5종 방제의 항비만 효과 비교 (The Anti-obesity Effect of 5 Herbal Formulas in High Fat Diet Induced Obese Mice)

  • Objectives : Samhwangsasim-tang (SST), Hwangryeonhaedok-tang (HHT), Ukgan-san (UGS), Onjunghwadam-hwan (OHH) and Samul-tang (SMT) have been used for the treatment of various diseases. This study was performed to compare the anti-obesity effects of 5 herbal formulas in high fat diet-(HFD) induced obese mice. Methods : The mice were randomly divided into seven groups that were fed a normal diet (ND), a HFD, a HFD plus SST (HFD + SST), a HFD plus HHT (HFD + HHT), a HFD plus UGS (HFD + UGS), a HFD plus OHH (HFD + OHH), or HFD plus SMT (HFD + SMT) at 300 mg/kg/day for 7 weeks. All groups were assayed for body weights, food efficiency ratio (FER), final liver and fat weight and blood biochemical parameters. Results : The increased body weights, food efficiency ratio (FER), and serum total triglyceride were decreased in HFD + OHH group relative to the same measurements in HFD group. Furthermore, the HFD + SST group significantly reduced FER, liver and abdominal subcutaneous fat weight gains, and serum total triglyceride, whereas HDL-cholesterol level was increased compared to HFD group. Conclusions : These results suggested that HFD + OHH and HFD + SST exert anti-obesity effects in HFD-induced obese mice.

제주 자생 식물들에 대한 항비만 효능에 대한 연구 (The Study on Anti-obesity Activity of the Wild Plants of Jeju Island)

  • 본 연구에서는 제주도에서 자생하는 30여 종의 식물추출물에 대한 항비만 효능을 조사하였다. 항비만 효능은 in vitro oil red-O staining 방법을 이용하여 지방전구세포 3T3-L1에서 분화억제력을 측정하였다. 그 결과, 30여 종의 식물 중에서 약도라지, 호장근, 유근피 등을 포함한 6종의 식물이 지방전구세포 3T3-L1의 분화를 억제하였다. 지방전구세포의 분화를 억제하는 물질 중 유근피, 약쑥, 호장근, 후박 4종은 우수한 항산화 효능도 동시에 가지고 있음을 확인하였다. 이러한 결과를 통해, 본 실험에서 확보된 추출물이 항비만 물질로 사용될 수 있는 가능성을 확인하였다.

괭생이모자반 추출물의 리파아제 저해 활성 및 3T3-L1 지방전구세포 분화억제 효과 (Inhibition of Lipase Activity and Preadipocyte Differentiation in 3T3-L1 Cells Treated with Sargassum horneri Extract)

  • In this study, in order to evaluate the anti-obesity effect of sargassum horneri extract, the effects of the extract on lipase activity and preadipocyte differentiation in 3T3-L1 cells were investigated. S. horneri extract between 0.0 and 1.0 mg/mL showed no cytotoxicity and inhibited lipase activity by 68.1%. When S. horneri extract was utilized at levels of 0.25, 0.5, and 1.0 mg/mL in 3T3-L1 cells, preadipocytes differentiation decreased by 11.4, 19.7, and 25.6%, respectively, showing anti-obesity effects. In addition, after treatment with 1.0 mg/mL S. horneri extract, the mRNA expression levels of sterol regulatory element binding proteins-1c (SREBP-1c), peroxisome proliferator activated receptor-γ (PPAR-γ), CCAAT enhancer binding protein-α (CEBP-α), fatty acid synthase (FAS), and stearoyl-CoA desaturase1 (SCD1) in 3T3-L1 cells were significantly decreased (p < 0.05) by 65.2, 54.9, 50.0, 33.8, and 33.8% respectively. These results showed that S. horneri extract suppresses lipase activity and prophylactic preadipocyte differentiation in 3T3-L1, and thus can be used as an anti-obesity agent in functional foods and medicines.

  • 본 연구에서는 솔잎 추출물(PDL)의 항비만활성을 검증하기 위해, PDL이 3T3-L1에서 DMI/insulin에 의해 유도되는 과도한 지질 형성을 억제하는지 평가하였다. 본 결과를 미루어 볼 때, PDL은 지방세포의 adipogenesis과 관련된 PPARγ, CEBPα 그리고 perilipin-1의 발현을 억제하여 과도한 지질형성을 억제하는 것으로 판단되며, PDL은 향후 비만 예방 및 치료를 위한 잠재적 소재로 활용이 가능할 것으로 판단된다. 그러나 본 연구는 PDL이 항비만 활성이 있는지 검증한 초기 단계 연구로 추가적인 기전연구와 동물기반 전임상 항비만활성 검증이 필요하다.

감귤피 효소적 추출물의 지방세포에서의 항비만 효과 (Anti-obesity effects of an enzymatic extract of mandarin (Citrus unshiu) peel in 3T3-L1 adipocytes)

  • 본 연구는 감귤 가공공정 중에 발생하는 가공부산물인 감귤피의 산업적 이용성 및 유용성을 증대시키고자 효소적 추출법을 적용하여 추출물을 제조하고 항비만 활성을 측정하였다. 감귤피 효소적 추출물(MPCE)의 폴리페놀 함량은 이전의 연구에서 보고한 유기용매 추출물보다 높은 함량을 나타내었다. MPCE의 항비만 활성을 알아보기 위해 지방세포 분화억제에 미치는 영향을 확인한 결과 MPCE는 농도 의존적으로 지방세포의 분화 및 지방축적을 감소시켰으며 이는 세포독성에 의한 것이 아님을 세포생존율 측정을 통해 확인하였다. 또한 MPCE는 adipogenesis 관련 전사인자인 C/EBPα, SREBP-1, PPARγ, FABP4의 단백질 발현을 감소시켰다. 이러한 결과들을 통해 MPCE는 adipogenesis 관련 전사인자의 단백질 발현 억제하여 지방세포의 분화 및 지방축적을 억제하였으며, 이를 통해 항비만 효과를 나타낸다는 것을 확인하였다. 따라서 MPCE는 비만 예방 효능을 가진 기능성 식품 소재로서의 활용 가치가 있는 것으로 판단되며, 보다 명확한 항비만 효능을 확인하기 위해 추후 동물 실험 등에 대한 연구가 필요할 것으로 생각된다.

Rosa acicularis Leaves Exert Anti-obesity Activity through AMPK-dependent Lipolysis and Thermogenesis in Mouse Adipocytes, 3T3-L1 Cells

  • It has been reported that Rosa acicularis has anti-obesity activity by inhibiting the digestive lipase activity. However, there is a lack of clear in vitro studies regarding the anti-obesity activity of Rosa acicularis. Therefore, in this study, we aimed to verify the anti-obesity activity of Rosa acicularis leaves (RAL) and elucidate its mechanism of action in 3T3-L1 preadipocytes. RAL dose-dependently inhibited the accumulation of lipid droplets and triacylglycerol. RAL had no effect on cell proliferation and survival in undifferentiated 3T3-L1 cells, but it inhibited cell proliferation in differentiating 3T3-L1 cells. RAL increased ATGL, p-HSL, and HSL, and decreased perilipin-1 in differentiating 3T3-L1 cells. In addition, RAL reduced lipid droplet accumulation and increased free glycerol content in differentiated 3T3-L1 cells. RAL increased ATGL and HSL in differentiated 3T3-L1 cells. Also, RAL increased p-AMPK, PPARγ, UCP-1, and PGC-1α in differentiating 3T3-L1 cells. AMPK inhibition by Compound C attenuated RAL-mediated increase of ATGL, HSL, PPARγ, and UCP-1 in 3T3-L1 cells. Taken together, it is thought that RAL may inhibit lipid accumulation through lipolysis and thermogenesis via the activation of AMPK in adipocytes.

알로에 베라 유래 식이섬유의 In Vitro 및 In Vivo 생리기능 특성 (In Vitro and In Vivo Physiological Characteristics of Dietary Fiber from By-product of Aloe vera Gel Processing)

  • A fiber fraction (Aloe cellulose), the by-product obtained from Aloe vera gel processing was freeze dried and investigated for in vitro glucose/ bile acid retarding effects of powdered sample (100 mesh) comparing with commercial $\alpha$-cellulose as a reference sample. We also examined the effectiveness of physiological functionality such as the antiobesity and anti-constipation on Sprague-Dawley (SD) rat. The Aloe cellulose powders during in vitro dialysis experiment for 2 hours exhibited the glucose and bile acid retarding index of 20.32-35.2% and 53.13-28.30%, respectively. Especially, freeze dried aloe cellulose showed the 2.5 and 1.2-6 times higher effect on in vitro glucose and bile acid retardation than those of $\alpha$-cellulose. These relatively good retarding effects on glucose and bile acid diffusion suggest a potential of preventing from diabetes and arteriosclerosis of some extent. Also, the results from animal experiments on SD rats fed a high-fat diet for 4 weeks suggested that Aloe cellulose might be used as a novel dietary fiber showing an effective anti-obesity and anti-constipation effect.

인진쑥 올리고당의 섭취가 비만쥐의 혈중 지질, 복강내 지방조직과 렙틴 수치에 미치는 영향 (Effects of Artemisia Iwayomogi Oligosaccharide on the Blood Lipids, Abdominal Adipose Tissues and Leptin Levels in the Obese Rats)

  • As obesity is known to be related to hyperlipidemia, insulin and leptin resistance, and other chronic diseases, much recent research has focused on functional food materials and their anti-obesity activity. This study was performed to study the effects of Artemisia Iwayomogi oligosaccharide AIPI on the anti-obesity function in normal rats and diet-induced obese (DIO) rats. F344 male rats were divided into four groups: Normal-control (CONT), normal-AIPI, DIO-CONT and DIO-AIPI. The groups were provided with water (in the CONT groups) or another drink for 4 weeks. The final body weights of rats in the DIO-AIPI group were lower than those in the CONT group. Abdominal adipose tissue weight per kg of body weight in the DIO-AIPI group was significantly lower than that in the DIO-CONT group. Also, the final levels of serum-triglyceride, serum-total cholesterol and serum-low density lipoprotein cholesterol in the DIO-AIPI group were lower than those in the DIO-CONT group. Moreover, the serum-high density lipoprotein cholesterol level in the normal-AIPI group was significantly higher than that in the normal-CONT group. Finally, the serum-leptin concentration was significantly lower in the DIO-AIPI group. Total lipid, triglyceride, and total cholesterol contents in the feces of the DIO-AIPI group were as high as 142%, 199%, and 165% of the respective values of the DIO-CONT GROUP. These results indicate that orally administered Artemisia Iwayomogi oligosaccharide not only has hypotriglycemic and hypocholesterolemic effects, but also has the effect of reducing the body weight and the abdominal adipose tissue weights obese rats. Therefore, we expect that Artemisia Iwayomogi oligosaccharide AIPI may have an anti-obesity function in F344 diet-induced obese rats. (Korean J Nutrition 36(5): 437∼445, 2003)

백출 및 발효백출의 장점막 투과성 개선 효과 및 항염증효과 (Effect of Unfermented and Fermented Atractylodes macrocephalae on Gut Permeability and Lipopolysaccharide-Induced Inflammation)

  • Objectives: The aim of this study is to investigate anti-imflammatory and protective effect for intestinal epithelial cells with Atractylodes macrocephae (AM), a traditional Korean Herbal medicine and fermented Atractylodes macrocephae (FAM) with Lactobacillus plantarum. Methods: HCT-116 and Raw 264.7 cells were used in this study. Using NO assay, we measured lipopolysaccharide (LPS)-induced anti-inflammatory effect. We measured permeability of intestinal epithelial cells with transepithelial electrical resistance and horseradish peroxide flux assay. Water soluble tetrazolium salt assay was used to see cell proliferation. All the results were presented in mean and standard deviation. We used Student's t-test for analyzing significance of results. Results: In Raw 264.7 cells NO production decreased 22.4% with pre-treatment of AM and FAM, especially with FAM in high concentration. In HCT-116 cells LPS-induced intestinal permeability had a protective effect with both AM and FAM, which was also tend to be proportional to the concentration. Cell viability increased up to 135.52% after treatment of high concentration of FAM in HCT-116, while there was no significant change in Raw 264.7 cells with herb treatments. Conclusions: These results show evidence that AM, especially fermented ones, significantly reduced intestinal membrane permeability. They also had a protective effect as well as an anti-inflammation effect for HCT-116 and Raw 264.7 cells. This suggest that FAM may be a therapeutic agent for Leaky gut syndrome by reducing intestinal permeability.

발효울금 50% 에탄올 추출물의 라디칼 소거능 및 지방형성 억제 효과 (Radical Scavenging and Anti-Obesity Effects of 50% Ethanol Extract from Fermented Curcuma longa L.)

  • 본 연구에서는 FCE50을 대상으로 다양한 항산화 실험을 실시한 결과, FCE50이 항산화능을 보유하고 있음을 확인하였다. 또한 FCE50은 3T3-L1 전지방세포의 분화과정 중 세포내 지방축적을 유의적으로 감소시키며 포도당 유입을 감소시켰다. 지방합성과 관련된 ACC 효소의 mRNA 발현량을 측정한 결과, 완전히 분화된 지방세포와 비교하여 FCE50의 처리 시 그 발현량이 현저히 감소됨을 확인하였다. 이상의 결과로부터 FCE50은 항산화 활성을 보유하고 있으며, 이는 FCE50이 지방세포로의 분화과정 중 지방형성을 유의적으로 감소시키는 결과에 영향을 미치는 것으로 사료된다.
