Effect of Cosurfactants on the Release Behavior of Zaltoprofen-loaded PLGA Microspheres in In Vitro : Preparation and Characterization (보조계면활성제 첨가에 따른 잘토프로펜을 함유한 PLGA 미립구의 생체외 방출 거동: 제조 및 특성)

  • Zaltoprofen, a propionic acid derivative non-steroidal anti-inflammatory drug (NSAID), is known to have powerful inhibitory effects on acute, subacute and chronic inflammation. We developed poly(lactide-co-glycolide)(PLGA) microspheres loaded with zaltoprofen for sustained controlled delivery using an oil-water solvent evaporation methods by varying PLGA molecular weight and cosurfactant contents. Physicochemical properties and morphology of zaltoprofen-loaded PLGA microspheres were investigated by scanning electron microscope, X-ray diffraction and differential scanning calorimeter. The size of microspheres increased with the molecular weight of PLGA and the content of cosurfactants. The increase of PLGA molecular weight and cosurfactant content decreased the porosity of microspheres, subsequently resulting in the slow drug release. The results demonstrated that the adjustment of PLGA molecular weight and the cosurfactant content allowed us to control the drug release profiles of drug-loaded microspheres.

The Effect of Lonicera Japonica Thunberg on Inflammatory Factor Expression Associated with Atherosclerosis (금은화가 HUVEC에서 죽상동맥경화증 관련 염증인자 발현 억제에 미치는 영향)

  • Objective: The purpose of this study was to investigate the effect of Lonicera Japonica Thunberg (LJT) on the inflammatory factor expression associated with atherosclerosis in human umbilical vein endothelial cells (HUVECs). Methods: After treatment with LJT in HUVEC which is treated with TNF-α, we measured the expression levels of biomarkers (MCP-1, ICAM-1, VCAM-1, KLF2, and eNOS), mRNA (CCL2, ICAM1, VCAM1, KLF2, and NOS3), and the proteins (MCP-1, ICAM-1, VCAM-1, KLF2, eNOS, ERK, JNK, and p38). Results: 1. Compared to the control, LJT significantly reduced MCP-1 and VCAM-1 levels at concentrations of 100, 200, and 400 ㎍/ml and ICAM-1 expression at 200 and 400 ㎍/ml compared to the control. It increased KLF2 levels at all three concentrations, but not significantly, while eNOS expression was significantly increased at 400 ㎍/ml. 2. LJT was seen to significantly reduce the expression of CCL2, ICAM1, and VCAM1 mRNA at concentrations of 100, 200, and 400 ㎍/ml compared to the control. In contrast, significantly increased KLF2 and NOS3 mRNA levels were observed at 400 ㎍/ml and at 200 and 400 ㎍/ml, respectively. 3. Compared to the control, LJT significantly reduced the protein expression of MCP-1 and VCAM-1 at 200 and 400 ㎍/ml and of ICAM-1 at 400 ㎍/ml. In addition, it increased both KLF2 and eNOS protein levels at 200 and 400 ㎍/ml. Although LJT did not have an effect on ERK expression in comparison with the control, it significantly reduced JNK levels at 200 and 400 ㎍/ml and p38 levels at 400 ㎍/ml. Conclusions: These results suggest that LJT has an effect on the inhibition of inflammatory factor expression associated with atherosclerosis in HUVECs which could contribute to the prevention of cardiovascular and cerebrovascular diseases.

Suppression of reactive oxygen species generation as a part of antioxidative effect of plant extracts (식물추출물 항산화효능 기전의 일부로서의 활성산소 발생 억제 효과)

  • Chemical scavenging of reactive oxygen species (ROS) is considered a major mechanism of antioxidant effects, but preventing ROS generation can be more efficient in attenuating oxidative damage. In this study, the extracts of plants, Solanum lycopersicum, Ailanthus altissima, Equisetum arvense, and Oenothera biennis, were tested to determine whether their antioxidative effects are driven by the prevention of superoxide generation from mitochondria, a major ROS generator. While all the extracts efficiently attenuated the elevation of ROS levels in human fibroblasts and inflammation-induced mice, those of S. lycopersicum, A. altissima, and O. laciniata only suppressed mitochondrial ROS generation and reduced levels of lipofuscin and lipid peroxidation. Furthermore, the extracts of A. altissima and O. laciniata extended the lifespan of fruit flies. Our results suggest that plant extracts with anti-oxidative effects differ in their ability to prevent ROS generation, which may be associated with the attenuation of oxidative damage in cells and animal tissues.

Improvement effect of cooked soybeans on HFD-deteriorated large intestinal health in rat model (쥐 모델에서 고지방사료로 악화된 대장 건강에 대한 콩의 개선 효과)

  • Obesity is associated with impaired intestinal epithelial barrier function, which contribute to host systemic inflammation and metabolic dysfunction. Korean traditional foods, fiber-rich bean products, have been various biological activities in anti-inflammatory responses, but has not reported the large intestinal health. In this study, we investigated the intestinal health promoting effect of cooked soybeans (CSB) on high fat diet (HFD)-induced obesity model. SD rat were fed either a HFD or HFD supplemented with 10.6% CSB (HFD+CSB) for animal experimental period. CSB treatment significantly decreased the HFD-induced weights of body and fat. Also, CSB treatment improved HFD-reduced tight junction components (ZO-1, Claudin-1, and Occludin-1) mRNA expression in large intestine tissue. Additionally, histopathological evaluation showed that CSB treatment attenuated the HFD-increased inflammatory cells infiltration and epithelial damages in large intestine tissue. At the genus level, effects of CSB supplement not yet clear, while dietary effects showed differential abundance of several genera including Lactobacillus, Duncaniella, and Alloprevotella. NMDS analysis showed significant microbial shifts by HFD, while CSB did not shift gut microbiota. CSB increased the abundance of the genera Anaerotignum, Enterococcus, Clostridium sensu stricto, and Escherichia/Shigella by linear discriminant analysis effect size analysis, while reduced the abundance of Longicatena and Ligilactobacillus. These findings indicate that CSB supplement improves HFD-deteriorated large intestinal health by the amelioration of tight junction component, while CSB did not shift gut microbiotas.

Current Status and Prospects for the Hemp Bioindustry (대마 생물산업의 현황과 전망)

  • Cannabis sativa L. belongs to the Cannabaceae family and is an annual herbaceous flowing plant. The plants can be classified into narcotic marijuana and nonnarcotic hemp. Different parts of C. sativa L. have been used as food, medicine, cosmetics, fiber and textile. However, the use of leaf, flower, and seed of C. sativa L was forbidden in Korea in January 1977 as a result of the Cannabis Control Act due to the narcotic properties. The plant's mature stems have limited uses for the production of fiber and sheets. Recently, various cannabinoids, terpenes and essential fatty acids were identified from C. sativa L., and their safety and useful bio-activities, such as neuroprotective, anti-inflammation, antithrombosis, antiepileptic, and antimicrobial activities, and the relief of pain, have been highlighted. Furthermore, the process of reduction of tetrahydrocannabinol, a representative narcotic compound, and the isolation of cannabidiol, a nonnarcotic active compound in C. sativa L., have been determined. These findings resulted in the legalization of C. sativa L. in Korea for medical use in December 2018 and the exclusion of C. sativa L. from the narcotic list of the UN Commission on Narcotics Drugs (UNCND) in December 2020. Therefore, developments of various high-value added products have commenced worldwide. Additionally, in 2021, the Korean government deregulated special zones based on hemp. In this study, the current status and the prospect of the hemp industry, as well as essential techniques for developing new hemp products, are provided for the activation of the Korea Green-Rush.

Effects of combined exercise on the blood inflammatory factors, DHEA-s and arterial stiffness of elderly women (복합운동이 고령여성의 혈중 염증인자, DHEA-s 및 동맥경직도에 미치는 영향)

  • The purpose of this study was to investigate the effects of combined exercise on blood inflammatory factors, DHEA-s and arterial stiffness in elderly women. The subjects were 42 elderly females volunteers, aged 70 to 85 years, composed of the combined exercise group (n=21) and control group (n=21). The 60 minute combined exercise program(aquarobics 1 time/week, strength exercise 2 times/week) was performed 3 times per week for 12 weeks, and the intensity was progressively increased every 4 weeks(1-4 weeks: RPE 12 to 13, 5-8 weeks: RPE 13 to 14, 9-12weeks: RPE 14 to 15). The test data were analyzed by two-way repeated measures ANOVA, paired t-test, independent t-test and the alpha level of .05 was set for all tests of significance. As a result, the combined exercise for 12 weeks reduced the inflammatory response of elderly women, and DHEA-s was found to have a positive effect on aging hormone. The arterial stiffness decreased the central arterial blood pressure (systolic and diastolic), prevented the increase of the pulse pressure and the wave increase index, and decreased the pulse wave velocity. These results suggest that regular and continuous combined exercise may be recommended for the healthy aging and longevity of elderly women by inducing anti-inflammation effect and improving the aging hormonal function and the vascular health.

Repression of CCSP Expression by KLF4 (KLF4에 의한 CCSP 발현 억제)

  • Clara cell secretory protein (CCSP) plays an important role in protecting the lungs from inflammation. This research focuses on identifying the cis-element for binding the repressor of CCSP gene expression. A DNase I footprinting experiment revealed three protected regions between -812 and -768 bp (45 bp) of the mCCSP promoter. One motif (D3: GCCTGGGAA) was 100% conserved across rat, hamster, and human. The addition of excess amounts of the D3 motif exhibited high competition within that 45 bp range in an electrophoretic mobility shift assay. However, when mutated D3 ($G{\underline{AA}}TG{\underline{TT}}AA$) was used, the competition was significantly reduced. This demonstrates that the D3 motif within that 45 bp region of the mCCSP promoter is an important site for the protein-DNA interaction. Transient transfection assays with -756 Luc resulted in highly decreased expression of CCSP than those with -812 Luc, suggesting that the 45 bp could function as a binding site for the repressor. Co-transfection of KLF4 exhibited significant repression of the -812 Luc but not the -768 Luc which clearly shows that KLF4 might function as a repressor for the CCSP gene and also suggests that the D3 motif is strongly involved in the binding of KLF4. In addition, when anti-KLF4 antibody was added, super-shifted bands were observed. This result demonstrates that KLF4 could function as a repressor by binding to this 45 bp region of the CCSP promoter and that the D3 motif might be involved in the specific binding of KLF4.

Protective Effects of Individual Extract and Mixture of Forsythia Fructus and Corydalis Tuber on Chronic Pancreatitis in Mice (마우스 만성 췌장염 모델에서 연교(連翹)와 현호색(玄胡索) 단일 추출물 및 추출 혼합물의 보호 효과)

  • Objectives : Forsythiae Fructus has been reported to have anti-inflammatory effects in many diseases, and Corydalis Tuber has been used as a pain suppressor in Eastern Asia. However, the protective effects of individual water extract of Forsythiae Fructus (FF) and Corydalis Tuber (CT) and the mixture of FF and CT (FC) on chronic pancreatitis (CP) were not well-investigated. Therefore, we investigated the protective effects of FF, CT, and FC on CP in mice. Methods : To induce CP, cerulein was injected 6 times a day, 4 times a week for 3 weeks. 1 h before the every cerulein injection, 200 mg/kg of FF, CT, or FC was intraperitoneally injected to mice. Histological analysis of pancreas was examined by hematoxylin and eosin stain and collagen deposition was examined by Masson's trichrome stain. Fibrogenic parameters such as α-smooth muscle actin (α-SMA), extracellular matrix (ECM) deposition, and fibrotic cytokines such as transforming growth factor (TGF)-β1 and platelet-derived growth factor (PDGF) were also analyzed by immunofluorescence stain and real-time PCR. Results : Histological damages in pancreas were inhibited by pre-treatment of FF or FC but not CT. α-SMA and ECM in pancreas were inhibited by pre-treatment of CT or FC but not FF. Moreover, the expression of TGF-β1 and PDGF in pancreas were inhibited by FF, CT or FC. Conclusions : Our results suggest that FC have protective effect on CP in mice through inhibition of α-SMA, ECM, TGF-β1 and PDGF in pancreas, and these findings could suggest new clinical strategy for CP.

Optimization of extraction conditions for functional ingredients from Tremella fuciformis Berk. using response surface methodology (반응표면분석법을 이용한 흰목이버섯의 기능성 성분 추출 조건 최적화)

  • Snow fungus (Tremella fuciformis) with functional contents has satisfactory effects on various diseases, including atherosclerosis, high cholesterol, healthy skin, cancer, diabetes, and anti-inflammation. In this study, the extraction yield and functional contents (ergothioneine and 𝛽-glucan) of white jelly fungus were compared based on the extraction conditions using response surface methodology. Results revealed the extraction conditions for optimization of the dependent variables to be 60℃ and 4.33 h, when 16.6 mg/mL of sample concentration was used. Under these conditions, the extraction yield was 24.9%, including ergothioneine (66.8 ㎍/g) and 𝛽-glucan (34.9 g/100 g). These results can be useful in understanding the functional ingredients and mass extraction process in mushroom.

Effect of Youngyanggak-san against Thioacetamide Induced Acute Liver Damage in Rat (영양각산이 Thioacetamide 유발 간손상에 미치는 영향)

  • Objectives : The current study is to evaluate the hepatoprotective effect of youngyanggak-san (YGS) on thioacetamide (TAA)-induced acute liver injury in rats. Methods : YGS is composed of Glycyrrhizae Radix, Asiasari Radix, Cimicifugae Rhizoma, Saigae Tataricae Cornu. While N-YGS (non-youngyanggak-san) doesn't include Saigae Tataricae Cornu. Two samples were administrated TAA together for 3 days. Thirty-six rats were divided into four groups. Rats except for the normal group were received TAA (200 mg/kg of body weight, I.P) were divided into three groups (n=9/group) : Group 1 (TAA only), Group 2 (TAA + 200 mg/kg YGS) and Group 3 (TAA + 200 mg/kg N-YGS). Acute liver damage confirmed using histological examination, The factors associated with oxidative stress and liver function activity measured in serum. Also, expressions of inflammation related proteins were investigated by western blot analysis. Results : Oxidative stress factors such as ROS and $ONOO^-$ in the Group 2 was manifested by a significant rise compared with Group 1. YGS markedly decreased the elevated ROS and $ONOO^-$. Furthermore, YGS significantly reduced the levels of serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) The nuclear $factor-{\kappa}B$ ($NF-{\kappa}B$) activation induced by TAA led to increase both inflammatory mediators and cytokines. While YGS administration remarkably suppressed such the overexpression. In addition, the histopathological analysis showed that the liver tissue lesions were improved obviously in YGS treatment. Conclusion : YGS provided a hepatoprotective effect on acute liver damage through the suppression of oxidative stress. Especially, this effect enhanced markedly when Saigae Tataricae Cornu is included.
