채취시기별 보검선인장 줄기의 항산화, 항당뇨 및 항알츠하이머 활성평가 (Antioxidant and Anti-Diabetic, Anti-Alzheimer Activities of Stem from Opuntia ficus-indica var. saboten Cultivated in Jeju at Harvest Time)

  • In this study, we assessed antioxidant, anti-diabetic, and anti-Alzheimer activities of Opuntia ficus-indica var. saboten (OFI) at harvest time. OFIs were cultivated December 2015~November 2016 in Jeju island. The 70% ethanol extracts of OFI were used to investigate total polyphenol and flavonoid contents, antioxidant(DPPH and ABTS radical scavenging assay), anti-diabetic(yeast ${\alpha}$-glucosidase and rat ${\alpha}$-glucosidase inhibition assay), and anti-Alzheimer(Acetylcholinesterase and butyrylcholinesterase inhibition assay) activities. Total polyphenol and flavonoid contents of OFIs were $17.40{\sim}23.11{\mu}g$ garlic acid/mg Ex and 2.17~6.22 ug (+)-catechine/mg Ex, respectively. DPPH and ABTS radical scavenging activity of OFIs were 131.98~184.90 mg ascorbic acid(AA) eq/100 g and 63.60~101.83 mg AA eq/100 g, respectively. In the anti-diabetic and anti-Alzheimer activities, 70% ethanol extracts of OFI exhibited moderate inhibition activity, compared to control (acarbose and beberine). Total polyphenol and flavonoid contents, antioxidant, anti-diabetic, and anti-Alzheimer activities were no significant differences by season, respectively. Therefore, information on comparative biological evaluations of OFI may be a beneficial in exploring functional food and drug development.

조구등이 $\beta$APP 과발현 인간 신경아세포암에서의 항치매 효과에 관한 연구 (Study of Repair Effect of Anti-Alzheimer on $\beta$APP Overexpression In Neuroblastoma cell line by Ramulus et Uncus Uncariae)

  • Ramulus et Uncus Uncariae (JGD) has sweet in flavour and slightly cold in property, acting on the liver and pericardium channels. This drug was described in a medical classic as having the ability to remove 'heat', check hyperfunction of the liver and relieve dizziness, tremors, and convulsions, and subdue 'endogenous wind'. So this study was estimated to check the anti-neuropathological effect of JGD on the Alzheimer in βAPP overexpression in neuroblastoma cell line and JGD extract was showed significantly anti-alzheimer effects (50 and 100 μg/㎖ of JGD extracts) compared with control group. Ramulus et Uncus Uncariae has anti-alzheimer effects on the βAPP overexpression in neuroblastoma cell line. So we expect that Ramulus et Uncus Uncariae may be used as a drug for neurodegenerative disease, such as stroke, Alzheimer's disease (AD). These results indicate that Ramulus et Uncus Uncariae possess strong inhibitory effect in the nervous system of apoptosis and repair effect against the degeneration of Neuroblastoma cells by βAPP expression.

Optogenetic neuromodulation with gamma oscillation as a new strategy for Alzheimer disease: a narrative review

  • The amyloid hypothesis has been considered a major explanation of the pathogenesis of Alzheimer disease. However, failure of phase III clinical trials with anti-amyloid-beta monoclonal antibodies reveals the need for other therapeutic approaches to treat Alzheimer disease. Compared to its relatively short history, optogenetics has developed considerably. The expression of microbial opsins in cells using genetic engineering allows specific control of cell signals or molecules. The application of optogenetics to Alzheimer disease research or clinical approaches is increasing. When applied with gamma entrainment, optogenetic neuromodulation can improve Alzheimer disease symptoms. Although safety problems exist with optogenetics such as the use of viral vectors, this technique has great potential for use in Alzheimer disease. In this paper, we review the historical applications of optogenetic neuromodulation with gamma entrainment to investigate the mechanisms involved in Alzheimer disease and potential therapeutic strategies.

민박쥐나물 추출물의 Caffeoylquinic Acid 정량 및 Acetylcholinesterase 활성과 Peroxynitrite 농도에 대한 효과 (Quantification of Caffeoylquinic Acids and the Effect of the Cacalia hastata var. orientalis Extract on Acetylcholinesterase Activity and Peroxynitrite Concentration)

  • To find the anti-Alzheimer's activity of Cacalia hastata var. orientalis (Compositae), caffeoylquinic acids were analyzed in this plant by HPLC and the inhibitory activities of the extract on cholinesterases including acetylcholinesterase (AChE) and butyrylcholinesterse (BChE) and peroxynitrite ($ONOO^-$) were assayed. Sum of caffeoylquinic acids in the 80% MeOH extract was quantitatively higher in leaves (35.8% of the extract) than in stems (3.7%). The compound, 3,5-dicaffeoylquinic acid, was contained in the highest amount in the leaf (44.32 mg/g dry weight). The $IC_{50}s$ of the 80% MeOH extract were shown to be $67.45{\mu}g/ml$ for AChE, and $8.59{\mu}g/ml$ (for $ONOO^-$), respectively, suggesting that the leaves would have the anti-Alzheimer's activity due to the high content of caffeoylquinic acids.

Panax Ginseng in the treatment of Alzheimer's disease and vascular dementia

  • Dementia has become one of the most important diseases threatening human health. Alzheimer's disease (AD) and vascular dementia (VaD) have the highest incidence rates among the types of dementia, but until now, therapeutic methods have been limited. Panax ginseng has been used in China for thousands of years to treat dementia, and modern medical studies have found that it contains multiple active components, such as ginsenosides, polysaccharides, amino acids, volatile oils and polyacetylenes, many of which have therapeutic effects in treating AD and VaD. Studies have found that ginsenosides have multitarget therapeutic effects in treating dementia, such as regulation of synaptic plasticity and the cholinergic system, inhibition of Aβ aggravation and tau hyperphosphorylation, anti-neuroinflammation, anti-oxidation effects and anti-apoptosis effects. Other active components of Panax ginseng, such as gintonin, oligosaccharides, polysaccharides and ginseng proteins, also have therapeutic effects on AD and VaD. The effectiveness of ginseng-containing Chinese medicine compounds has also been confirmed by clinical and basic investigations in treating AD and VaD. In this review, we summarized the potential therapeutic effects and related mechanisms of Panax ginseng in treating AD and VaD to provide some examples for further studies.

단풍취 추출물의 Acetylcholinesterase 저해 및 Peroxynitrite 소거에 대한 활성과 Caffeoylquinic Acid 성분의 분석 (Effect of Ainsliaea acerifolia Extract on Anti-acetylcholinesterase and Peroxynitrite Scavenging and Analysis of Caffeoylquinic Acids)

  • 국화과에 속하는 단풍취의 추출물이 가지는 알츠하이머에 대한 효과를 알기 위하여 AChE에 대한 억제효과와 peroxynitrite 소거효과를 연구하였다. 단풍취 추출물은 AChE에 대한 억제효과로서 그 $IC_{50}$$73.4{\mu}g/ml$이었으며 peroxynitrite 소거효과에 관한 $IC_{50}$$8.60{\mu}g/mL$이었으므로 알츠하이머 질환에 유익할 것으로 판단된다. 단풍취에 가장 많이 함유된 성분인 3,5-dicaffeoylquinic acid 성분의 활성도 이와 유사한 활성을 보였으므로 단풍취 추출물의 효과는 주로 이 화합물의 활성에 따른 것으로 예상된다. Caffeoylquinic acid가 알츠하이머 질환에 효과있다는 보고가 있으므로 이 식물의 잎과 줄기 추출물에서 6종의 지표물질을 이용하여 정량법을 확립한 후 정량하였다. 그 결과 잎에서는 성분 함량이 25.1%를 차지하여 줄기의 함량보다 훨씬 높았다. 특히 잎 추출물 중 3,5-dicaffeoylquinic acid의 함량이 145.6 mg/g에 달하여 알츠하이머 질환 치료에 유익할 것으로 예상된다.

A Comparison between Extract Products of Magnolia officinalis on Memory Impairment and Amyloidogenesis in a Transgenic Mouse Model of Alzheimer's Disease

  • The components of Magnolia officinalis have well known to act anti-inflammatory, anti-oxidative and neuroprotective activities. These efficacies have been sold many products as nutritional supplement extracted from bark of Magnolia officinalis. Thus, to assess and compare neuroprotective effect in the nutritional supplement (Magnolia $Extract^{TM}$, Health Freedom Nutrition LLC, USA) and our ethanol extract of Magnolia officinalis (BioLand LTD, Korea), we investigated memorial improving and anti-Alzheimer's disease effects of extract products of Magnolia officinalis in a transgenic AD mice model. Oral pretreatment of two extract products of Magnolia officinalis (10 mg/kg/day in 0.05% ethanol) into drinking water for 3 months ameliorated memorial dysfunction and prevented $A{\beta}$ accumulation in the brain of Tg2576 mice. In addition, extract products of Magnolia officinalis also decreased expression of ${\beta}$-site APP cleaving enzyme 1 (BACE1), amyloid precursor protein (APP) and its product, C99. Although both two extract products of Magnolia officinalis could show preventive effect of memorial dysfunction and $A{\beta}$ accumulation, our ethanol extract of Magnolia officinalis (BioLand LTD, Korea) could be more effective than Magnolia $Extract^{TM}$ (Health Freedom Nutrition LLC, USA). Therefore, our results showed that extract products of Magnolia officinalis were effective for prevention and treatment of AD through memorial improving and anti-amyloidogenic effects via down-regulating ${\beta}$-secretase activity, and neuroprotective efficacy of Magnolia extracts could be differed by cultivating area and manufacturing methods.

  • A series of tetrahydroaminoacridine and their analogues were synthesized. Tetrahydroaminoacridine(tacrine) is an anticholinesterase agent used in the treatment of Alzheimer's disease. Introduction of piperidine group at the para position enhanced anti-inflamatory activity for Alzheimer's disease. We investigated their ability to inhibit cyclooxygenase-1 and 2 isoforms. (omitted)

Diagnosis of Alzheimer's disease through measurement of anti-beta amyloid antibody in patient's serum

  • Amyloid beta (A$\beta$) is believed one of the major pathogens of Alzheimer's disease (AD), and the reduction of A$\beta$ is considered a primary therapeutic target. Immunization with A$\beta$ can reduce A$\beta$ burden and pathological features in transgenic AD model mouse. This means anti-A$\beta$ autoantibodies may have a role in AD pathology. Recent findings suggest anti-A$\beta$ autoantibodies level decrease in AD patients. The early detection of AD is important for treatment of this disease. However, diagnosis on AD has only been possible through limited methods such as neuropsychological examination or MRI. To investigate whether it was possible to detect the presence and different levels of naturally occurring anti-A$\beta$ autoantibodies in the plasma of patients with AD and age-matched controls. An advanced ELISA was performed to detect levels of naturally occurring anti-A$\beta$ autoantibodies in the plasma. The level of anti-A$\beta$ auto-antibodies from patients with idiopathic Parkinson's disease or stroke and from normal controls were compared to that of AD patients. Our results showed a significantly lower anti-A$\beta$ autoantibodies level in AD compared to those with other neurological diseases or control. The level of anti-A$\beta$ autoantibodies in the serum may be used to diagnose the presence of AD.
