg/kg or 300mg/kg of Rhizoma Atractylodis(RA) subcutaneously onto both Sinsu($BL_{23}$) alternately on the same time everyday for 30days. Body weight, gross appearance of epididymal fat area, blood glucose, insulin, insulin resistance(HOMA-IR), non-esterified fatty acid, cholesterol, triglyceride, AST, ALT, histological analysis of white adipose tissue, gene expression responsible for adipocyte differentiation and AMPK activation were analyzed. Results : RA herbal acupuncture inhibited the development of weight gain, hyperglycemia, hyperinsulinemia, hyperlipidemia, increases of AST and ALT, and the enlargement of fat cell size induced by HFD. Also, RA herbal acupuncture inhibited the expression of PPAR-${\gamma}$, C/$EBP{\alpha}$, aP2, LPL, FAS, SCD-1 and enhanced the activation of AMP-activated protein kinase. Conclusions : The results of this study demonstrate that RA herbal acupuncture can exert the anti-obesity effect and it is partially mediated by activation of AMPK and inhibition of the gene expressions responsible for adipocyte differentiation. Further studies will be required to ascertain the nti-obesity effect and mechanisms of action of RA herbal acupuncture in animal models and human for aclinical application.

Effect of Herbal Acupuncture with Sang-hwang(Phellinus linteus) on High Fat Diet-induced Obesity in Rats (지방식이로 유도된 비만동물모델에서의 약침의 효과)

  • Acupuncture has fairly good weight-reducing effect in treating simple obesity due to the neuroendocrine regulation. In this study, the antiobesity effects of herbal acupuncture(HA) with Sang-hwang(phellinus linteus) at Fuai(SP16) were investigated in the rat fed on high-fat(HF) diet. Sang-hwang mushroom has been proven to have anti-carcinogenic effects and Sang-hwang extracts are highly effective in treatment and preventive treatment of AIDS, diabetes and high blood-pressure. To determine whether the Sang-hwang herbal acupuncture may have the anti-obesity effect, male Sprague-Dawley(4-wk-old) rats were fed a HF diet for 5 wk, which produced significant weight gain compared to rats were fed a normal diet, and then herbal acupuncture were treated for 3 wk in HF diet group. The body weight, food consumption, food effeciency ratio(FER), body fat mass, plasma nitric oxide(NO) were investigated in rats fed on normal diet, HF diet, and HF diet with HA(HF-diet-HA) groups. NO has been proposed to be involved in the regulation of food intake. In addition, the expression of appetite peptides such as orexigenic peptide neuropeptide Y(NPY) and the anorectic peptide cholecystokinin(CCK) were observed in the hypothalamus. HF-HA group reduced body weight gain, FER, body fat contents and NO concentration compared to HF diet group. The expression of NPY was reduced in arcuate nucleus(ARC), and CCK was increased in the paraventricular nucleus(PVN) after treatment of HA. In conclusion, Sang-hwang HA reduced adipocity, plasma NO and hypothalamic NPY, but increased CCK expression in the HF dietinduced obesity rat, therefore HA may have anti-obesity action through regulating body weight and appetite peptide of the central nervous system.

The Effects in Metabolism and Adipose Tissue Inflammation Induced by the Massa Medicata Fermentata on Obese Type 2 Diabetes Mouse Model (신국(神麯) 투여가 비만형 제 2형 당뇨병 동물모델의 대사인자와 지방조직 염증반응에 미치는 영향)

  • Objectives: Recent data have revealed that the plasma concentration of inflammatory mediators is increased in the insulin-resistant states of obesity and type 2 diabetes. The purpose of this study was to investigate the antidiabetic and anti-obesity effect of Massa Medicata Fermentata on obese type 2 diabetes mice. Methods: In order to examine the effects of Massa Medicata Fermentata, obese type 2 diabetes mice induced by Surwit's high fat, high sucrose diet. Mice were divided into 4 groups of ND (normal diet), HFD (high fat and high sucrose diet), Met (high fat and high sucrose diet with metformin) and MMF (high fat and high sucrose diet with Massa Medicata Fermentata) and investigated over 8 weeks. Diabetic and obese clinical markers, including body weight, glucose level, lipid level, leptin concentration, epididymal fat pad and liver weights and adipose tissue macrophage (ATM) were determined. Results: Compared with the HFD group, body weight, fructosamine, triglyceride, epididymal fat pad weight and ATM were significantly reduced in the MMF group. Conclusions: From the above results, the intake of Massa Medicata Fermentata may be effective in anti-hyperglycemia and anti-obesity by the attenuation of glucose and lipid levels and also inflammation state. Massa Medicata Fermentata may be beneficial for controlling diabetes mellitus type 2 in humans.

The effects of genistein and exercise on obesity and adipose tissue inflammation in ovariectomized mice (난소절제 쥐에서 비만과 지방조직 염증에 대한 제니스테인과 운동의 영향)

  • This study investigated the effects of exercise on genistein-induced adipose tissue reduction and anti-inflammation of adipose tissue in ovariectomized mice fed a high-fat diet. Compared to mice fed a high-fat diet (C), both genistein (G) or exercise only (Ex) not only decreased adipose tissue weight and adipocyte size, but also glucose and free fatty acid levels in serum, which were more effectively reduced when genistein and exercise were treated simultaneously (G/Ex). The expression of lipogenesis and inflammatory cytokine genes of both G and Ex in adipose tissue was decreased compared to C. In particular, the decrease of these genes expression in G/Ex was more effective than each treatment alone. Thus, this study revealed that simultaneous treatment of genistein and exercise resulted in more effective improvement of obesity and adipose tissue inflammation than genistein single treatment in ovariectomized mice fed a high-fat diet, and it is a result of positive regulation of lipogenesis genes. This study suggests that the exercise has a beneficial effect on anti-obesity and adipose tissue anti-inflammation of genistein.

Anti-Obesity Effects of Red Onions Extract (적양파 추출물의 항비만 활성)

  • Obesity is known as a metabolic disease caused by abnormal differentiation of fat tissue due to an imbalance between energy intake and consumption.. The purpose of this study was to confirm the changes in the genes associated with pancreatic lipase activity and pre-adipocyte cell differentiation by treatment of red onion extract treatment. The effect of red onion extract treatment on pre-adipocyte differentiation was evaluated using 3T3-L1 adipocytes, and the activity of related genes was confirmed through Real-Time PCR. As a result of the experiment, the red onion extract inhibit pancreatic lipidase activity by concentration dependent manner. In addition, it was found to inhibit adipocyte differentiation and inhibit the activity of genes(C/EBP-α, C/EBP-β, PPAR-γ) associated with adipocyte differentiation. Through the results of this experiment, it is suggested that the red onion extract can be developed as a high potential material with anti-obesity efficacy by suppressing adipocytic differentiation by controlling genes related to adipocyte differentiation.

Anti-Obesity Effects of Menthae Herba Hydrosol on High-Fat Diet Induced Obese Mice (고지방 식이로 유도된 비만 생쥐에서 박하 Hydrosol의 항비만 효과)

  • Objectives We investigated anti-obesity effects of Menthae Herba hydrosol in obese mice. Methods Animals were divided into four groups, and treatments were performed for 7 weeks. After the treatment, serum lipid profiles, weight and pathological morphology in liver, kidney, adipose tissue were measured. Also, hepatic protein and gene expression levels of lipid metabolism-related factors were analyzed. Results Body weight was decreased in P3% group. In P1% (group fed high-fat diet and 1% Menthae Herba hydrosol) and P3% (group fed high-fat diet and 3% Menthae Herba hydrosol) group, weight of white adipose tissue, serum levels of triglyceride and blood urea nitrogen were decreased, and weight of muscle was increased. Also, liver, kidney and epididymal adipocyte size were reduced in P1% and P3% group. Adenosine monophosphate-activated protein kinase was increased and sterol regulatory element binding protein-1c (SREBP-1c) was decreased in P3% group. mPeroxisome proliferator-activator receptor-γ, mMonocyte chemotactic protein-1 were decreased in P1% and P3% group. In P3% group, mSREBP-1c was decreased and mCarnitine palmitoyl transferase-1 was increased. And mUncoupling protein 1 in brown adipose tissue was increased. Conclusions These results suggest that Menthae Herba hydrosol has a worthy effect on anti-obesity.

Antioxidant activity and anti-obesity effect of Coprinus comatus in Zucker rat (fa/fa) (먹물버섯(Coprinus comatus )의 항산화 활성 및 Zucker rat에 대한 항비만 효과)

  • The aim of the study was to investigate the anti-obesity effect of Coprinus comatus (CC) in high-fat diet-fed Zucker rat (fa/fa). Obesity was induced by feeding on high-fat diet (HFD) containing 60% kcal fat for 10 weeks, in which CC extracts were administrated through the gastrointestinal tract at a concentration of 200 mg/kg BW/day for 10 weeks. The total polyphenol and flavonoid contents of CC extracts were found to be $18.5{\pm}1.1mg$ of catechin equivalent/g, and $5.24{\pm}0.54mg$ of quercetin equivalent/g extract, respectively. The DPPH, ABTS, and hydroxyl radical scavenging activities of CC extracts were 15.34 %, 17.25%, and 16.18%, respectively. In animal study, CC administration significantly reduced the body weight, while there were no significant differences in the daily food intake between the HFD-fed Zucker rats and HFD plus CC-fed rats. CC treatment decreased epididymal and perirenal fat weights in HFD-fed Zucker rats. Significant decreases in the levels of triglyceride and total cholesterol in the serum and liver were observed in the CC-treated group compared with HFD-fed Zucker rats. Serum HDL-cholesterol levels in the CC-treated group were increased compared with the HFD-fed groups. Serum AST and ALT activities in the CC group were significantly lower than those of the HFD-fed group. Taken together, these data demonstrated that CC has potential in preventing high-fat diet induced obesity and is a good candidate for an anti-obesity agent.

  • Objectives : This study wad designed to elucidate the short term effect of Rossa rugosae Radix on proliferation, differentiation and maturation of 3T3-L1 Preadipocyte. Methods : 3T3-L1 preadipocytes obtained from Korean Cell line Bank were cultured in a Dulbecco's modified eagle medium(DMED) culture colution containing 10% fetal bovine serum(FBS) and various concentration of aqueous extract of Rossa rugosae Radix on proliferation, differentiation and maturation of 3T3-L1 preadipocytes were investigate after treatment for 24 hours b measuring MTT, Oil Red O and latate dehydrogenase activity.. Results : The Rossa rugosae Radix extract inhibited significantly the proliferation of 3T3-L1 preadipocytes and tended to increase latate dehydrogenase activity in the media of differentiated 3T3-L1 preadipocytes & matured 3T3-L1 preadipocytes. the extract also inhibit the lipid accumulation of differentiated and maturaion of 3T3-L1 preadipocytes, suggesting that Rossa rugosae Radix has anti-obesity effect: however further in vivo study is needed to demonstrate its pharmacological effects.
