Suppressive effects of $Schizandra$ $chinensis$ Baillon water extract on allergy-related cytokine generation and degranulation in IgE-antigen complex-stimulated RBL-2H3 cells

  • $Schizandra$ $chinensis$ Baillon is a traditional folk medicine plant that is used to treat and prevent several inflammatory diseases and cancer in Korea, but the underlying mechanisms involved in its anti-allergic activity are not fully understood. This study was designed to investigate mechanisms of anti-allergic activity of a $Schizandra$ $chinensis$ Baillon water extract (SCWE) in immunoglobulin E (IgE)-antigen complex-stimulated RBL2H3 cells and to assess whether gastric and intestinal digestion affects the anti-allergic properties of SCWE. Oxidative stress is an important consequence of the allergic inflammatory response. The antioxidant activities of SCWE increased in a concentration-dependent manner. RBL-2H3 cells were sensitized with monoclonal anti-dinitrophenol (DNP) specific IgE, treated with SCWE, and challenged with the antigen DNP-human serum albumin. SCWE inhibited ${\beta}$-hexosaminidase release and expression of interleukin (IL)-4, IL-13, and tumor necrosis factor-alpha (TNF-${\alpha}$) mRNA and protein in IgE-antigen complex-stimulated RBL2H3 cells. We found that digested SCWE fully maintained its antioxidant activity and anti-allergic activity against the IgE-antigen complex-induced activation of RBL-2H3 cells. SCWE may be useful for preventing allergic diseases, such as asthma. Thus, SCWE could be used as a natural functional ingredient for allergic diseases in the food and/or pharmaceutical industries.

  • We investigated whether silk fibroin peptide derived from the silkworm, Bombyx mori, could inhibit inflammation and enhance the anti-inflammatory activity of Tat-superoxide dismutase (Tat-SOD), which was previously reported to effectively penetrate various cells and tissues and exert anti-oxidative activity in a mouse model of inflammation. Inflammation was induced by topical treatment of mouse ears with 12-O-tetradecanoylphorbol-13-acetate (TPA). Histological, Western blot, and reverse transcription-polymerase chain reaction data demonstrated that silk fibroin peptide or Tat-SOD alone could suppress elevated levels of cyclooxygenase-2, interleukin-6, interleukin-1beta, and tumor necrosis factor-alpha induced by TPA. Moreover, silk fibroin peptide significantly enhanced the anti-inflammatory activity of Tat-SOD, although it had no influence on in vitro and in vivo transduction of Tat-SOD. Silk fibroin peptide exhibited anti-inflammatory activity in a mice model of inflammation. Therefore, silk fibroin peptide alone or in combination with Tat-SOD might be used as a therapeutic agent for various inflammatory diseases.

  • This study was designed to assess the change of the antioxidative and biological enzyme activities [tyrosinase, elastase, collagenase, and hyaluronidase (HAase)] of extracts from elicitor-treated Oplismenus undulatifolius. Elicitor-treated O. undulatifolius showed higher total phenolic content than the non-treated extract. As a result of comparing the anti-oxidant activity of elicitor-treated O. undulatifolius extract and non-treated extract, the elicitor-treated group showed high activity. Elicitor-treated O. undulatifolius showed higher elastase and collagenase inhibitory activities as anti-wrinkle effect, tyrosinase inhibitory activity as whitening effect, and anti-inflammation effect to induced as HAase inhibition than the non-treated extract. Therefore, elicitor-treatment during O. undulatifolius cultivation in outdoors will elevate total phenolics content in the plant and elevate of various bioactivities, which will yield high quality for industrialization.

Anti-oxidant and Anti-inflammatory Effects of Ethanol Extracts from Leonurus japonicus Houtt. on LPS-induced RAW 264.7 Cells (익모초 에탄올 추출물의 항산화 및 항염증 활성)

  • Leonurus japonicus (L. japonicus) Houtt., a biennial plant in the Lamiaceae family is broadly distributed in Asia such as Korea, China, Japan. The aerial part of L. japonicus is used as a traditional medicine to treat uterine disease including dysmenorrhea, amenorrhea, sterility. In this study, we examined the antioxidant and anti-inflammatory effects of L. japonicus ethanol extracts. The antioxidant activity of L. japonicus was measured by total polyphenol and flavonoid content, and DPPH, ABTS scavenging, reducing power activity, and intracellular ROS expression assay. The anti-inflammatory effects were measured by nitric oxide (NO), cytokines (TNF-α and IL-1β) production and inflammatory protein expression in LPS-induced RAW 264.7 cells. Total polyphenol and flavonoid content of L. japonicus were 51.40 ± 0.47 mg of gallic acid equivalents/g and 73.28 ± 0.10 mg of rutin equivalents/g respectively. DPPH, ABTS radical scavenging activity and reducing power activity tended to increase concentration-dependent and treatment L. japonicus with 400 ㎍/mL reduced ROS production by 69.5%. Furthermore, L. japonicus inhibited NO, TNF-α and IL-1β production in a concentration-dependant manner and reduced the expression levels of inflammatory proteins via regulating NF-κB, MAPK pathway. Therefore, we suggest that L. japonicus could be a natural antioxidants and medicinal source to treat oxidative stress and inflammation-related disease.

Antimicrobial Activities and Free Radical Scavenging Effect of Korean Folk Plants (민속식물의 항균활성 및 산화적 스트레스 개선 효과)

  • We investigated the antioxidative and antimicrobial activities of the methanol extracts from Korean folk plants (MKs) in Chungcheong Province. Among 30 MKs, 16 plants at $100{\mu}g/ml$ showed over 90% scavenging activity of 1,1-diphenyl-2-picrylhydrazy (DPPH) and 30 plants exerted the hydroxyl radical scavenging effect over 55%. Fourteen plants at the concentration $50{\mu}g/30{\mu}l$ showed strong microbial inhibition activity against Escherichia coli and Staphylococcus aureus, with clear zone greater than 11 mm in disc assays. Furthermore, the protective effect against anti-inflammatory system using RAW 264.7 macrophage cell was also studied. The treatment of LPS & INF-${\gamma}$ to RAW 264.7 cell induced nitric oxide (NO), however inhibit the formation of NO less than 50% of 5 plants. The present result indicates that the 30 species of MKs exerts protective effect of oxidative stress, antimicrobial activities and anti-inflammatory. In particular, Rhus javanica and Cornus controversa showed stronger effect on not only radical scavenging activity and inhibits growth of S. aureus but also highest protective effects from inflammation.

Antimicrobial Activity of High Density Polyethylene Fabric Containing Scutellaria Baicalensis Extract-Loaded Zeolite Microparticles (황금추출물 담지 제올라이트 마이크로입자를 함유한 고밀도 폴리에틸렌 원단의 항균 특성)

  • Scutellaria Baicalensis(SB) is widely used in traditional and modern oriental medicine. It possesses several biology activities such as anti-oxidative, anti-inflammatory, antimicrobial and antiviral activities. In this study, a functional high density polyethylene (HDPE) fabric with antimicrobial properties was developed using zeolite microparticles as a SB extract delivery carrier. Zeolites loaded with SB extract were prepared by immersing in an SB extract aqueous solution. The average size of the SB extract-loaded zeolites was about 0.1 to $2.0{\mu}m$, and the morphology of the zeolites was not altered after SB extract binding. The resulting SB extract-loaded zeolites were then immobilized homogeneously onto the HDPE fabric using acrylic binder. The encapsulation efficiency of SB extract to the zeolite was more than 45%. The in vitro release test of SB extract-loaded zeolites containing HDPE fabrics showed release of 35% of the total SB extract by day 1 in a 24hours immersion study. Moreover, the SB extract-loaded zeolites containing HDPE fabrics showed effective antimicrobial activity against Streptococcus mutans, Staphylococcus aureus, and Klebsiella pneumoniae, indicating that this innovative delivery platform potently imparted antimicrobial activity to the HDPE fabric. In conclusion, the current study suggests that the HDPE fabric containing the SB extract-loaded zeolites microparticle carrier system has potential as an effective antimicrobial textile such as safety gloves, protective gloves etc.

  • Matrix metalloproteinases (MMPs) are crucial extracellular matrices degrading enzymes that have important roles in metastasis of cancer progression as well as other significant conditions such as oxidative stress and hepatic fibrosis. Marine plants are on the rise for their potential to provide natural products that exhibit remarkable health benefits. In this context, brown algae species have been of much interest in the pharmaceutical field with reported instances of isolation of bioactive compounds against tumor growth and MMP activity. In this study, eight different brown algae species were harvested, and their extracts were compared in regard to their anti-MMP effects. According to gelatin zymography results, Ecklonia cava, Ecklonia bicyclis, and Ishige okamurae showed higher inhibitory effects than the other samples on MMP-2 and -9 activity at the concentrations of 10, 50, and $100{\mu}g/mL$. However, only I. okamurae was able to regulate the MMP activity through the expression of MMP and tissue inhibitor of MMP observed by mRNA levels. Overall, brown algae species showed to be good sources for anti-MMP agents, while I. okamurae needs to be further studied for its potential to yield pharmaceutical molecules that can regulate MMP-activity through cellular pathways as well as enzymatic inhibition.

Modulative Effect of Human Hair Dermal Papilla Cell Apoptosis by Oregonin from the Braches of Alnus japonica (오리나무 가지 유래 Oregonin의 인체 모유두 세포 Apoptosis 조절 효능)

  • A diarylheptanoid, (5S)-1,7-bis-(3,4-dihydroxyphenyl)-5-hydroxyheptane-3-one-5-O-${\beta}$-D-xylopyranoside, named oregonin (1), was isolated from the of Alnus japonica (A. japonica), which is a species of the genus Betulaceae, growing throughout Korea, Japan and China. The structure was elucidated by various spectroscopic methods including negative and positive LC/MS, 1H-NMR, and 13C-NMR techniques or by comparison with authentic samples. In order to evaluate the anti-oxidative activities of oregonin (1) isolated from A. japonica, 1,1-diphenyl-2-picryhydrazyl (DPPH) radical scavenging activity and 2,2'-azino-bis[3-ethylbenzothiazoline-6-sulphonic acid] (ABTS) radical scavenging activity were measured in vitro. Oregonin from A. japonica exhibited potent DPPH and ABTS radical scavenging activities. A. japonica shows not only 1,1-diphenyl-2-picryhydrazyl (DPPH) radical scavenging activity and ABTS radical scavenging activity, but also apoptosis modulative effects. The present results indicate that A. japonica could be a hair-growth-promoting agent for cosmetic products.

Anti-Oxidative and Anti-Inflammatory Effect of Water Extract from Perillae Semen in RBL-2H3 Cells (RBL-2H3 세포에서 차조기씨 물 추출물의 항산화 및 항염증 효과)

  • The present study was to investigate anti-oxidative and anti-inflammatory activity of Perillae semen in RBL-2H3 basophilic leukemia cells. Inhibitory effect of Perillae semen onto free radical generation was determined by measuring DPPH and hydroxyl radical scavenging activities in vitro. Anti-inflammatory actions of Perillae semen extracts (100, 250, $500\;{\mu}g/mL$) were assessed by testing their effects on the degranulation of mast cells. For this, $\beta$-hexosaminidase released from RBL-2H3 cells was used and proinflammatory cytokines were measured by an ELISA kit. Our results indicated that Perillae semen water extracts effectively inhibited free radical generation. At the concentration of $500\;{\mu}g/mL$ of water extract, the degranulation of RBL-2H3 cells were inhibited by 42.1%. The IgE-antigen complex increased the accumulation of IL-4 and TNF-$\alpha$ secretion in RBL-2H3 cells and treatments with 250 and $500\;{\mu}g/mL$ of Perillae semen extracts suppressed the IgE induced secretion of IL-4 and TNF-$\alpha$ protein by 20.5, 26.9% and 14.5, 16.5% respectively. We observed that Perillae semen water extract reduced $\beta$-hexosaminidase, IL-4, and TNF-$\alpha$ secretion in RBL-2H3 cells. These results provide that Perillae semen may be beneficial in the treatment of allergic inflammatory disease.

Anti-oxidative and Anti-inflammatory Effect of Fractionated Extracts of Smilacis Glabrae Rhizoma in Human Umbilical Vein Endothelial Cell (혈관내피세포에서 토복령(土茯苓)의 항산화 및 항염증 효과)

  • Objectives : Smilacis glabrae rhizoma (SG) has been traditionally used as a herbal medication of musculoskeletal disorders like arthritis, pain, convulsions, and syphilis in traditional Korean medicine. This study was investigated anti-oxidative and anti-inflammatory effect of fractionated extracts of Smilacis Glabrae Rhizoma in Human Umbilical Vein Endothelial Cell (HUVEC). Methods : SG extract prepared with methanol, and then fractionated with hexane, dichloromethane, ethylacetate, n-butanol and water. Inhibitory effect of SG onto free radical generation was determined by measuring DPPH, superoxide anions and nitric oxide scavenging activities in vitro. Cytotoxic activity of extracts on RAW 264.7 cells was measured using 5-(3-caroboxymethoxyphenyl)-2H-tetra-zolium inner salt (MTS) assay. Intracelluar oxidation was analysed by DCF-DA assay. The nitric oxide (NO) production was measured by Griess reagent system. The levels of ICAM-1 and VCAM-1 expression were confirmed by western blot. And proinflammatory cytokines were measured by ELISA kit. Results : Our results indicated that fractionated extracts, especially ethyl acetate (EA) extract, significantly inhibited free radical generation, the TNF-$\alpha$-induced intracellular oxidation. Furthermore, the EA extract protected TNF-$\alpha$-induced adhesion to THP-1, expression of adhesion molecules accompanied by an attenuation of IL-6 and IL-8 formation in HUVEC. Conclusions : These results indicate that EA extract of SG have potential as an agent of atherosclerosis and other chronic inflammatory diseases including diabetes, hypertension, and arthritis.
